The enigma of Calonectria species occurring on leaves of Ilex aquifolium in Europe.

ABSTRACT: Species of Calonectria are common saprobes and plant pathogens on a wide range of hosts occurring in subtropical to tropical regions of the world. The aim of the present study was to resolve the status of new Calonectria collections obtained on Ilex leaves from France. Based on DNA sequence data of their β-tubulin and histone gene regions, as well as morphology, the new collections matched the ex-type strain of Cylindrocladium ilicicola. On the host and in culture, yellow to brownish-yellow perithecia were observed that did not strain red in 3 % KOH. Based on these results, C. ilicicola and its purported teleomorph, Ca. pyrochroa, were shown to represent two distinct species, as the latter has bright red perithecia that strain purple in KOH. A new combination, Ca. lauri, based on Tetracytumlauri, is subsequently proposed for C. ilicicola. Calonectria lauri is distinct from Ca. ilicicola, a pathogen commonly associated with Cylindrocladium black rot of peanut. Finally, Ca. canadiana is proposed as new name for Cy. canadiense, which is a nursery pathogen involved with root rot of several tree genera in Quebec, Canada.

Project description:The leaves of Ilex paraguariensis (known as Yerba mate), used as a popular beverage, are a very well-recognized plant material with various biological activities, including analeptic (because of caffeine), anti-obesity (phenolics, saponins), antimicrobial, and antiviral (phenolics, saponins). Here, the chemical compositions of the leaves of two European Ilex species (× meserveae and aquifolium) with three varieties each were investigated. The terpenoid, saponin, and polyphenolic fractions were submitted for LC-MS or GC-MS analysis against a standard Mate leaf. In addition, the aroma profiles of all the species were analysed using HS-SPME-Arrow prior to GC-MS analysis. All fractions were subjected to antiviral and cytotoxic assays. We found 86 compounds in all accessions, with limonene, linalool, and p-cymene being predominant. There were minor similarities between the volatile compositions of the European and South American species. We found ursolic and oleanolic acid to be the main compounds in the terpenoid fraction. Mono-caffeoylquinic acids and di-caffeoylquinic acids were the main constituents of the polar fractions. About 180 compounds from the saponin group were tentatively identified, of which 9 and 3 were selected as distinctive markers for I. meserveae and I. aquifolium, respectively. Based on chemical screening, I. aquifolium Silver Queen was chosen as the source of terpenoid and saponin fractions and polyphenol extracts. The most substantial inhibition of cancer cell growth was observed with saponin in the case of the MCF7 (human breast cancer) cell line, while for LoVo and L929 cell lines (human colorectal cancer and reference mouse fibroblasts), it was slightly weaker. These results should be analysed further as a promising chemoprevention of colorectal and gastrointestinal cancers. Saponin and polyphenolic extracts exhibited similar activities against HSV-1 and HAdV-5, with 4-log reduction in virus titres. This study focuses our attention on a field of potential antiviral formulations derived from European holly.

Project description:The cuticle is a lipid-rich layer that protects aerial plant organs against multiple stress factors such as dehydration. In this study, cuticle composition and structure in relation to water loss are examined in a broad ecophysiological context, taking into consideration leaf age and side from Ilex aquifolium (holly) in comparison with Eucalyptus globulus (eucalypt) and Prunus laurocerasus (cherry laurel). Enzymatically isolated cuticular membranes from holly leaves were studied under three treatment conditions: natural (no chemical treatment), after dewaxing, and after methanolysis, and the rate of water loss was assessed. Structural and chemical changes were evaluated using different microscopy techniques and by Fourier transform infrared (FTIR) spectroscopy. The potential mechanisms of solute absorption by holly leaves were additionally evaluated, also testing if its prickly leaf margin may facilitate uptake. The results indicate that the treatment conditions led to structural changes, and that chemical composition was hardly affected because of the occurrence of cutan. Structural changes led to more hydrophilic adaxial surfaces, which retained more water and were more efficient than natural cuticles, while changes were not significant for abaxial surfaces. Across natural cuticles, age was a significant factor for eucalypt but not for holly. Young eucalypt cuticles were the group that absorbed more water and had the lowest water loss rate. When comparing older leaf cuticles of the three species, cherry laurel was found to absorb more water, which was, however, lost more slowly, compared with the other species. Evidence was gained that holly leaves can absorb foliar-applied solutes (traced after calcium chloride application) through the adaxial and abaxial surfaces, the adaxial mid veins, and to a lower extent, the spines. In conclusion, for the species examined, the results show variations in leaf cuticle composition and structure in relation to leaf ontogeny, and water sorption and desorption capacity.

Project description:Many Calonectria species are causal agents of diseases on several forestry, agricultural and horticultural crops. Calonectria leaf blight is one of the most important diseases associated with Eucalyptus plantations and nurseries in Asia and South America. Recently, symptoms of leaf rot and leaf blight caused by Calonectria species were observed in a one-year-old Eucalyptus experimental plantation in GuangXi Province, southern China. To better understand the species diversity, mating strategy and pathogenicity of Calonectria species isolated from diseased tissues and soils, diseased leaves and soils under the trees from ten Eucalyptus urophylla hybrid genotypes were collected. Three hundred and sixty-eight Calonectria isolates were obtained from diseased Eucalyptus leaves and soils under these trees, and 245 representative isolates were selected based on the sampling substrates and Eucalyptus genotypes and identified by DNA sequence analyses based on the translation elongation factor 1-alpha (tef1), β-tubulin (tub2), calmodulin (cmdA) and histone H3 (his3) gene regions, as well as a combination of morphological characteristics. These isolates were identified as Calonectria hongkongensis (50.2%), C. pseudoreteaudii (47.4%), C. aconidialis (1.6%), C. reteaudii (0.4%) and C. auriculiformis (0.4%). This is the first report of C. reteaudii and C. auriculiformis occurrence in China. Calonectria pseudoreteaudii was isolated from both Eucalyptus diseased leaves and soils; the other four species were only obtained from soils. MAT1-1-1 and MAT1-2-1 gene amplification and mating type assignment results showed that C. pseudoreteaudii is heterothallic and an asexual cycle represents the primary reproductive mode, C. reteaudii and C. auriculiformis are likely to be heterothallic and C. hongkongensis and C. aconidialis are homothallic. Based on the genetic diversity comparisons for C. pseudoreteaudii isolates from diseased leaves and soils, we hypothesize that C. pseudoreteaudii in soils was spread from diseased leaves. Both the mycelia plug and conidia suspension inoculations indicated that all five Calonectria species were pathogenic to the two Eucalyptus genotypes tested and the tolerance of the two genotypes differed. It is necessary to understand the ecological niche and epidemiological characteristics of these Calonectria species and to select disease resistant Eucalyptus genotypes in southern China in the future.
