An illustrated key to the genera of Thripinae (Thysanoptera, Thripidae) from Iran.

ABSTRACT: An illustrated key is provided for the identification of 35 genera of Thripinae (Thysanoptera: Thripidae) from Iran with comments for each genus. Chirothrips maximi Ananthakrishnan and Limothrips cerealium Haliday are recorded from Iran for the first time. A checklist is provided of Thripinae recorded from this country.

Reproductive Behavior of Echinothrips americanus (Thysanoptera: Thripidae).

Project description:Most Thysanoptera possess a haplo-diploid reproductive mode and reproduce via arrhenotoky. Females can mature eggs successively throughout almost their entire life, but in most terebrantian thrips spermiogenesis is complete by adult male eclosion, and testes contain only mature spermatids. In parasitoid wasps this phenomenon of preadult spermiogenesis is described as prospermatogeny. It is unclear if prospermatogeny and this predetermined sperm quantity have implications for mating strategy and fitness. In this study, we give a detailed description of mating behavior of the thripine species Echinothrips americanus, which largely corresponds with the only available data of another species of this family, Frankliniella occidentalis (Thysanoptera: Thripidae). With investigations using light microscopy, we describe for the first time the chronological sequence of internal processes during copulation. The release of male accessory gland material followed subsequently by spermatozoa indicates production of a female-determined type 1 spermatophore. Despite prospermatogeny, males are able to inseminate 10 females with an equal amount of spermatozoa. It is only the quantity of glandular material that decreases with the number of previous copulations. Based on these new findings, the reproductive strategy of this species is discussed.

An illustrated key to the genera and subgenera of the Alysiini (Hymenoptera, Braconidae, Alysiinae), with three genera new for China.

Project description:An illustrated key to the genera and subgenera of the Alysiini (Hymenoptera, Braconidae, Alysiinae) from China is presented. Three genera new for China are reported: Adelurola Strand, 1924, Anisocyrta Foerster, 1863, and Pentapleura Foerster, 1863. The total for China is 26 genera of Alysiini and an additional seven subgenera (excluding the nominal subgenera, which are included in the total of genera). The known Chinese species are listed under each genus and the biology is summarised. Separatatus sinicus (Zheng, Chen & Yang, 2012) and Grammospila eurys (Chen & Wu, 1994) are new combinations. Regetus Papp, 1999, and Adelphenaldis Fischer, 2003, are new synonyms of Eusynaldis Zaykov & Fischer, 1982. In addition, Eusynaldis Zaykov & Fischer and Synaldis Foerster, 1863, are treated as subgenera of Aspilota Foerster, 1863, and Dinotrema Foerster, 1863, respectively. An aberrant species of Separatatus Chen & Wu, 1994, S. parallelussp. n., is described from Yunnan and Hainan.

Microbiome profiling of the onion thrips, Thrips tabaci Lindeman (Thysanoptera: Thripidae).

Project description:The gut microbial community structure of adult Thrips tabaci collected from 10 different agro-climatically diverse locations of India was characterized by using the Illumina MiSeq platform to amplify the V3 region of the 16S rRNA gene of bacteria present in the sampled insects. Analyses were performed to study the bacterial communities associated with Thrips tabaci in India. The complete bacterial metagenome of T. tabaci was comprised of 1662 OTUs of which 62.25% belong to known and 37.7% of unidentified/unknown bacteria. These OTUs constituted 21 bacterial phyla of 276 identified genera. Phylum Proteobacteria was predominant, followed by Actinobacteria, Firmicutes, Bacteroidetes and Cyanobacteria. Additionally, the occurrence of the reproductive endosymbiont, Wolbachia was detected at two locations (0.56%) of the total known OTUs. There is high variation in diversity and species richness among the different locations. Alpha-diversity metrics indicated the higher gut bacterial diversity at Bangalore and lowest at Rahuri whereas higher bacterial species richness at T. tabaci samples from Imphal and lowest at Jhalawar. Beta diversity analyses comparing bacterial communities between the samples showed distinct differences in bacterial community composition of T. tabaci samples from different locations. This paper also constitutes the first record of detailed bacterial communities associated with T. tabaci. The location-wise variation in microbial metagenome profile of T. tabaci suggests that bacterial diversity might be governed by its population genetic structure, environment and habitat.

Detection and Localization of Wolbachia in Thrips palmi Karny (Thysanoptera: Thripidae).

Project description:Thrips palmi Karny is a globally distributed polyphagous agricultural pest. It causes huge economic loss by its biological behaviors like feeding, reproduction and transmission of tospoviruses. Since T. palmi shows close morphological similarities with other thrips species, we employed mitochondrial cytochrome oxidase 1 (mtCO1) gene as a molecular marker. BLAST analysis of this sequence helped us to identify the collected specimen as T. palmi. We observed the female to male ratio of about 3:1 from collected samples and suspected the presence of Wolbachia. The presence of Wolbachia was detected by PCR using genus specific primers of 16S rRNA gene. Further confirmation of Wolbachia strain was achieved by conducting PCR amplification of three ubiquitous genes ftsZ, gatB and groEL. A phylogenetic tree was constructed with concatenated sequences of ftsZ and gatB gene to assign supergroup to Wolbachia. Finally, we localized Wolbachia in abdominal region of the insect using fluorescent in situ hybridization with the help of confocal microscope. Our result confirmed the presence of Wolbachia supergroup B strain for the first time in T. palmi.

Cypermethrin resistance and reproductive types in onion thrips, Thrips tabaci (Thysanoptera: Thripidae).

Project description:Cypermethrin resistance and reproductive types were examined for T. tabaci strains. Some arrhenotokous and thelytokous strains encoded the sodium channel mutation (T929I) involved in cypermethrin resistance. However, the resistance levels varied to some degree among the strains. A cytochrome P450 inhibitor, piperonyl butoxide, showed different synergistic effects on the strains examined. These results suggest that fundamental and additional levels of cypermethrin resistance in T. tabaci are conferred respectively by reduced sensitivity of the sodium channel and by cytochrome P450-mediated detoxification.

Chromosome-level genome assembly of bean flower thrips Megalurothrips usitatus (Thysanoptera: Thripidae).

Project description:Bean flower thrips Megalurothrips usitatus is a staple pest of cowpea and other legumes and causes dramatic economic losses. Its small size allows for easy concealment, and large reproductive capacity easily leads to infestations. Despite the importance of a genome in developing novel management strategies, genetic studies on M. usitatus remain limited. Thus, we generated a chromosome-level M. usitatus genome using a combination of PacBio long read and Hi-C technologies. The assembled genome was 238.14 Mb with a scaffold N50 of 13.85 Mb. The final genome was anchored into 16 pseudo-chromosomes containing 14,000 genes, of which 91.74% were functionally annotated. Comparative genomic analyses revealed that expanded gene families were enriched in fatty acid metabolism and detoxification metabolism (ABC transporters), and contracted gene families were strongly associated with chitin-based cuticle development and sensory perception of taste. In conclusion, this high-quality genome provides an invaluable resource for us to understand the thrips' ecology and genetics, contributing to pest management.

Onion Thrips (Thysanoptera: Thripidae) Feeding Promotes Infection By Pantoea ananatis in Onion.

Project description:Onion thrips, Thrips tabaci Lindeman, is a primary insect pest of onions (Allium cepa) worldwide. Onion thrips cause feeding damage by destroying epidermal tissue. They are also vectors of Pantoea ananatis (Serrano) Mergaert, the bacteria that causes center rot. Onions with center rot develop white streaks with water-soaked margins along the onion leaves, which turn necrotic and lead to bulb rot during storage. The role of thrips feeding on the establishment and progression of bacterial infection in onions has not been investigated. Onions infested with thrips and inoculated with P. ananatis had more necrotic tissue and symptoms were more severe with increasing thrips density. We conducted a fluorescence microscopy study that examined how P. ananatis (expressing a fluorescence protein gene) colonized a control group of onions without thrips in comparison to a test group of onions with thrips. We found that P. ananatis colonized some onions in the control group because of naturally existing wounds in the epidermal tissue but more colonization was found in the thrips infested group because of the increased presence of entry points caused by thrips feeding. Overall, our results demonstrate that wounds caused by thrips feeding facilitate center rot development by providing entry sites for the bacteria into leaf tissue.

Complete mitochondrial genome sequence for the <i>Thrips hawaiiensis</i> (Thysanoptera: Thripidae).

Project description:Thrips hawaiiensis (Morgan) (Thysanoptera: Thripidae) is a common Thysanoptera insect widely distributed in Asia and the Pacific, it damages various plants. In this study the complete mitochondrial genome of T. hawaiiensis was sequenced and characterized by using next-generation sequencing technique. The total length of the complete genome is 15,357 bp and A + T content of 77.8% (GeneBank accession No. MW582621). The T. hawaiiensis mitochondrial genome consists of 13 protein-coding genes (PCGs), 2 ribosomal RNA genes, 22 transfer RNA genes (tRNAs) and 2 non-coding control regions (Dloop region). According to previous studies, only a few complete mitochondrial genomes from Order Thysanoptera have been reported. Thus, T. hawaiiensis complete mitochondrial genome sequence reported will provide molecular information for mitochondrial genome research on Thysanoptera.

(type species: Opius (Agnopius) daghoides Zaykov & Fischer, 1983) from West and East Palaearctic regions and Pseudosteresgen. nov. (type species: Biosteresadanaensis Fischer & Beyarslan, 2005) from West Palaearctic region. Two new species are described and illustrated: Cephaloplitesgijswijtisp. nov. from Greece and Cavopiusdepressoriussp. nov. from S. Korea. Opius (Hypocynodus) kilisanus Fischer & Beyarslan, 2005 is a new synonym of Cephaloplitesmocsaryi Szépligeti, 1897. Repellency of Plant Extracts against the Legume Flower Thrips Megalurothrips sjostedti (Thysanoptera: Thripidae).

Project description:Megalurothrips sjostedti Trybom is an important pest of cowpea (Vigna unguiculata L.) in Africa. To propose an alternative to chemical control, the repellency of 24 plant extracts was evaluated against adult female thrips of M. sjostedti in the laboratory. Plant extracts in ethanol were separately applied on a filter paper disk in a still air visual cue olfactometer. The results showed highly significant differences in repellency among extract type, concentration and their interactions. We classified the level of repellency into four categories as strong, good, moderate and weak or non- repellent based on hierarchical ascendant classification. We identified Piper nigrum, Cinnamomum zeylanicum, Cinnamomum cassia as strong repellents. Five extracts were classified as good, eight as moderate and the remaining eight extracts were weak or non-repellent. Repellency of the extracts increased with the concentration suggesting that the behavioral response of M. sjostedti was dose-dependent. Mono- and sesquiterpene hydrocarbon compounds from seven highly repellent extracts were identified by gas chromatography-mass spectrometry (GC/MS). The use of repellent extracts could be useful in developing integrated pest management strategies for thrips on legume crops. In this regard, the specific modes of action of the identified compounds need to be investigated to incorporate them into the existing crop protection strategies.
