Optimum periodicity of repeated contractile actions applied in mass transport.

ABSTRACT: Dynamically repeated periodic patterns are abundant in natural and artificial systems, such as tides, heart beats, stock prices, and the like. The characteristic repeatability and periodicity are expected to be optimized in effective system-specific functions. In this study, such optimum periodicity is experimentally evaluated in terms of effective mass transport using one-valve and multi-valve systems working in contractile fluid flows. A set of nanoscale gating functions is utilized, operating in nanocomposite networks through which permeates selectively pass under characteristic contractile actions. Optimized contractile periodicity exists for effective energy impartment to flow in a one-valve system. In the sequential contractile actions for a multi-valve system, synchronization with the fluid flow is critical for effective mass transport. This study provides fundamental understanding on the various repeated periodic patterns and dynamic repeatability occurring in nature and mechanical systems, which are useful for broad applications.

Sediment transport drives tidewater glacier periodicity.

Project description:Most of Earth's glaciers are retreating, but some tidewater glaciers are advancing despite increasing temperatures and contrary to their neighbors. This can be explained by the coupling of ice and sediment dynamics: a shoal forms at the glacier terminus, reducing ice discharge and causing advance towards an unstable configuration followed by abrupt retreat, in a process known as the tidewater glacier cycle. Here we use a numerical model calibrated with observations to show that interactions between ice flow, glacial erosion, and sediment transport drive these cycles, which occur independent of climate variations. Water availability controls cycle period and amplitude, and enhanced melt from future warming could trigger advance even in glaciers that are steady or retreating, complicating interpretations of glacier response to climate change. The resulting shifts in sediment and meltwater delivery from changes in glacier configuration may impact interpretations of marine sediments, fjord geochemistry, and marine ecosystems.The reason some of the Earth's tidewater glaciers are advancing despite increasing temperatures is not entirely clear. Here, using a numerical model that simulates both ice and sediment dynamics, the authors show that internal dynamics drive glacier variability independent of climate.

6-year periodicity and variable synchronicity in a mass-flowering plant.

Project description:Periodical organisms, such as bamboos and periodical cicadas, are very famous for their synchronous reproduction. In bamboos and other periodical plants, the synchronicity of mass-flowering and withering has been often reported indicating these species are monocarpic (semelparous) species. Therefore, synchronicity and periodicity are often suspected to be fairly tightly coupled traits in these periodical plants. We investigate the periodicity and synchronicity of Strobilanthes flexicaulis, and a closely related species S. tashiroi on Okinawa Island, Japan. The genus Strobilanthes is known for several periodical species. Based on 32-year observational data, we confirmed that S. flexicaulis is 6-year periodical mass-flowering monocarpic plant. All the flowering plants had died after flowering. In contrast, we found that S. tashiroi is a polycarpic perennial with no mass-flowering from three-year individual tracking. We also surveyed six local populations of S. flexicaulis and found variation in the synchronicity from four highly synchronized populations (>98% of plants flowering in the mass year) to two less synchronized one with 11-47% of plants flowering before and after the mass year. This result might imply that synchrony may be selected for when periodicity is established in monocarpic species. We found the selective advantages for mass-flowering in pollinator activities and predator satiation. The current results suggest that the periodical S. flexicaulis might have evolved periodicity from a non-periodical close relative. The current report should become a key finding for understanding the evolution of periodical plants.

Mitigation Actions Scenarios Applied to the Dairy Farm Management Systems.

Project description:The environmental impacts of the dairy industry, particularly global warming, are heavily influenced by milk production. Thus, there is an urgent need for farm-level actions and opportunities for improvement, implying mitigation strategies. The aim of this paper is to investigate five possible mitigation actions at the dairy farm and which one the farmers were willing to adopt: management and distribution of livestock manure and fertilizers, anaerobic manure treatment, optimization of the herd composition, feed quality, and heat recovery. A life cycle assessment was conducted on 63 farms using the product environmental footprint approach. The latter was divided into four quartiles, from which four representative farms were selected. For each farm, three scenarios have been analyzed considering the reference impact (reference scenario), the application of the mitigation actions (best-case scenario), and what farmers would implement (realistic scenario). Overall, the most effective mitigation actions in the best-case scenario were anaerobic manure treatment and the management and distribution of livestock manure and fertilizers, showing a potential reduction in total environmental impacts of 7-9% and 6-7%, respectively. Farmers' responses indicated a willingness to implement the latter mitigation strategy better. The optimization of the herd composition, feed quality, and heat recovery reported a range impact reduction between 0.01-5%.

Filament transport supports contractile steady states of actin networks.

Project description:In all eukaryotic cells, the actin cytoskeleton is maintained in a dynamic steady-state. Actin filaments are continuously displaced from cell periphery, where they assemble, towards the cell's center, where they disassemble. Despite this constant flow and turnover, cellular networks maintain their overall architecture constant. How such a flow of material can support dynamic yet steady cellular architectures remains an open question. To investigate the role of myosin-based forces in contractile steady-states of actin networks, we used a reconstituted in vitro system based on a minimal set of purified proteins, namely actin, myosin and actin regulators. We found that, contrary to previous bulk experiments, when confined in microwells, the actin network could self-organize into ordered arrangements of contractile bundles, flowing continuously without collapsing. This was supported by three-dimensional fluxes of actin filaments, spatially separated yet balancing each other. Unexpectedly, maintaining these fluxes did not depend on filament nucleation or elongation, but solely on filament transport. Ablation of the contractile bundles abolished the flux balance and led to network collapse. These findings demonstrate that the dynamic steady state of actin networks can be sustained by filament displacement and recirculation, independently of filament assembly and disassembly.

A methodology for identifying optimum vibration absorbers with a reaction mass.

Project description:Tuned mass dampers (TMDs), in which a reaction mass is attached to a structural system via a spring-parallel-damper connection, are commonly used in a wide range of applications to suppress deleterious vibrations. Recently, a mass-included absorber layout with an inerter element, termed the tuned mass damper inerter (TMDI), was introduced, showing significant performance benefits on vibration suppression. However, there are countless mass-included absorber layouts with springs, dampers and inerters, which could potentially provide more preferred dynamic properties. Currently, because there is no systematic methodology for accessing them, only an extremely limited number of mass-included absorber layouts have been investigated. This paper proposes an approach to identify optimum vibration absorbers with a reaction mass. Using this approach, a full class of absorber layouts with a reaction mass and a pre-determined number of inerters, dampers and springs connected in series and parallel, can be systematically investigated using generic Immittance-Function-Networks. The advan- tages of the proposed approach are demonstrated via a 3 d.f. structure example.

Opposing actions of septins and Sticky on Anillin promote the transition from contractile to midbody ring.

Project description:During cytokinesis, closure of the actomyosin contractile ring (CR) is coupled to the formation of a midbody ring (MR), through poorly understood mechanisms. Using time-lapse microscopy of Drosophila melanogaster S2 cells, we show that the transition from the CR to the MR proceeds via a previously uncharacterized maturation process that requires opposing mechanisms of removal and retention of the scaffold protein Anillin. The septin cytoskeleton acts on the C terminus of Anillin to locally trim away excess membrane from the late CR/nascent MR via internalization, extrusion, and shedding, whereas the citron kinase Sticky acts on the N terminus of Anillin to retain it at the mature MR. Simultaneous depletion of septins and Sticky not only disrupted MR formation but also caused earlier CR oscillations, uncovering redundant mechanisms of CR stability that can partly explain the essential role of Anillin in this process. Our findings highlight the relatedness of the CR and MR and suggest that membrane removal is coordinated with CR disassembly.

Dynamics of hydraulic and contractile wave-mediated fluid transport during Drosophila oogenesis.

Project description:From insects to mice, oocytes develop within cysts alongside nurse-like sister germ cells. Prior to fertilization, the nurse cells' cytoplasmic contents are transported into the oocyte, which grows as its sister cells regress and die. Although critical for fertility, the biological and physical mechanisms underlying this transport process are poorly understood. Here, we combined live imaging of germline cysts, genetic perturbations, and mathematical modeling to investigate the dynamics and mechanisms that enable directional and complete cytoplasmic transport in Drosophila melanogaster egg chambers. We discovered that during "nurse cell (NC) dumping" most cytoplasm is transported into the oocyte independently of changes in myosin-II contractility, with dynamics instead explained by an effective Young-Laplace law, suggesting hydraulic transport induced by baseline cell-surface tension. A minimal flow-network model inspired by the famous two-balloon experiment and motivated by genetic analysis of a myosin mutant correctly predicts the directionality, intercellular pattern, and time scale of transport. Long thought to trigger transport through "squeezing," changes in actomyosin contractility are required only once NC volume has become comparable to nuclear volume, in the form of surface contractile waves that drive NC dumping to completion. Our work thus demonstrates how biological and physical mechanisms cooperate to enable a critical developmental process that, until now, was thought to be mainly biochemically regulated.

Kindlin-2 modulates PTH1R to regulate bone mass and PTH actions in bone

Project description:To investigation the role of PTH and Kindlin-2 in bone development, we performed single-cell RNA-sequencing. From Con-veh, Con-PTH, cKO-veh, cKO-PTH, we profiled more than 20k single cells, including multi-potent mesenchymal stromal cells (MSC), osteoprogenitors, osteoblasts, chondrocytes, fibroblasts, endothelial cells, smooth muscle cells, skeletal muscle cells, pericytes, and schwann cells. We found proportion of part of these cells were significant altered by PTH or Kindlin-2 loss, especially for MSC, osteoblast, chondrocyte, and fibroblast. Transcriptomic analysis revealed gene expression was dramatically regulated by PTH or Kindlin-2 loss.

PERK regulates skeletal muscle mass and contractile function in adult mice.

Project description:Skeletal muscle mass is regulated by the coordinated activation of several anabolic and catabolic pathways. The endoplasmic reticulum (ER) is a major site of protein folding and a reservoir for calcium ions. Accretion of misfolded proteins or depletion in calcium concentration causes stress in the ER, which leads to the activation of a signaling network known as the unfolded protein response (UPR). In the present study, we investigated the role of the protein kinase R-like endoplasmic reticulum kinase (PERK) arm of the UPR in the regulation of skeletal muscle mass and function in naive conditions and in a mouse model of cancer cachexia. Our results demonstrate that the targeted inducible deletion of PERK reduces skeletal muscle mass, strength, and force production during isometric contractions. Deletion of PERK also causes a slow-to-fast fiber type transition in skeletal muscle. Furthermore, short hairpin RNA-mediated knockdown or pharmacologic inhibition of PERK leads to atrophy in cultured myotubes. While increasing the rate of protein synthesis, the targeted deletion of PERK leads to the increased expression of components of the ubiquitin-proteasome system and autophagy in skeletal muscle. Ablation of PERK also increases the activation of calpains and deregulates the gene expression of the members of the FGF19 subfamily. Furthermore, the targeted deletion of PERK increases muscle wasting in Lewis lung carcinoma tumor-bearing mice. Our findings suggest that the PERK arm of the UPR is essential for the maintenance of skeletal muscle mass and function in adult mice.-Gallot, Y. S., Bohnert, K. R., Straughn, A. Symbioses are restructured by repeated mass coral bleaching.

Project description:Survival of symbiotic reef-building corals under global warming requires rapid acclimation or adaptation. The impact of accumulated heat stress was compared across 1643 symbiont communities before and after the 2016 mass bleaching in three coral species and free-living in the environment across ~900 kilometers of the Great Barrier Reef. Resilient reefs (less aerial bleaching than predicted from high satellite sea temperatures) showed low variation in symbioses. Before 2016, heat-tolerant environmental symbionts were common in ~98% of samples and moderately abundant (9 to 40% in samples). In corals, heat-tolerant symbionts were at low abundances (0 to 7.3%) but only in a minority (13 to 27%) of colonies. Following bleaching, environmental diversity doubled (including heat-tolerant symbionts) and increased in one coral species. Communities were dynamic (Acropora millepora) and conserved (Acropora hyacinthus and Acropora tenuis), including symbiont community turnover and redistribution. Symbiotic restructuring after bleaching occurs but is a taxon-specific ecological opportunity.
