First Description of Infection of Caprine Herpesvirus 1 (CpHV-1) in Goats in Mainland France.

ABSTRACT: The purpose of this study was to investigate the epidemiological situation of the caprine herpesvirus 1 (CpHV-1) infection in nine districts in mainland France, mostly in the south, near Italy or Spain, where high seroprevalence has been observed. Two more central areas were also included in the study. The serosurvey was carried out in 9564 goats (275 herds) using bovine herpesvirus 1 (BoHV-1) glycoprotein B and E ELISAs. To confirm the presence of specific CpHV-1 antibodies, some of the samples were tested in neutralization assay. Results demonstrate, for the first time, CpHV-1 infection in goat herds on the French mainland. The analysis found cases of alphaherpesviruses infection in each district studied, with different levels of seroprevalence observed within each district (ranging from 0.2% to 31.56% at an individual level and from 9% to 46.2% for herd seroprevalence). Moreover, in the Alpes-Maritimes district, the seroprevalence seemed to be higher in older goats (79.45% of animals 6 years old or more) than in younger animals (40.99% of one-year-olds). This result suggests frequent virus re-excretion and circulation in herds. Results analysis also shows that the seroprevalence was higher when the herd size increased. In addition, the first French CpHV-1 strain was isolated from nasal swabs taken on an infected goat. The data reported herein demonstrate that CpHV-1 circulates in mainland France, which should henceforth be taken into consideration in cases of unexplained abortion in goats.

Project description:BackgroundBiting midges of the genus Culicoides Latreille (Diptera: Ceratopogonidae) are involved in the transmission of several viruses affecting humans and livestock, particularly bluetongue (BTV). Over the last decade, Culicoides surveillance has been conducted discontinuously and at various temporal and spatial scales in mainland France following the BTV epizootics in 2008-2009 and its reemergence and continuous circulation since 2015. The ability to predict seasonal dynamics and spatial abundance of Culicoides spp. is a key element in identifying periods and areas at high risk of transmission in order to strengthen surveillance for early detection and to establish seasonally disease-free zones. The objective of this study was to model the abundance of Culicoides spp. using surveillance data.MethodsA mixed-effect Poisson model, adjusted for overdispersion and taking into account temperature data at each trap location, was used to model the weekly relative abundance of Culicoides spp. over a year in 24 vector zones, based on surveillance data collected during 2009-2012. Vector zones are the spatial units used for Culicoides surveillance since 2016 in mainland France.ResultsThe curves of the predicted annual abundance of Culicoides spp. in vector zones showed three different shapes: unimodal, bimodal or plateau, reflecting the temporal variability of the observed counts between zones. For each vector zone, the model enabled to identify periods of vector activity ranging from 25 to 51 weeks.ConclusionsAlthough the data were collected for surveillance purposes, our modeling approach integrating vector data with daily temperatures, which are known to be major drivers of Culicoides spp. activity, provided areas-specific predictions of Culicoides spp. abundance. Our findings provide decisions makers with essential information to identify risk periods in each vector zone and guide the allocation of resources for surveillance and control. Knowledge of Culicoides spp. dynamics is also of primary importance for modeling the risk of establishment and spread of midge-borne diseases in mainland France.

Project description:A total of 232 goat fecal samples (124 diarrheic and 108 nondiarrheic) collected from 12 farms in Southwest China were tested for astrovirus using RT-PCR. A total of 16.9% (21/124) of diarrheic and 20.4% (22/108) of nondiarrheic samples were astrovirus-positive, and no statistical difference was found in the detection rate between healthy and sick goats. Furthermore, 28 obtained complete ORF2 sequences could be classified into six genotypes according to the species classification criteria of the International Committee on Taxonomy of Viruses (ICTV). It is worth noting that, in addition to four known caprine astrovirus genotypes (MAstV-33, MAstV-34, Caprine Astrovirus G5.1, and Caprine Astrovirus G3.1), MAstV-13 and MAstV-24 genotypes were identified in goats. Interestingly, five of 19 ORF2 sequences in the Caprine Astrovirus G3.1 genotype showed possible intragenotypic recombination events. Furthermore, nearly complete caprine astrovirus genomes of MAstV-13 and MAstV-24 genotypes were obtained. The genome of the SWUN/ECJK3/2021 strain shared the highest similarity (62.0% to 73.9%) with astrovirus in MAstV-13, and clustered in the so-called human-mink-ovine (HMO) clade, which contained the majority of the neurotropic astrovirus strains. Moreover, the SWUN/LJK2-2/2020 strain showed the highest similarity (69.7% to 78.6%) and the closest genetic relationship to the known porcine and bovine astroviruses in MAstV-24. In conclusion, this study confirmed six genotypes of astrovirus circulating among goats in Southwest China, including MAstV-13 and MAstV-24 genotypes. These findings enhance our knowledge of the prevalence and diversity of astroviruses. IMPORTANCE Caprine astrovirus is a newly emerging virus, and information regarding its prevalence and molecular characteristics remains limited. In this study, six genotypes of astrovirus, including MAstV-13 and MAstV-24, were identified in goats, adding two novel caprine astrovirus genotypes to the four previously known genotypes, thereby enriching the diversity of the caprine astrovirus. Moreover, genomes of MAstV-13 SWUN/ECJK3/2021 and MAstV-24 SWUN/LJK2-2/2020 strains were obtained from goats, which aids in the understanding of the infection spectrum and host range of the two genotypes. This study is the first to demonstrate the presence of neurotropic-like astrovirus (MAstV-13) in goats, which has significant implications for the diagnosis of neurological diseases in goats.
