Obstacle optimization for panic flow--reducing the tangential momentum increases the escape speed.

ABSTRACT: A disastrous form of pedestrian behavior is a stampede occurring in an event involving a large crowd in a panic situation. To deal with such stampedes, the possibility to increase the outflow by suitably placing a pillar or some other shaped obstacles in front of the exit has been demonstrated. We present a social force based genetic algorithm to optimize the best design of architectural entities to deal with large crowds. Unlike existing literature, our simulation results indicate that appropriately placing two pillars on both sides but not in front of the door can maximize the escape efficiency. Human experiments using 80 participants correspond well with the simulations. We observed a peculiar property named tangential momentum, the escape speed and the tangential momentum are found to be negatively correlated. The idea to reduce the tangential momentum has practical implications in crowd architectural design.

Dynamic pricing optimization for high-speed railway based on passenger flow assignment.

Project description:In order to improve the operation efficiency and market competitiveness, how to optimize the ticket pricing strategy of high-speed railway to match the dynamic supply-demand relationship was an urgent problem to be studied. Taking differentiated passenger demand and supply trains as the research object, the space-time service network based on train timetable was constructed. The generalized cost formula and travel utility formula of passenger travel were proposed, which contained economy, rapidity, convenience, comfort, and route correlation cost. A multi-objective dynamic pricing model was proposed. The model aimed at maximize the corporate revenue and maximize passenger travel benefit, and was solved by large neighborhood search heuristic algorithm and path size logit assignment based on capacity constraint-passenger flow increment accurate algorithm. Based on real data, the Shandong circular high-speed railway case compared the average total revenue under different ticket price adjustment ranges and the ticket price for different classes of trains under different OD levels. The case proved the practicability of dynamic pricing adjustment strategy considering train classification, which could provide a reference for the ticket price management of high-speed railway.

Purification of hemoglobin by tangential flow filtration with diafiltration.

Project description:A recent study by Palmer, Sun, and Harris (Biotechnol. Prog., 25:189-199, 2009) demonstrated that tangential flow filtration (TFF) can be used to produce HPLC-grade bovine and human hemoglobin (Hb). In this current study, we assessed the quality of bovine Hb (bHb) purified by introducing a 10 L batch-mode diafiltration step to the previously mentioned TFF Hb purification process. The bHb was purified from bovine red blood cells (RBCs) by filtering clarified RBC lysate through 50 nm (stage I) and 500 kDa (stage II) hollow fiber (HF) membranes. The filtrate was then passed through a 100 kDa (stage III) HF membrane with or without an additional 10 L diafiltration step to potentially remove additional small molecular weight impurities. Protein assays, SDS-PAGE, and LC-MS of the purified bHb (stage III retentate) reveal that addition of a diafiltration step has no effect on bHb purity or yield; however, it does increase the methemoglobin level and oxygen affinity of purified bHb. Therefore, we conclude that no additional benefit is gained from diafiltration at stage III and a three stage TFF process is sufficient to produce HPLC-grade bHb.

Continuous integrated antibody precipitation with two-stage tangential flow microfiltration enables constant mass flow.

Project description:Continuous precipitation is a new unit operation for the continuous capture of antibodies. The capture step is based on continuous precipitation with PEG6000 and Zn++ in a tubular reactor integrated with a two-stage continuous tangential flow filtration unit. The precipitate cannot be separated with centrifugation, because a highly compressed sediment results in poor resolubilization. We developed a new two-stage tangential flow microfiltration method, where part of the concentrated retentate of the first stage was directly fed to the second stage, together with the wash buffer. Thus, the precipitate was concentrated and washed in a continuous process. We obtained 97% antibody purity, a 95% process yield during continuous operation, and a fivefold reduction in pre-existing high-molecular-weight impurities. For other unit operations, surge tanks are often required, due to interruptions in the product mass flow out of the unit operation (e.g., the bind/elute mode in periodic counter-current chromatography). Our setup required no surge tanks; thus, it provided a truly continuous antibody capture operation with uninterrupted product mass flow. Continuous virus inactivation and other flow-through unit operations can be readily integrated downstream of the capture step to create truly continuous, integrated, downstream antibody processing without the need for hold tanks.

Process intensification for lentiviral vector manufacturing using tangential flow depth filtration.

Project description:For gene therapies to become more accessible and affordable treatment options, process intensification is one possible strategy to increase the number of doses generated per batch of viral vector. Process intensification for lentiviral vector manufacturing can be achieved by enabling perfusion in the production bioreactor when applied in tandem with a stable producer cell line, allowing for significant expansion of cells and production of lentiviral vectors without the need for transfer plasmids. Tangential flow depth filtration was used to achieve an intensified lentiviral vector production by enabling perfusion to expand cell density and allow for continuous separation of lentiviral vectors from producer cells. Hollow-fiber depth filters made of polypropylene with 2- to 4-μm channels demonstrated high filter capacity, extended functional life, and efficient separation of lentiviral vectors from producer cells and debris when used for this intensified process. We anticipate that process intensification with tangential flow depth filtration at 200-L scale from a suspension culture can produce on the order of magnitude of 10,000 doses per batch of lentiviral vectors required for CAR T or TCR cell and gene therapy that would require approximately 2 × 109 transducing units per dose.

Genetically driven brain serotonin deficiency facilitates panic-like escape behavior in mice.

Project description:Multiple lines of evidence implicate brain serotonin (5-hydroxytryptamine; 5-HT) system dysfunction in the pathophysiology of stressor-related and anxiety disorders. Here we investigate the influence of constitutively deficient 5-HT synthesis on stressor-related anxiety-like behaviors using Tryptophan hydroxylase 2 (Tph2) mutant mice. Functional assessment of c-Fos after associated foot shock, electrophysiological recordings of GABAergic synaptic transmission, differential expression of the Slc6a4 gene in serotonergic neurons were combined with locomotor and anxiety-like measurements in different contextual settings. Our findings indicate that constitutive Tph2 inactivation and consequential lack of 5-HT synthesis in Tph2 null mutant mice (Tph2-/-) results in increased freezing to associated foot shock and a differential c-Fos activity pattern in the basolateral complex of the amygdala. This is accompanied by altered GABAergic transmission as observed by recordings of inhibitory postsynaptic currents on principal neurons in the basolateral nucleus, which may explain increased fear associated with hyperlocomotion and escape-like responses in aversive inescapable contexts. In contrast, lifelong 5-HT deficiency as observed in Tph2 heterozygous mice (Tph+/-) is able to be compensated through reduced GABAergic transmission in the basolateral nucleus of the amygdala based on Slc6a4 mRNA upregulation in subdivisions of dorsal raphe neurons. This results in increased activity of the basolateral nucleus of the amygdala due to associated foot shock. In conclusion, our results reflect characteristic syndromal dimensions of panic disorder and agoraphobia. Thus, constitutive lack of 5-HT synthesis influence the risk for anxiety- and stressor-related disorders including panic disorder and comorbid agoraphobia through the absence of GABAergic-dependent compensatory mechanisms in the basolateral nucleus of the amygdala.

Liquid Fraction Effect on Foam Flow through a Local Obstacle.

Project description:An experimental study of quasi-two-dimensional liquid foams with varying liquid fractions is presented. Experiments are conducted in a Hele-Shaw cell with a local permeable obstacle placed in the center and filling 35, 60 and 78% of the cell gap. Foam velocity is calculated using a standard cross-correlation algorithm. Estimations of the liquid fraction of the foam are performed using a new simplified method based on a statistical analysis of foam cell structures. The pattern of the foam velocity field varies with increasing liquid fraction, responsible for significant variation of the foam's rheology. The local permeability decreases with increasing obstacle height and liquid fraction. In case of high liquid fraction (5.8×10-2), the permeability coefficient tends to zero for obstacles filling more than 78% of the cell gap.

Mass flow and momentum flux in nanoporous membranes in the transitional flow region.

Project description:An experimental study of momentum transfer in nanoporous polymeric track-etched membranes with pore diameters ranging from 100 to 1300 nm and nanochannel lengths of 12-20 μm was performed using He, N2, CO2, and SF6 propellants in a wide range of plenum and background pressures. Mass flux through the membranes was elaborated as a combination of Knudsen diffusion and viscous flow at Knudsen numbers above 0.1 and become choked at lower Knudsen numbers. The discharge coefficient for the membranes attained was 0.6, making the permeation rate similar to that of thin orifices. The effect is attributed to the mirror reflection of the molecules from the pore walls at low angles of incidence. The exhaust gas velocity is found to be dependent on the plenum to background pressure ratio and channel length-to-diameter ratio, reaching 0.9 of the velocity of the gas expanded to vacuum (up to 2 M). Close to an isothermal expansion occurs in nanochannels of all sizes. A general quantitative description for gas expansion in nanochannels is provided. The highest thrust is generated in the choked flow regime with the SF6 propellant and a value of 4.5 N cm-2 is attained at a propellant consumption of 0.165 kg (cm2 s)-1 for the membranes with 1300 nm nanochannels. The specific impulse of 138 s is reached for helium. The results show the prospects of the utilization of nanoporous membranes in cold gas propulsion systems.

Signatures of magnetism control by flow of angular momentum.

Project description:Exploring new strategies to manipulate the order parameter of magnetic materials by electrical means is of great importance not only for advancing our understanding of fundamental magnetism but also for unlocking potential applications. A well-established concept uses gate voltages to control magnetic properties by modulating the carrier population in a capacitor structure1-5. Here we show that, in Pt/Al/Fe/GaAs(001) multilayers, the application of an in-plane charge current in Pt leads to a shift in the ferromagnetic resonance field depending on the microwave frequency when the Fe film is sufficiently thin. The experimental observation is interpreted as a current-induced modification of the magnetocrystalline anisotropy ΔHA of Fe. We show that (1) ΔHA decreases with increasing Fe film thickness and is connected to the damping-like torque; and (2) ΔHA depends not only on the polarity of charge current but also on the magnetization direction, that is, ΔHA has an opposite sign when the magnetization direction is reversed. The symmetry of the modification is consistent with a current-induced spin6-8 and/or orbit9-13 accumulation, which, respectively, act on the spin and/or orbit component of the magnetization. In this study, as Pt is regarded as a typical spin current source6,14, the spin current can play a dominant part. The control of magnetism by a spin current results from the modified exchange splitting of the majority and minority spin bands, providing functionality that was previously unknown and could be useful in advanced spintronic devices.

Stokes flow around an obstacle in viscous two-dimensional electron liquid.

Project description:The electronic analog of the Poiseuille flow is the transport in a narrow channel with disordered edges that scatter electrons in a diffuse way. In the hydrodynamic regime, the resistivity decreases with temperature, referred to as the Gurzhi effect, distinct from conventional Ohmic behaviour. We studied experimentally an electronic analog of the Stokes flow around a disc immersed in a two-dimensional viscous liquid. The circle obstacle results in an additive contribution to resistivity. If specular boundary conditions apply, it is no longer possible to detect Poiseuille type flow and the Gurzhi effect. However, in flow through a channel with a circular obstacle, the resistivity decreases with temperature. By tuning the temperature, we observed the transport signatures of the ballistic and hydrodynamic regimes on the length scale of disc size. Our experimental results confirm theoretical predictions.

High frequency of CD8 escape mutations in elite controllers as new obstacle for HIV cure.

Project description:Accumulation of mutations in epitopes of cytolytic-T-lymphocytes immune response (CTL) in HIV-reservoir seems to be one of the reasons for shock-and-kill strategy failure. Ten non-controller patients on successful cART (TX) and seven elite controllers (EC) were included. HIV-Gag gene from purified resting memory CD4+ T-cells was sequenced by Next-Generation-Sequencing. HLA class-I alleles were typed to predict optimal HIV-Gag CTL epitopes. For each subject, the frequency of mutated epitopes in the HIV-Gag gene, the proportion of them considered as CTL-escape variants as well as their effect on antigen recognition by HLA were assessed. The proportion (%) of mutated HIV-Gag CTL epitopes in the reservoir was high and similar in EC and TX (86%[50-100] and 57%[48-82] respectively, p=0.315). Many of them were predicted to negatively impact antigen recognition. Moreover, the proportion of mutated epitopes considered to be CTL-escape variants was also similar in TX and EC (77%[49-92] vs. 50%[33-75] respectively, p=0.117). Thus, the most relevant finding of our study was the high and similar proportions of HIV-Gag CTL-escape mutations in the reservoir of both HIV-noncontroller patients with cART (TX) and patients with spontaneous HIV-control (EC). Our findings suggest that escape mutations of CTL-response may be another obstacle to eliminate the HIV reservoir and constitute a proof of concept that challenges HIV cure strategies focused on the reactivation of reservoirs. Due to the small sample size that could impact the robustness of the study, further studies with larger cohorts of elite controller patients are needed to confirm these results.
