A new family of non-toxic biocides based on low melting point (1250°C) transparent glasses with high content of ZnO (15-40wt%) belonging to the miscibility region of the B2O3-SiO2-Na2O-ZnO system has been developed. Project description:Novel CaO-P2 O5 -Nb2 O5 -Ag2 O invert glasses with substitution Ag2 O for Nb2 O5 were successfully prepared using a melt-quenching method. Ag2 O in the glasses act as a network modifier oxide, playing the same role as Na2 O, which breaks the phosphate chains. Analysis of the ultraviolet-visible absorption spectra of the glasses showed that the glass matrix contained ionic silver species and silver nanoparticles. Approximately 0.05 mM of Nb5+ ions released from the glasses, which would be expected to stimulate osteoblast differentiation. A glass containing 1 mol % Ag2 O showed a linear increase in the releasing amount of Ag+ ions with increasing soaking time, whereas glasses containing 3-5 mol % Ag2 O showed Ag+ ion concentrations of around 13 μM at day 3, and then maintained similar values until day 7. When the solution was replaced with fresh solution every 2 days, the Ag+ ion dissolution amounts indicated almost constantly 13 μM due to AgCl formation. There were no differences in the numbers of primary osteoblast cells on silver-free and silver-containing glasses after cultivation for 1-7 days. The silver-containing calcium phosphate invert glasses showed cytocompatibility with simultaneous antibacterial activity to Escherichia coli and Staphylococcus aureus. © 2017 Wiley Periodicals, Inc. Project description:BackgroundPlant fungi (e.g., Pellicularia sasakii, Gibberella zeae, Fusarium oxysporum, and Cytospora mandshurica and Phytophthora infestans) and bacteria (e.g., Ralstonia solanacearum) are extremely difficult to manage in agricultural production. The high incidence of plant mortality and the lack of effective control methods make P. sasakii and R. solanacearum two of the world's most destructive plant pathogens. Pathogenic fungi and bacteria are responsible for billions of dollars in economic losses worldwide each year. Thus, we designed an active amide structure and synthesized a series of novel amide derivatives containing a triazole moiety to discover new bioactive molecules and pesticides that can act against fungi and bacteria.ResultsA series of amide derivatives containing a triazole moiety were synthesized. All the obtained compounds were characterized through proton and carbon nuclear magnetic resonance spectroscopy, infrared spectroscopy, and elemental analysis. Preliminary antifungal activity test showed that some of the synthesized compounds exhibited moderate antifungal activity against P. sasakii, G. azeae, F. oxysporum, C. mandshurica, and P. infestans at 50 mg/L. Compound 4u displayed more potent antifungal activity against P. sasakii and G. azeae than hymexazol. Preliminary antibacterial activity results showed that some of the synthesized compounds exhibited high anti-bacterial activity against R. solanacearum at 200 mg/L. Compounds 4m and 4q displayed high antibacterial activity against R. solanacearum, with 71% and 65% inhibitory rates, respectively.ConclusionsA series of novel amide derivatives containing 1,2,4-triazole moiety were synthesized through the reaction of intermediate 3 with different acyl chlorides and anhydrous potassium carbonates in anhydrous tetrahydrofuran at 50°C, using 2,4-dichloroacetophenoneas as a starting material. The title compounds exhibited high inhibitory effects against P. sasakii, R. solanacearum, and G. azeae.

Project description:The sterilization of the entire root canal system represents the main goal of every endodontist, given the fact that the control of the microbial flora is the key point of every root canal treatment. The diversity of microorganisms found inside the root canal and also the resistance of some bacterial species to intracanal medications led to a continuous development of new endodontic products. The present study focuses on the comparison of the antibacterial and antifungal properties of different endodontic products, two commercially available, one experimental plant based extract, and two control substances.The disc diffusion assay was used to determine the antibacterial and antifungal properties of chlorhexidine, calcium hydroxide, a mix extract between Arctium lappa root powder and Aloe barbadensis Miller gel, Amoxicillin with clavulanic acid and Fluconazole (as control substances). Two of the most common microorganisms found in endodontic infections were chosen: Enterococcus faecalis (ATCC 29212) and Candida albicans ATCC(10231).All tested substances showed inhibition zones around the discs, for Enterococcus faecalis and Candida albicans, including the experimental mix extract of Arctium lappa root powder with Aloe vera gel.The experimental mix extract of Arctium lappa root powder and Aloe vera gel is able to inhibit very resistant microorganisms, like Enterococcus faecalis and Candida albicans.

Project description:The use of a combination of nanoparticles as antimicrobial agents can be one strategy to overcome the tendency of microbes to become resistant to antibiotic action. Also, the optimization of nano-photocatalysts to efficiently remove persistent pollutants from wastewater is a hot topic. In this study, two composites ZnO/Au (1% wt.) and ZnO/Ag (1% wt.) were synthesized by simple aqueous solution methods. The structure and morphology of the r nanocomposites were analyzed by structural and optical characterization methods. The formation of AuNPs and AgNPs in these experiments was also discussed. The antimicrobial properties of ZnO, ZnO/Au, and ZnO/Ag nanomaterials were investigated against Gram-negative bacteria (Pseudomonas aeruginosa) and Gram-positive bacteria (Staphylococcus aureus). The results showed an increase of 80% in the antimicrobial activity of ZnO/Au against Pseudomonas aeruginosa compared with 30% in the case of ZnO/Ag. Similarly, in the case of the S. aureus strain tests, ZnO/Au increased the antimicrobial activity by 55% and ZnO/Ag by 33%. The photocatalytic tests indicated an improvement in the photocatalytic degradation of methylene blue (MB) under UV irradiation using ZnO/Au and ZnO/Ag nanocomposites compared to bare ZnO. The photocatalytic degradation efficiency of ZnO after 60 min of UV irradiation was ∼83%, while the addition of AuNPs enhanced the degradation rate to ∼95% (ZP2), and AgNP presence enhanced the efficiency to ∼98%. The introduction of noble metallic nanoparticles into the ZnO matrix proved to be an effective strategy to increase their antimicrobial activity against P. aeruginosa and S. aureus, and their photocatalytic activity was evaluated through the degradation of MB dye. Comparing the enhancing effects of Au and Ag, it was found that ZnO/Au was a better antimicrobial agent while ZnO/Ag was a more effective photocatalyst under UV irradiation.

Sesquiterpenoids, with abundant structural skeleton types and a wide range of bioactivities, are considered as good candidates to be antibacterial and antifungal agents. In the past decades, many sesquiterpenoids were isolated from plants and fungi that exhibited good antibacterial and antifungal activities. In this review, the names, source, structures, antibacterial and antifungal degrees, and mechanisms of sesquiterpenoids with antibacterial and antifungal activity from 2012 to 2022 are summarized, and the structure-activity relationship of these sesquiterpenoids against bacteria and fungi is also discussed.

Project description:The rapid emergence of drug-resistant bacteria and fungi poses a threat for healthcare worldwide. The development of novel effective small molecule therapeutic strategies in this space has remained challenging. Therefore, one orthogonal approach is to explore biomaterials with physical modes of action that have the potential to generate antimicrobial activity and, in some cases, even prevent antimicrobial resistance. Here, to this effect, we describe an approach for forming silk-based films that contain embedded selenium nanoparticles. We show that these materials exhibit both antibacterial and antifungal properties while crucially also remaining highly biocompatible and noncytotoxic toward mammalian cells. By incorporating the nanoparticles into silk films, the protein scaffold acts in a 2-fold manner; it protects the mammalian cells from the cytotoxic effects of the bare nanoparticles, while also providing a template for bacterial and fungal eradication. A range of hybrid inorganic/organic films were produced and an optimum concentration was found, which allowed for both high bacterial and fungal death while also exhibiting low mammalian cell cytotoxicity. Such films can thus pave the way for next-generation antimicrobial materials for applications such as wound healing and as agents against topical infections, with the added benefit that bacteria and fungi are unlikely to develop antimicrobial resistance to these hybrid materials.

Project description:Edible flowers have been used since ancient times, but their potential for improving human health has not been explored. This study aimed to evaluate the profile of bioactive compounds (organic acids, phenolics, and carotenoids) and the antioxidant and antimicrobial activity of nine flower varieties with high concentrations of carotenoids or total phenolic compounds. Ninety-three edible flowers were analysed for physicochemical characteristics, total phenolic and carotenoid concentrations, and antioxidant activity (ABTS). Bioactive profiles were determined by rapid resolution liquid chromatography (RRLC), and antimicrobial activity was determined against Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa and Streptococcus mutans, and Candida albicans and Candida tropicalis. Chrysanthemum x hybrid orange, Helianthus annuus yellow, Tagetes patula orange, Canna indica red, and Hibiscus rosa-sinensis (orange1 and yellow) showed significant concentrations of total carotenoids. In contrast, Pelargonium hortorum orange2, Hibiscus rosa-sinensis red1, and Rosa x hybrid variety medium yellow showed high levels of total phenolics. The predominant compounds in these species were citric acid (991.4 mg/g DW in Hibiscus rosa-sinensis red1), 4-hydroxybenzoic acid (936.2 mg/100 g DW in P. hortorum orange2), kaempferol (971. 9 mg/100 g DW in T. patula orange), quercetin glucoside (958.8 in C. x hybrid), quercetin (919.3 mg/100 g DW in T. patula), α-carotene, and β-carotene in T. patula orange (989.5 and 601.2 mg/100 g DW, respectively). Regarding antimicrobial activity, T. patula orange and P. hortorum orange2 inhibited bacterial growth, while C. x hybrid orange and P. hortorum orange2 inhibited Candida albicans, and the latter inhibited Candida tropicalis. These results indicate the potential of edible flowers as a natural source of bioactive compounds and as a tool in the fight against antimicrobial resistance.

Project description:Microbial infections remains a serious challenge in food industries due to their resistance to some of the well-known antibacterial and antifungal agents. In this work, a novel monomyristoyl ester (fructosyl monomyristate) and two other derivatives (i.e., glucosyl and galactosyl monomyristates) were successfully synthesized from myristic acid and monosaccharides in two-step reactions. First, the myristic acid was converted to myristoyl chloride, and then the myristoyl chloride was reacted with fructose, glucose and galactose separately to produce the corresponding monosaccharide monomyristate derivatives. The structures of the synthesized products were confirmed by Fourier transform infrared (FTIR), proton and carbon nuclear magnetic resonance (1H- and 13C-NMR), and mass spectral (MS) data. The monomyristates esters were obtained in reaction yields of 45.80%-79.49%. The esters were then evaluated for their antimicrobial activity using the disc diffusion test. It was found that the esters exhibited a medium antibacterial activity against gram-positive bacteria; however, they showed a weak antibacterial activity against gram-negative bacteria. Amongst the esters, galactosyl myristate yielded the highest antibacterial activity against Salmonella typhimurium, Staphylococcusaureus and Bacillussubtilis, while glucosyl monomyristate exhibited the highest antibacterial activity only against Escherichia coli. Additionally, all products showed remarkable antifungal activity against Candida albicans. These findings demonstrate that monosaccharide monomyristate derivatives are promising for use as biocompatible antimicrobial agents in the future.

Project description:To explore more efficient and less toxic antibacterial and antifungal pesticides, we utilized 2,6-difluorobenzamide as a starting material and ultimately synthesized 23 novel benzoylurea derivatives containing a pyrimidine moiety. Their structures were characterized and confirmed by 1H NMR, 13C NMR, 19F NMR, and HRMS. The bioassay results demonstrated that some of the title compounds exhibited moderate to good in vitro antifungal activities against Botrytis cinerea in cucumber, Botrytis cinerea in tobacco, Botrytis cinerea in blueberry, Phomopsis sp., and Rhizoctonia solani. Notably, compounds 4j and 4l displayed EC50 values of 6.72 and 5.21 μg/mL against Rhizoctonia solani, respectively, which were comparable to that of hymexazol (6.11 μg/mL). Meanwhile, at 200 and 100 concentrations, the target compounds 4a-4w exhibited lower in vitro antibacterial activities against Xanthomonas oryzae pv. oryzicola and Xanthomonas citri subsp. citri, respectively, compared to those of thiodiazole copper. Furthermore, the molecular docking simulation demonstrated that compound 4l formed hydrogen bonds with SER-17 and SER-39 of succinate dehydrogenase (SDH), providing a possible explanation for the mechanism of action between the target compounds and SDH. This study represents the first report on the antifungal and antibacterial activities of novel benzoylurea derivatives containing a pyrimidine moiety.

Project description:The emergence and re-emergence of antibiotic-resistant bacteria is a potential threat to treating infectious diseases. This study employed a nanometer-scale green synthesis using an extract of Solanum incanum leaves to obtain nanoparticles (NPs) and nanocomposites (NCs) possessing antibacterial properties. The FESEM-EDS elemental mapping analysis proved the novelty of the green synthesis approach in synthesizing a copper-doped ZnO NCs with good dopant distribution. The crystallinity and ZnO bandgap were adjusted by extrinsic copper doping in the ZnO lattice. The optical property adjustments from 3.04 to 2.97 eV for indirect Kubelka-Munk functions were confirmed from DRS-UV-vis analysis. The dopant inclusion in the host lattice was also confirmed by the angle shift on the XRD pattern analysis relative to single ZnO. In addition to doping, the XRD pattern analysis also showed the development of CuO crystals. The lattice fringe values from HRTEM analysis confirmed the existence of both CuO and ZnO crystals with local heterojunctions. Doping and heterojunctions have crucial values in charge transfer and visible light harvesting behaviour, as proved by the PL analysis. The synergistic effects of the doped NCs showed greater antibacterial activity against both Gram-positive and Gram-negative bacteria as a result of more ROS generation through the bacteria-cell-catalyst interaction and release of metal ions. The antioxidant potential of the doped NCs was found to be higher than that of single NPs, using the 2,2-diphenyl-1-picrylhydrazyl free radical scavenging assay and is expected to impart protective effects to the host cells by scavenging destructive free radicals. Thus, the overall analysis leads to the conclusion that the potentiality of synthesized materials has a future outlook for biological applications, especially in the development of antimicrobials to combat antibiotic-resistant bacteria and microbes.
