Hydrogel Nanofilaments via Core-Shell Electrospinning.

ABSTRACT: Recent biomedical hydrogels applications require the development of nanostructures with controlled diameter and adjustable mechanical properties. Here we present a technique for the production of flexible nanofilaments to be used as drug carriers or in microfluidics, with deformability and elasticity resembling those of long DNA chains. The fabrication method is based on the core-shell electrospinning technique with core solution polymerisation post electrospinning. Produced from the nanofibers highly deformable hydrogel nanofilaments are characterised by their Brownian motion and bending dynamics. The evaluated mechanical properties are compared with AFM nanoindentation tests.

Native collagen hydrogel nanofibres with anisotropic structure using core-shell electrospinning.

Project description:Collagen hydrogel is a popular extracellular matrix (ECM) material in regenerative medicine and has an isotropic structure. In contrast, native ECM has an anisotropic structure. Electrospinning of collagen dissolved in organic solvents is widely used for fabricating anisotropic collagen nanofibres; however, such fibres are water-soluble and require cross-linking before use as scaffolds for cell culture. Herein, electrospinning using a core-shell nozzle was employed to spin an aqueous acidic solution of collagen and encapsulate it within a shell of polyvinylpyrrolidone (PVP). Subsequently, the core collagen was gelled, and the shell PVP was washed away using a basic ethanol solution to yield anisotropic collagen hydrogel nanofibres. Immunostaining and Fourier transform infrared spectroscopy revealed that the obtained fibres were composed of collagen, and surface PVP was removed completely. Circular dichroism measurements confirmed that the fibres exhibited the triple helical structure characteristic of collagen. Human umbilical vein endothelial cells cultured on the collagen hydrogel fibres were oriented along the fibre direction. Hence, this method is suitable for fabricating fibrous anisotropic collagen hydrogels without chemical and thermal cross-linking, and can facilitate the development of safe medical materials with anisotropy similar to that of native ECM.

Amyloid fibril-directed synthesis of silica core-shell nanofilaments, gels, and aerogels.

Project description:Amyloid fibrils have evolved from purely pathological materials implicated in neurodegenerative diseases to efficient templates for last-generation functional materials and nanotechnologies. Due to their high intrinsic stiffness and extreme aspect ratio, amyloid fibril hydrogels can serve as ideal building blocks for material design and synthesis. Yet, in these gels, stiffness is generally not paired by toughness, and their fragile nature hinders significantly their widespread application. Here we introduce an amyloid-assisted biosilicification process, which leads to the formation of silicified nanofibrils (fibril-silica core-shell nanofilaments) with stiffness up to and beyond ∼20 GPa, approaching the Young's moduli of many metal alloys and inorganic materials. The silica shell endows the silicified fibrils with large bending rigidity, reflected in hydrogels with elasticity three orders of magnitude beyond conventional amyloid fibril hydrogels. A constitutive theoretical model is proposed that, despite its simplicity, quantitatively interprets the nonmonotonic dependence of the gel elasticity upon the filaments bundling promoted by shear stresses. The application of these hybrid silica-amyloid hydrogels is demonstrated on the fabrication of mechanically stable aerogels generated via sequential solvent exchange, supercritical [Formula: see text] removal, and calcination of the amyloid core, leading to aerogels of specific surface area as high as 993 [Formula: see text]/g, among the highest values ever reported for aerogels. We finally show that the scope of amyloid hydrogels can be expanded considerably by generating double networks of amyloid and hydrophilic polymers, which combine excellent stiffness and toughness beyond those of each of the constitutive individual networks.

Significant Electromechanical Characteristic Enhancement of Coaxial Electrospinning Core-Shell Fibers.

Project description:Electrospinning is a low-cost and straightforward method for producing various types of polymers in micro/nanofiber form. Among the various types of polymers, electrospun piezoelectric polymers have many potential applications. In this study, a new type of functional microfiber composed of poly(γ-benzyl-α,L-glutamate) (PBLG) and poly(vinylidene fluoride) (PVDF) with significantly enhanced electromechanical properties has been reported. Recently reported electrospun PBLG fibers exhibit polarity along the axial direction, while electrospun PVDF fibers have the highest net dipole moment in the transverse direction. Hence, a combination of PBLG and PVDF as a core-shell structure has been investigated in the present work. On polarization under a high voltage, enhancement in the net dipole moment in each material and the intramolecular conformation was observed. The piezoelectric coefficient of the electrospun PBLG/PVDF core-shell fibers was measured to be up to 68 pC N-1 (d33), and the voltage generation under longitudinal extension was 400 mVpp (peak-to-peak) at a frequency of 60 Hz, which is better than that of the electrospun homopolymer fibers. Such new types of functional materials can be used in various applications, such as sensors, actuators, smart materials, implantable biosensors, biomedical engineering devices, and energy harvesting devices.

Core-Shell Polyvinyl Alcohol (PVA) Base Electrospinning Microfibers for Drug Delivery.

Project description:In this study, electrospun membranes were developed for controlled drug release applications. Both uniaxial Polyvinyl alcohol (PVA) and coaxial fibers with a PVA core and a poly (L-lactic acid) (PLLA) and polycaprolactone (PCL) coating were produced with different coating structures. The best conditions for the manufacture of the fibers were also studied and their morphology was analyzed as a function of the electrospinning parameters. Special attention was paid to the fiber surface morphology of the coaxial fibers, obtaining both porous and non-porous coatings. Bovine serum albumin (BSA) was used as the model protein for the drug release studies and, as expected, the uncoated fibers were determined to have the fastest release kinetics. Different release rates were obtained for the coated fibers, which makes this drug release system suitable for different applications according to the release time required.

All-organic, conductive and biodegradable yarns from core-shell nanofibers through electrospinning.

Project description:Electrically conductive and biodegradable materials are desired for a range of applications in wearable electronics to address the growing ecological problem of e-waste. Herein, we report on the design and fabrication of all-organic, conductive and biodegradable nanofibrous core-shell yarn produced by in situ polymerization of aniline on the surface of electrospun poly(ε-caprolactone) nanofibers. The effect of concentration of aniline monomer on the morphology and resistivity of deposited polyaniline layer was investigated. The electrical resistance changed almost instantaneously with the strain for multiple stretch and recovery cycles. This rapid and sensitive response to mechanical loading and unloading is promising to validate the possibility of using the conductive yarns as strain sensors for monitoring human motion. Increasing the number of plies of yarn to three resulted in a three-fold reduction of the resistance. The twisted plied yarns were incorporated into fabric by stitching to demonstrate their use as a wearable electrode for capacitive sensors. This approach presents an early step in realizing all-organic conductive biodegradable nanofibrous yarns for biodegradable smart textiles.

Core-shell hydrogel microcapsules for improved islets encapsulation.

Project description:Islets microencapsulation holds great promise to treat type 1 diabetes. Currently used alginate microcapsules often have islets protruding outside capsules, leading to inadequate immuno-protection. A novel design of microcapsules with core-shell structures using a two-fluid co-axial electro-jetting is reported. Improved encapsulation and diabetes correction is achieved in a single step by simply confining the islets in the core region of the capsules.

Aqueous-Based Coaxial Electrospinning of Genetically Engineered Silk Elastin Core-Shell Nanofibers.

Project description:A nanofabrication method for the production of flexible core-shell structured silk elastin nanofibers is presented, based on an all-aqueous coaxial electrospinning process. In this process, silk fibroin (SF) and silk-elastin-like protein polymer (SELP), both in aqueous solution, with high and low viscosity, respectively, were used as the inner (core) and outer (shell) layers of the nanofibers. The electrospinnable SF core solution served as a spinning aid for the nonelectrospinnable SELP shell solution. Uniform nanofibers with average diameter from 301 ± 108 nm to 408 ± 150 nm were obtained through adjusting the processing parameters. The core-shell structures of the nanofibers were confirmed by fluorescence and electron microscopy. In order to modulate the mechanical properties and provide stability in water, the as-spun SF-SELP nanofiber mats were treated with methanol vapor to induce β-sheet physical crosslinks. FTIR confirmed the conversion of the secondary structure from a random coil to β-sheets after the methanol treatment. Tensile tests of SF-SELP core-shell structured nanofibers showed good flexibility with elongation at break of 5.20% ± 0.57%, compared with SF nanofibers with an elongation at break of 1.38% ± 0.22%. The SF-SELP core-shell structured nanofibers should provide useful options to explore in the field of biomaterials due to the improved flexibility of the fibrous mats and the presence of a dynamic SELP layer on the outer surface.

Chitin Nanofibrils Enabled Core-Shell Microcapsules of Alginate Hydrogel.

Project description:An engineered 3D architectural network of the biopolymeric hydrogel can mimic the native cell environment that promotes cell infiltration and growth. Among several bio-fabricated hydrogel structures, core-shell microcapsules inherit the potential of cell encapsulation to ensure the growth and transport of cells and cell metabolites. Herein, a co-axial electrostatic encapsulation strategy is used to create and encapsulate the cells into chitin nanofibrils integrated alginate hydrogel microcapsules. Three parameters that are critical in the electrostatic encapsulation process, hydrogel composition, flow rate, and voltage were optimized. The physicochemical characterization including structure, size, and stability of the core-shell microcapsules was analyzed by scanning electron microscope (SEM), FTIR, and mechanical tests. The cellular responses of the core-shell microcapsules were evaluated through in vitro cell studies by encapsulating NIH/3T3 fibroblast cells. Notably, the bioactive microcapsule showed that the cell viability was found excellent for more than 2 weeks. Thus, the results of this core-shell microcapsule showed a promising approach to creating 3D hydrogel networks suitable for different biomedical applications such as in vitro tissue models for toxicity studies, wound healing, and tissue repair.

Coaxial Electrospinning Construction Si@C Core-Shell Nanofibers for Advanced Flexible Lithium-Ion Batteries.

Project description:Silicon (Si) is expected to be a high-energy anode for the next generation of lithium-ion batteries (LIBs). However, the large volume change along with the severe capacity degradation during the cycling process is still a barrier for its practical application. Herein, we successfully construct flexible silicon/carbon nanofibers with a core-shell structure via a facile coaxial electrospinning technique. The resultant Si@C nanofibers (Si@C NFs) are composed of a hard carbon shell and the Si-embedded amorphous carbon core framework demonstrates an initial reversible capacity of 1162.8 mAh g-1 at 0.1 A g-1 with a retained capacity of 762.0 mAh g-1 after 100 cycles. In addition, flexible LIBs assembled with Si@C NFs were hardly impacted under an extreme bending state, illustrating excellent electrochemical performance. The impressive performances are attributed to the high electric conductivity and structural stability of the porous carbon fibers with a hierarchical porous structure, indicating that the novel Si@C NFs fabricated using this electrospinning technique have great potential for advanced flexible energy storage.

Emulsion electrospinning of sodium alginate/poly(ε-caprolactone) core/shell nanofibers for biomedical applications.

Project description:Electrospun nanofibers have shown great potential as drug vehicles and tissue engineering scaffolds. However, the successful encapsulation of multiple hydrophilic/hydrophobic therapeutic compounds is still challenging. Herein, sodium alginate/poly(ε-caprolactone) core/shell nanofibers were fabricated via water-in-oil emulsion electrospinning. The sodium alginate concentration, water-to-oil ratio, and surfactant concentration were optimized for the maximum stability of the emulsion. The results demonstrated that an increasing water-to-oil ratio results in more deviation from Newtonian fluid and leads to a broader distribution of the fibers' diameters. Moreover, increasing poly(ε-caprolactone) concentration increases loss and storage moduli and increases the diameter of the resulting fibers. The nanofibers' characteristics were investigated by scanning electron microscopy, transmission electron microscopy, confocal laser scanning microscopy, Fourier transform infrared spectroscopy, X-ray diffraction, and water contact angle measurements. It was observed that using an emulsion composition of 10% (w/v) PCL and a water-to-oil ratio of 0.1 results in smooth, cylindrical, and uniform core/shell nanofibers with PCL in the shell and ALG in the core. The in vitro cell culture study demonstrated the favorable biocompatibility of nanofibers. Overall, this study provides a promising and trustworthy material for biomedical applications.
