Isolation of a 2-hydroxytetrahydrofuran complex from copper-promoted hydroxylation of THF.

ABSTRACT: A complex of a binucleating macrocyclic ligand comprising a [Cu(II)(?-OH)Na](2+) core reacts with CuI in THF/CH3CN to yield a novel species with a deprotonated 2-hydroxytetrahydrofuran (THF-2-ol) bridging between Cu(II) and Na(I) ions. The complexes were characterized by X-ray crystallography, electron paramagnetic resonance spectroscopy, and electrospray ionization mass spectrometry. (18)O-labeling studies support incorporation of the O atom from ?-OH into the coordinated THF-2-ol ligand.

Copper-Promoted One-Pot Approach: Synthesis of Benzimidazoles.

Project description:A facile, one-pot, and proficient method was developed for the production of various 2-arylaminobenzimidazoles. This methodology is based for the first time on a copper catalyst promoted domino C-N cross-coupling reaction for the generation of 2-arylaminobenzimidazoles. Mechanistic investigations revealed that the synthetic pathway involves a copper-based desulphurization/nucleophilic substitution and a subsequent domino intra and intermolecular C-N cross-coupling reactions. Some of the issues typically encountered during the synthesis of 2-arylaminobezimidazoles, including the use of expensive catalytic systems and the low reactivity of bromo precursors, were addressed using this newly developed copper-catalyzed method. The reaction procedure is simple, generally with excellent substrate tolerance, and provides good to high yields of the desired products.

The method employs disulfide reagents, copper(II) acetate, and DMSO solvent at 90-130 °C. Application of this methodology to the direct trifluoromethylsulfenylation of C-H bonds was demonstrated.

A Dinuclear Copper(II) Complex Electrochemically Obtained via the Endogenous Hydroxylation of a Carbamate Schiff Base Ligand: Synthesis, Structure and Catalase Activity.

Project description:In the present work, we report a neutral dinuclear copper(II) complex, [Cu2(L1)(OH)], derived from a new [N,O] donor Schiff base ligand L1 that was formed after the endogenous hydroxylation of an initial carbamate Schiff base H2L coordinated with copper ions in an electrochemical cell. The copper(II) complex has been fully characterized using different techniques, including X-ray diffraction. Direct current (DC) magnetic susceptibility measurements were also performed at variable temperatures, showing evidence of antiferromagnetic behavior. Its catalase-like activity was also tested, demonstrating that this activity is affected by temperature.

Mechanism of O2 activation and substrate hydroxylation in noncoupled binuclear copper monooxygenases.

Project description:Peptidylglycine α-hydroxylating monooxygenase (PHM) and dopamine β-monooxygenase (DβM) are copper-dependent enzymes that are vital for neurotransmitter regulation and hormone biosynthesis. These enzymes feature a unique active site consisting of two spatially separated (by 11 Å in PHM) and magnetically noncoupled copper centers that enables 1e- activation of O2 for hydrogen atom abstraction (HAA) of substrate C-H bonds and subsequent hydroxylation. Although the structures of the resting enzymes are known, details of the hydroxylation mechanism and timing of long-range electron transfer (ET) are not clear. This study presents density-functional calculations of the full reaction coordinate, which demonstrate: (i) the importance of the end-on coordination of superoxide to Cu for HAA along the triplet spin surface; (ii) substrate radical rebound to a CuII hydroperoxide favors the proximal, nonprotonated oxygen; and (iii) long-range ET can only occur at a late step with a large driving force, which serves to inhibit deleterious Fenton chemistry. The large inner-sphere reorganization energy at the ET site is used as a control mechanism to arrest premature ET and dictate the correct timing of ET.

Copper-promoted synthesis of 1,4-benzodiazepinones via alkene diamination.

Project description:A new method for the synthesis of 2-aminomethyl functionalized 1,4-benzodiazepin-5-ones is presented. The benzodiazepine core is well-known to interact with biological receptors and many pharmaceutical drugs are derived from this structure. The alkene diamination strategy is employed for the first time for the synthesis of 1,4-benzodiazepinones. In this reaction, copper(2-ethylhexanoate)2 serves as promoter and a range of external amines can be coupled with 2-sulfonamido-N-allyl benzamides to generate the 1,4-benzodiazepinones in good yields.

Project description:The title compound, di-μ-chlorido-dichlorido-1κ(2)Cl-tetra-kis-(tetra-hydro-furan)-1κ(2)O,2κ(2)O-lithiumytterbium(III), [LiYbCl(4)(C(4)H(8)O)(4)], was prepared by the reaction of YbCl(3)(THF)(3) with LiCl in THF (THF is tetra-hydro-furan). The central motif of the structure is a Yb(μ-Cl)(2)Li ring. The Yb atom is hexa-coordinated to four Cl atoms and two THF mol-ecules oriented in a trans fashion. The Li atom has a tetra-hedral environment and is coordinated to two Cl atoms and two THF mol-ecules. No inter-molecular inter-actions other than van der Waals forces were observed. Two of the THF mol-ecules are disordered over two positions.
