Adjusting the balance between effective loading and vector migration of macrophage vehicles to deliver nanoparticles.

ABSTRACT: The nature of macrophage allows the possibility that this cell type could be used as drug delivery system to track therapeutic drug nanoparticles (NPs) in cancer. However, there is no existing research on the regulation between effective loading of NPs and targeted delivery of macrophages. Here, we investigated the important parameters of intracellular NP quantity and the vector migration rate. Macrophage loading capacity was obtained by comparing the uptake quantity of varisized NPs, and the delivery ability of loaded cells was determined by measuring vector migration rates. We observed a positive correlation between the size of NPs and directed macrophage migration. Our findings suggest that the molecular mechanism of migration vector rate regulation involved increased expression levels of colony-stimulating factor-1 (CSF-1) receptor and integrin induced by 100-nm and 500-nm particles. The ability of macrophages uptake to varisized NPs showed the opposite trend, with the increased vector rate of cell migration influenced by NPs. We are able to demonstrate the important balance between effective macrophage loading and targeted delivery. By adjusting the balance parameters, it will be possible to utilize NPs in macrophage-mediated disease diagnosis and therapy.

Project description:The design of adaptable drug delivery systems able to encapsulate and release drugs with different wettability has been attracting widespread interest. Additionally, many attempts have been made to tune hydrophobic/hydrophilic drug release kinetics over time, avoiding the so-called burst release. In this scenario, hydrogels resulting from the assembly of micellar structures showing a hydrophobic core and a hydrophilic shell could represent a promising alternative to design versatile drug vehicles. In this regard, this work aimed at designing new thermosensitive micellar hydrogels starting from a custom-made amphiphilic poly(ether urethane) (PEU). Specifically, a commercial triblock copolymer (Poloxamer® 407), selected to ensure the temperature-driven chain arrangement into micelles, was reacted with 1,6-diisocyanatohexane and 1,4-cyclohexanedimethanol. The successful PEU synthesis was proved by size-exclusion chromatography ( M ¯ w 50000 Da) and infrared spectroscopy. Subsequently, the wettability-driven drug arrangement within the micelle network as well as the influence of drug-loading on the resultant formulation thermosensitivity was investigated by selecting ibuprofen (IBU) and ibuprofen sodium salt (IBUSS) as hydrophobic and hydrophilic drugs, respectively. Specifically, growing drug amounts were loaded into PEU solutions, and the average hydrodynamic micelle diameters and the critical micellar temperatures (CMT) were measured. Systems containing IBU at the highest tested concentration (i.e., 20 mg/mL) showed a significantly higher micelle average diameter (58.2 ± 4.7 nm) and a remarkably lower CMT (8.9°C) with respect to both the control (40.1 ± 1.4 nm and 21.6°C) and IBUSS-loaded formulations (37.3 ± 2.1 nm and 22.4°C). Then, the influence of drug encapsulation on the temperature at which micelles begin to aggregate was rheologically assessed, showing that IBU-loading induced a decrease in this parameter (14.6, 8.7, and 13.7°C for virgin, IBU-loaded, and IBUSS-loaded hydrogel, respectively). Finally, IBU and IBUSS releasing mechanism was analysed using the Korsmayer-Peppas model (n value of 0.63 ± 0.007 and 0.89 ± 0.003 for IBU- and IBUSS-loaded gels, respectively). Thanks to their micellar organisation, the here-developed hydrogel platform allowed the encapsulation of a high number of molecules with different wettability. Additionally, these systems exhibited tunable payload-releasing time without burst release and open the way toward the engineering of smart systems for the sustained co-delivery of multiple drugs in a target tissue/organ.

Project description:Macrophages are critical regulators of the tumor microenvironment and often present an immuno-suppressive phenotype, supporting tumor growth and immune evasion. Promoting a robust pro-inflammatory macrophage phenotype has emerged as a therapeutic modality that supports tumor clearance, including through synergy with immune checkpoint therapies. Polyglucose nanoparticles (macrins), which possess high macrophage affinity, are useful vehicles for delivering drugs to macrophages, potentially altering their phenotype. Here, we examine the potential of functionalized macrins, synthesized by crosslinking carboxymethyl dextran with L-lysine, as effective carriers of immuno-stimulatory drugs to tumor-associated macrophages (TAMs). Azide groups incorporated during particle synthesis provided a handle for click-coupling of propargyl-modified β-cyclodextrin to macrins under mild conditions. Fluorescence-based competitive binding assays revealed the ability of β-cyclodextrin to non-covalently bind to hydrophobic immuno-stimulatory drug candidates (Keq ~ 103 M-1), enabling drug loading within nanoparticles. Furthermore, transcriptional profiles of macrophages indicated robust pro-inflammatory reprogramming (elevated Nos2 and Il12; suppressed Arg1 and Mrc1 expression levels) for a subset of these immuno-stimulatory agents (UNC2025 and R848). Loading of R848 into the modified macrins improved the drug's effect on primary murine macrophages by three-fold in vitro. Intravital microscopy in IL-12-eYFP reporter mice (24 h post-injection) revealed a two-fold enhancement in mean YFP fluorescence intensity in macrophages targeted with R848-loaded macrins, relative to vehicle controls, validating the desired pro-inflammatory reprogramming of TAMs in vivo by cell-targeted drug delivery. Finally, in an intradermal MC38 tumor model, cyclodextrin-modified macrin NPs loaded with immunostimulatory drugs significantly reduced tumor growth. Therefore, efficient and effective repolarization of tumor-associated macrophages to an M1-like phenotype-via drug-loaded macrins-inhibits tumor growth and may be useful as an adjuvant to existing immune checkpoint therapies.

Project description:BackgroundGene therapy has held promises for treating specific genetic diseases. However, the key to clinical application depends on effective gene delivery.MethodsUsing a large animal model, we developed two pharmaceutical formulations for gene delivery in the pigs' vagina, which were made up of poly (β-amino ester) (PBAE)-plasmid polyplex nanoparticles (NPs) based two gel materials, modified montmorillonite (mMMT) and hectorite (HTT).FindingsBy conducting flow cytometry of the cervical cells, we found that PBAE-GFP-NPs-mMMT gel was more efficient than PBAE-GFP-NPs-HTT gel in delivering exogenous DNA intravaginally. Next, we designed specific CRISPR/SpCas9 sgRNAs targeting porcine endogenous retroviruses (PERVs) and evaluated the genome editing efficacy in vivo. We discovered that PERV copy number in vaginal epithelium could be significantly reduced by the local delivery of the PBAE-SpCas9/sgRNA NPs-mMMT gel. Comparable genome editing results were also obtained by high-fidelity version of SpCas9, SpCas9-HF1 and eSpCas9, in the mMMT gel.
