Biogenic silver nanoparticles using Rhinacanthus nasutus leaf extract: synthesis, spectral analysis, and antimicrobial studies.

ABSTRACT: Nanotechnology is gaining momentum due to its ability to transform metals into nanoparticles. The synthesis, characterization, and applications of biologically synthesized nanomaterials have become an important branch of nanotechnology. Plant extracts are a cost-effective, ecologically friendly, and efficient alternative for the large-scale synthesis of nanoparticles. In this study, silver nanoparticles (AgNps) were synthesized using Rhinacanthus nasutus leaf extract. After exposing the silver ions to the leaf extract, the rapid reduction of silver ions led to the formation of AgNps in solution. The synthesis was confirmed by ultraviolet-visible spectroscopy, Fourier transform infrared spectroscopy, and transmission electron microscopy. The in vitro antimicrobial activity of the AgNps synthesized using R. nasutus leaf extract was investigated against Bacillus subtilis, Staphylococcus aureus, Pseudomonas aeruginosa, Klebsiella pneumonia, Escherichia coli, Aspergillus niger, and Aspergillus flavus using a disc diffusion method. The AgNps showed potential activity against all of the bacterial strains and fungal colonies, indicating that R. nasutus has the potential to be used in the development of value-added products in the biomedical and nanotechnology-based industries.

Project description:Agro-industrial wastes are rich in polyphenols and other bioactive compounds, and valorizing these wastes is a crucial worldwide concern for saving health and the environment. In this work, olive leaf waste was valorized by silver nitrate to produce silver nanoparticles (OLAgNPs), which exhibited various biological, antioxidant, anticancer activities against three cancer cell lines, and antimicrobial activity against multi-drug resistant (MDR) bacteria and fungi. The obtained OLAgNPs were spherical, with an average size of 28 nm, negatively charged at -21 mV, and surrounded by various active groups more than the parent extract based on FTIR spectra. The total phenolic and total flavonoid contents significantly increased in OLAgNPs by 42 and 50% over the olive leaf waste extract (OLWE); consequently, the antioxidant activity of OLAgNPs increased by 12% over OLWE, recording an SC50 of OLAgNPs of 5 µg/mL compared to 30 µg/mL in the extract. The phenolic compound profile detected by HPLC showed that gallic acid, chlorogenic acid, rutin, naringenin, catechin, and propyl gallate were the main compounds in the HPLC profile of OLAgNPs and OLWE; the content of these compounds was higher in OLAgNPs than OLWE by 16-fold. The higher phenolic compounds in OLAgNPs are attributable to the significant increase in biological activities of OLAgNPs than that of OLWE. OLAgNPs successfully inhibited the proliferation of three cancer cell lines, MCF-7, HeLa, and HT-29, by 79-82% compared to 55-67% in OLWE and 75-79% in doxorubicin (DOX). The preliminary worldwide problem is multi-drug resistant microorganisms (MDR) because of the random use of antibiotics. Therefore, in this study, we may find the solution in OLAgNPs with concentrations of 2.5-20 µg/mL, which significantly inhibited the growth of six MDR bacteria L. monocytogenes, B. cereus, S. aureus, Y. enterocolitica, C. jejuni, and E. coli with inhibition zone diameters of 25-37 mm and six pathogenic fungi in the range of 26-35 mm compared to antibiotics. OLAgNPs in this study may be applied safely in new medicine to mitigate free radicals, cancer, and MDR pathogens.

Project description:BackgroundOxidative stress and nonenzymatic protein glycation lead to serious diabetic complications that increase the risk of mortality. Rhinacanthus nasutus leaf crude extracts are previously reported for their antidiabetic, antiglycation, and antioxidant potential.ObjectiveThe present study was performed to prepare a standardized rhinacanthins-rich extract (RRE) and evaluate its superoxide scavenging and antiglycation effects as compared to its marker compounds, namely, rhinacanthin-C (RC), rhinacanthin-D (RD), and rhinacanthin-N (RN).Materials and methodsRRE was obtained by microwave-assisted green extraction along with a simple step of fractionation using Amberlite® column. RC, RD, and RN were isolated from the RRE using silica gel column chromatography. Superoxide scavenging activity was performed by cyclic voltammetry, and fructose-mediated human serum albumin glycation model was used for antiglycation activity. In silico studies were conducted to identify the structure-activity relationships of rhinacanthins.ResultsOn the basis of kinetic measurements, RRE exhibited the most potent antioxidant activity via ErCi mechanism, with a 50% inhibitory concentration (IC50) value of 8.0 μg/mL, antioxidant capacity of 39439 M-1, and binding constant of 45709 M-1. Project description:BackgroundViscum orientale is a largely used parasitic plant with traditional medicinal properties. They are considered to possess the medicinal properties of host tree which they grow on. It's a least explored plant with ethanopharmacological importance. As a result, the current work aimed to investigate the biological effects of Viscum orientale extract and silver nanoparticles (AgNPs) generated from it.MethodsAgNPs synthesized using Viscum orientale plant extract and analysed on time dependent series and was characterized using Ultra Violet UV-visible spectra, Fourier Transform Infrared Spectroscopy FTIR, X-ray diffraction (XRD), Energy Dispersive X-ray Spectroscopy (EDX), Scanning Electron Microscopy (SEM). Further using disc method anti-microbial assay was performed following antioxidation screening using 1,1-diphenyl-2-picryl-hydrazyl (DPPH), reducing power and nitric oxide content and heamgglutination with human blood.ResultsOn green synthesis using silver, the phyto contituents of plant Viscum orientale effectively reduced silver ions at 3-4 h of continuous stirring to form AgNPs. UV-vis spectra showed a typical peak of AgNPs at 480 nm. The FTIR analysis confirmed the covering of silver layers to bio-compounds of the extract. SEM analysis represented AgNPs as spherical morphologies ranging from 119-222 nm. AgNPs exhibited impressive zone of inhibition against Escherichia coli (8.1 ± 0.3 mm), Staphylococcus aureus (10.3 ± 0.3 mm), Bacillus subtilis (7.3 ± 0.3 mm), Bacillus cereus (8.2 ± 0.3 mm), Salmonella typhi (7.1 ± 0.2 mm). AgNps exhibited efficiency against DPPH at EC50 value of 57.60 µg/ml. and reducing power at EC50 of 53.42 µg/ml and nitric oxide scavenging of EC50 of 56.01 µg/ml concentration. Further, anthelmintic activity results showed synthesized nanoparticles significant reduction in the paralysis time to 5.4 ± 0.3 min and death time to 6.5 ± 0.6 min in contrast to the individual factors. On hemagglutination using AgNPs, above 80 µg/ml of concentration showed very significant effect on comparison with water extract.ConclusionSynthesized AgNPs using Viscum orientale water extract displayed versatile biological activity than individual extract. This study has forecasted a new path to explore more on this AgNPs for further research.
