Outbreak of Rust Caused by Coleosporium asterum on Solidago virgaurea var. gigantea in Ulleung-do.

ABSTRACT: From June to October 2012, severe outbreaks of rust disease on Solidago virgaurea var. gigantea were observed in 5 different regions of Ulleung-do, Korea. Typical rust symptoms appeared as yellow spots, uredinia on leaves, and massive urediniospores covering the entire plant. Severely infected plants became discolored and died. On the basis of its morphological and molecular characteristics, the causal agent was identified as Coleosporium asterum. This pathogen has been reported as rust disease on S. virgaurea in the Canada and United States. In Korea, rust disease on S. virgaurea var. gigantea was reported as Uromyces rudbeckia in 1992. To the best of our knowledge, C. asterum is a novel rust pathogen on S. virgaurea var. gigantea in Korea.

Project description:New substances with antimicrobial properties are needed to successfully treat emerging human, animal, or plant pathogens. Seven clerodane diterpenes, previously isolated from giant goldenrod (Solidago gigantea) root, were tested against Gram-positive Bacillus subtilis, Bacillus spizizenii and Rhodococcus fascians by measuring minimal bactericidal concentration (MBC), minimal inhibitory concentration (MIC) and half-maximal inhibitory concentration (IC50). Two of them, Sg3a (a dialdehyde) and Sg6 (solidagoic acid B), were proved to be the most effective and were selected for further study. Bacillus spizizenii was incubated with the two diterpenes for shorter (1 h) or longer (5 h) periods and then subjected to genome-wide transcriptional analyses. Only a limited number of common genes (28 genes) were differentially regulated after each treatment, and these were mainly related to the restoration of cell membrane integrity and to membrane-related transports. Changes in gene activity indicated that, among other things, K+ and Na+ homeostasis, pH and membrane electron transport processes may have been affected. Activated export systems can be involved in the removal of harmful molecules from the bacterial cells. Inhibition of bacterial chemotaxis and flagellar assembly, as well as activation of genes for the biosynthesis of secondary metabolites, were observed as a general response. Depending on the diterpenes and the duration of the treatments, down-regulation of the protein synthesis-related, oxidative phosphorylation, signal transduction and transcription factor genes was found. In other cases, up-regulation of the genes of oxidation-reduction processes, sporulation and cell wall modification could be detected. Comparison of the effect of diterpenes with the changes induced by different environmental and nutritional conditions revealed several overlapping processes with stress responses. For example, the Sg6 treatment seems to have caused a starvation-like condition. In summary, there were both common and diterpene-specific changes in the transcriptome, and these changes were also dependent on the length of treatments. The results also indicated that Sg6 exerted its effect more slowly than Sg3a, but ultimately its effect was greater.

Project description:Studies of in situ plant response and adaptation to complex environmental stresses, are crucial for understanding the mechanisms of formation and functioning of ecosystems of anthropogenically transformed habitats. We study short- and long-term responses of photosynthetic apparatus (PSA) and anti-oxidant capacity to complex abiotic stresses of common plants Calamagrostis epigejos and Solidago gigantea in semi-natural (C) and heavy metal contaminated habitats (LZ). We found significant differences in leaf pigment content between both plant species growing on LZ plots and their respective C populations. The average values of leaf chlorophyll indexes were 27% lower in the LZ populations of both species and significantly lower in Sg plants in comparison to Ce ones. The average values of the anthocyanin index in CeLZ and SgLZ populations were significantly higher (by 18%) than in their respective controls. In both Ce and Sg plants occurring on LZ plots, the average leaf flavonol indexes were higher than on their controls by 31% and 15% and this index was significantly higher in SgLZ population than CeLZ and CeC plants (by 34% and 54%, respectively). Both Ce and Sg populations growing on LZ plots showed significantly lower photosynthetic rate (A), transpiration rate (E) and stomatal conductance (gs) in comparison to controls. On the other hand, a significantly higher photosynthetic rate was detected in SgLZ than in CeLZ populations. The catalase activities were significantly higher than recorded in Sg than in Ce tissues, irrespective of the plot type. They were also higher in LZ populations than those in controls for both species. Moreover, the H2O2 content in Sg tissues was significantly higher than those in Ce. Hydrogen peroxide content in CeLZ and SgLZ were respectively 39% (non-significant) and 57% higher, compared to their controls. The reverse pattern was found in the case of MDA, whose concentration was significantly higher in the leaves of Ce population compared to the control population. The average MDA concentration in CeLZ populations was 17% higher than in the CeC. In the case of Sg no significant differences were found. Mechanisms of plant species adaptation to industrial areas are crucial for species selection and planning effective reclamation of them. The analysis of chlorophyll fluorescence induction curves as well as well as the results of JIP test revealed the decreased of Fj value despite positive ΔK-band in SgLZ and CeLZ plants suggesting the increased rate of electron transfer from QA to QB at the acceptor side of PSII, thus a high quantity of P680+ and/or effective quenching by exogenous molecules. The increase in the I-P part of the induction curve typically attributed to the reduction of electron transporters (ferredoxin, intermediary acceptors, and NADP) of the PSI acceptor side was observed in both SgC and SgLZ but not in CeLZ populations. These changes demonstrate species-specific effects on electron transport during the light phase of photosynthesis under complex environmental stress. Our results show that Sg and Ce individuals developed a range of structural and functional adaptations to protect PSA against complex environmental stresses (possible combination of heavy metals, water deficiency, temperature, nutrient deficiency and salinity). Both species from LZ plots could tolerate high levels of Cd, Zn and Pb in leaf tissues. Therefore they can be potential candidates for use in phytoremediation of HM contaminated areas. However, further long-term field and experimental research on plant traits response and adaptation to complex environmental stresses on industrial habitats are needed.

Project description:The late goldenrod (Soldiago gigantea Aiton; Asteraceae) is one of the most abundant invasive species in various types of habitats. Its long-creeping plagiotropic rhizomes enable the plant to build up dense, monospecific stands within a short time. Particularly in nature conservation areas, the invasion of goldenrod can cause severe disruptions in the naturally occuring mutualims between plants, insects and higher trophic levels, subsequently impeding the achievement of nature conservation goals. As management options of goldenrod in nature conservation areas are limited, this three-year study aimed to test the effectiveness of three management treatments (two-time mowing, triticale cultivation, and reverse rotary cutting) on four different sites in the Austrian Donau-Auen National Park. The number and height of goldenrod shoots were recorded three times a year on twelve permanent trial plots on each site to test for the effectiveness of the treatments. In addition, vegetation surveys were performed to observe the recovery potential of native plant species. Even though the three-years mowing and the triticale cultivation reduced goldenrod by 95.6% and 97.2% resp., we could find no relation between the effectiveness of the treatment and the intensity of disturbance created by the control option. On the contrary, with a reduction of only 5.4% in goldenrod density the most intensive treatment, the rotary cutting, showed the lowest efficiency. The highest positive effect on the re-establishment of native plant species was recorded with two mowing events per year. Even though the study revealed that certain management options have the potential to effectively reduce goldenrod and to simultaneously increase the establishment success of native species, results can only be seen as so-called snapshots. For example, as shown on site EJW one unforeseeable wild boar digging event transformed a 84.5% reduction into a 4.7% increase in goldenrod density. Therefore, a proper and regular monitoring is essential to be able to react to the effects of unpredictable events that can have severe impact on vegetation dynamics.
