A highly selective fluorescent probe for direct detection and isolation of mouse embryonic stem cells.

ABSTRACT: Stem cell research has gathered immense attention in the past decade due to the remarkable ability of stem cells for self-renewal and tissue-specific differentiation. Despite having numerous advancements in stem cell isolation and manipulation techniques, there is a need for highly reliable probes for the specific detection of live stem cells. Herein we developed a new fluorescence probe (CDy9) with high selectivity for mouse embryonic stem cells. CDy9 allows the detection and isolation of intact stem cells with marginal impact on their function and capabilities.

Project description:A novel fluorescent quinolizinium-based turn-off probe has been developed for selective detection of cysteine. The probe showed high selectivity and sensitivity towards cysteine over other amino acids including the similarly structured homocysteine and glutathione with a detection limit of 0.18 μM (S/N = 3). It was successfully applied to cysteine detection in living cells with low cytotoxicity and quantitative analysis of spiked mouse serum samples with moderate to good recovery (96-109%).

A Highly Selective Turn-On Fluorescent Probe for the Detection of Zinc.

Project description:A novel turn-on fluorescence probe L has been designed that exhibits high selectivity and sensitivity with a detection limit of 9.53 × 10-8 mol/L for the quantification of Zn2+. 1H-NMR spectroscopy and single crystal X-ray diffraction analysis revealed the unsymmetrical nature of the structure of the Schiff base probe L. An emission titration experiment in the presence of different molar fractions of Zn2+ was used to perform a Job's plot analysis. The results showed that the stoichiometric ratio of the complex formed by L and Zn2+ was 1:1. Moreover, the molecular structure of the mononuclear Cu complex reveals one ligand L coordinates with one Cu atom in the asymmetric unit. On adding CuCl2 to the ZnCl2/L system, a Cu-Zn complex was formed and a strong quenching behavior was observed, which inferred that the Cu2+ displaced Zn2+ to coordinate with the imine nitrogen atoms and hydroxyl oxygen atoms of probe L.

Fluorescent Mechanism of a Highly Selective Probe for Copper(II) Detection: A Theoretical Study.

Project description:A highly selective probe for copper(II) detection based on the dansyl group was theoretically studied by means of (time-dependent) density functional theory. The calculated results indicated that the oscillator strength of the fluorescent process for the probe molecule is considerably large, but the counterpart of its copper(II) complex is nearly zero; therefore, the predicted radiative rate kr of the probe is several orders of magnitude larger than that of its complex; however, the predicted internal conversion rate kic of both the probe and its complex is of the same order of magnitude. In addition, the simulated intersystem crossing rate kisc of the complex is much greater than that of the probe due to the effect of heavy atom from the copper atom in the complex. Based on the above information, the calculated fluorescence quantum yield of the probe is 0.16% and that of the complex becomes 10-6%, which implies that the first excited state of the probe is bright state and that of the complex is dark state. For the complex, the hole-electron pair analysis indicates that the process of S0 → S1 belongs to metal-to-ligand charge transfer; its density-of-state diagram visually illustrates that the highest occupied molecular orbital (HOMO) contains the ingredient of the s orbital from the copper atom, which decreases the frontier orbital energy level and the overlap integral of HOMO and LUMO.

Novel Coumarin-Substituted Cyclophosphazene as a Fluorescent Probe for Highly Selective Detection of 2,4,6-Trinitrophenol.

Project description:Nitroaromatic compounds (NACs) such as 2,4,6-trinitrophenol (TNP), commonly known as picric acid (PA), hold widespread application in industries such as dyestuff production, wood preservation, explosives manufacturing, insect control, and photographic development. In this study, the organic-inorganic hybrid 4,4',4″,4‴,4″″,4‴″-((1,3,5,2λ5,4λ5,6λ5-triazatriphosphinine-2,2,4,4,6,6-hexayl)hexakis(oxy))hexakis(2H-chromen-2-one) (Cpz-4-HC) was synthesized via the nucleophilic substitution reaction of 4-hydroxycoumarin (4-HC) with hexachlorocyclotriphosphazene (HCCP). The structure of Cpz-4-HC was fully characterized by Fourier transform infrared (FT-IR), 1H-, 13C-, and 31P NMR, and HRMS. Cpz-4-HC is used as a chemical fluorescence sensor for the detection of TNP, with a K SV value of 4.71 × 104 M-1 and a low limit of detection (LOD) of 0.334 ppm over some other analytes such as 2,4-DNP, 4-NP, 2-NP, 1,3-DNB, 2,4-DNT, and 2,6-DNT in water. The sensing mechanism was elucidated through spectral overlap analysis, indicating the resonance energy transfer as the dominant quenching process. Dynamic quenching was established through fluorescence lifetime studies, further affirming Cpz-4-HC capability for environmental monitoring. Experimental and theoretical analyses underscored TNP's strong interaction with Cpz-4-HC, corroborating its suitability for sensing applications. Their recyclable nature and ultrafast response time make them highly suitable for detecting TNP, even in the presence of other interfering nitroaromatics. This study provides novel perspectives on the development and formulation of a chemical fluorescent sensor for TNP, utilizing a straightforward synthesis method.

A highly selective TPE-based AIE fluorescent probe is developed for the detection of Ag.

Project description:The detection of Ag+ in the environment is very important to determine the level of pollution from silver complexes, which have caused various human health problems. Herein, an aggregation-induced emission (AIE) chromophore (tetraphenylethane, TPE) attached to a benzimidazole group (tetra-benzimidazole, TBI-TPE) is synthesized and utilized to detect Ag+ in the environment. The strong chelating effect between the benzimidazole group and Ag+ leads to the formation of aggregates, and strong yellow fluorescence signals were observed after adding Ag+ into a TBI-TPE solution. The stoichiometry of the complex of TBI-TPE and Ag+ was established to be 1 : 2 using photochemical and mass spectra measurements. The detection limit of the Ag+ assay is 90 nM with a linear range from 100 nM to 6 μM. This study provides a facile method to determine Ag+ in real environmental samples with satisfactory results.

Sensitive Detection of Various Forms of Hydrogen Sulfide via Highly Selective Naphthalimide-Based Fluorescent Probe.

Project description:Hydrogen sulfide (H2S) is an important gasotransmitter, but only a few methods are available for real-time detection. Fluorescent probes are attractive tools for biological applications because of their high sensitivity, convenience, rapid implementation, noninvasive monitoring capability, and simplicity in fluorescent imaging of living cells and tissues. Herein, we report on a pro-fluorescent probe, NAP-Py-N3 based on naphthalimide derivative, which was found to show high selectivity toward H2S over various other analytes, including biothiols, making it feasible to detect H2S. After reaction with H2S, this probe showed rapid and significant turn-on green fluorescent enhancement at 553 nm (about 54-fold, k2 = 9.62 M-1s-1), high sensitivity (LOD: 15.5 nM), significant Stokes shift (118 nm), and it was found that the fluorescence quantum yield of fluorescence product can reach 0.36. Moreover, the probe has also been successfully applied to detect the gaseous H2S and to confirm the presence of H2S released from modern organic donors, which in recent years have been commonly used to investigate the role of H2S in biological systems. All the results indicate that this probe is excellent and highly valuable.

A New Ratiometric Fluorescent Probe Based on BODIPY for Highly Selective Detection of Hydrogen Sulfide.

Project description:Hydrogen sulfide (H2S) as small molecular signal messenger plays key functions in numerous biological processes. The imaging detection of intracellular hydrogen sulfide is of great significance. In this work, a ratiometric fluorescent probe BH based on an asymmetric BODIPY dye for detection of H2S was designed and synthesized. After the interaction with hydrogen sulfide, probe display colorimetric and ratiometric fluorescence response, with its maximum emission fluorescence wavelength red-shifted from 542 nm to 594 nm, which is attributed to the sequential nucleophilic reaction of H2S leading to enhanced molecular conjugation after ring formation of the BODIPY skeleton. A special response mechanism has been fully investigated by NMR titration and MS, so that the probe has excellent detection selectivity. Furthermore, probe BH has low cytotoxicity and fluorescence imaging experiments indicate that it can be used to monitor hydrogen sulfide in living cells.

A Novel NIR Fluorescent Probe for Highly Selective Detection of Nitroreductase and Hypoxic-Tumor-Cell Imaging.

Project description:Nitroreductase as a potential biomarker for aggressive tumors has received extensive attention. In this work, a novel NIR fluorescent probe for nitroreductase detection was synthesized. The probe Py-SiRh-NTR displayed excellent sensitivity and selectivity. Most importantly, the confocal fluorescence imaging demonstrated that HepG-2 cells treated with Py-SiRh-NTR under hypoxic conditions showed obvious enhanced fluorescence, which means that the NTR was overexpressed under hypoxic conditions. Moreover, the probe showed great promise that could help us to study related anticancer mechanisms research.

Novel Amino-pillar[5]arene as a Fluorescent Probe for Highly Selective Detection of Au3+ Ions.

Project description:A novel fluorescent probe, amino-pillar[5]arene (APA), was prepared via a green, effective, and convenient synthetic method, which was characterized by nuclear magnetic resonance (NMR), infrared (IR), and high-resolution mass spectrometry. The fluorescence sensing behavior of the APA probe toward 22 metal ions in aqueous solutions were studied by fluorescence spectroscopy. The results showed that APA could be used as a selective fluorescent probe for the specificity detection of Au3+ ions. Moreover, the detection characteristics were investigated by fluorescence spectral titration, pH effect, fluorescence competitive experiments, Job's plot analysis, 1H NMR, and IR. The results indicated that detection of Au3+ ions by the APA probe could be achieved in the range of pH 1-13.5 and that other coexisting metal ions did not cause any marked interference. The titration analysis results indicated that the fluorescence intensity decreased as the concentration of Au3+ ions increased, with an excellent correlation (R 2 = 0.9942). The detection limit was as low as 7.59 × 10-8 mol·L-1, and the binding ratio of the APA probe with Au3+ ions was 2:1. Therefore, the APA probe has potential applications for detecting Au3+ ions in the environment and in living organisms.

Highly sensitive and selective detection of triphosgene with a 2-(2'-hydroxyphenyl)benzimidazole derived fluorescent probe.

Project description:In this work, a 2-(2'-hydroxyphenyl)benzimidazole derived fluorescent probe, 2-(2'-hydroxy-4'-aminophenyl)benzimidazole (4-AHBI), was synthesized and its fluorescent behavior toward triphosgene were evaluated. The results showed that 4-AHBI exhibited high sensitivity (limit of detection, 0.08 nM) and excellent selectivity for triphosgene over other acyl chlorides including phosgene in CH2Cl2 solution. Moreover, 4-AHBI loaded test strips were prepared for the practical sensing of triphosgene.
