Three New Humulane Sesquiterpenes from Cultures of the Fungus Antrodiella albocinnamomea.

ABSTRACT: Three new humulane-type sesquiterpenes, antrodols A-C (1-3), were isolated from cultures of the fungus Antrodiella albocinnamomea. Their structures were elucidated on the basis of extensive spectroscopic analysis. Antrodols A-C (1-3) are first examples of humulane-type sesquiterpenes isolated from cultures of higher fungi, and antrodol A (1) was the first report of humulane-type sesquiterpene with a methyl rearranged at C-3. All compounds were evaluated in the enzyme inhibition assay against two protein-tyrosine phosphatases (PTPs): MEG2 and PTP1Bc.

Steroids and Sesquiterpenes From Cultures of the Fungus Phellinus igniarius.

Project description:Two new steroids, 3α,17α,19,20-tetrahydroxy-4α-methylpregn-8-ene (1) and 3α,12α,17α,20-tetrahydroxy-4α-methylpregn-8-ene (2) and three new sesquiterpenoids, 12-hydroxy-α-cadinol (3), 3α,12-dihydroxy-δ-cadinol (4), and 3α,6α-dihydroxyspiroax-4-ene (5), have been isolated from cultures of the fungus Phellinus igniarius. Their structures were characterized based on extensive spectroscopic data. In preliminary in vitro assays, compounds 3 and 4 exhibited the vascular-activities against phenylephrine-induced vasoconstriction with the relaxing rates of 11.0 % and 7.0 % at 3 × 10-4 M, respectively.

Irlactane and Tremulane Sesquiterpenes from the Cultures of the Medicinal Fungus Irpex lacteus HFG1102.

Project description:A new irlactane-type, namely irlactin K (1), and 22 tremulane-type sesquiterpenes including fourteen previously undescribed ones, namely irpexolactins A-N (2-15), and a known irlactane-type sesquiterpenoid, were isolated from the fermentation broth of the medicinal fungus Irpex lacteus HFG1102. The structures of all the isolates were characterized by extensive spectroscopic methods, including 1D and 2D NMR and MS spectroscopic analysis. The absolute configurations of irlactin K and the known compound conocenol B (20) were established by single-crystal X-ray diffraction analysis. The vasorelaxant effects of irlactin K (1), irpexolactins A (2), C (4), K (12), and irlactam (22) were evaluated.

Project description:A new chloro-trinoreremophilane sesquiterpene 1, three new chlorinated eremophilane sesquiterpenes 2-4, together with a known compound, eremofortine C (5), were isolated from an Antarctic deep-sea derived fungus, Penicillium sp. PR19N-1. Structures were established using IR, HRMS, 1D and 2D NMR techniques. In addition, the plausible metabolic network of these isolated products is proposed. Compound 1 showed moderate cytotoxic activity against HL-60 and A549 cancer cell lines.

Project description:Five new guanacastane-type diterpenes, named guanacastepenes P-T (1-5), were isolated from cultures of the fungus Psathyrella candolleana. Their structures were elucidated on the basis of extensive spectroscopic methods. All of the compounds were tested for their 11β-hydroxysteroid dehydrogenase (11β-HSD1) inhibitory activity. Compound 3 exhibited inhibitory activity against both human and mouse isozymes of 11β-HSD1 with IC50 values of 6.2 and 13.9 μM, respectively.

The structures were elucidated on the basis of NMR spectroscopic and mass spectrometry data. The absolute configuration of marasmiellin B was determined by application of the modified Mosher's method.
