Brightening gold nanoparticles: new sensing approach based on plasmon resonance energy transfer.

ABSTRACT: Scattering recovered plasmonic resonance energy transfer (SR-PRET) was reported by blocking the plasmon resonance energy transfer (PRET) from gold nanoparticle (GNP) to the adsorbed molecules (RdBS). Due to the selective cleavage of the Si-O bond by F- ions, the quenching is switched off causing an increase in the brightness of the GNPs,detected using dark-field microscopy (DFM) were brightened. This method was successfully applied to the determination of fluoride ions in water. The SR-PRET provides a potential approach for a vitro/vivo sensing with high sensitivity and selectivity.

Lighting up ultraviolet fluorescence from chicken albumen through plasmon resonance energy transfer of gold nanoparticles.

Project description:Au nanoparticles (AuNPs), which easily aggregate in organic thin film, are observed to well-disperse in chicken albumen thin films. The incorporated AuNPs is distributed uniformly inside the thin film and is as dense as 650 (particles/μm(3)). In addition, enhanced ultraviolet (UV) fluorescence centered at 350 nm is observed from the AuNPs-containing chicken albumen thin film. The enhancement is proposed to be originated from the plasmon resonance energy transfer (PRET) from the d-band absorption of AuNPs to the chicken albumen protein. The enhanced fluorescence is further verified by the shorter fluorescence lifetime from the time-resolved fluorescence spectra. These results indicate that d-band transition of AuNPs can be used to interface with other UV-emitting biomolecules. Results in this study demonstrate that AuNPs exhibit future potentials in applications for both the organic thin-film technology and nano-biotechnology.

Forster Resonance Energy Transfer-Mediated Globular Protein Sensing Using Polyelectrolyte Complex Nanoparticles.

Project description:Polyelectrolyte complex nanoparticles (PEC NPs) are synthesized using two oppositely charged polyelectrolytes, i.e., anionic poly(sodium 4-styrene sulfonate) (PSS) and cationic poly(diallyldimethylammoniumchloride), at molar mixing ratios (n -/n +) of ≈0.4, 0.67, 0.75, and 1.5 by applying consecutive centrifugation to modify the optical property of PSS. However, for n -/n + ≈ 0.75, PEC NPs exhibit a larger blue shift and a specific emission peak occurs at ≈278 nm for the 225 nm excitation. The mechanism of such modification of PSS emission after complex formation is proposed. This specific emission by PEC NPs nearly matches with the optical absorption wavelength of globular proteins. The emission intensity of PEC NPs is therefore quenched in the presence of globular proteins (bovine serum albumin, human serum albumin, lysozyme, and hemoglobin) through resonance energy transfer between the donor (PEC NPs) and acceptor (globular proteins). The spectral overlap integral and the variation of the separation distance from 1.8 to 2.5 nm between the donor and acceptor confirm the resonance energy transfer. Sensing of proteins by the PEC NPs is possible within the detection limit of 5 nM and therefore such PEC NPs can be used as an efficient and promising protein sensing material.

Multispectral Localized Surface Plasmon Resonance (msLSPR) Reveals and Overcomes Spectral and Sensing Heterogeneities of Single Gold Nanoparticles.

Project description:Metal nanoparticles can be sensitive molecular sensors due to enhanced absorption and scattering of light near a localized surface plasmon resonance (LSPR). Variations in both intrinsic properties such as the geometry and extrinsic properties such as the environment can cause heterogeneity in nanoparticle LSPR and impact the overall sensing responses. To date, however, few studies have examined LSPR and sensing heterogeneities, due to technical challenges in obtaining the full LSPR spectra of individual nanoparticles in dynamic assays. Here, we report multispectral LSPR (msLSPR), a wide-field imaging technique for real-time spectral monitoring of light scattering from individual nanoparticles across the whole field of view (FOV) at ∼0.5 nm spectral and ∼100 ms temporal resolutions. Using msLSPR, we studied the spectral and sensing properties of gold nanoparticles commonly used in LSPR assays, including spheres, rods, and bipyramids. Complemented with electron microscopy imaging, msLSPR analysis revealed that all classes of gold nanoparticles exhibited variations in LSPR peak wavelengths that largely paralleled variations in morphology. Compared with the rods and spheres, gold nanobipyramids exhibited both more uniform and stronger sensing responses as long as the bipyramids are structurally intact. Simulations incorporating the experimental LSPR properties demonstrate the negative impact of spectral heterogeneity on the overall performance of conventional, intensity-based LSPR assays and the ability of msLSPR in overcoming both particle heterogeneity and measurement noise. These results highlight the importance of spectral heterogeneity in LSPR-based sensors and the potential advantage of performing LSPR assays in the spectral domain.

Plasmonic Gold Decorated MWCNT Nanocomposite for Localized Plasmon Resonance Sensing.

Project description:The synergism of excellent properties of carbon nanotubes and gold nanoparticles is used in this work for bio-sensing of recombinant bovine growth hormones (rbST) by making Multi Wall Carbon Nanotubes (MWCNT) locally optically responsive by augmenting it optical properties through Localized Surface Plasmon Resonance (LSPR). To this purpose, locally gold nano particles decorated gold-MWCNT composite was synthesized from a suspension of MWCNT bundles and hydrogen chloroauric acid in an aqueous solution, activated ultrasonically and, then, drop-casted on a glass substrate. The slow drying of the drop produces a "coffee ring" pattern that is found to contain gold-MWCNT nanocomposites, accumulated mostly along the perimeter of the ring. The reaction is studied also at low-temperature, in the vacuum chamber of the Scanning Electron Microscope and is accounted for by the local melting processes that facilitate the contact between the bundle of tubes and the gold ions. Biosensing applications of the gold-MWCNT nanocomposite using their LSPR properties are demonstrated for the plasmonic detection of traces of bovine growth hormone. The sensitivity of the hybrid platform which is found to be 1 ng/ml is much better than that measuring with gold nanoparticles alone which is only 25 ng/ml.

Surface Plasmon Resonance-Based Immunosensor for Igm Detection with Gold Nanoparticles.

Project description:In this work, a surface plasmon resonance (SPR) based immunosensor was prepared by the immobilization of the amine-functionalized gold nanoparticles (N-AuNPs) on the sensing surface to sense immunoglobulin M (IgM) antibodies in the aqueous solution and artificial plasma. The characterization studies of SPR based immunosensor for IgM detection were performed with scanning electron microscope (SEM), contact angle measurements, and ellipsometry. Kinetic studies for the IgM immunosensor were carried out in the range of 1.0 to 200 ng/mL IgM concentrations in an aqueous solution. The total IgM analysis time including adsorption, desorption, and regeneration cycles was nearly 10 min for the prepared immunosensor. The limit of detection (LOD) and limit of quantification (LOQ) were found as 0.08 and 0.26 ng/mL, respectively. The reusability of the proposed immunosensor was tested with 6 consecutive adsorption-desorption, and regeneration cycles. Also, enzyme-linked immunosorbent assay (ELISA) method was utilized in the validation of the immunosensor.

Long-Range and High-Efficiency Plasmon-Assisted Forster Resonance Energy Transfer.

Project description:The development of a long-range and efficient Förster resonance energy transfer (FRET) process is essential for its application in key enabling optoelectronic and sensing technologies. Via controlling the delocalization of the donor's electric field and Purcell enhancements, we experimentally demonstrate long-range and high-efficiency Förster resonance energy transfer using a plasmonic nanogap formed between a silver nanoparticle and an extended silver film. Our measurements show that the FRET range can be extended to over 200 nm while keeping the FRET efficiency over 0.38, achieving an efficiency enhancement factor of ∼108 with respect to a homogeneous environment. Reducing Purcell enhancements by removing the extended silver film increases the FRET efficiency to 0.55, at the expense of the FRET rate. We support our experimental findings with numerical calculations based on three-dimensional finite difference time-domain calculations and treat the donor and acceptor as classical dipoles. Our enhanced FRET range and efficiency structures provide a powerful strategy to develop novel optoelectronic devices and long-range FRET imaging and sensing systems.

Molecular-Fluorescence Enhancement via Blue-Shifted Plasmon-Induced Resonance Energy Transfer.

Project description:We report molecular-fluorescence enhancement via the blue-shifted plasmon-induced resonance energy transfer (PIRET) from single Au nanorods (AuNRs) to merocyanine (MC) dye molecules. The blue-shifted PIRET occurs when there is a proper spectral overlap between the scattering of AuNRs and the absorption of MC molecules. Along with the quenching of scattering from AuNRs, the blue-shifted PIRET enhances the fluorescence of nearby molecules. On the basis of the fluorescence enhancement, we conclude that AuNRs can be used as donors with clear advantages to excite the fluorescence of molecules as acceptors in AuNR-molecule hybrids. On the one hand, compared to conventional molecular donors in Förster resonance energy transfer (FRET), AuNRs have much larger absorption cross sections at the plasmon resonance frequencies. On the other hand, energy-transfer efficiency of PIRET decreases at a lower rate than that of FRET when the donor-acceptor distance is increased. Besides, the blue-shifted PIRET allows excitation with incident light of lower energy than the acceptor's absorption, which is difficult to achieve in FRET because of the Stokes shift. With the capability of enhancing molecular fluorescence with excitation light of low intensity and long wavelength, the blue-shifted PIRET will expand the applications of nanoparticle- molecule hybrids in biosensing and bioimaging by increasing signal-to-noise ratio and by reducing photodamage to biological cells and organelles at the targeted areas.

Nano Sensing and Energy Conversion Using Surface Plasmon Resonance (SPR).

Project description:Nanophotonic technique has been attracting much attention in applications of nano-bio-chemical sensing and energy conversion of solar energy harvesting and enhanced energy transfer. One approach for nano-bio-chemical sensing is surface plasmon resonance (SPR) imaging, which can detect the material properties, such as density, ion concentration, temperature, and effective refractive index in high sensitivity, label-free, and real-time under ambient conditions. Recent study shows that SPR can successfully detect the concentration variation of nanofluids during evaporation-induced self-assembly process. Spoof surface plasmon resonance based on multilayer metallo-dielectric hyperbolic metamaterials demonstrate SPR dispersion control, which can be combined with SPR imaging, to characterize high refractive index materials because of its exotic optical properties. Furthermore, nano-biophotonics could enable innovative energy conversion such as the increase of absorption and emission efficiency and the perfect absorption. Localized SPR using metal nanoparticles show highly enhanced absorption in solar energy harvesting. Three-dimensional hyperbolic metamaterial cavity nanostructure shows enhanced spontaneous emission. Recently ultrathin film perfect absorber is demonstrated with the film thickness is as low as ~1/50th of the operating wavelength using epsilon-near-zero (ENZ) phenomena at the wavelength close to SPR. It is expected to provide a breakthrough in sensing and energy conversion applications using the exotic optical properties based on the nanophotonic technique.

Thermal deformation of gold nanostructures and its influence on surface plasmon resonance sensing.

Project description:Plasmonic nanostructures have been recently used in elevated temperature applications such as sensing of high-energy systems and localized heat generation for heat-assisted magnetic recording, thermophotovaltaics, and photothermal therapy. However, plasmonic nanostructures exposed to elevated temperature often experience permanent deformations, which could significantly degrade performance of the plasmonic devices. Therefore, understanding of thermal deformation of plasmonic nanostructures and its influence on the device performance is essential to the development of robust high-performance plasmonic devices. Here, we report thermal deformation of lithographic planar gold nanopatch and nanohole arrays and its influence on surface plasmon resonance sensing. The gold nanostructures are fabricated on a silicon substrate and on the end-face of an optical fiber using electron-beam lithography and focused-ion-beam lithography, respectively. The fabricated gold nanostructures are exposed to cyclic thermal loading in the range of 25 °C to 500 °C. Through experimental and numerical studies, we investigate (i) thermal deformation modes of the gold nanostructures, (ii) influence of the gold nanostructure geometry on the degree and mechanism of the thermal deformation, and (iii) influence of the thermal deformation on performance of surface plasmon resonance sensing. The obtained understanding from these studies is expected to help guide the development of robust high-performance plasmonic sensors for monitoring in elevated temperature environments. Although the current work is focused on gold nanostructures, it can be extended to provide useful insights on thermal deformation of refractory plasmonic nanostructures at extreme temperature.

Highly Sensitive Detection of Melamine Based on the Fluorescence Resonance Energy Transfer between Conjugated Polymer Nanoparticles and Gold Nanoparticles.

Project description:Adding melamine as additives in food products will lead to many diseases and even death. However, the present techniques of melamine detection require time-consuming steps, complicated procedures and expensive analytical apparatus. The fluorescent assay method was facile and highly sensitive. In this work, a fluorescence resonance energy transfer (FRET) system for melamine detection was constructed based on conjugated polymer nanoparticles (CPNs) and gold nanoparticles (AuNPs). The energy transfer efficiency is up to 82.1%, and the system is highly selective and sensitive to melamine detection with a lower detection limit of 1.7 nmol/L. Moreover, the interaction mechanism was explored. The results showed that the fluorescence of CPNs were firstly quenched by AuNPs, and then restored after adding melamine because of reducing FRET between CPNs and AuNPs. Lastly, the proposed method was carried out for melamine detection in powdered infant formula with satisfactory results.
