Biocompatible and photostable AIE dots with red emission for in vivo two-photon bioimaging.

ABSTRACT: Bioimaging systems with cytocompatibility, photostability, red fluorescence, and optical nonlinearity are in great demand. Herein we report such a bioimaging system. Integration of tetraphenylethene (T), triphenylamine (T), and fumaronitrile (F) units yielded adduct TTF with aggregation-induced emission (AIE). Nanodots of the AIE fluorogen with efficient red emission were fabricated by encapsulating TTF with phospholipid. The AIE dots enabled three-dimensional dynamic imaging with high resolution in blood vessels of mouse brain under two-photon excitation.

Long-term two-photon neuroimaging with a photostable AIE luminogen.

Project description:In neuroscience, fluorescence labeled two-photon microscopy is a promising tool to visualize ex vivo and in vivo tissue morphology, and track dynamic neural activities. Specific and highly photostable fluorescent probes are required in this technology. However, most fluorescent proteins and organic fluorophores suffer from photobleaching, so they are not suitable for long-term imaging and observation. To overcome this problem, we utilize tetraphenylethene-triphenylphosphonium (TPE-TPP), which possesses aggregation-induced emission (AIE) and two-photon fluorescence characteristics, for neuroimaging. The unique AIE feature of TPE-TPP makes its nanoaggregates resistant to photobleaching, which is useful to track neural cells and brain-microglia for a long period of time. Two-photon fluorescence of TPE-TPP facilitates its application in deep in vivo neuroimaging, as demonstrated in the present paper.

Photostable fluorescent organic dots with aggregation-induced emission (AIE dots) for noninvasive long-term cell tracing.

Project description:Long-term noninvasive cell tracing by fluorescent probes is of great importance to life science and biomedical engineering. For example, understanding genesis, development, invasion and metastasis of cancerous cells and monitoring tissue regeneration after stem cell transplantation require continual tracing of the biological processes by cytocompatible fluorescent probes over a long period of time. In this work, we successfully developed organic far-red/near-infrared dots with aggregation-induced emission (AIE dots) and demonstrated their utilities as long-term cell trackers. The high emission efficiency, large absorptivity, excellent biocompatibility, and strong photobleaching resistance of the AIE dots functionalized by cell penetrating peptides derived from transactivator of transcription proteins ensured outstanding long-term noninvasive in vitro and in vivo cell tracing. The organic AIE dots outperform their counterparts of inorganic quantum dots, opening a new avenue in the development of fluorescent probes for following biological processes such as carcinogenesis.

Highly Photostable Carbon Dots from Citric Acid for Bioimaging.

Project description:Bioimaging supported by nanoparticles requires low cost, highly emissive and photostable systems with low cytotoxicity. Carbon dots (C-dots) offer a possible solution, even if controlling their properties is not always straightforward, not to mention their potentially simple synthesis and the fact that they do not exhibit long-term photostability in general. In the present work, we synthesized two C-dots starting from citric acid and tris (hydroxymethyl)-aminomethane (tris) or arginine methyl ester dihydrochloride. Cellular uptake and bioimaging were tested in vitro using murine neuroblastoma and ovine fibroblast cells. The C-dots are highly biocompatible, and after 24 h of incubation with the cells, 100% viability was still observed. Furthermore, the C-dots synthesized using tris have an average dimension of 2 nm, a quantum yield of 37%, high photostability and a zeta potential (ζ) around -12 mV. These properties favor cellular uptake without damaging cells and allow for very effective bioimaging.

One-Pot Synthesis of Customized Metal-Phenolic-Network-Coated AIE Dots for In Vivo Bioimaging.

Project description:The integration of aggregation-induced emission luminogens (AIEgens) and inorganic constituents to generate multifunctional nanocomposites has attracted much attention because it couples the bright aggregate-state fluorescence of AIEgens with the diverse imaging modalities of inorganic constituents. Herein, a facile and universal strategy to prepare metal-phenolic-network (MPN)-coated AIE dots in a high encapsulation efficiency is reported. Through precise control on the nucleation of AIEgens and deposition of MPNs in tetrahydrofuran/water mixtures, termed as coacervation, core-shell MPN-coated AIE dots with bright emission are assembled in a one-pot fashion. The optical properties of MPN-coated AIE dots can be readily tuned by varying the incorporated AIEgens. Different metal ions, such as Fe3+ , Ti4+ , Cu2+ , Ni2+ , can be introduced to the nanoparticles. The MPN-coated AIE dots with a red-emissive AIEgen core are successfully used to perform magnetic resonance/fluorescence dual-modality imaging in a tumor-bearing mouse model and blood flow visualization in a zebrafish larva. It is believed that the present study provides a tailor-made nanoplatform to meet the individual needs of in vivo bioimaging.

Aggregation tailored emission of a benzothiazole based derivative: photostable turn on bioimaging.

Project description:Herein, we report a benzothiazole based probe which exhibits aggregation induced emission in mixed solvents, v/v THF : water system. The blue emission observed in the solution is ascribed to the enolic emission of the excited state intramolecular proton transfer (ESIPT) prone benzothiazole chromophore, as the aggregation induced by the protic solvent, water, is expected to restrict the phototautomerization of the probe to the keto form which generally emits in the red region. However, the green emission observed in the solid aggregated state is ascribed to its keto emission, as in the solid state the ESIPT process is activated owing to stable intramolecular hydrogen bonding, giving a keto-aggregation induced emission (AIE) coupled emission. The aggregation process is evident from the particle size and the change in morphology predicted by dynamic light scattering (DLS) and scanning electron microscopy (SEM) respectively, in the aggregated state. Interestingly, the emission in the solution, as well as the solid state, is convincingly mimicked in the fluorescence imaging of various live cancer cells and plant roots.

Novel near-infrared II aggregation-induced emission dots for <i>in vivo</i> bioimaging.

Project description:Near-infrared II fluorescence imaging holds great promise for in vivo imaging and imaging-guided surgery with deep penetration and high spatiotemporal resolution. However, most NIR-II aromatic luminophores suffer from the notorious aggregation-caused quenching (ACQ) effect in the aqueous solution, which largely hinders their biomedical application in vivo. In this study, the first NIR-II organic aggregation-induced emission (AIE) fluorophore (HLZ-BTED), encapsulated as nanoparticles (HLZ-BTED dots) for in vivo biomedical imaging, was designed and synthesized. The NIR-II AIE HLZ-BTED dots showed high temporal resolution, high photostability, outstanding water-solubility and biocompatibility in vitro and in vivo. The HLZ-BTED dots were further used for long-term breast tumor imaging and visualizing tumor-feeding blood vessels, long-term hind limb vasculature and incomplete hind limb ischemia. More importantly, as a proof-of-concept, this is the first time that non-invasive and real-time NIR-II imaging of the gastrointestinal tract in health and disease has been performed, making the AIE dots a promising tool for gastrointestinal (GI) tract research, such as understanding the healthy status of GI peristalsis, diagnosing and evaluating intestinal motility dysfunction, and assessing drug effects on intestinal obstruction.

One step synthesis of efficient red emissive carbon dots and their bovine serum albumin composites with enhanced multi-photon fluorescence for in vivo bioimaging.

Project description:Efficient red emissive carbon dots (CDs) in aqueous solutions are very scarce for high performance bioimaging applications. In this work, we report a one-step solvothermal treatment to synthesize pure red emissive CDs (FA-CDs) from citric acid and urea in formic acid without complicated purification procedures. Photoluminescence quantum yield (PLQY) of 43.4% was observed in their dimethyl sulfoxide solutions. High PLQY up to 21.9% in aqueous solutions was achieved in their bovine serum albumin (BSA) composites (FA-CDs@BSA) with significantly enhanced multi-photon fluorescence. The strong surface electron-withdrawing structure of FA-CDs caused by the high content of C = O groups contributes for their pure red emission. Owing to the significantly enhanced single and multi-photon red fluorescence and enlarged particle sizes after composing with BSA, in vivo tumor imaging and two-photon fluorescence imaging of blood vessels in mouse ear have been realized via intravenous injection of FA-CDs@BSA aqueous solutions.

Using AIE Luminogen for Long-term and Low-background Three-Photon Microscopic Functional Bioimaging.

Project description:Fluorescent probes are one of the most popularly used bioimaging markers to monitor metabolic processes of living cells. However, long-term light excitation always leads to photobleaching of fluorescent probes, unavoidable autofluorescence as well as photodamage of cells. To overcome these limitations, we synthesized a type of photostable luminogen named TPE-TPP with an aggregation induced emission (AIE) characteristic, and achieved its three-photon imaging with femtosecond laser excitation of 1020 nm. By using TPE-TPP as fluorescent probes, three-photon microscopy under 1020 nm excitation showed little photo-damage, as well as low autofluorescence to HeLa cells. Due to the AIE effect, the TPE-TPP nanoaggregates uptaken by cells were resistant to photobleaching under three-photon excitation for an extended period of time. Furthermore, we demonstrated that for the present TPE-TPP AIE the three-photon microscopy (with 1020 nm excitation) had a better signal to noise ratio than the two-photon microscopy (with 810 nm excitation) in tissue imaging.

Enhanced trion emission from colloidal quantum dots with photonic crystals by two-photon excitation.

Project description:For colloidal quantum dots, the ongoing biggest problem is their fluorescence blinking. Until now, there is no generally accepted model for this fluorescence blinking. Here, two-photon excited fluorescence from CdSe/ZnS nanocrystals on silicon nitride photonic crystals is studied using a femtosecond laser. From analysis of the spectra and decay processes, most of the relative trion efficiency is larger than 10%, and the largest relative trion efficiency reaches 46.7%. The photonic crystals enhance the trion emission of CdSe/ZnS nanocrystals, where the enhancement is due to the coupling of the trion emission to the leaky mode of the photonic crystal slab. Moreover, the photonic crystals enhance the Auger-assisted trapping efficiency of electrons/holes to surface states, and then enhance the efficiency of the generations of charge separation and DC electric field, which modifies the trion spectrum. Therefore, a model is present for explaining the mechanism of fluorescence blinking including the effect of the environment.

Folate receptor-targeted aggregation-enhanced near-IR emitting silica nanoprobe for one-photon in vivo and two-photon ex vivo fluorescence bioimaging.

Project description:A two-photon absorbing (2PA) and aggregation-enhanced near-infrared (NIR) emitting pyran derivative, encapsulated in and stabilized by silica nanoparticles (SiNPs), is reported as a nanoprobe for two-photon fluorescence microscopy (2PFM) bioimaging that overcomes the fluorescence quenching associated with high chromophore loading. The new SiNP probe exhibited aggregate-enhanced emission producing nearly twice as strong a signal as the unaggregated dye, a 3-fold increase in two-photon absorption relative to the DFP in solution, and approximately 4-fold increase in photostability. The surface of the nanoparticles was functionalized with a folic acid (FA) derivative for folate-mediated delivery of the nanoprobe for 2PFM bioimaging. Surface modification of SiNPs with the FA derivative was supported by zeta potential variation and (1)H NMR spectral characterization of the SiNPs as a function of surface modification. In vitro studies using HeLa cells expressing a folate receptor (FR) indicated specific cellular uptake of the functionalized nanoparticles. The nanoprobe was demonstrated for FR-targeted one-photon in vivo imaging of HeLa tumor xenograft in mice upon intravenous injection of the probe. The FR-targeting nanoprobe not only exhibited highly selective tumor targeting but also readily extravasated from tumor vessels, penetrated into the tumor parenchyma, and was internalized by the tumor cells. Two-photon fluorescence microscopy bioimaging provided three-dimensional (3D) cellular-level resolution imaging up to 350 ?m deep in the HeLa tumor.
