Camera-enabled techniques for organic synthesis.

ABSTRACT: A great deal of time is spent within synthetic chemistry laboratories on non-value-adding activities such as sample preparation and work-up operations, and labour intensive activities such as extended periods of continued data collection. Using digital cameras connected to computer vision algorithms, camera-enabled apparatus can perform some of these processes in an automated fashion, allowing skilled chemists to spend their time more productively. In this review we describe recent advances in this field of chemical synthesis and discuss how they will lead to advanced synthesis laboratories of the future.

Functional Optimization of Designer Cardiac Organoids Enabled by Machine Learning Techniques

Project description:Stem cell organoids are powerful models for studying organ development, disease modeling, drug screening, and regenerative medicine applications. The convergence of organoid technology, tissue engineering, and artificial intelligence (AI) could potentially enhance our understanding of the design principle for organoid engineering. In this study, we utilized micropatterning techniques to create a designer library of 230 cardiac organoids with 7 geometric designs (Circle 200, Circle 600, Circle 1000, Rectangle 1:1, Rectangle 1:4, Star 1:1, and Star 1:4). We employed manifold learning techniques to analyze single organoid heterogeneity based on 10 physiological parameters. We successfully clustered and refined our cardiac organoids based on their functional similarity using unsupervised machine learning approaches, thus elucidating unique functionalities associated with geometric designs. We also highlighted the critical role of calcium rising time in distinguishing organoids based on geometric patterns and clustering results. This innovative integration of organoid engineering and machine learning enhances our understanding of structure-function relationships in cardiac organoids, paving the way for more controlled and optimized organoid design.

Table-salt enabled interface-confined synthesis of covalent organic framework (COF) nanosheets.

Project description:Two-dimensional covalent organic frameworks (COFs) are gaining tremendous interest for their potential applications in a diversity of fields. However, synthesis of COF nanosheets (CONs) usually suffers from tedious exfoliation processes and low yields. Herein, we present an exfoliation-free and scalable strategy to prepare few-layered CONs based on interface-confined synthesis, in which cheap and recyclable table salt (NaCl) is used as the sacrificial substrate. Salt particles are introduced into the reaction system, creating billions of solid-liquid interfaces. Oligomers formed upon the reaction between monomers are immediately adsorbed on salt surfaces, and the following polymerization leading to crystalline CONs is exclusively confined to salt surfaces. Salts can be easily removed by water washing, producing CONs with the thickness down to a few nanometers and lateral sizes up to hundreds of micrometers depending on the size of salt particles and the concentration of monomers. Four different kinds of CONs, both imine-linked and boron-containing, are synthesized from this generic method. As a demonstration, we prepare highly permeable and selective membranes using resultant CONs as building blocks. Thanks to the defect-free stacking of CONs with thin thicknesses and large lateral sizes on porous substrates, the membranes precisely separate similarly sized dyes while allowing ultrafast water permeation. This interface-confined strategy opens a new platform for the controllable and scalable synthesis of COF nanosheets and is essential for the burgeoning real-world applications of COFs in various fields.

An Automated Toolchain for Camera-Enabled Sensing of Drinking Water Chlorine Residual.

Project description:Chlorine residual concentration is an important parameter to prevent pathogen growth in drinking water. Disposable color changing test strips that measure chlorine in tap water are commercially available to the public; however, the color changes are difficult to read by eye, and the data are not captured for water service providers. Here we present an automated toolchain designed to process digital images of free chlorine residual test strips taken with mobile phone cameras. The toolchain crops the image using image processing algorithms that isolate the areas relevant for analysis and automatically white balances the image to allow for use with different phones and lighting conditions. The average red, green, and blue (RGB) color values of the image are used to predict a free chlorine concentration that is classified into three concentration tiers (<0.2 mg/L, 0.2-0.5 mg/L, or >0.5 mg/L), which can be reported to water users and recorded for utility use. The proposed approach was applied to three different phone types under three different lighting conditions using a standard background. This approach can discriminate between concentrations above and below 0.5 mg/L with an accuracy of 90% and 94% for training and testing data sets, respectively. Furthermore, it can discriminate between concentrations of <0.2 mg/L, 0.2-0.5 mg/L, or >0.5 mg/L with weighted-averaged F1 scores of 79% and 88% for training and testing data sets, respectively. This tool sets the stage for tap water consumers and water utilities to gather frequent measurements and high-resolution temporal and spatial data on drinking water quality.

Single 5-centimeter-aperture metalens enabled intelligent lightweight mid-infrared thermographic camera.

Project description:Existing mid-infrared thermographic cameras rely on a stack of refractive lenses, resulting in bulky and heavy imaging systems that restrict their broader utility. Here, we demonstrate a lightweight metalens-based thermographic camera (MTC) enabled by a single 0.5-mm-thick, 3.7-g-weight, flat, and mass-producible metalens. The large aperture size (5 cm) of our metalens, when combined with an uncooled focal plane array, enables thermal imaging at distances of tens of meters. By computationally removing the veiling glare, our MTC realizes the temperature mapping with an inaccuracy of less than ±0.7% within the range of 35° to 700°C and shows exceptional environmental adaptability. Furthermore, by using intelligent algorithms and spectral filtering, our uncooled MTC enables visualization and quantification of the SF6 gas leakage at a long distance of 5 m, with a remarkable minimum detectable leak rate of 0.2 sccm. Our work opens the door to the lightweight and multifunctional intelligent thermal imaging systems.

Ultrathin two-dimensional conjugated metal-organic framework single-crystalline nanosheets enabled by surfactant-assisted synthesis.

Project description:Two-dimensional conjugated metal-organic frameworks (2D c-MOFs) have recently emerged for potential applications in (opto-)electronics, chemiresistive sensing, and energy storage and conversion, due to their excellent electrical conductivity, abundant active sites, and intrinsic porous structures. However, developing ultrathin 2D c-MOF nanosheets (NSs) for facile solution processing and integration into devices remains a great challenge, mostly due to unscalable synthesis, low yield, limited lateral size and low crystallinity. Here, we report a surfactant-assisted solution synthesis toward ultrathin 2D c-MOF NSs, including HHB-Cu (HHB = hexahydroxybenzene), HHB-Ni and HHTP-Cu (HHTP = 2,3,6,7,10,11-hexahydroxytriphenylene). For the first time, we achieve single-crystalline HHB-Cu(Ni) NSs featured with a thickness of 4-5 nm (∼8-10 layers) and a lateral size of 0.25-0.65 μm2, as well as single-crystalline HHTP-Cu NSs with a thickness of ∼5.1 ± 2.6 nm (∼10 layers) and a lateral size of 0.002-0.02 μm2. Benefiting from the ultrathin feature, the synthetic NSs allow fast ion diffusion and high utilization of active sites. As a proof of concept, when serving as a cathode material for Li-ion storage, HHB-Cu NSs deliver a remarkable rate capability (charge within 3 min) and long-term cycling stability (90% capacity retention after 1000 cycles), superior to the corresponding bulk materials and other reported MOF cathodes.

Quantum correlation enhanced super-resolution localization microscopy enabled by a fibre bundle camera.

Project description:Despite advances in low-light-level detection, single-photon methods such as photon correlation have rarely been used in the context of imaging. The few demonstrations, for example of subdiffraction-limited imaging utilizing quantum statistics of photons, have remained in the realm of proof-of-principle demonstrations. This is primarily due to a combination of low values of fill factors, quantum efficiencies, frame rates and signal-to-noise characteristic of most available single-photon sensitive imaging detectors. Here we describe an imaging device based on a fibre bundle coupled to single-photon avalanche detectors that combines a large fill factor, a high quantum efficiency, a low noise and scalable architecture. Our device enables localization-based super-resolution microscopy in a non-sparse non-stationary scene, utilizing information on the number of active emitters, as gathered from non-classical photon statistics.

Machine learning enabled orthogonal camera goniometry for accurate and robust contact angle measurements.

Project description:Characterization of surface wettability plays an integral role in physical, chemical, and biological processes. However, the conventional fitting algorithms are not suitable for accurate estimation of wetting properties, especially on hydrophilic surfaces, due to optical distortions triggered by changes in the focal length of the moving drops. Therefore, here we present an original setup coupled with Convolutional Neural Networks (CNN) for estimation of Contact Angle (CA). The developed algorithm is trained on 3375 ground truth images (at different front-lit illuminations), less sensitive to the edges of the drops, and retains its stability for images that are synthetically blurred with higher Gaussian Blurring (GB) values (GB: 0-22) if compared to existing goniometers (GB: 0-12). Besides, the proposed technique can precisely analyze drops of various colors and chemistries on different surfaces. Finally, our automated orthogonal camera goniometer has a significantly lower average standard deviation (6.7° vs. 14.6°) and coefficient of variation (14.9 vs. 29.2%) than the existing techniques and enables wettability assessment of non-spherical drops on heterogeneous surfaces.

Millimeter-precision positioning for wide-angle indoor area enabled by metalens-integrated camera.

Project description:Due to signal shielding caused by building structures, conventional mature positioning technologies such as the Global Positioning System (GPS) are only suitable for outdoor navigation and detection. However, there are many scenarios that urgently require high-precision indoor positioning technologies, such as indoor wireless optical communications (OWCs), navigation in large buildings, and warehouse management. Here, we proposed a millimeter-precision indoor positioning technology based on metalens-integrated camera, which determines the position of the device through imaging of beacon LEDs. Thanks to the wide-angle imaging design of our metalens, the camera can accurately capture images of beacon LEDs even when it is situated in distant corner locations. Consequently, our localization scheme achieves millimeter-level positioning accuracy across majority of wide-angle (∼120°) indoor area. Compared to traditional positioning schemes by photodiode (PD), our imaging-based approach demonstrates superior resistance to interference, thereby safeguarding positioning precision from the external signals influence. Furthermore, the compact dimensions and high performances of the positioning device make it suitable for integration into highly portable devices, such as smartphones and drones, revealing its broad potential applications in the future.

mRNA delivery enabled by metal-organic nanoparticles.

Project description:mRNA therapeutics are set to revolutionize disease prevention and treatment, inspiring the development of platforms for safe and effective mRNA delivery. However, current mRNA delivery platforms face some challenges, including limited organ tropism for nonvaccine applications and inflammation induced by cationic nanoparticle components. Herein, we address these challenges through a versatile, noncationic nanoparticle platform whereby mRNA is assembled into a poly(ethylene glycol)-polyphenol network stabilized by metal ions. Screening a range of components and relative compositional ratios affords a library of stable, noncationic, and highly biocompatible metal-organic nanoparticles with robust mRNA transfection in vitro and in mice. Intravenous administration of the lead mRNA-containing metal-organic nanoparticles enables predominant protein expression and gene editing in the brain, liver, and kidney, while organ tropism is tuned by varying nanoparticle composition. This study opens an avenue for realizing metal-organic nanoparticle-enabled mRNA delivery, offering a modular approach to assembling mRNA therapeutics for health applications.

Silicon solar cell-enabled organic photoelectrochemical transistor optoelectronics.

Project description:Organic electrochemical transistors (OECTs) have been increasingly explored for innovative electronic devices. However, they inherently demand two power suppliers, which is unfavorable for the utilization of portable and wearable systems with strict energy requirements. Herein, by assembling a monocrystalline silicon solar cell into the OECT circuit with light as fuel, we demonstrated the possibility of a self-powered and light-modulated operation of organic photoelectrochemical transistor (OPECT) optoelectronics. Exemplified by poly(3,4-ethylenedioxythiophene):poly(styrenesulfonate) (PEDOT:PSS)-based depletion-mode and accumulation-mode OECTs, different light-addressable configurations were constructed, and the corresponding characteristics were systematically studied and compared. Different device behaviors with distinct characteristics could be achieved with the appropriate usage of light stimulation. Toward applications, optologics were designed with various parameters depending on the incident irradiance. Light-controlled OPECT unipolar inverters were further demonstrated and optimized with respect to the power source and resistance. This work features new OPECT optoelectronics combined with proper flexible substrates and solar cells for potential applications in portable and wearable devices.Electronic supplementary materialSupplementary material is available in the online version of this article at 10.1007/s40843-022-2295-8.
