Screening of ligands for the Ullmann synthesis of electron-rich diaryl ethers.

ABSTRACT: In the search for new ligands for the Ullmann diaryl ether synthesis, permitting the coupling of electron-rich aryl bromides at relatively low temperatures, 56 structurally diverse multidentate ligands were screened in a model system that uses copper iodide in acetonitrile with potassium phosphate as the base. The ligands differed largely in their performance, but no privileged structural class could be identified.

Synthesis and Antiproliferative Activity of Steroidal Diaryl Ethers.

Project description:Novel 13α-estrone derivatives have been synthesized via direct arylation of the phenolic hydroxy function. Chan-Lam couplings of arylboronic acids with 13α-estrone as a nucleophilic partner were carried out under copper catalysis. The antiproliferative activities of the newly synthesized diaryl ethers against a panel of human cancer cell lines (A2780, MCF-7, MDA-MB 231, HeLa, SiHa) were investigated by means of MTT assays. The quinoline derivative displayed substantial antiproliferative activity against MCF-7 and HeLa cell lines with low micromolar IC50 values. Disturbance of tubulin polymerization has been confirmed by microplate-based photometric assay. Computational calculations reveal significant interactions of the quinoline derivative with the taxoid binding site of tubulin.

Synthesis of axially chiral diaryl ethers via NHC-catalyzed atroposelective esterification.

Project description:Axially chiral diaryl ethers bearing two potential axes find unique applications in bioactive molecules and catalysis. However, only very few catalytic methods have been developed to construct structurally diverse diaryl ethers. We herein describe an NHC-catalyzed atroposelective esterification of prochiral dialdehydes, leading to the construction of enantioenriched axially chiral diaryl ethers. Mechanistic studies indicate that the matched kinetic resolutions play an essential role in the challenging chiral induction of flexible dual-axial chirality by removing minor enantiomers via over-functionalization. This protocol features mild conditions, excellent enantioselectivity, broad substrate scope, and applicability to late-stage functionalization, and provides a modular platform for the synthesis of axially chiral diaryl ethers and their derivatives.

Cu-catalyzed arylation of phenols: synthesis of sterically hindered and heteroaryl diaryl ethers.

Project description:Cu-catalyzed O-arylation of phenols with aryl iodides and bromides can be performed under mild condition in DMSO/K(3)PO(4) with use of picolinic acid as the ligand for copper. This method tolerates a variety of functional groups and is effective in the synthesis of hindered diaryl ethers and heteroaryl ethers.

Mechanistic Investigations of the Hydrogenolysis of Diaryl Ethers Catalyzed by Nickel Complexes of N-Heterocyclic Carbene Ligands.

Project description:Recent interest in the valorization of lignin has led to reactions involving the cleavage of strong aromatic C-O bonds. However, few experimental mechanistic studies of these reactions have been published. We report detailed mechanistic analysis of the hydrogenolysis of diaryl ethers catalyzed by the combination of Ni(COD)2 (COD = 1,5-cyclooctadiene) and an N-heterocyclic carbene (NHC). Experiments on the catalytic reaction indicated that NaOt-Bu was necessary for catalysis, but kinetic analysis showed that the base is not involved in the rate-limiting C-O bond cleavage. The resting state of the catalyst is an NHC-Ni(η6-arene) complex. Substitution of the coordinated solvent with diaryl ether allowed isolation of a diaryl ether-bound Ni complex. Rate-limiting C-O bond cleavage occurs to generate a three-coordinate product of oxidative addition, a metallacyclic version of which has been prepared independently. Stoichiometric studies show that arene and phenol products are released following reaction with H2. NaOt-Bu was found to deprotonate the phenol product and to prevent formation of inactive NiI dimers.

Electrochemical oxidative decarboxylation and 1,2-aryl migration towards the synthesis of 1,2-diaryl ethers.

Project description:Carboxylic acid compounds are important chemicals and are widely present in various natural products. They are not only nucleophiles, but also radical precursors. Classic transition-metal-catalyzed and photochemical decarboxylation have shown their excellent site selectivity in radical chemistry. However, electrochemical decarboxylation with a long history hasn't got enough attention in recent years. In this work, the electrochemical oxidative decarboxylation and 1,2-aryl migration of 3,3-diarylpropionic acids have been introduced to construct C-O bonds with alcohols. Remarkably, this transformation can proceed smoothly without metal catalysts and external oxidants.

Catalytic Enantioselective Synthesis of Axially Chiral Diaryl Ethers Via Asymmetric Povarov Reaction Enabled Desymmetrization.

Project description:Axially chiral diaryl ethers represent a distinct class of atropisomers, characterized by a unique dual C─O axes system, which have been found in a variety of natural products, pharmaceuticals, and ligands. However, the catalytic enantioselective synthesis of these atropoisomers poses significant challenges, due to the difficulty in controlling both chiral C─O axes, and their more flexible conformations. Herein, an efficient protocol for catalytic enantioselective synthesis of axially chiral diaryl ethers is presented using organocatalyzed asymmetric Povarov reaction-enabled desymmetrization, followed by aromatizations. This method yields a wide range of novel quinoline-based diaryl ether atropoisomers in good yields and high enantioselectivities. Notably, various aromatization protocols are developed, resulting in a diverse set of polysubstituted quinoline-containing diaryl ether atropisomers. Thermal racemization studies suggested excellent configurational stabilities for these novel diaryl ether atropisomers (with racemization barriers up to 38.1 kcal mol-1). Moreover, this research demonstrates for the first time that diaryl ether atropisomers lacking the bulky t-Bu group can still maintain a stable configuration, challenging the prior knowledge in the field. The fruitful derivatizations of the functional group-rich chiral products further underscore the value of this method.

A new biarylphosphine ligand for the Pd-catalyzed synthesis of diaryl ethers under mild conditions.

Project description:A new bulky biarylphosphine ligand (L8) has been developed that allows the Pd-catalyzed C-O cross-coupling of a wide range of aryl halides and phenols under milder conditions than previously possible. A direct correlation between the size of the ligand substituents in the 2', 4', and 6' positions of the nonphosphine containing ring and the reactivity of the derived catalyst system was observed. Specifically, the rate of coupling increased with the size of these substituents.

Copper-catalysed dynamic kinetic asymmetric C-O cross-coupling to access chiral aryl oxime ethers and diaryl ethers.

Project description:Dynamic kinetic resolution (DKR) has emerged as an elegant and powerful tool for enantioselective synthesis, enabling the transformation of racemic compounds into enantiomerically enriched products with theoretically quantitative yields. Despite its widespread success, the dynamic kinetic asymmetric C-O cross-coupling has presented significant challenges and remains unexplored. In this study, we report a dynamic kinetic asymmetric C-O cross-coupling of oximes and phenols via copper/BOX-catalysed enantioselective O-arylation with diaryliodonium salts. This method efficiently produces a wide range of inherently chiral oxime ethers, as well as axially chiral styrenes, with high yields and excellent regio- and enantioselectivities. Through controlled experiments and Density Functional Theory (DFT) studies, we have elucidated the dynamic kinetic resolution process and gained insights into the origins of regio- and enantioselectivity.

Tetradentate copper complex supported on boehmite nanoparticles as an efficient and heterogeneous reusable nanocatalyst for the synthesis of diaryl ethers.

Project description:In this work boehmite nanoparticles (BNPs) were prepared through addition of aqueous solution of NaOH to solution of Al(NO3)3·9H2O. Then, the surface of BNPs was modified by (3-chloropropyl)trimethoxysilane (CPTMS) and further tetradentate ligand (MP-bis(AMP)) was anchored on its surface. At final step, a tetradentate organometallic complex of copper was stabilized on the surface of modified BNPs (Cu(II)-MP-bis(AMP)@boehmite). These obtained nanoparticles were characterized using SEM imaging, WDX, EDS, AAS and TGA analysis, BET method, FT-IR spectroscopy, and XRD pattern. In continue, the catalytic activity of Cu(II)-MP-bis(AMP)@boehmite has been used as a much efficient, reusable and hybrid of organic-inorganic nanocatalyst in the synthesis of ether derivatives through C-O coupling reaction under palladium-free and phosphine-free conditions. Cu(II)-MP-bis(AMP)@boehmite catalyst has been recovered and reused again for several times in the synthesis of ether derivatives.

Towards a Catalytic Atroposelective Synthesis of Diaryl Ethers via C(sp 2)-H Alkylation Using Nitroalkanes.

Project description:Herein we report studies towards a small molecule catalytic approach to access atropisomeric diaryl ethers that proceeds via a C(sp 2)-H alkylation using nitroalkanes as the alkyl source. A quaternary ammonium salt derived from quinine containing a sterically hindered urea at the C-9 position was found to effect atroposelective C(sp 2)-H alkylation with moderate to good enantioselectivities across several naphthoquinone-containing diaryl ethers. Products can then be isolated in greater than 95:5 er after one round of trituration. For several substrates that were evaluated we observed a 'nitroethylated' product in similar yields and selectivities.
