The first consistent and reproducible evaluation of the effect of aging on dynamic signature is reported. Project description:The signature has long been in use for the user verification. These signatures have user specific features that differentiate the individual for authentication. The signature verification can be offline or online. The offline verification considers only the static features of the signatures through the signature image, while the online verification considers various dynamic features associated with the signature such as pen pressure, pen tilt angle, velocity, acceleration, pen up and pen down, etc at various time stamps which are recorded using special digitizing tablets such as Wacom devices (STU-500, STU-530 and DTU-1031) [1,14] etc. In todays scenario, smartphones are widely used world-wide, and come equipped with various sensors e.g. accelerometer, gyroscope, magnetometer, GPS, etc. able to capture sensor logs and have been used widely in the literature to capture the dynamics of users' behaviour while a signer signs on his smartphone. However, there is scarcity of publicly available databases for the online signatures collected using smartphone. In the present work, we describe biometric signature dataset iSignDB captured using smartphone. The iSignDB [6,10] consists of the genuine signature samples of a user as well as the skilled forgery samples where imposter was given multiple attempts to mimic the mannerisms of the original signer before giving skilled forgery samples. A total of 30 samples towards the genuine signature over 3 sessions with 10 samples per session while 15 samples of the skilled forgery with 5 samples per session were collected. Each of the session were at least 15 days apart. The iOS and Android based smartphones (namely iPhone7 and Redmi Note 7) were used for the data collection. The sensors used to collect this data, present in the smartphone are the gyroscope, magnetometer, GPS, and accelerometer. Smartphones having sensors any one lesser than these four, were not used for data collection, in order to have consistent number of features under each sample. They generate the following sensor readings: angular velocity, acceleration, orientation, geomagnetic field in the x, y, and z directions, position, which is collected using the MATLAB Mobile App installed in the smartphone, that sends the data to a licensed MathWorks cloud account in the form of a multitude of sensor logs. Each sample has image of the signature along with sensor readings. Some of the publicly available smartphone biometric signature databases are DooDB [2], MOBISIG [3], eBioSign DS 2 [7], etc. in which at least acceleration sensor reading is present but the iSignDB ensures these five of the sensor readings (acceleration, angular velocity, magnetic field, orientation, position) under each sample. This dataset can be successfully used to design smartphone biometric signature authentication system which is robust against a number of spoof attacks [11], [12], [13], [14]. As every user has a unique way of handling his/her smartphone which varies over different level of emotional intelligence of the user over a time period, this dataset can also be used for behavioural analysis of the users.

Project description:A principled approach to the analysis of eye movements for behavioural biometrics is laid down. The approach grounds in foraging theory, which provides a sound basis to capture the uniqueness of individual eye movement behaviour. We propose a composite Ornstein-Uhlenbeck process for quantifying the exploration/exploitation signature characterising the foraging eye behaviour. The relevant parameters of the composite model, inferred from eye-tracking data via Bayesian analysis, are shown to yield a suitable feature set for biometric identification; the latter is eventually accomplished via a classical classification technique. A proof of concept of the method is provided by measuring its identification performance on a publicly available dataset. Data and code for reproducing the analyses are made available. Overall, we argue that the approach offers a fresh view on either the analyses of eye-tracking data and prospective applications in this field.

Project description:In traditional astronomies across the world, groups of stars in the night sky were linked into constellations-symbolic representations rich in meaning and with practical roles. In some sky cultures, constellations are represented as line (or connect-the-dot) figures, which are spatial networks drawn over the fixed background of stars. We analyse 1802 line figures from 56 sky cultures spanning all continents, in terms of their network, spatial, and brightness features, and ask what associations exist between these visual features and culture type or sky region. First, an embedded map of constellations is learnt, to show clusters of line figures. We then form the network of constellations (as linked by their similarity), to study how similar cultures are by computing their assortativity (or homophily) over the network. Finally, we measure the diversity (or entropy) index for the set of constellations drawn per sky region. Our results show distinct types of line figures, and that many folk astronomies with oral traditions have widespread similarities in constellation design, which do not align with cultural ancestry. In a minority of sky regions, certain line designs appear universal, but this is not the norm: in the majority of sky regions, the line geometries are diverse.

Project description:We analyzed expression of 81 normal muscle samples from humans of varying ages, and have identified a molecular profile for aging consisting of 250 age-regulated genes. This molecular profile correlates not only with chronological age but also with a measure of physiological age. We compared the transcriptional profile of muscle aging to previous transcriptional profiles of aging in the kidney and the brain, and found a common signature for aging in these diverse human tissues. The common aging signature consists of six genetic pathways; four pathways increase expression with age (genes in the extracellular matrix, genes involved in cell growth, genes encoding factors involved in complement activation, and genes encoding components of the cytosolic ribosome), while two pathways decrease expression with age (genes involved in chloride transport and genes encoding subunits of the mitochondrial electron transport chain). We also compared transcriptional profiles of aging in humans to those of the mouse and fly, and found that the electron transport chain pathway decreases expression with age in all three organisms, suggesting that this may be a public marker for aging across species.

Project description:Comparative biogerontology has much to contribute to the study of aging. A broad range of aging rates has evolved to meet environmental challenges, and understanding these adaptations can produce valuable insights into aging. The supra Phylum Lophotrochozoa is particularly understudied and has several groups that have intriguing patterns of aging. Members of the lophotrochozoan phylum Rotifera are particularly useful for aging studies because cohort life tables can be conducted with them easily, and biochemical and genomic tools are available for examining aging mechanisms. This paper reviews a variety of caloric restriction regimens, small molecule inhibitors, and dietary supplements that extend rotifer lifespan, as well as important interactions between caloric restriction and genotype, antioxidant supplements, and TOR and JNK pathways, and the use of RNAi to identify key genes involved in modulating the aging response. Examples of how rapamycin and JNK inhibitor exposure keeps mortality rates low during the reproductive phase of the life cycle are presented, and the ease of conducting life table experiments to screen natural products from red algae for life extending effects is illustrated. Finally, experimental evolution to produce longer-lived rotifer individuals is demonstrated, and future directions to determine the genetic basis of aging are discussed.

Project description:Chemoresistance mediated by insulin resistance (IR) in HCC has already been validated. However, the underlying mechanism, especially the involvement of microRNAs (miRNAs) was unelucidated. In this study, miRNA microarrays and bioinformatics methods were employed to determine the dysregulation of miRNA by IR in HCC cells, and quantitative RT-PCR (qRT-PCR) was applied to valid the miRNA array data. Of all the 2006 miRNAs screened, 32 miRNAs were found up or down regulated between the HepG2/IR cells and its parental cells. Further literature mining revealed that some of these miRNAs may function as oncogenes or tumor suppressors that contribute to tumor progression, recurrence, and metastasis which eventually lead to chemotherapeutic resistance. Interestingly, bioinformatics analysis by Gene Ontology (GO) enrichment pathway indicating that function of the predicted target genes of these dysregulated miRNAs were significantly enriched in the processes related with biosynthesis, catabolism, modification etc., and Kyoto Encyclopedia of Genes and Genomes (KEGG) mapping showed that the biological regulatory mechanisms were integrated in cancer-related pathways. Moreover, we also constructed a network which connected the differentially expressed miRNAs to target genes, GO enrichments and KEGG pathways to reveal the hub miRNAs, genes and pathways. Collectively, our present study demonstrated the possible miRNAs and predicted target genes involving in the pathophysiology of insulin resistant HCC, providing novel insights into the molecular mechanisms of multidrug resistance in the insulin resistant HepG2 cells.

Project description:BackgroundColorectal cancer (CRC) is the third most common cancer worldwide. The opening of the TCGA and GEO databases has promoted the progress of CRC prognostic assessment, while the aging-related risk signature has never been mentioned.MethodsR software packages, GSEA software, Venn diagram, Metascape, STRING, Cytoscape, cBioPortal, TIMER and GeneMANIA website were used in this study.ResultsAging-related gene sets, GO_AGING, GO_CELL_AGING and GO_CELLULAR_SENESCENCE, were activated significantly in CRC tissues. We constructed an aging-related risk signature using LASSO COX regression in training group TCGA and validated in testing group GSE39582. The risk score was significantly associated with the overall survival of CRC patients, whose stability was clarified by stratified survival analysis and accuracy was demonstrated using the ROC curve. The risk score was significantly increased in the advanced stage, T3-4, N1-3 and M1 and positively correlated with the richness of immune cell infiltration in the tumor microenvironment. We further investigated the molecular characteristics of 15 hub genes at the DNA and protein levels and performed GSEA between high- and low-risk groups.ConclusionsThe aging-related signature is a reliable prognostic analysis model and can predict the severity and immune cell infiltration of CRC patients.

Project description:Aging is a major risk factor for age-related ocular diseases including age-related macular degeneration in the retina and retinal pigment epithelium (RPE), cataracts in the lens, glaucoma in the optic nerve, and dry eye syndrome in the cornea. We used targeted metabolomics to analyze metabolites from young (6 weeks) and old (73 weeks) eyes in C57 BL6/J mice. Old mice had diminished electroretinogram responses and decreased number of photoreceptors in their retinas. Among the 297 detected metabolites, 45-114 metabolites are significantly altered in aged eye tissues, mostly in the neuronal tissues (retina and optic nerve) and less in cornea, RPE/choroid, and lens. We noted that changes of metabolites in mitochondrial metabolism and glucose metabolism are common features in the aged retina, RPE/choroid, and optic nerve. The aging retina, cornea, and optic nerve also share similar changes in Nicotinamide adenine dinucleotide (NAD), 1-methylnicotinamides, 3-methylhistidine, and other methylated metabolites. Metabolites in taurine metabolism are strikingly influenced by aging in the cornea and lens. In conclusion, the aging eye has both common and tissue-specific metabolic signatures. These changes may be attributed to dysregulated mitochondrial metabolism, reprogrammed glucose metabolism and impaired methylation in the aging eye. Our findings provide biochemical insights into the mechanisms of age-related ocular changes.

Project description:microRNA (miRNA) expression profiles varied greatly among current studies due to different technological platforms and small sample size. Systematic and integrative analysis of published datesets that compared the miRNA expression profiles between hepatocellular carcinoma (HCC) tissue and paired adjacent noncancerous liver tissue was performed to determine candidate HCC associated miRNAs. Moreover, we further validated the confirmed miRNAs in a clinical setting using qRT-PCR and Tumor Cancer Genome Atlas (TCGA) dataset. A miRNA integrated-signature of 5 upregulated and 8 downregulated miRNAs was identified from 26 published datesets in HCC using robust rank aggregation method. qRT-PCR demonstrated that miR-93-5p, miR-224-5p, miR-221-3p and miR-21-5p was increased, whereas the expression of miR-214-3p, miR-199a-3p, miR-195-5p, miR-150-5p and miR-145-5p was decreased in the HCC tissues, which was also validated on TCGA dataset. A miRNA based score using LASSO regression model provided a high accuracy for identifying HCC tissue (AUC = 0.982): HCC risk score = 0.180E_miR-221 + 0.0262E_miR-21 - 0.007E_miR-223 - 0.185E_miR-130a. E_miR-n = Log 2 (expression of microRNA n). Furthermore, expression of 5 miRNAs (miR-222, miR-221, miR-21 miR-214 and miR-130a) correlated with pathological tumor grade. Cox regression analysis showed that miR-21 was related with 3-year survival (hazard ratio [HR]: 1.509, 95%CI: 1.079-2.112, P = 0.016) and 5-year survival (HR: 1.416, 95%CI: 1.057-1.897, P = 0.020). However, none of the deregulated miRNAs was related with microscopic vascular invasion. This study provides a basis for further clinical application of miRNAs in HCC.
