Photoinduced Brook-Type Rearrangement of Acylcyclopolysilanes.

ABSTRACT: Previously unknown 1,1,4-tris(trimethylsilyl)-4-acyldodecamethylcyclohexasilanes (Me₃Si)₂Si₆Me₁₂(Me₃Si)COR (16a, R = tert-butyl; 16b, R = 1-adamantyl) have been synthesized by the reaction of the potassium silanides (Me₃Si)₂Si₆Me₁₂(Me₃Si)K with acid chlorides ClCOR, and their photochemical rearrangement reactions have been studied. The molecular structures of 16a,b as determined by single-crystal X-ray diffraction analysis exhibit an unusual twist-boat conformation of the cyclohexasilane ring. When 16a,b were photolyzed with λ >300 nm radiation, they underwent Brook type 1,3-Si → O migration reactions to generate the cyclohexasilanes 17a,b with exocyclic Si=C bonds along with smaller amounts of the ring-enlarged species 19a,b with endocyclic Si=C double bonds. While 17a,b were stable enough to allow characterization by NMR and UV absorption spectroscopy, the less stable products 19a,b could only be observed in the form of their methanol adducts.

Synthesis of constrained bicycloalkanes through bibase-promoted brook rearrangement/radical-polar crossover cyclization.

Project description:Highly constrained bicyclic scaffolds are ubiquitous and attracting increasing interest in pharmaceutical and biotechnology discoveries owing to the enhanced activities. Herein, we report a protocol to access highly substituted constrained bicycloalkanes from readily accessible α-silyl alcohols and olefins through a bibase-promoted Brook rearrangement/radical-polar crossover cyclization (RPCC) process. Of note, the practical procedure features broad substrate scope and good group tolerance under mild and operationally simple conditions, using an inexpensive organic photocatalyst. Gram-scale preparation and diverse synthetic transformations demonstrate opportunities to rapidly construct molecular complexity. Mechanistic studies have indicated that the transformation involves a bibase-promoted radical transfer rearrangement addition/radical-polar crossover cyclization relay sequence, which differs from traditional solitary RPCC reactions.

Photo-Brook rearrangement of acyl silanes as a strategy for photoaffinity probe design.

Project description:Photoaffinity labeling (PAL) is a powerful tool for the identification of non-covalent small molecule-protein interactions that are critical to drug discovery and medicinal chemistry, but this approach is limited to only a small subset of robust photocrosslinkers. The identification of new photoreactive motifs capable of covalent target capture is therefore highly desirable. Herein, we report the design, synthesis, and evaluation of a new class of PAL warheads based on the UV-triggered 1,2-photo-Brook rearrangement of acyl silanes, which hitherto have not been explored for PAL workflows. Irradiation of a series of probes in cell lysate revealed an iPr-substituted acyl silane with superior photolabeling and minimal thermal background labeling compared to other substituted acyl silanes. Further, small molecule (+)-JQ1- and rapamycin-derived iPr acyl silanes were shown to selectively label recombinant BRD4-BD1 and FKBP12, respectively, with minimal background. Together, these data highlight the untapped potential of acyl silanes as a novel, tunable scaffold for photoaffinity labeling.

Photoinduced Skeletal Rearrangement of N-Substituted Colchicine Derivatives.

Project description:Colchicine is an active pharmaceutical ingredient widely used for treating gout, pericarditis, and familial Mediterranean fever with high antimitotic activity. The photoisomerization of colchicine deactivates its anti-inflammatory and antimitotic properties. However, despite numerous reports on colchicine derivatives, their photostability has not been investigated in detail. This report reveals the effects of UV-induced rearrangement on the structure and reports the biological activity of new N-substituted colchicine derivatives.

Me3SiSiMe2(O n Bu): a disilane reagent for the synthesis of diverse silacycles via Brook- and retro-Brook-type rearrangement.

Project description:Herein, a readily available disilane Me3SiSiMe2(O n Bu) has been developed for the synthesis of diverse silacycles via Brook- and retro-Brook-type rearrangement. This protocol enables the incorporation of a silylene into different starting materials, including acrylamides, alkene-tethered 2-(2-iodophenyl)-1H-indoles, and 2-iodobiaryls, via the cleavage of Si-Si, Si-C, and Si-O bonds, leading to the formation of spirobenzosiloles, fused benzosiloles, and π-conjugated dibenzosiloles in moderate to good yields. Preliminary mechanistic studies indicate that this transformation is realized by successive palladium-catalyzed bis-silylation and Brook- and retro-Brook-type rearrangement of silane-tethered silanols.

Stable Silenolates and Brook-Type Silenes with Exocyclic Structures.

Project description:The first silenolates with exocyclic structures [(Me3Si)2Si(Si2Me4)2SiC(R)O]-K+ (2a: R = 1-adamantyl; 2b: mesityl; 2c: o-tolyl) were synthesized by the reaction of the corresponding acylcyclohexasilanes 1a-c with KOtBu. NMR spectroscopy and single-crystal X-ray diffraction analysis suggest that the aryl-substituted silenolates 2b,c exhibit increased character of functionalized silenes as compared to the alkyl-substituted derivative 2a due to the different coordination of the K+ counterion to the SiC(R)O moiety. 2b,c, thus, reacted with ClSiiPr3 to give the exocyclic silenes (Me3Si)2Si(Si2Me4)2Si=C(OSiiPr3)R (3b: R = Mes; 3c: o-Tol), while 2a afforded the Si-silylated acylcyclohexasilane 1d. The thermally remarkably stable compound 3b, which is the first isolated silene with the sp2 silicon atom incorporated into a cyclopolysilane framework, could be fully characterized structurally and spectroscopically.

Density Functional Theory Evaluation of a Photoinduced Intramolecular Aryl Ether Rearrangement.

Project description:Recently, a new approach of converting (hetero)aryl ethers to C-C coupled products via a photoinduced intramolecular rearrangement has been reported. Although this reaction is photocatalyst-free, it requires excitation in the ultraviolet (UV) range. To help refine this process, three different 2-(hetero)aryloxybenzaldehydes are selected from the available substrate scope in which the general mechanism based on experimental results is evaluated using density functional theory calculations. The reaction takes place in the triplet state after photoexcitation and includes three main steps: the addition of carbonyl carbon to the ipso carbon of the aryl ether followed by the C-O cleavage of the resulting spirocyclic intermediates and then the transfer of the formyl proton to afford 2-hydroxybenzophenone-type products. This agrees with the experiments, but the calculated pathways show considerable differences between the three substrates. Above all, either the first or the second step can be rate-determining but not the C-H activation. The important factor behind the differences is the spin-density rearrangement, which is mainly responsible for the barrier of the ether cleavage. Based on the obtained insights, the strategy to improve the ∼250 nm excitation has been briefly discussed, and promising molecules are proposed to improve the scope of this process.

Driving Protein Conformational Changes with Light: Photoinduced Structural Rearrangement in a Heterobimetallic Oxidase.

Project description:The heterobimetallic R2lox protein binds both manganese and iron ions in a site-selective fashion and activates oxygen, ultimately performing C-H bond oxidation to generate a tyrosine-valine cross-link near the active site. In this work, we demonstrate that, following assembly, R2lox undergoes photoinduced changes to the active site geometry and metal coordination motif. Through spectroscopic, structural, and mass spectrometric characterization, the photoconverted species is found to consist of a tyrosinate-bound iron center following light-induced decarboxylation of a coordinating glutamate residue and cleavage of the tyrosine-valine cross-link. This process occurs with high quantum efficiencies (Φ = 3%) using violet and near-ultraviolet light, suggesting that the photodecarboxylation is initiated via ligand-to-metal charge transfer excitation. Site-directed mutagenesis and structural analysis suggest that the cross-linked tyrosine-162 is the coordinating residue. One primary product is observed following irradiation, indicating potential use of this class of proteins, which contains a putative substrate channel, for controlled photoinduced decarboxylation processes, with relevance for in vivo functionality of R2lox as well as application in environmental remediation.
