Biochemical nature of Russell Bodies.

ABSTRACT: Professional secretory cells produce and release abundant proteins. Particularly in case of mutations and/or insufficient chaperoning, these can aggregate and become toxic within or amongst cells. Immunoglobulins (Ig) are no exception. In the extracellular space, certain Ig-L chains form fibrils causing systemic amyloidosis. On the other hand, Ig variants lacking the first constant domain condense in dilated cisternae of the early secretory compartment, called Russell Bodies (RB), frequently observed in plasma cell dyscrasias, autoimmune diseases and chronic infections. RB biogenesis can be recapitulated in lymphoid and non-lymphoid cells by expressing mutant Ig-μ, providing powerful models to investigate the pathophysiology of endoplasmic reticulum storage disorders. Here we analyze the aggregation propensity and the biochemical features of the intra- and extra-cellular Ig deposits in human cells, revealing β-aggregated features for RB.

Russell-Like Bodies in Plant Seeds Share Common Features With Prolamin Bodies and Occur Upon Recombinant Protein Production.

Project description:Although many recombinant proteins have been produced in seeds at high yields without adverse effects on the plant, endoplasmic reticulum (ER) stress and aberrant localization of endogenous or recombinant proteins have also been reported. The production of murine interleukin-10 (mIL-10) in Arabidopsis thaliana seeds resulted in the de novo formation of ER-derived structures containing a large fraction of the recombinant protein in an insoluble form. These bodies containing mIL-10 were morphologically similar to Russell bodies found in mammalian cells. We confirmed that the compartment containing mIL-10 was enclosed by ER membranes, and 3D electron microscopy revealed that these structures have a spheroidal shape. Another feature shared with Russell bodies is the continued viability of the cells that generate these organelles. To investigate similarities in the formation of Russell-like bodies and the plant-specific protein bodies formed by prolamins in cereal seeds, we crossed plants containing ectopic ER-derived prolamin protein bodies with a line accumulating mIL-10 in Russell-like bodies. This resulted in seeds containing only one population of protein bodies in which mIL-10 inclusions formed a central core surrounded by the prolamin-containing matrix, suggesting that both types of protein aggregates are together removed from the secretory pathway by a common mechanism. We propose that, like mammalian cells, plant cells are able to form Russell-like bodies as a self-protection mechanism, when they are overloaded with a partially transport-incompetent protein, and we discuss the resulting challenges for recombinant protein production.

Mycoparasite Hypomyces odoratus infests Agaricus xanthodermus fruiting bodies in nature.

Project description:Mycopathogens are serious threats to the crops in commercial mushroom cultivations. In contrast, little is yet known on their occurrence and behaviour in nature. Cobweb infections by a conidiogenous Cladobotryum-type fungus identified by morphology and ITS sequences as Hypomyces odoratus were observed in the year 2015 on primordia and young and mature fruiting bodies of Agaricus xanthodermus in the wild. Progress in development and morphologies of fruiting bodies were affected by the infections. Infested structures aged and decayed prematurely. The mycoparasites tended by mycelial growth from the surroundings to infect healthy fungal structures. They entered from the base of the stipes to grow upwards and eventually also onto lamellae and caps. Isolated H. odoratus strains from a diseased standing mushroom, from a decaying overturned mushroom stipe and from rotting plant material infected mushrooms of different species of the genus Agaricus while Pleurotus ostreatus fruiting bodies were largely resistant. Growing and grown A. xanthodermus and P. ostreatus mycelium showed degrees of resistance against the mycopathogen, in contrast to mycelium of Coprinopsis cinerea. Mycelial morphological characteristics (colonies, conidiophores and conidia, chlamydospores, microsclerotia, pulvinate stroma) and variations of five different H. odoratus isolates are presented. In pH-dependent manner, H. odoratus strains stained growth media by pigment production yellow (acidic pH range) or pinkish-red (neutral to slightly alkaline pH range).

Predicting biochemical oxygen demand in European freshwater bodies.

Project description:Biochemical Oxygen Demand (BOD) is an indicator of organic pollution in freshwater bodies correlated to microbiological contamination. High BOD concentrations reduce oxygen availability, degrade aquatic habitats and biodiversity, and impair water use. High BOD loadings to freshwater systems are mainly coming from anthropogenic sources, comprising domestic and livestock waste, industrial emissions, and combined sewer overflows. We developed a conceptual model (GREEN+BOD) to assess mean annual current organic pollution (BOD fluxes) across Europe. The model was informed with the latest available European datasets of domestic and industrial emissions, population and livestock densities. Model parameters were calibrated using 2008-2012 mean annual BOD concentrations measured in 2157 European monitoring stations, and validated with other 1134 stations. The most sensitive model parameters were abatement of BOD by secondary treatment and the BOD decay exponent of travel time. The mean BOD concentrations measured in monitored stations was 2.10 mg O2/L and predicted concentrations were 2.54 mg O2/L; the 90th percentile of monitored BOD concentration was 3.51 mg O2/L while the predicted one was 4.76 mg O2/L. The model could correctly classify reaches for BOD concentrations classes, from high to poor quality, in 69% of cases. High overestimations (incorrect classification by 2 or more classes) were 2% and large underestimations were 5% of cases. Across Europe about 12% of freshwater network was estimated to be failing good quality due to excessive BOD concentrations (>5 mg O2/L). Dominant sources of BOD to freshwaters and seas were point sources and emissions from intensive livestock systems. Comparison with previous assessments confirms a decline of BOD pollution since the introduction of EU legislation regulating water pollution.

Hot bodies protect amphibians against chytrid infection in nature.

Project description:Environmental context strongly affects many host-pathogen interactions, but the underlying causes of these effects at the individual level are usually poorly understood. The amphibian chytrid fungus has caused amphibian population declines and extinctions in many parts of the world. Many amphibian species that have declined or have been extirpated by the pathogen in some environments coexist with it in others. Here we show that in three species of rainforest frogs in nature, individuals' probability of infection by the amphibian chytrid fungus was strongly related to their thermal history. Individuals' probability of infection declined rapidly as they spent more time above the pathogen's upper optimum temperature. This relationship can explain population-level patterns of prevalence in nature, and suggests that natural or artificial selection for higher thermal preferences could reduce susceptibility to this pathogen. Similar individual-level insights could improve our understanding of environmental context-dependence in other diseases.

Student satisfaction in the Russell Group and Non-Russell Group Universities in UK.

Project description:Undeniably, student satisfaction in higher education is one of the important key factors for university ranking and league table. Accordingly, this article describes the student satisfaction data between Russell Group and Non-Russell Group universities in United Kingdom (UK). The data includes 19 Russell Group and 102 Non-Russell Group universities. We present some descriptive statistics of the variables included in the dataset. The results of the article are based on the two-Sample t-Test and CI. The findings from the data suggest that there is no statistically significant difference on student satisfaction rating between Russell Group and Non-Russell Group universities. This study has policy implications for the higher education in the UK.

Distribution and Clinicopathological Features of Mott Cells (Plasma Cells Containing Russell Bodies) in Gastric Cancer: Presence of Mott Cells Is Associated with Favorable Prognosis.

Project description:Gastric cancer (GC) is still one of the leading causes of cancer-related mortality. We previously reported the relationship between histological heterogeneity of tumor cells and molecular features in GC. The tumor microenvironment also has a crucial role in GC progression and therapeutic resistance. In this study, we focused on the tumor microenvironment, especially inflammatory cells in GC. Using GC tissue slides, we investigated the distribution and clinicopathological significance of inflammatory cell counts including eosinophils, neutrophils, lymphocytes, and plasma cells. Additionally, we investigated the relationship between Mott cells (plasma cells containing Russell bodies) and clinicopathological features. In neoplastic gastric mucosa, a high number of plasma cells was associated with low T-grade, early stage, and good prognosis. We then focused on Mott cells and found that their presence in neoplastic gastric mucosa was associated with lower T and N grades, early stage, and Helicobacter pylori infection and was inversely associated with CD44 and EGFR expression. Additionally, the presence of Mott cells was associated with good prognosis in advanced GC and was an independent favorable prognostic predictor. The presence of Mott cells in GC might be one useful prognostic predictor, and Mott cells might have an important role in the carcinogenesis of H. pylori infection.
