Long-duration animal tracking in difficult lighting conditions.

ABSTRACT: High-throughput analysis of animal behavior requires software to analyze videos. Such software typically depends on the experiments' being performed in good lighting conditions, but this ideal is difficult or impossible to achieve for certain classes of experiments. Here, we describe techniques that allow long-duration positional tracking in difficult lighting conditions with strong shadows or recurring "on"/"off" changes in lighting. The latter condition will likely become increasingly common, e.g., for Drosophila due to the advent of red-shifted channel rhodopsins. The techniques enabled tracking with good accuracy in three types of experiments with difficult lighting conditions in our lab. Our technique handling shadows relies on single-animal tracking and on shadows' and flies' being accurately distinguishable by distance to the center of the arena (or a similar geometric rule); the other techniques should be broadly applicable. We implemented the techniques as extensions of the widely-used tracking software Ctrax; however, they are relatively simple, not specific to Drosophila, and could be added to other trackers as well.

Project description:Parameters of illumination including the spectra, intensity, and photoperiod play an important role in the cultivation of plants under greenhouse conditions, especially for vegetables such as lettuce. We previously showed that illumination by a combination of red, blue, and white LEDs with a high red light intensity, was optimal for lettuce cultivation; however, the effect of the photoperiod on lettuce cultivation was not investigated. In the current work, we investigated the influence of photoperiod on production (total biomass and dry weight) and parameters of photosynthesis, respiration rate, and relative chlorophyll content in lettuce plants. A 16 h (light):8 h (dark) illumination regime was used as the control. In this work, we investigated the effect of photoperiod on total biomass and dry weight production in lettuce plants as well as on photosynthesis, respiration rate and chlorophyll content. A lighting regime 16:8 h (light:dark) was used as control. A shorter photoperiod (8 h) decreased total biomass and dry weight in lettuce, and this effect was related to the suppression of the linear electron flow caused by the decreasing content of chlorophylls and, therefore, light absorption. A longer photoperiod (24 h) increased the total biomass and dry weight, nevertheless an increase in photosynthetic processes, light absorption by leaves and chlorophyll content was not recorded, nor were differences in respiration rate, thus indicating that changes in photosynthesis and respiration are not necessary conditions for stimulating plant production. A simple model to predict plant production was also developed to address the question of whether increasing the duration of illumination stimulates plant production without inducing changes in photosynthesis and respiration. Our results indicate that increasing the duration of illumination can stimulate dry weight accumulation and that this effect can also be induced using the equal total light integrals for day (i.e., this stimulation can be also caused by increasing the light period while decreasing light intensity). Increasing the duration of illumination is therefore an effective approach to stimulating lettuce production under artificial lighting.

Project description:BackgroundThere is increasing recognition of the need to stabilize sleep-wake cycles in individuals with major mental disorders. As such, clinicians and researchers advocate the use of interventions targeted at sleep and circadian dysrhythmias as an adjunct to the standard treatments offered for acute illness episodes of a broad range of diagnoses. To determine the trans-diagnostic generalizability of chronotherapy, we explore the benefits of admitting individuals with an acute illness episode to a psychiatric inpatient unit where changes in light exposure are integrated into the therapeutic environment.Methods/designA two-arm, pragmatic effectiveness, randomized controlled treatment trial, where individuals admitted for acute inpatient psychiatric care will be allocated to a ward with blue-depleted evening light or to a ward with the same layout and facilities but lacking the new lighting technology. The trial will test whether the experimental lighting conditions offer any additional benefits beyond those associated with usual treatment in an acute psychiatric inpatient unit. The main objectives are to examine any differences between groups in the mean duration of hospitalization in days. Additional analyses will compare group differences in symptoms, functioning, medication usage, and side effects and whether length of stay is associated with stability of sleep-wake cycles and circadian rhythms. Ancillary investigations should determine any benefits according to diagnostic subgroups and potential drawbacks such as any adverse effects on the well-being of professionals working across both wards.DiscussionThis unit offers a unique opportunity to explore how exposure to different lighting conditions may modify sleep-wake cycles and how any changes in sleep-wake cycle may impact on the clinical and functional outcomes of individuals experiencing an acute episode of a severe mental disorder that requires inpatient care. The findings could influence the future design of hospital units offering care to patients with mental or physical disorders.Trial registrationClinicalTrials.gov, ID: NCT03788993 . Project description:The objective of this study was to determine the duration of the main phenological stages, plant growth development, yield, and cone quality of hop cultivars grown under artificial light (17 h per day) during vegetative development (early season) in a subtropical climate region. The study was conducted in Palotina, Paraná, Brazil (24° S), during the 2021 and 2022 growing seasons. The plants were cultivated in a 5.5 m high trellis system with artificial light supplementation during vegetative development. The hop cultivars Hallertau Mittelfrüher, Mapuche, Northern Brewer, Spalter, and Yakima Gold were used in the treatments. The duration of the phenological stages, vegetative growth (plant height, fresh mass of the plants, number of lateral branches per plant), components of productive yield (number of cones per side branch, number of cones per plant, fresh mass, length, and width of the cone, production of fresh cones per plant, and yield), and chemical components of the cones (alpha- and beta-acid contents, and essential oil concentrations) were recorded. The duration of the phenological stages was visually evaluated, and plant growth was analyzed using non-linear log-logistic regression. The remaining data were subjected to analysis of variance and the means were compared using Tukey's test. The data were also subjected to multivariate analysis using the principal components test, correlation analysis, and hierarchical grouping. The cultivar Mapuche was considered an early hop in both seasons, and the cultivars Spalter and Yakima Gold were considered early cultivars in the second season. In both seasons, the productive yield components were positively correlated with the precocity of the cultivars, in which Mapuche in the 2021 season and Mapuche, Spalter, and Yakima Gold in the 2022 season had the highest mean of the number of cones per side branch and per plant, production per plant, and productivity. The cultivar Yakima Gold had a positive correlation with the chemical quality of cones, alpha and beta acid contents, and essential oil concentrations, for both seasons.

Project description:BackgroundFuture long-duration space exploration missions, such as traveling to Mars, will create an increase in communication time delays and disruptions and remove the viability of emergency returns to Earth for timely medical treatment. Thus, higher levels of medical autonomy are necessary. Crew selection is proposed as the first line of defense to minimize medical risk for future missions; however, the second proposed line of defense is medical preparedness and crew member autonomy. In an effort to develop a decision support system, the Canadian Space Agency mandated a team of scientists from Thales Research and Technology Canada (Québec, QC) and Université Laval (Québec, QC) to create an evidence-based medical condition database linking mission-critical human conditions with key causal factors, diagnostic and treatment information, and probable outcomes.ObjectiveTo complement this database, we are currently conducting a scoping review to better understand the depth and breadth of evidence about managing medical conditions in space.MethodsThis scoping review will adhere to quality standards for scoping reviews, employing Levac, Colquhoun, and O'Brien's 6-stage methodology; the reported results will follow the PRISMA (Preferred Reporting Items for Systematic Reviews and Meta-Analyses) extension for scoping reviews. In stage 1, we identified the research question in collaboration with the Canadian Space Agency (CSA), the main knowledge user. We prioritized 10 medical conditions: (1) acute coronary syndrome, (2) atrial fibrillation, (3) eye penetration, (4) herniated disk, (5) nephrolithiasis, (6) pulmonary embolism, (7) retinal detachment, (8) sepsis, (9) stroke, and (10) spaceflight associated neuro-ocular syndrome. In stage 2, with the help of an information specialist from Cochrane Canada Francophone, papers were identified through searches of the following databases: ARC, Embase, IeeeXplore, Medline Ovid, PsychINFO, and Web of Science. In stage 3, studies will be selected and assessed using a 3-step process and emerging, refined exclusion criteria. In stage 4, the data will be charted in a table based on parameters required by the CSA and developed using Google spreadsheets for shared access. In stage 5, evidence-based descriptive summaries will be produced for each condition, as well as descriptive analyses of collected data. Finally, in stage 6, the findings will be shared with the CSA to guide the completion of this project.ResultsThis study was planned in December 2018. Stage 1 has been completed. The initial database search strategy with all target conditions combined identified a total of 10,403 citations to review through title and abstract screening and after duplicate removal. We plan to complete stages 2-6 by the beginning of 2021.ConclusionsThis scoping review will map the literature on the management of 10 priority medical conditions in space. It will also enable us to identify knowledge gaps that must be addressed in future research, ensuring successful and medically safe future missions as humankind embarks upon new frontiers of space exploration.International registered report identifier (irrid)DERR1-10.2196/24323.
