Revision of the southeast Asian soldier-fly genus Parastratiosphecomyia Brunetti, 1923 (Diptera, Stratiomyidae, Pachygastrinae).

ABSTRACT: The genus Parastratiosphecomyia Brunetti is revised with the description of two new species: Parastratiosphecomyia freidbergisp. n. from India and Parastratiosphecomyia rozkosnyisp. n. from Laos and Thailand. All four species in the genus are illustrated and a key to species is provided. Type localities of previously described taxa are briefly discussed.

Proteomics and Lipidomics of Black Soldier Fly (Diptera: Stratiomyidae) and Blow Fly (Diptera: Calliphoridae) Larvae.

Project description:Farming insects has recently emerged as a new source of protein and lipid production. To date, research has mostly focused on food applications of insects. Focusing on nonfood potential of oil and proteins of insects, high-throughput studies of insect lipids and proteins are needed. We performed proteomics and lipidomics investigation on black soldier fly (Hermitia illucens) and blow fly (Lucilia sericata) larvae to investigate new potential and applications. We used mass spectrometry for proteomics and lipidomics analysis of control and treated larvae. Treatment was performed by incubation with a biological decomposer. We provide the list of all fatty acids with their concentration in control and treated larvae. This result showed high levels of lauric acid in black soldier fly, which could even increase after biological decomposition. Proteomics analysis showed the presence of proteins like collagen of cosmetic interest, and proteins with antimicrobial properties such as phenoloxidases and enzymatic activities, such as amylase and trypsin. Insects harbor high potential for nonfood usage as additives, antimicrobial effects, and even pharmaceuticals and cosmetics. These data open avenues for future research in pharmacological and cosmetic approaches to find new molecules of interests.

Starvation Alters Gut Microbiome in Black Soldier Fly (Diptera: Stratiomyidae) Larvae.

Project description:Unlike for vertebrates, the impact of starvation on the gut microbiome of invertebrates is poorly studied. Deciphering shifts in metabolically active associated bacterial communities in vertebrates has led to determining the role of the associated microbiome in the sensation of hunger and discoveries of associated regulatory mechanisms. From an invertebrate perspective, such as the black soldier fly, such information could lead to enhanced processes for optimized biomass production and waste conversion. Bacteria associated with food substrates of black soldier fly are known to impact corresponding larval life-history traits (e.g., larval development); however, whether black soldier fly larval host state (i.e., starved) impacts the gut microbiome is not known. In this study, we measured microbial community structural and functional shifts due to black soldier fly larvae starvation. Data generated demonstrate such a physiological state (i.e., starvation) does in fact impact both aspects of the microbiome. At the phylum level, community diversity decreased significantly during black soldier fly larval starvation (p = 0.0025). Genus level DESeq2 analysis identified five genera with significantly different relative abundance (q < 0.05) across the 24 and 48 H post initiation of starvation: Actinomyces, Microbacterium, Enterococcus, Sphingobacterium, and Leucobacter. Finally, we inferred potential gene function and significantly predicted functional KEGG Orthology (KO) abundance. We demonstrated the metabolically active microbial community structure and function could be influenced by host-feeding status. Such perturbations, even when short in duration (e.g., 24 H) could stunt larval growth and waste conversion due to lacking a full complement of bacteria and associated functions.

Bacteria mediate oviposition by the black soldier fly, Hermetia illucens (L.), (Diptera: Stratiomyidae).

Project description:There can be substantial negative consequences for insects colonizing a resource in the presence of competitors. We hypothesized that bacteria, associated with an oviposition resource and the insect eggs deposited on that resource, serve as a mechanism regulating subsequent insect attraction, colonization, and potentially succession of insect species. We isolated and identified bacterial species associated with insects associated with vertebrate carrion and used these bacteria to measure their influence on the oviposition preference of adult black soldier flies which utilizes animal carcasses and is an important species in waste management and forensics. We also ascertained that utilizing a mixture of bacteria, rather than a single species, differentially influenced behavioral responses of the flies, as did bacterial concentration and the species of fly from which the bacteria originated. These studies provide insight into interkingdom interactions commonly occurring during decomposition, but not commonly studied.

On the Korean Peninsula, the species diversity of this subfamily has been poorly examined to date, as is reflected in the National species list of Korea by the National Institute of Biological Resources.New informationIn this paper, the soldier fly genus Beris Latreille is reported for the first time from Korea, based on observations of the following three species: B.fuscipes Meigen, 1820, B.hildebrandtae Pleske, 1930 and B.hirotui Ôuchi, 1943. Additionally, the authors propose two nomenclatural acts: 1) B.angustifacies Nagatomi and Tanaka, 1972 syn. nov. = B.hildebrandtae Pleske, 1930; 2) B.liaoningana Cui, Li and Yang, 2010 syn. nov. = B.hildebrandtae Pleske, 1930. The authors also provide an identification key, descriptions, photographs and distributional notes on these Korean species.

Updated Italian checklist of Soldier Flies (Diptera, Stratiomyidae).

Project description:An updated checklist for Stratiomyidae of Italy is presented. Previous knowledge and information are put together in a comprehensive way, integrated also with results obtained by sampling with Malaise traps in some of the test areas of the LIFE+ project ManFor C.BD. At the time of writing, with 91 known species, the Italian fauna of Stratiomyidae is the richest in Europe. Neopachygaster meromelas (Dufour, 1841) and Zabrachia minutissima (Zetterstedt, 1838) are new to the Italian fauna. A comprehensive key to the European species of Chorisops Rondani, 1856 is given.

A bioinformatic study of antimicrobial peptides identified in the Black Soldier Fly (BSF) Hermetia illucens (Diptera: Stratiomyidae).

Project description:Antimicrobial peptides (AMPs) play a key role in the innate immunity, the first line of defense against bacteria, fungi, and viruses. AMPs are small molecules, ranging from 10 to 100 amino acid residues produced by all living organisms. Because of their wide biodiversity, insects are among the richest and most innovative sources for AMPs. In particular, the insect Hermetia illucens (Diptera: Stratiomyidae) shows an extraordinary ability to live in hostile environments, as it feeds on decaying substrates, which are rich in microbial colonies, and is one of the most promising sources for AMPs. The larvae and the combined adult male and female H. illucens transcriptomes were examined, and all the sequences, putatively encoding AMPs, were analysed with different machine learning-algorithms, such as the Support Vector Machine, the Discriminant Analysis, the Artificial Neural Network, and the Random Forest available on the CAMP database, in order to predict their antimicrobial activity. Moreover, the iACP tool, the AVPpred, and the Antifp servers were used to predict the anticancer, the antiviral, and the antifungal activities, respectively. The related physicochemical properties were evaluated with the Antimicrobial Peptide Database Calculator and Predictor. These analyses allowed to identify 57 putatively active peptides suitable for subsequent experimental validation studies.

How could climate change influence the distribution of the black soldier fly, <i>Hermetiaillucens</i> (Linnaeus) (Diptera, Stratiomyidae)?

Project description:The black soldier fly, Hermetiaillucens (Linnaeus, 1758), is a saprophagous species used to decompose organic matter. This study proposes a distribution model of H.illucens to illustrate its current and future distribution. The methodology includes data collection from the Global Biodiversity Information Facility (GBIF), complemented with iNaturalist, manual expert curation of occurrence records, six species distribution models algorithms and one ensemble model. The average temperature of the driest annual quarter and the precipitation of the coldest annual quarter were the key variables influencing the potential distribution of H.illucens. The distribution range is estimated to decrease progressively and their suitable habitats could change dramatically in the future due to global warming. On the other hand, current optimal habitats would become uninhabitable for the species, mainly at low latitudes. Under this scenario, the species is projected to move to higher latitudes and elevations in the future. The results of this study provide data on the distribution of H.illucens, facilitating its location, management and sustainable use in current and future scenarios.

Life-History Traits of the Black Soldier Fly, Hermetia illucens (L.) (Diptera: Stratiomyidae), Reared on Three Manure Types.

Project description:Structural changes and growth of animal production systems have resulted in greater volumes of manure. Current manure storage methods pose a potential environmental threat. Lessening these issues is a key concern for the animal production industry. The primary aim of this research was to evaluate black soldier fly (BSF) performance when fed poultry, swine, or dairy manure at different rates (18 or 27 g/2 d until 40% prepupation). The results indicated that larvae fed with the control diet (Gainesville diet) were the heaviest (+31-70%); however, for other life-history traits, those fed the higher feed rate of poultry manure produced comparable results to the control. Larvae fed more resource, regardless of manure type, weighed more as larvae (+3-9%), pupae (+22-48%), and adults (+18-42%), developed faster (up to 3-4 d), had a higher percentage reach the prepupal stage (+2-16%), lived longer as adults (+1 d), and converted more resource to biomass (up to 1% more) than those fed at the lower rate. Yet, no difference was detected in dry matter (DM) reduction across feed rate for a given manure type. Based on these results, all three manure types can be digested by black soldier fly larvae, thus demonstrating their potential for waste management.

Assessment of the N- and P-Fertilization Effect of Black Soldier Fly (Diptera: Stratiomyidae) By-Products on Maize.

Project description:To meet the growing demand for an alternative animal protein source, the Black Soldier Fly (BSF) (Hermetia illucens) industry is expanding. Thus, the valuation of its byproducts, foremost BSF frass, is getting more economic and ecological weight. Three different residues, BSF frass, larval skins, and dead adult flies, were compared with a mineral and an organic commercial fertilizer in a pot trial with maize (Zea mays L., [Poales: Poaceae]). byproducts were applied in three nutrient-based application rates (180; 215 kg N/ha; 75 kg P2O5/ha), and plant nutrients, physiological and yield parameters were measured at harvest date. Ground flies had the highest N-fertilizing effect of all byproducts, similar to commercial mineral and organic fertilizers used as controls, whereas its proportion of the BSF production systems' output is low. Frass as the abundant byproduct showed comparably low N-fertilization effects. Its low N availability was attributed to volatilization losses, mainly driven by high pH and ammonium contents. BSF frass as the main byproduct output is more suited as a basic fertilizer or potting substrate amendment than as a short-term organic fertilizer. Postprocessing of frass seems reasonable. For a profound assessment of frass as fertilizer, several aspects (e.g., the overall impact of postprocessing, plant strengthening and plant protection potential, effects on microbial processes) must be clarified.
