Emotional imagery: assessing pleasure and arousal in the brain's reward circuitry.

ABSTRACT: Research on emotional perception and learning indicates appetitive cues engage nucleus accumbens (NAc) and medial prefrontal cortex (mPFC), whereas amygdala activity is modulated by the emotional intensity of appetitive and aversive cues. This study sought to determine patterns of functional activation and connectivity among these regions during narrative emotional imagery. Using event-related fMRI, we investigate activation of these structures when participants vividly imagine pleasant, neutral, and unpleasant scenes. Results indicate that pleasant imagery selectively activates NAc and mPFC, whereas amygdala activation was enhanced during both pleasant and unpleasant imagery. NAc and mPFC activity were each correlated with the rated pleasure of the imagined scenes, while amygdala activity was correlated with rated emotional arousal. Functional connectivity of NAc and mPFC was evident throughout imagery, regardless of hedonic content, while correlated activation of the amygdala with NAc and mPFC was specific to imagining pleasant scenes. These findings provide strong evidence that pleasurable text-driven imagery engages a core appetitive circuit, including NAc, mPFC, and the amygdala.

Project description:Reward motivation and emotion share common dimensions of valence and arousal, but the nature of interactions between the two constructs is relatively unclear. On the one hand, based on the common valence dimension, valence-compatible interactions are expected where reward motivation would facilitate the processing of compatible (i.e., positive) emotion and hamper the processing of incompatible (i.e., negative) emotion. On the other hand, one could hypothesize valence-general interactions driven by the arousal dimension, where the processing of both positive and negative emotions would be facilitated under reward motivation. Currently, the evidence for valence-compatible vs. valence-general type interactions between reward motivation and goal-relevant emotion is relatively mixed. Moreover, as most of the previous work focused primarily on appetitive motivation, the influence of aversive motivation on goal-relevant emotion is largely unexplored. To address these important gaps, in the present study, we investigated the interactions between motivation and categorization of facial emotional expressions by manipulating the valence dimension of motivation (appetitive and aversive motivation levels) together with that of emotion (positive and negative valence stimuli). Specifically, we conducted two behavioral experiments to separately probe the influence of appetitive and aversive motivation (manipulated via an advance cue signaling the prospect of monetary gains in Experiment 1 and losses in Experiment 2, respectively) on the categorization of happy, fearful, and neutral faces. We tested the two competing hypotheses regarding the interactions between appetitive/aversive motivation and emotional face categorization: Valence-compatible vs. Valence-general. We found evidence consistent with valence-general interactions where both appetitive and aversive motivation facilitated the categorization of happy and fearful faces relative to the neutral ones. Our findings demonstrate that interactions between reward motivation and categorization of emotional faces are driven by the arousal dimension, not by valence.

Project description:ObjectiveAlterations in serotonin signalling within the brain-gut axis have been implicated in the pathophysiology of irritable bowel syndrome (IBS) and is a treatment target. Acute tryptophan depletion (ATD) decreases brain serotonin (5-hydroxytryptamine; 5-HT) levels, and increases visceral perception and negative emotional bias in patients with IBS. The aim of the present study was to determine the effect of ATD on brain activity and connectivity during visceral stimuli in healthy women, and to compare the ATD-induced brain connectivity of an arousal circuit in female patients with IBS without ATD.Methods12 healthy females (19-25 years) were studied under placebo (PLA) conditions and ATD. Functional MRI measurements were performed during a rectal barostat protocol, consisting of random non-painful and maximal tolerable distensions. Partial least squares analyses and structural equation modelling were used to evaluate the effect of ATD on functional and effective brain connectivity during distension. Results in healthy controls under ATD were compared with the effective connectivity of brain responses to 45 mm Hg rectal distension in 14 female patients with constipation-predominant IBS (IBS-C) (24-50 years).ResultsIn healthy controls, ATD resulted in increased response of an extensive brain network to balloon distension, including the amygdala and nodes of emotional arousal and homeostatic afferent networks. The effect was greater during high inflation, suggesting greater engagement of the central serotonion system with more aversive visceral stimuli. Effective connectivity analysis revealed a profound effect of ATD on coupling between emotional arousal network nodes, resulting in loss of negative feedback inhibition of the amygdala. A near-identical pattern was identified in the patients with IBS-C.ConclusionsThe findings are consistent with an ATD-induced disinhibition of and increased connectivity within an emotional arousal network during aversive stimulation. Together with the previous demonstration of ATD-induced visceral hyperalgesia in healthy controls, and the near-identical effective connectivity pattern observed in patients with IBS-C, these findings suggest that dysregulation of this brain network may play a role in central pain amplification and IBS pathophysiology.
