An illustrated guide to the identification of the known species of Diatraea Guilding (Lepidoptera, Crambidae, Crambinae) based on genitalia.

ABSTRACT: The genus Diatraea Guilding is one of the most economically important groups of moths in the Western Hemisphere. The larvae are stem borers that feed on species of Poaceae, or grasses, such as sugarcane, corn, rice, and sorghum, as well as many other native grasses. Interest in this group has risen considerably since sugarcane and other grasses have been utilized and/or investigated as biofuels. This is the first modern study to treat all 41 valid described species. Most type specimens were examined and we provide a checklist with 19 new synonyms. We provide keys for the identification of most species in this genus based on morphology of the male and female genitalia and modern illustrations of male and female genitalia. We also provide an updated table of species distribution by country.

Identification of Diatraea spp. (Lepidoptera: Crambidae) based on cytochrome oxidase II.

Project description:Diatraea spp. (Lepidoptera: Crambidae) are a group of insects that are agriculture pests in many economically relevant crops such as sugarcane, sorghum, corn and rice. Recognized species for this genus respond differentially to natural enemies used in their biological control, emphasizing the importance of species in a regional approach. Currently, identification is based on the male genitalia. However, the availability of specimens collected from field and subjectivity based on the character recognition can seriously hamper species identification, and therefore result in inadequate pest management. To overcome this, individuals of Diatraea spp. preliminarily classified male genitalia and obtained from reared conditions and the field (both derived from natural populations occurring in Colombia) were analyzed using genitalic morphometry and molecular biology specifically using a fragment of the cytochrome oxidase subunit II (CO II) mitochondrial gene. Although morphometric analysis did not show any overriding results regarding genitalia morphology, the bioinformatics analyses of CO II sequences resulted in an adequate classification of the individuals within the recognized species. It also, revealed that the occurrence of clades associated with geographical distribution may be associated with cryptic species. The latter was also confirmed by a Single-Strand Conformation Polymorphism (SSCP) methodology evaluating the same fragment of CO II. This experimental approach allows properly recognizing each species and in consequence is proposed as an effective tool in Diatraea species identification.

Morphological and molecular characterization of Brazilian populations of Diatraea saccharalis (Fabricius, 1794) (Lepidoptera: Crambidae) and the evolutionary relationship among species of Diatraea Guilding.

Project description:The sugarcane borer or corn stalk borer, Diatraea Guilding is polyphagous insect pest of many important crops such as corn, sorghum and sugarcane. Losses arising from the attack of Diatraea species have been a serious problem, which may cause loss in sugarcane production around 0.25% in sugar, 0.20% in alcohol and 0.77% of body weight for every 1% infestation and up to 21% in corn production fields. In Brazil, the most commonly reported species are Diatraea saccharalis (Fabricius, 1794) and Diatraea impersonatella (Walker, 1863) (= D. flavipennella). However, multiple other species of Diatraea have been identified in Brazil according to the literature. Currently, little information exists on the presence of the other species causing injury to sugarcane and corn. The objectives of this study were to improve the accuracy of species assignment, evaluate the population genetic structure, and address many of the outstanding questions of systematics and evolution of Brazilian populations of D. saccharalis. To address these main questions, classical taxonomic methods were used, focused on morphological characterization of the reproductive organs, especially the male genitalia. In addition, genetic studies were performed using simple sequence repeats (SSR) and a fragment of cytochrome C oxidase subunit I (COI) gene. The data and findings from this research will contribute to the understanding of evolutionary aspects of insect pests in order to develop more effective and sustainable population management practices.

Within-Host Competition between Two Entomopathogenic Fungi and a Granulovirus in Diatraea saccharalis (Lepidoptera: Crambidae).

Project description:We provide insights into how the interactions of two entomopathogenic fungi and a virus play a role in virulence, disease development, and pathogen reproduction for an economically important insect crop pest, the sugarcane borer Diatraea saccharalis (Fabricius) (Lepidoptera: Crambidae). In our model system, we highlight the antagonistic effects of the co-inoculation of Beauveria bassiana and granulovirus (DisaGV) on virulence, compared to their single counterparts. By contrast, combinations of Metarhizium anisopliae and B. bassiana, or M. anisopliae and DisaGV, have resulted in additive effects against the insect. Intriguingly, most cadavers that were derived from dual or triple infections, produced signs/symptoms of only one species after the death of the infected host. In the combination of fungi and DisaGV, there was a trend where a higher proportion of viral infection bearing conspicuous symptoms occurred, except when the larvae were inoculated with M. anisopliae and DisaGV at the two highest inoculum rates. Co-infections with B. bassiana and M. anisopliae did not affect pathogen reproduction, since the sporulation from co-inoculated larvae did not differ from their single counterparts.

Geographic population structure of the sugarcane borer, Diatraea saccharalis (F.) (Lepidoptera: Crambidae), in the southern United States.

Project description:The sugarcane borer moth, Diatraea saccharalis, is widespread throughout the Western Hemisphere, and is considered an introduced species in the southern United States. Although this moth has a wide distribution and is a pest of many crop plants including sugarcane, corn, sorghum and rice, it is considered one species. The objective was to investigate whether more than one introduction of D. saccharalis had occurred in the southern United States and whether any cryptic species were present. We field collected D. saccharalis in Texas, Louisiana and Florida in the southern United States. Two molecular markers, AFLPs and mitochondrial COI, were used to examine genetic variation among these regional populations and to compare the sequences with those available in GenBank and BOLD. We found geographic population structure in the southern United States which suggests two introductions and the presence of a previously unknown cryptic species. Management of D. saccharalis would likely benefit from further investigation of population genetics throughout the range of this species.

Host-plant associated genetic divergence of two <i>Diatraea</i> spp. (Lepidoptera: Crambidae) stemborers on novel crop plants.

Project description:Diatraea lineolata and Diatraea saccharalis (Lepidoptera: Crambidae) are moths with stemboring larvae that feed and develop on economically important grasses. This study investigated whether these moths have diverged from a native host plant, corn, onto introduced crop plants including sorghum, sugarcane, and rice. Diatraea larvae were collected from these four host plants throughout the year in El Salvador and were reared on artificial diet until moths or parasitoids emerged. Adult moths were subsequently identified to species. Amplified fragment length polymorphisms (AFLPs) and mitochondrial DNA cytochrome oxidase I (COI) were used to examine whether or not there was genetic divergence of D. lineolata or D. saccharalis populations on the four host plants. Percent parasitism was also determined for each moth on its host plants. D. lineolata was collected from corn in the rainy season and sorghum in the dry season. D. saccharalis was most abundant on sugarcane in the rainy season and sorghum in the dry season. The AFLP analysis found two genetically divergent populations of both D. lineolata and D. saccharalis. Both moths had high levels of parasitism on their dominant host plant in the rainy season, yet had low levels of parasitism on sorghum in the dry season. The presence of two genotypes of both Diatraea spp. on sorghum suggest that host-associated differentiation is occurring on this novel introduced crop plant.

Development of a bioassay method to test activity of cry insecticidal proteins against Diatraea spp. (Lepidoptera: Crambidae) sugarcane stem borers.

Project description:The genus Diatraea (Lepidoptera: Crambidae) includes stem borers representing the most critical sugarcane pests in the Americas. Colombia's most widely distributed and damaging Diatraea species include Diatraea saccharalis, D. indigenella, D. busckella, and D. tabernella. The reduced efficacy of biological tools commonly used in controlling several species highlights the importance of evaluating alternative management strategies, such as transgenic plants expressing insecticidal proteins from the bacterium Bacillus thuringiensis (Bt). The selection of optimal Bt insecticidal proteins for Diatraea control depends on bioassays with purified Bt proteins. Because there is no described artificial diet for borer species other than D. saccharalis and availability of most purified Bt toxins is restricted, this study aimed at developing a bioassay method using fresh corn tissue and providing proof of concept by testing susceptibility to the Cry1Ac insecticidal protein from Bt. Toxicity was evaluated with a single Cry1Ac dose applied directly to corn discs. Stem borer mortality after seven days was higher than 90% for all four tested Diatraea species, while control mortality was below 8%. In addition, we observed that Cry1Ac caused more than 90% weight inhibition in all survivors and delayed development. These results validate the use of this method to determine mortality and growth inhibition due to the consumption of the Cry1Ac protein in each of the Diatraea species. Furthermore, this method could be used to assess other entomopathogenic substances to control these insect pests.

Project description:Herpetogramma sphingealissp. n., a new species of Crambidae (Lepidoptera), is described from Québec, Canada. The species is included in the genus Herpetogramma Led., 1863, a genus in the subfamily Spilomelinae. Adults and genitalia of this species are described and illustrated, as well as those of Herpetogramma aeglealis (Walker, 1859) and Herpetogramma thestealis (Walker, 1859), and adults of the semi-melanic form of Herpetogramma aeglealis are illustrated.

A new species of Patania from the Hainan Island, China (Lepidoptera, Crambidae).

Project description:Patania clava sp. n. is described from the Diaoluo Mountain of Hainan Island, China. The new species is distinguished from its most similar congener, Patania iopasalis (Walker, 1859), by the following features: wingspan 33.0-35.0 mm (vs. 21.0-30.0 mm), ventral cilia of the male antenna as long as the diameter of flagellomere (vs. 1/4), the thick finger-like gnathos (vs. the short broad sheet-like), a long thick needle-like cornutus stretching out from (vs. embedded in) a cluster of spicular cornuti near apex. Images of adult and genitalia of the new species are provided.
