Project description:A set of unsymmetrical heteroaryl 1,2-diketones were synthesized by a heteroarylation/oxidation sequence with up to 65% isolated yields. Palladium catalyst XPhos Pd G4 and SeO2 were the key reagents used in this methodology, and microwave irradiation was utilized to facilitate an efficient and ecofriendly process. The application of heteroaryl 1,2-diketones is demonstrated through the synthesis of an unsymmetrical 2-phenyl-3-(pyridin-3-yl)quinoxaline (5a) from 1-phenyl-2-(pyridin-3-yl)ethane-1,2-dione (4a). The lowest energy conformations of 4a and 5a were located using Density Functional Theory (DFT) at the M06-2X/def2-TZVP level of theory. Two lowest energy conformations of 4a differ with respect to the position of the N atom in the pyridyl ring and 0.27 kcal/mol energy difference between them corresponds to 60.4 and 39.6% at 50 °C in toluene. Four lowest energy conformations for 5a have the energy differences of 0.01, 0.03 and 0.07 kcal/mol that corresponds to 26.0, 25.7, 24.9 and 23.4%, respectively. A comparison of 4a and 5a to the less hindered analogs (oxalyl chloride and oxalic acid) is used to investigate the structural features and bonding using Natural Bond Orbital (NBO) analysis.

Project description:Diastereoselective synthesis of the trans-decalin-based α-hydroxyl butanone spirocarbocycles bearing all-carbon quaternary stereogenic centers has been achieved via Norrish-Yang photocyclization of trans-decalin-substituted-2,3-butanediones using daylight. Density functional theory (DFT) calculations suggest that this diastereoselective reaction is affected by both substrate conformation and intramolecular hydrogen bonds. The developed chemistry could be applied to synthesizing the derivatives of the trans-decalin-based biologically important natural products.

Project description:We report metal-free organic 1,2-diketones that exhibit fast and highly efficient room-temperature phosphorescence (RTP) with high colour purity under various conditions, including solutions. RTP quantum yields reached 38.2% in solution under Ar, 54% in a polymer matrix in air, and 50% in crystalline solids in air. Moreover, the narrowband RTP consistently dominated the steady-state emission, regardless of the molecular environment. Detailed mechanistic studies using ultrafast spectroscopy, single-crystal X-ray structure analysis, and theoretical calculations revealed picosecond intersystem crossing (ISC) followed by RTP from a planar conformation. Notably, the phosphorescence rate constant k p was unambiguously established as ∼5000 s-1, which is comparable to that of platinum porphyrins (representative heavy-metal phosphor). This inherently large k p enabled the high-efficiency RTP across diverse molecular environments, thus complementing the streamlined persistent RTP approach. The mechanism behind the photofunction has been elucidated as follows: (1) the large k p is due to efficient intensity borrowing of the T1 state from the bright S3 state, (2) the rapid ISC occurs from the S1 to the T3 state because these states are nearly isoenergetic and have a considerable spin-orbit coupling, and (3) the narrowband emission results from the minimal geometry change between the T1 and S0 states. Such mechanistic understanding based on molecular orbitals, as well as the structure-RTP property relationship study, highlighted design principles embodied by the diketone planar conformer. The fast RTP strategy enables development of organic phosphors with emissions independent of environmental conditions, thereby offering alternatives to precious-metal based phosphors.

Project description:Large bone defects cannot form a callus and exhibit high complication rates even with the best treatment strategies available. Tissue engineering approaches often use scaffolds designed to match the properties of mature bone. However, natural fracture healing is most efficient when it recapitulates development, forming bone via a cartilage intermediate (endochondral ossification). Because mechanical forces are critical for proper endochondral bone development and fracture repair, we hypothesized that recapitulating developmental mechanical forces would be essential for large bone defect regeneration in rats. Here, we engineered mesenchymal condensations that mimic the cellular organization and lineage progression of the early limb bud in response to local transforming growth factor-β1 presentation from incorporated gelatin microspheres. We then controlled mechanical loading in vivo by dynamically tuning fixator compliance. Mechanical loading enhanced mesenchymal condensation-induced endochondral bone formation in vivo, restoring functional bone properties when load initiation was delayed to week 4 after defect formation. Live cell transplantation produced zonal human cartilage and primary spongiosa mimetic of the native growth plate, whereas condensation devitalization before transplantation abrogated bone formation. Mechanical loading induced regeneration comparable to high-dose bone morphogenetic protein-2 delivery, but without heterotopic bone formation and with order-of-magnitude greater mechanosensitivity. In vitro, mechanical loading promoted chondrogenesis and up-regulated pericellular matrix deposition and angiogenic gene expression. In vivo, mechanical loading regulated cartilage formation and neovascular invasion, dependent on load timing. This study establishes mechanical cues as key regulators of endochondral bone defect regeneration and provides a paradigm for recapitulating developmental programs for tissue engineering.

A simple and high-yielding protocol was developed for the synthesis of a wide range of 1,2-diaryl diketones, which are the backbone for a variety of medicinally important molecules.

Project description:While the use of visible light to drive chemical reactivity is of high importance to the development of environmentally benign chemical transformations, the concomitant use of a stoichiometric electron donor or acceptor is often required to steer the desired redox behavior of these systems. The low-cost and ubiquity of tertiary amine bases has led to their widespread use as reductive additives in photoredox catalysis. Early use of trialkylamines in this context was focused on their role as reductive excited state quenchers of the photocatalyst, which in turn provides a more highly reducing catalytic intermediate. In this Account, we discuss some of the observations and thought processes that have led from our use of amines as reductive additives to their use as complex substrates and intermediates for natural product synthesis. Early attempts by our group to construct key carbon-carbon bonds via free-radical intermediates led to the observation that some trialkylamines readily behave as efficient hydrogen atom donors under redox-active photochemical conditions. In the wake of in-depth mechanistic studies published in the 1970s, 1980s and 1990s, this understanding has in turn allowed for a systematic approach to the design of a number of photochemical methodologies through rational tuning of the amine component. Minimization of the C-H donicity of the amine additive was found to promote desired C-C bond formation in a number of contexts, and subsequent elucidation of the amine's redox fate has sparked a reevaluation of the amine's role from that of reagent to that of substrate. The reactivity of tertiary amines in these photochemical systems is complex, and allows for a number of mechanistic possibilities that are not necessarily mutually exclusive. A variety of combinations of single-electron oxidation, C-H abstraction, deprotonation, and β-scission result in the formation of reactive intermediates such as α-amino radicals and iminium ions. These processes have been explored in depth in the photochemical literature and have resulted in a firm mechanistic grasp of the behavior of amine radical cations in fundamental systems. Harnessing the synthetic potential of these transient species represents an ongoing challenge for the controlled functionalization of amine substrates, because these mechanistic possibilities may result in undesired byproduct formation or substrate decomposition. The presence of tertiary amines in numerous alkaloids, pharmaceuticals, and agrochemicals lends credence to the potential utility of this chemistry in natural product synthesis, and herein we will discuss how these transformations might be controlled for synthetic purposes.

Project description:In this work, we exploit the electronic features of tetrathiafulvalene (TTF) as a backbone in synthesizing chiral derivatives. The aim is to make use of TTF's well-known and unique redox and semiconducting properties in the fields of enantio-selective recognition and chiral charge transfer (CT) complex preparation, with the ultimate objective of obtaining devices with various potential applications, ranging from plasmonics to quantum computing. In particular, both cyclohexane-bis (TTF-amide)-based enantiomers 1-(S,S) and 1-(R,R), stable under an oxidation regime, have been selected, and under these conditions, the electrochemical enantiospecific response of the four possible systems, coming from the combination with L- and D-tartaric acid, respectively, was tested. The 1:tartaric acid adducts show lower oxidation potentials than the pristine 1, together with clear enantio-discrimination demonstrated by sizeable potential differences in the range of 29-46 mV between the diastereomeric adducts. Because the oxidation potential of 1 suggests the possibility of the formation of CT complexes, impedance and FT-IR spectra were recorded to confirm this hypothesis in the case of the CT complex 1:I2. The experimental results obtained through the FT-IR analysis were also compared with the theoretical results deriving from the DFT-based calculations.

Project description:Replacing a faulty gene with a correct copy has become a viable therapeutic option as a result of recent progress in gene editing protocols. Targeted integration of therapeutic genes in hematopoietic stem cells has been achieved for multiple genes using Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR)/Cas9 system and Adeno-Associated Virus (AAV) to carry a donor template. Although this is a promising strategy to correct genetic blood disorders, it is associated with toxicity and loss of function in CD34+ hematopoietic stem and progenitor cells, which has hampered clinical application. Balancing the maximum achievable correction against deleterious effects on the cells is critical. However, multiple factors are known to contribute, and the optimization process is laborious and not always clearly defined. We have developed a flexible multidimensional Response Surface Methodology approach for optimization of gene correction. Using this approach, we could rapidly investigate and select editing conditions for CD34+ cells with the best possible balance between correction and cell/colony-forming unit (CFU) loss in a parsimonious one-shot experiment. This method revealed that using relatively low doses of AAV2/6 and CRISPR/Cas9 ribonucleoprotein complex, we can preserve the fitness of CD34+ cells and, at the same time, achieve high levels of targeted gene insertion. We then used these optimized editing conditions for the correction of p67phox-deficient chronic granulomatous disease (CGD), an autosomal recessive disorder of blood phagocytic cells resulting in severe recurrent bacterial and fungal infections and achieved rescue of p67phox expression and functional correction of CD34+-derived neutrophils from a CGD patient.

Project description:BackgroundMental health problems during pregnancy are multifaceted major health issues.AimTo develop and validate a short screening questionnaire that covers important domains including antenatal depression, tokophobia, fetal bonding disorder, suicidality, and emesis.MethodsA total of 321 pregnant women who were less than 36 weeks gestation responded to a cross-sectional web survey and filled in the 33-item Dimensional Assessment of Mother Baby Organization Questionnaire (DAMBO Q33, available as an e-book). Out of these women, 111 (35%) responded to our invitation to an online research interview (Dimensional Assessment of Mother Baby Organization-Research Version). Using theory-driven or cluster analysis-driven categories of the five mental health domains as a gold standard, we identified the most powerful DAMBO Q33 questionnaire items for each domain and calculated psychometric properties of cut-off points of the domain scores for the questionnaire, including sensitivity, specificity, and positive and negative predictive values. We proposed the best cut-off value for each domain.ResultsWe identified 11 DAMBO Q33 items as the best predictors for five mental health domains. The psychometric properties of the best cut-off points were acceptable for each domain. By these cut-off points, we could identify 57%, 19%, 100%, 46%, and 46% of true cases of antenatal depression, tokophobia, fetal bonding disorder, suicidality, and emesis, respectively. Positive cases of the five domains substantially coexisted with each other.ConclusionThe final 11-item questionnaire (DAMBO Q11) may be a useful screening tool for the five major mental health problems among pregnant women.
