Molecular characterization of two Bangladeshi infectious bursal disease virus isolates using the hypervariable sequence of VP2 as a genetic marker.

ABSTRACT: Two Bangladeshi infectious bursal disease virus (IBDV) isolates collected in 2007, termed GB1 and GB3, were subjected to comparative sequencing and phylogenetic analyses. Sequence analysis of a 474-bp hypervariable region in the VP2 gene revealed that among four major amino acid substitutions observed in the strains, two were unique to GB1 and GB3 (Ser217Leu and Ala270Thr) while one substitution was only found in GB1 (Asn299Ser). Among IBDVs from Bangladesh including GB1 and GB3, the rate of identity and homology was around 97~99%. The amino acid sequences of GB1 and GB3 differ from those of previous Bangladeshi IBDV isolates and contain amino acid substitutions Pro222Ala and Asn299Ser (in GB3 only). Phylogenetic analysis revealed that GB1 and GB3 are grouped with other very virulent IBDVs of European and American origin in contrast to two previously isolated Bangladeshi IBDV strains (GenBank accession Nos. AF362776 and AF260317), which belong to the Asian group. It was concluded that GB1 and GB3 belong to a very virulent group of IBDVs. However, amino acid sequences of GB1 and GB3 differ from those of the other Bangladeshi IBDVs by one or two amino acids encoded in the hypervariable region of the VP2 gene.

Project description:Infectious bursal disease (IBD) is an acute and fatal immunosuppressive disease caused by infectious bursal disease virus (IBDV). As an obligate intracellular parasite, IBDV infection is strictly regulated by host factors. Knowledge on the antiviral activity and possible mechanism of host factors might provide the theoretical basis for the prevention and control of IBD. In this study, RNA-sequencing results indicated that many host factors were induced by IBDV infection, among which the expression levels of OASL (2´,5´-oligadenylate synthetase-like protein) was significantly upregulated. OASL overexpression significantly inhibited IBDV replication, whereas OASL knockdown promoted IBDV replication. Interestingly, the antiviral ability of OASL was independent of its canonical enzymatic activity, i.e., OASL targeted viral protein VP2 for degradation, depending on the autophagy receptor p62/SQSTM1 in the autophagy pathway. Additionally, the 316 lysine (K) of VP2 was the key site for autophagy degradation, and its replacement with arginine disrupted VP2 degradation induced by OASL and enhanced IBDV replication. Importantly, our results for the first time indicate a unique and potent defense mechanism of OASL against double-stranded RNA virus by interaction with viral proteins, which leads to their degradation.ImportanceOASL (2´,5´-oligadenylate synthetase-like protein) exhibits broad-spectrum antiviral effects against single-stranded RNA viruses in mammals, potentially serving as a promising target for novel antiviral strategies. However, its role in inhibiting the replication of double-stranded RNA viruses (dsRNA viruses), such as infectious bursal disease virus (IBDV), in avian species remains unclear. Our findings indicated a unique and potent defense mechanism of OASL against dsRNA viruses. It has been previously shown in mammals that OASL inhibits virus replication through increasing interferon production. The groundbreaking aspect of our study is the finding that OASL has the ability to interact with IBDV viral protein VP2 and target it for degradation and thus exerts its antiviral effect. Our results reveal the interaction between avian natural antiviral immune response and IBDV infection. Our study not only enhances our understanding of bird defenses against viral infections but can also inform strategies for poultry disease management.

Project description:Infectious Bursal Disease (IBD) is a well-described disease in young chickens. It is caused by the Infectious Bursal Disease Virus (IBDV), which has a bi-segmented, double-strand RNA genome. The absence of a lipidic envelope makes IBDV highly resistant to environmental conditions. Consequently, it is widely reported around the world. Fourteen samples retrieved from chickens exhibiting apparent alterations of the bursa of Fabricius between 2017 and 2021 were included in the study. These samples were passaged into embryonated eggs and the presence of IBD was confirmed through RT-PCR. The PCR products were sequenced and analyzed to characterize the Chilean IBDV isolates for comparison with GenBank sequences, including vaccines sequences currently used in Chile.Phylogenetic analysis classified the Chilean sequences as A1B1, except the sample 15002_CL_2021 which was classified as A2B1. On the other hand, all Chilean viruses were grouped as B1, based on viral segment B. Estimated evolutionary divergence between different genogroups supports these clustering. Moreover, samples 13936_CL_2017, 14038_CL_2017, 14083_CL_2017, 14145_CL_2018, 14431_CL_2019, and 14459_CL_2019 showed high similitude with the D78 and ViBursa CE vaccines (both currently used in Chile). Viruses 14010_CL_2018, 14040_CL_2017, 14514_CL_2019 and 14019_CL_2017 exhibited patterns that do not exactly fit either vaccine. Finally, viruses 15,041 N-_CL_2021, 15,041 N+_CL_2021, and 15004_CL_2021 showed even more differences regarding both vaccines.This is the first study in Chile to analyze the genetic sequences of IBDV isolates. The different assessments conducted as part of the study suggest a close relationship with vaccines currently in use. Interestingly, one of the viruses exhibited a reassortment in its genome segments, which could confer new characteristics to the virus. However, new approaches would be required to establish the origin of the isolated viruses, as well as how the recombination is changing its virulence or morbidity.
