The effect of direct and indirect monitoring on generosity among preschoolers.

ABSTRACT: The purpose of this study is to examine the effect of direct and indirect monitoring on generosity among five-year-old preschoolers and to reveal the primary motivation for their generosity. Forty-two preschoolers completed one-shot dictator games in Condition 1 while being monitored by the experimenter (the direct monitoring condition). In Condition 2, an image of staring eyes was displayed on the computer monitor (the indirect monitoring condition). In Condition 3, the computer monitor showed a picture of flowers (the non-monitoring condition). The results showed that while there was no difference between the mean levels of allocation in the indirect and non-monitoring conditions, the mean level of allocation in the direct monitoring condition was significantly higher than in the non-monitoring condition. These results showed that five-year-old preschoolers concerned with being monitored by, and receiving direct responses from, others tend to be more generous.

Relationship between Salivary Oxytocin Levels and Generosity in Preschoolers.

Project description:This study examined the association between salivary oxytocin (sOT) levels and generosity in preschoolers. Fifty preschoolers played two dictator games (DG) by deciding how to allocate 10 chocolates between themselves and another child, who was either from the same class as the participant (ingroup member), or an unknown child from another class (outgroup member). sOT levels were assessed in saliva collected from the children immediately prior to the DG tasks. While sOT levels were negatively associated with allocations made to both ingroup and outgroup members by boys, among girl sOT levels were positively related to allocations made to ingroup members, and unrelated to allocations made to outgroup members. These results suggest sex differences in the association between salivary oxytocin and generosity.

The evolution of indirect reciprocity under action and assessment generosity.

Project description:Indirect reciprocity is a mechanism for the evolution of cooperation based on social norms. This mechanism requires that individuals in a population observe and judge each other's behaviors. Individuals with a good reputation are more likely to receive help from others. Previous work suggests that indirect reciprocity is only effective when all relevant information is reliable and publicly available. Otherwise, individuals may disagree on how to assess others, even if they all apply the same social norm. Such disagreements can lead to a breakdown of cooperation. Here we explore whether the predominantly studied 'leading eight' social norms of indirect reciprocity can be made more robust by equipping them with an element of generosity. To this end, we distinguish between two kinds of generosity. According to assessment generosity, individuals occasionally assign a good reputation to group members who would usually be regarded as bad. According to action generosity, individuals occasionally cooperate with group members with whom they would usually defect. Using individual-based simulations, we show that the two kinds of generosity have a very different effect on the resulting reputation dynamics. Assessment generosity tends to add to the overall noise and allows defectors to invade. In contrast, a limited amount of action generosity can be beneficial in a few cases. However, even when action generosity is beneficial, the respective simulations do not result in full cooperation. Our results suggest that while generosity can favor cooperation when individuals use the most simple strategies of reciprocity, it is disadvantageous when individuals use more complex social norms.

The Effect of Direct and Indirect EZH2 Inhibition in Rhabdomyosarcoma Cell Lines.

Project description:Enhancer of Zeste homolog 2 (EZH2) is involved in epigenetic regulation of gene transcription by catalyzing trimethylation of histone 3 at lysine 27. In rhabdomyosarcoma (RMS), increased EZH2 protein levels are associated with poor prognosis and increased metastatic potential, suggesting EZH2 as a therapeutic target. The inhibition of EZH2 can be achieved by direct inhibition which targets only the enzyme activity or by indirect inhibition which also affects activities of other methyltransferases and reduces EZH2 protein abundance. We assessed the direct inhibition of EZH2 by EPZ005687 and the indirect inhibition by 3-deazaneplanocin (DZNep) and adenosine dialdehyde (AdOx) in the embryonal RD and the alveolar RH30 RMS cell line. EPZ005687 was more effective in reducing the cell viability and colony formation, in promoting apoptosis induction, and in arresting cells in the G1 phase of the cell cycle than the indirect inhibitors. DZNep was more effective in decreasing spheroid viability and size in both cell lines than EPZ005687 and AdOx. Both types of inhibitors reduced cell migration of RH30 cells but not of RD cells. The results show that direct and indirect inhibition of EZH2 affect cellular functions differently. The alveolar cell line RH30 is more sensitive to epigenetic intervention than the embryonal cell line RD.

Natural direct and indirect effects on the exposed: effect decomposition under weaker assumptions.

Project description:We define natural direct and indirect effects on the exposed. We show that these allow for effect decomposition under weaker identification conditions than population natural direct and indirect effects. When no confounders of the mediator-outcome association are affected by the exposure, identification is possible under essentially the same conditions as for controlled direct effects. Otherwise, identification is still possible with additional knowledge on a nonidentifiable selection-bias function which measures the dependence of the mediator effect on the observed exposure within confounder levels, and which evaluates to zero in a large class of realistic data-generating mechanisms. We argue that natural direct and indirect effects on the exposed are of intrinsic interest in various applications. We moreover show that they coincide with the corresponding population natural direct and indirect effects when the exposure is randomly assigned. In such settings, our results are thus also of relevance for assessing population natural direct and indirect effects in the presence of exposure-induced mediator-outcome confounding, which existing methodology has not been able to address.

Estimating Direct and Indirect Protective Effect of Influenza Vaccination in the United States.

Project description:With influenza vaccination rates in the United States recently exceeding 45% of the population, it is important to understand the impact that vaccination is having on influenza transmission. In this study, we used a Bayesian modeling approach, combined with a simple dynamical model of influenza transmission, to estimate this impact. The combined framework synthesized evidence from a range of data sources relating to influenza transmission and vaccination in the United States. We found that, for seasonal epidemics, the number of infections averted ranged from 9.6 million in the 2006-2007 season (95% credible interval (CI): 8.7, 10.9) to 37.2 million (95% CI: 34.1, 39.6) in the 2012-2013 season. Expressed in relative terms, the proportion averted ranged from 20.8% (95% CI: 16.8, 24.3) of potential infections in the 2011-2012 season to 47.5% (95% CI: 43.7, 50.8) in the 2008-2009 season. The percentage averted was only 1.04% (95% CI: 0.15, 3.2) for the 2009 H1N1 pandemic, owing to the late timing of the vaccination program in relation to the pandemic in the Northern hemisphere. In the future, further vaccination coverage, as well as improved influenza vaccines (especially those offering better protection in the elderly), could have an even stronger effect on annual influenza epidemics.

Preschoolers and the endowment effect.

Project description:We show that preschoolers exhibit the endowment effect as evidenced by experiments where children generally chose to keep their own toys rather than trading them for similar ones. Furthermore, we relate the emergence of this effect to children's innate psychobiological traits--emotional state, gender, handedness, and digit ratio. The trials were conducted with 141 children across 6 kindergartens. We also found support that children, like adults, exhibit a preference for physical possession as opposed to ownership. As with adults, emotions also seem to matter, as children who were described as quiet and calm were more likely to present the endowment effect. Also of note, right-handed children described as quiet were more likely to exhibit the phenomenon. Furthermore, female children were generally found to be calmer than males, while males tended to be more fearful than females. This result was also previously found in teenagers.

A three-way decomposition of a total effect into direct, indirect, and interactive effects.

Project description:Recent theory in causal inference has provided concepts for mediation analysis and effect decomposition that allow one to decompose a total effect into a direct and an indirect effect. Here, it is shown that what is often taken as an indirect effect can in fact be further decomposed into a "pure" indirect effect and a mediated interactive effect, thus yielding a three-way decomposition of a total effect (direct, indirect, and interactive). This three-way decomposition applies to difference scales and also to additive ratio scales and additive hazard scales. Assumptions needed for the identification of each of these three effects are discussed and simple formulae are given for each when regression models allowing for interaction are used. The three-way decomposition is illustrated by examples from genetic and perinatal epidemiology, and discussion is given to what is gained over the traditional two-way decomposition into a direct and an indirect effect.

Direct and Indirect Effect of TGFβ on Treg Transendothelial Recruitment in HCC Tissue Microenvironment.

Project description:The balance between anti-tumor and tumor-promoting immune cells, such as CD4+ Th1 and regulatory T cells (Tregs), respectively, is assumed to dictate the progression of hepatocellular carcinoma (HCC). The transforming growth factor beta (TGFβ) markedly shapes the HCC microenvironment, regulating the activation state of multiple leukocyte subsets and driving the differentiation of cancer associated fibroblasts (CAFs). The fibrotic (desmoplastic) reaction in HCC tissue strongly depends on CAFs activity. In this study, we attempted to assess the role of TGFβ on transendothelial migration of Th1-oriented and Treg-oriented CD4+ T cells via a direct or indirect, CAF-mediated mechanisms, respectively. We found that the blockage of TGFβ receptor I-dependent signaling in Tregs resulted in impaired transendothelial migration (TEM) of these cells. Interestingly, the secretome of TGFβ-treated CAFs inhibited the TEM of Tregs but not Th1 cells, in comparison to the secretome of untreated CAFs. In addition, we found a significant inverse correlation between alpha-SMA and FoxP3 (marker of Tregs) mRNA expression in a microarray analysis involving 78 HCCs, thus suggesting that TGFβ-activated stromal cells may counteract the trafficking of Tregs into the tumor. The apparent dual behavior of TGFβ as both pro- and anti-tumorigenic cytokines may add a further level of complexity to the mechanisms that regulate the interactions among cancerous, stromal, and immune cells within HCC, as well as other solid tumors, and contribute to better manipulation of the TGFβ signaling as a therapeutic target in HCC patients.

Quantifying direct and indirect effect for longitudinal mediator and survival outcome using joint modeling approach.

Project description:Longitudinal biomarkers are widely used in biomedical and translational researches to monitor the progressions of diseases. Methods have been proposed to jointly model longitudinal data and survival data, but its causal mechanism is yet to be investigated rigorously. Understanding how much of the total treatment effect is through the biomarker is important in understanding the treatment mechanism and evaluating the biomarker. In this work, we propose a causal mediation analysis method to compute the direct and indirect effects, when a joint modeling approach is used to take the longitudinal biomarker as the mediator and the survival endpoint as the outcome. Such a joint modeling approach allows us to relax the commonly used "sequential ignorability" assumption. We demonstrate how to evaluate longitudinally measured biomarkers using our method with two case studies, an AIDS study and a liver cirrhosis study.

No evidence that economic inequality moderates the effect of income on generosity.

Project description:A landmark study published in PNAS [Côté S, House J, Willer R (2015) Proc Natl Acad Sci USA 112:15838-15843] showed that higher income individuals are less generous than poorer individuals only if they reside in a US state with comparatively large economic inequality. This finding might serve to reconcile inconsistent findings on the effect of social class on generosity by highlighting the moderating role of economic inequality. On the basis of the importance of replicating a major finding before readily accepting it as evidence, we analyzed the effect of the interaction between income and inequality on generosity in three large representative datasets. We analyzed the donating behavior of 27,714 US households (study 1), the generosity of 1,334 German individuals in an economic game (study 2), and volunteering to participate in charitable activities in 30,985 participants from 30 countries (study 3). We found no evidence for the postulated moderation effect in any study. This result is especially remarkable because (i) our samples were very large, leading to high power to detect effects that exist, and (ii) the cross-country analysis employed in study 3 led to much greater variability in economic inequality. These findings indicate that the moderation effect might be rather specific and cannot be easily generalized. Consequently, economic inequality might not be a plausible explanation for the heterogeneous results on the effect of social class on prosociality.
