Two new species of Bryobia (Acarina, Prostigmata, Tetranychidae) from South France.

ABSTRACT: Two new species of Tetranychidae belonging to the genus Bryobia are reported from France. Bryobiabelliloci sp. n. and Bryobiagigas sp. n. collected on Genistacinerea and Bituminariabituminosa, respectively, are described and illustrated in the present work. Additional data to the original description of Bryobiacinereae are given and an identification key to known Bryobia species from France is also provided.

Origins of asexuality in Bryobia mites (Acari: Tetranychidae).

Project description:BackgroundObligate asexual reproduction is rare in the animal kingdom. Generally, asexuals are considered evolutionary dead ends that are unable to radiate. The phytophagous mite genus Bryobia contains a large number of asexual species. In this study, we investigate the origin and evolution of asexuality using samples from 111 populations in Europe, South Africa and the United States, belonging to eleven Bryobia species. We also examine intraspecific clonal diversity for one species, B. kissophila, by genotyping individuals from 61 different populations. Knowledge on the origin of asexuality and on clonal diversity can contribute to our understanding of the paradox of sex.ResultsThe majority (94%) of 111 sampled populations reproduces asexually. Analysis of part of nuclear 28S rDNA shows that these asexuals do not form a monophyletic clade. Analysis of the mitochondrial COI region shows that intraspecific variation is extensive (up to 8.8%). Within B. kissophila, distinct clades are found, which are absent at the nuclear 28S rDNA level. Moreover, paraphyletic patterns are found at the mitochondrial DNA.ConclusionAsexuality is widespread in the genus Bryobia, signifying that some animal taxa do contain a high number of asexuals. We argue that asexuality originated multiple times within Bryobia. Wolbachia bacteria cause asexuality in at least two Bryobia species and may have infected different species independently. The high intraspecific clonal diversity and the patterns of paraphyly at the mitochondrial DNA in B. kissophila might be explained by a high mutation fixation rate and past hybridization events. Reproductive parasites like Wolbachia and Cardinium might influence these processes. We discuss the role these bacteria could play in the evolutionary success of asexual species.

World species of the subgenus Oligonychus (Reckiella) Tuttle and Baker (Acari, Prostigmata, Tetranychidae), diagnostic keys, taxonomic notes, and a new species.

Project description:Oligonychus Berlese (Acari, Prostigmata, Tetranychidae) is an agriculturally important and the largest genus of spider mites, comprised of 211 species (including new species), two subgenera, four species groups, and 11 species subgroups. The present study comprehensively addressed the morphotaxonomic-based identification of world species of the subgenus Reckiella Tuttle and Baker. Five diagnostic keys were developed for identifying Oligonychus (Reckiella) species belonging to five species subgroups: iseilemae, pritchardi, biharensis, gossypii, and exsiccator. Taxonomic notes are provided on intraspecific variations and some closely related Oligonychus (Reckiella) species representing six species complexes, viz. the afrasiaticus complex, the litchii complex, the pratensis complex, the plegas complex, the sacchari complex, and the tylus complex. One new spider mite species, Oligonychus bahaensis sp. nov., is described and illustrated from Grasses (Poaceae) under the subgenus Reckiella.

Species groups, subgroups, and key to world species of the genus Schizotetranychus Tragardh, 1915 (Acari, Prostigmata, Tetranychidae).

Project description:After a comprehensive taxonomic assessment of descriptions/ illustrations of all known (118) species of the spider mite genus Schizotetranychus Trägårdh (Acari: Prostigmata: Tetranychidae), five species groups are proposed based on the number of tactile setae on tibia II in female, i.e., vermiculatus with four setae (four spp.), schizopus with five setae (52 spp.), spireafolia with six setae (10 spp.), asparagi with seven setae (20 spp.), and bambusae with eight setae on tibia II (22 spp.). The species group schizopus is further divided into three species subgroups based on tactile setae on tibia I: schizopus with eight/ nine setae (21 spp.), andropogoni with seven setae (26 spp.), and taquarae with six tactile setae excluding the solenidion on tibia I (five spp.). Eight Schizotetranychus species were not assigned to any species group because of brief descriptions and/ or illustration and without information on the number of tactile setae on tibiae I and II. Moreover, two Schizotetranychus species, S.gausus Baker & Pritchard and S.luculentus Tseng that have six setae/ structures including a spinneret and a solenidion on the palp tarsus, are provisionally transferred to the genus Stigmaeopsis Banks. Finally, keys to species groups and subgroups of the world species of Schizotetranychus are provided.

Molecular-Based Taxonomic Inferences of Some Spider Mite Species of the Genus Oligonychus Berlese (Acari, Prostigmata, Tetranychidae).

Project description:DNA barcoding technology using short DNA sequences has emerged as an efficient and reliable tool for identifying, confirming, and resolving closely related taxa. This study used ITS2-rDNA and mtCOI DNA sequences to confirm the identity of eight Oligonychus species, representing 68 spider mite samples, collected mainly from Saudi Arabia (SA) and some from Mexico, Pakistan, USA, and Yemen. The intraspecific nucleotide divergences of the studied Oligonychus species ranged from 0% to 1.2% for ITS2 and 0% to 2.9% for COI. However, the interspecific nucleotide divergences were distinctly higher than the intraspecific ones and ranged from 3.7% to 51.1% for ITS2 and 3.2% to 18.1% for COI. Furthermore, molecular data correctly confirmed the species identity of 42 Oligonychus samples lacking males, including a previously claimed sample of O. pratensis from SA. High genetic variations were detected in two Oligonychus species: O. afrasiaticus (McGregor) (nine ITS2 and three COI haplotypes) and O. tylus Baker and Pritchard (four ITS2 and two COI haplotypes). In addition, ITS2- and COI-based phylogenetic trees confirmed the subdivision of the genus Oligonychus. In conclusion, integrative taxonomic approaches are vital to resolve the closely related Oligonychus species, identify the samples lacking male specimens, and assess phylogenetic relationships within and among species.

Taxonomy of the Genus Bryobia Koch (Acari: Tetranychidae): Reconsideration of Subgenera and Updated Species Groups.

Project description:The present study aimed to revise the subgeneric divisions of the genus Bryobia based on persistent characteristics and to propose species groups in each subgenus to further ease the identification of species. As a result, only three subgenera, based on literature, are recognized, viz, subgenus Bryobia Koch (duplex setae present on leg tarsi III-IV), subgenus Allobia Livschits and Mitrofanov (duplex setae absent on leg tarsi III-IV), and subgenus Lyobia Livschits and Mitrofanov (duplex setae present only on leg tarsus III). The analysis based on morphological characters and available COI sequences on the GenBank database did not resolve the phylogenetic relationship of Bryobia species, and likewise, no support could be found for the proposed subgeneric divisions. The species in each subgenus were further categorized into species groups based on the position of the fourth pair of dorsocentral setae f1, viz, three species groups, praetiosa, osterloffi, and neoephedrae in subgenus Bryobia; three species groups, pritchardi, abbatielloi, and deserticola in subgenus Allobia; and three species groups, eurotiae, sarothamni, and rubrioculus in subgenus Lyobia. Furthermore, detailed notes are provided on eight Bryobia species which were described by Meyer, having morphological characteristics of both the tribes Bryobiini and Hystrichonychini. The generic status of two bryobine species, B. pseudorubrioculus Smiley and Baker and B. tuttlie Smiley and Baker, is also discussed and considered as species inquirendae.

The genus Oligonychus Berlese (Acari, Prostigmata, Tetranychidae): taxonomic assessment and a key to subgenera, species groups, and subgroups.

Project description:A comprehensive taxonomic assessment of the most agriculturally important and highly diverse spider mite genus, Oligonychus Berlese (Acari: Tetranychidae) was performed. The sub-generic division, species groups, doubtful species, species complexes and the interpretation of a key generic character are discussed. Based on the orientation of the male aedeagus, only two subgenera, namely Oligonychus Berlese (aedeagus downturned) and Reckiella Tuttle & Baker (aedeagus upturned), are valid in the genus Oligonychus. The subgenera Homonychus Wainstein, Metatetranychoides Wainstein, and Wainsteiniella Tuttle & Baker are considered to be synonyms of the subgenus Oligonychus, whereas the subgenus Pritchardinychus Wainstein is proposed as a synonym of the subgenus Reckiella. Moreover, based on female morphological characters, four species groups (coffeae, exsiccator, iseilemae, and peruvianus) and 11 species subgroups (aceris, biharensis, coffeae, comptus, exsiccator, gossypii, iseilemae, peruvianus, pritchardi, smithi, and subnudus) are suggested in the subgenera Oligonychus and Reckiella. Fourteen Oligonychus species are proposed as species inquirendae, and potential cryptic species complexes in the genus Oligonychus are briefly highlighted. It is agreed that the clunal seta h1 is always absent, while the para-anal setae h2 and h3 are always present in the genus Oligonychus. A key to subgenera, species groups, and species subgroups of the genus Oligonychus is provided.

Evidence for Reconsidering the Taxonomic Status of Closely Related Oligonychus Species in punicae Complex (Acari: Prostigmata: Tetranychidae).

Project description:To elucidate the taxonomic problems in species delineation within the Oligonychus punicae complex (O. punicae, O. mangiferus, and O. vitis) (Acari: Prostigmata: Tetranychidae), we performed morphological and molecular investigations on mite samples, collected from different hosts/countries. Thirty-nine samples of punicae complex, collected from Egypt, Pakistan, and Saudi Arabia (SA), did not show any considerable morphological differences in females and males. All 39 samples of the punicae complex resembled the original description of O. punicae, while the claimed Mexican O. punicae was distinctively different based on male aedeagus. Molecularly, the low nucleotide diversity ranged from 0% to 2.1% (ITS2-rDNA) and 0% to 1% (COI-mtDNA), and was observed among various DNA sequences of the punicae complex from Egypt, India, Israel, Pakistan, and SA, confirming their identity as one species. The high genetic divergence ranged from 17.2% to 18.8% (ITS2) and 9.2% to 10.2% (COI), observed between the claimed Mexican O. punicae and all other sequences of the punicae complex, indicating that the Mexican sample do not belong to O. punicae. Basing our findings on both morphological and molecular data, we can conclude that O. mangiferus and O. vitis are synonymized with O. punicae. Additionally, this study reveals that the claimed Mexican O. punicae needs to be re-identified.

Comparative Transcriptome Analysis Reveals the Mechanism Related to Fluazinam Stress of Panonychus citri (Acarina: Tetranychidae).

Project description:The use of a large number of chemical acaricides to control these pest mites has led to an increasing problem of pesticide resistance, which has always been the difficulty in integrated pest management (IPM). Fluazinam has a good control effect on Panonychus citri, the serious pest on citrus; however, we only know the mechanism of action of fluazinam as a fungicide and its mechanism of action on mites remains unclear. Through analysis using Illumina high-throughput transcriptomic sequencing and differential expression genes in P. citri treated with fluazinam, 59 cytochrome P450 genes, 23 glutathione s-transferase genes, five carboxylate esterase genes, 11 superoxide dismutase genes and 15 catalase genes were identified. The Gene Ontology enrichment and the enrichment of KEGG results showed that the treatment were enrichment for redox enzyme pathways. Evaluating the efficacy of fluazinam, and analyzing the transcriptome data of P. citri under fluazinam stress, potentially provide a new agent for prevention and control of P. citri, and also preliminary research results for exploring the mechanism of action of fluazinam on P. citri. Given the up-regulated expression levels of genes for Mn-superoxide dismutase and catalase, we speculate that they play an important role in fluazinam-stress action on P. citri.

Australian Bryobia mites (Trombidiformes: Tetranychidae) form a complex of cryptic taxa with unique climatic niches and insecticide responses.

Project description:BackgroundBryobia (Koch) mites belong to the economically important spider mite family, the Tetranychidae, with >130 species described worldwide. Due to taxonomic difficulties and most species being asexual, species identification relies heavily on genetic markers. Multiple putative Bryobia mite species have been identified attacking pastures and grain crops in Australia. In this study, we collected 79 field populations of Bryobia mites and combined these with 134 populations that were collected previously. We characterised taxonomic variation of mites using 28S rDNA amplicon-based DNA metabarcoding using next-generation sequencing approaches and direct Sanger sequencing. We then undertook species distribution modelling of the main genetic lineages and examined the chemical responses of multiple field populations.ResultsWe identified 47 unique haplotypes across all mites sampled that grouped into four distinct genetic lineages. These lineages have different distributions, with three of the four putative lineages showing different climatic envelopes, as inferred from species distribution modelling. Bryobia mite populations also showed different responses to a widely used insecticide (the organophosphate, omethoate), but not to another chemical (the pyrethroid, bifenthrin) when examined using laboratory bioassays.ConclusionOur findings indicate that cryptic diversity is likely to complicate the formulation of management strategies for Bryobia mites. Although focussed on Australia, this study demonstrates the challenges of studying Bryobia and highlights the importance of further research into this complex group of mites across the world. © 2022 The Authors.
