Project description:Various enzymes can be used to modify the structure of hemicelluloses directly in vivo or following extraction from biomass sources, such as wood and agricultural residues. Generally, these enzymes can contribute to designer hemicelluloses through four main strategies: (1) enzymatic hydrolysis such as selective removal of side groups by glycoside hydrolases (GH) and carbohydrate esterases (CE), (2) enzymatic cross-linking, for instance, the selective addition of side groups by glycosyltransferases (GT) with activated sugars, (3) enzymatic polymerization by glycosynthases (GS) with activated glycosyl donors or transglycosylation, and (4) enzymatic functionalization, particularly via oxidation by carbohydrate oxidoreductases and via amination by amine transaminases. Thus, this Perspective will first highlight enzymes that play a role in regulating the degree of polymerization and side group composition of hemicelluloses, and subsequently, it will explore enzymes that enhance cross-linking capabilities and incorporate novel chemical functionalities into saccharide structures. These enzymatic routes offer a precise way to tailor the properties of hemicelluloses for specific applications in biobased materials, contributing to the development of renewable alternatives to conventional materials derived from fossil fuels.

Project description:The rapid diagnosis of Pseudomonas aeruginosa infection is very important because this bacterium is one of the main sources of healthcare-associated infections. Pseudomonas quinolone signal (PQS) is a specific molecule for quorum sensing (QS) in P. aeruginosa, a form of cell-to-cell bacterial communication and its levels can allow the determination of the bacterial population. In this study, the development of the first electrochemical detection of PQS using screen-printed electrodes modified with carbon nanotubes (CNT-SPE) is reported. The electrochemical fingerprint of PQS was determined using different electrode materials and screen-printed electrodes modified with different nanomaterials. The optimization of the method in terms of electrolyte, pH, and electrochemical technique was achieved. The quantification of PQS was performed using one of the anodic peaks in the electrochemical fingerprint of the PQS on the CNT-SPE. The sensor exhibited a linear range from 0.1 to 15 µM, with a limit of detection of 50 nM. The sensor allowed the selective detection of PQS, with low interference from other QS molecules. The sensor was successfully applied to analysis of real samples (spiked urine and human serum samples, spiked microbiological growth media, and microbiological cultures).

Project description:In this study, natural clay (NC) was collected from Saudi Arabia and modified by cocamidopropyl betaine (CAPB) at different conditions (CAPB concentration, reaction time, and reaction temperature). NC and modified clay (CAPB-NC) were characterized using X-ray diffraction, thermogravimetric analysis, Fourier transform infrared spectroscopy, field-emission scanning electron microscopy, and N2 adsorption at 77 K. The adsorption efficiency of NC and CAPB-NC toward Pb2+ and reactive yellow 160 dye (RY160) was evaluated. The adsorption process was optimized in terms of solution initial pH and adsorbent dosage. Finally, the adsorption kinetics and isotherms were studied. The results indicated that NC consists of agglomerated nonporous particles composed of quartz and kaolinite. CAPB modification reduced the specific surface area and introduced new functional groups by adsorbing on the NC surface. The concentration of CAPB affects the adsorption of RY160 tremendously; the optimum concentration was 2 times the cation exchange capacity of NC. The equilibrium adsorption capacity of CAPB-NC toward RY160 was about 6 times that of NC and was similar for Pb2+. The adsorption process followed the pseudo-second-order kinetics for both adsorptive. RY160 adsorption on CAPB-NC occurs via multilayer formation while Pb2+ adsorption on NC occurs via monolayer formation..

Project description:Novel drug delivery systems are developed to improve the biological behavior of poorly soluble drugs and to improve therapeutic outcomes. In melanoma therapy, the goal is efficient drug delivery and mitigation of drug resistance. Melphalan (Mel), a currently used therapeutic agent for melanoma, requires solvent system for solubilization, leading to poor chemical stability. Moreover, drug resistance often renders the drug inefficient in clinical setting. A novel β-cyclodextrin-modified gemini surfactant (CDgemini) delivery system was developed to incorporate Mel in order to improve its physicochemical and biological behavior. Melphalan nanoparticles (Mel-NP) showed optimal particle size in the 200-250 nm range for endocytosis and induced significantly higher cell death compared with Mel (50% of inhibitory concentration [IC50] of 36 µM for the complexes vs 82 µM for Mel). The CDgemini delivery system did not alter the pathway of the cellular death triggered by Mel and caused no intrinsic toxicity to the cells. The Mel-NP complexes induced significant cell death in melanoma cells that were rendered resistant to Mel. These findings demonstrate in principle the applicability of the CDgemini delivery system as safe and efficient alternative to the current melanoma therapy, especially in chemoresistant cases.

Project description:Tuning surface structures by bottom-up synthesis has been demonstrated as an effective strategy to improve the catalytic performances of nanoparticle catalysts. Nevertheless, the surface modification of three-dimensional nanoporous metals, fabricated by a top-down dealloying approach, has not been achieved despite great efforts devoted to improving the catalytic performance of three-dimensional nanoporous catalysts. Here we report a surfactant-modified dealloying method to tailor the surface structure of nanoporous gold for amplified electrocatalysis toward methanol oxidation and oxygen reduction reactions. With the assistance of surfactants, {111} or {100} faceted internal surfaces of nanoporous gold can be realized in a controllable manner by optimizing dealloying conditions. The surface modified nanoporous gold exhibits significantly enhanced electrocatalytic activities in comparison with conventional nanoporous gold. This study paves the way to develop high-performance three-dimensional nanoporous catalysts with a tunable surface structure by top-down dealloying for efficient chemical and electrochemical reactions.

Project description:The Pseudomonas aeruginosa MvfR-dependent QS regulatory pathway controls the expression of key virulence genes; and is activated via the extracellular signals 4-hydroxy-2-heptylquinoline (HHQ) and 3,4-dihydroxy-2-heptylquinoline (PQS). Our findings reveal a multi-layered homeostatic regulation of PQS and HHQ and the MvfR regulon component PqsE which is a key mediator in orchestrating this homeostatic regulation.

Project description:The opportunistic pathogen Pseudomonas aeruginosa employs quorum sensing to govern the production of many virulence factors. Interference with quorum sensing signaling has therefore been put forward as an attractive approach to disarm this pathogen. Here, we analyzed the quorum quenching properties of natural and engineered (2-alkyl-)3-hydroxy-4(1H)-quinolone 2,4-dioxygenases (HQDs) that inactivate the P. aeruginosa signal molecule PQS (Pseudomonas quinolone signal; 2-heptyl-3-hydroxy-4(1H)-quinolone). When added exogenously to P. aeruginosa cultures, all HQDs tested significantly reduced the levels of PQS and other alkylquinolone-type secondary metabolites deriving from the biosynthetic pathway, such as the respiratory inhibitor 2-heptyl-4-hydroxyquinoline N-oxide. HQDs from Nocardia farcinica and Streptomyces bingchenggensis, which combine low KM values for PQS with thermal stability and resilience in the presence of P. aeruginosa exoproducts, respectively, attenuated production of the virulence factors pyocyanin and pyoverdine. A delay in mortality was observed when Galleria mellonella larvae were infected with P. aeruginosa suspensions treated with the S. bingchenggensis HQD or with inhibitors of alkylquinolone biosynthesis. Our data indicate that quenching of PQS signaling has potential as an anti-virulence strategy; however, an efficient anti-virulence therapy against P. aeruginosa likely requires a combination of agents addressing multiple targets.

Project description:In the present work, we reported the simple way to fabricate an electrochemical sensing platform to detect Bisphenol A (BPA) using galvanostatic deposition of Au on a glassy carbon electrode covered by cetyltrimethylammonium bromide (CTAB). This material (CTAB) enhances the sensitivity of electrochemical sensors with respect to the detection of BPA. The electrochemical response of the modified GCE to BPA was investigated by cyclic voltammetry and differential pulse voltammetry. The results displayed a low detection limit (22 nm) and a linear range from 0.025 to 10 µm along side with high reproducibility (RSD = 4.9% for seven independent sensors). Importantly, the prepared sensors were selective enough against interferences with other pollutants in the same electrochemical window. Notably, the presented sensors have already proven their ability in detecting BPA in real plastic water drinking bottle samples with high accuracy (recovery range = 96.60%-102.82%) and it is in good agreement with fluorescence measurements.

Project description:It is necessary to develop novel and efficient alternatives to fluorocarbon surfactant and prepare fluorine-free environmentally-friendly fire extinguishing agent. The carboxyl modified polyether polysiloxane surfactant (CMPS) with high surface activity was synthesized via the esterification reaction using hydroxyl-containing polyether modified polysiloxane (HPMS) and maleic anhydride (MA) as raw materials. The process conditions of the esterification reaction were optimized by orthogonal tests, and the optimum process parameters were determined as follows: reaction temperature of 85 °C, reaction time of 4.5 h, isopropyl alcohol content of 20% and the molar ratio of HPMS/MA of 1/1. The chemical structure, surface activity, aggregation behavior, foam properties, wetting properties and electron distribution were systematically investigated. It was found that the carboxyl group was successfully grafted into silicone molecule, and the conjugated system was formed, which changed the interaction force between the molecules and would affect the surface activity of the aqueous solution. The CMPS exhibited excellent surface activity and could effectively reduce the water's surface tension to 18.46 mN/m. The CMPS formed spherical aggregates in aqueous solution, and the contact angle value of CMPS is 15.56°, illustrating that CMPS had excellent hydrophilicity and wetting performance. The CMPS can enhance the foam property and has good stability. The electron distribution results indicate that the introduced carboxyl groups are more inclined towards the negative charge band, which would be conducive to weak the interaction between molecules and improve the surface activity of the solution. Consequently, new foam fire extinguishing agents were prepared by using CMPS as a key component and they exhibited excellent fire-fighting performance. The prepared CMPS would be the optimal alternative to fluorocarbon surfactant and could be applied in foam extinguishing agents.

Project description:The modification of the Tetraselmis sp. algae material (Tetra-Alg) with surfactant Cethyltrimethylammonium Bromide (CTAB) yielded adsorbent Tetra-Alg-CTAB as an adsorbent of methyl orange (MO) and methylene blue (MB) solutions. The characterization of the adsorbent used an infrared (IR) spectrometer to identify functional groups and Scanning Electron Microscopy with Energy Dispersive X-ray (SEM-EDX FEI Inspect-S50, Midland, ON, Canada) to determine the surface morphology and elemental composition. Methyl orange and methylene blue adsorption on the adsorbent Tetra-Alg, Tetraselmis sp. algae-modified Na+ ions (Tetra-Alg-Na), and Tetra-Alg-CTAB were studied, including variations in pH, contact time, concentration, and reuse of adsorbents. The adsorption of MO and MB by Tetra-Alg-CTAB at pH 10, during a contact time of 90 min, and at a concentration of 250 mg L-1 resulted in MO and MB being absorbed in the amounts of 128.369 and 51.013 mg g-1, respectively. The adsorption kinetics and adsorption isotherms of MO and MB and Tetra-Alg, Tetra-Alg-Na, and Tetra-Alg-CTAB tend to follow pseudo-second-order kinetics models and Freundlich adsorption isotherms with each correlation coefficient value (R2) approaching 1. Due to the modification with the cationic surfactant CTAB, anionic dyes can be strongly sorbed in alkaline pH due to strong electrostatic attraction, while MB is more likely to involve cation exchange and hydrogen bonding. The reuse of Tetra-Alg-CTAB was carried out four times with adsorption percent > 70%, and the adsorbent was very effective in the adsorption of anionic dyes such as MO.
