Project description:We can regard the wider incentive structures that operate across science, such as the priority given to novel findings, as an ecosystem within which scientists strive to maximise their fitness (i.e., publication record and career success). Here, we develop an optimality model that predicts the most rational research strategy, in terms of the proportion of research effort spent on seeking novel results rather than on confirmatory studies, and the amount of research effort per exploratory study. We show that, for parameter values derived from the scientific literature, researchers acting to maximise their fitness should spend most of their effort seeking novel results and conduct small studies that have only 10%-40% statistical power. As a result, half of the studies they publish will report erroneous conclusions. Current incentive structures are in conflict with maximising the scientific value of research; we suggest ways that the scientific ecosystem could be improved.

Project description:More than 110 000 publications have used microarrays to decipher phenotype-associated genes, clinical biomarkers and gene functions. Microarrays rely on digital assaying the fluorescence signals of arrays. In this study, we retrospectively constructed raw images for 37 724 published microarray data, and developed deep learning algorithms to automatically detect systematic defects. We report that an alarming amount of 26.73% of the microarray-based studies are affected by serious imaging defects. By literature mining, we found that publications associated with these affected microarrays have reported disproportionately more biological discoveries on the genes in the contaminated areas compared to other genes. 28.82% of the gene-level conclusions reported in these publications were based on measurements falling into the contaminated area, indicating severe, systematic problems caused by such contaminations. We provided the identified published, problematic datasets, affected genes and the imputed arrays as well as software tools for scanning such contamination that will become essential to future studies to scrutinize and critically analyze microarray data.

Project description:Grifols' recent Alzheimer Management by Albumin Replacement ("AMBAR") study investigated the effects of plasmapheresis with albumin replacement, plus intravenous immunoglobulin (IVIG) in some subjects, in patients with mild-to-moderate Alzheimer's disease (AD). AMBAR was a phase IIb trial in the United States and a phase III trial in Europe. There were three treatment groups (plasmapheresis with albumin replacement; plasmapheresis with low dose albumin and IVIG; plasmapheresis with high dose albumin and IVIG) and sham-treated controls. Disease progression in pooled treated patients was 66% less than control subjects based on ADAS-Cog scores (p = 0.06) and 52% less based on ADCS-ADL scores (p = 0.03). Moderate AD patients had 61% less progression, based on both ADAS-Cog and ADCS-ADL scores, than their sham-treated counterparts (p-values 0.05 and 0.002), and their CDR-Sb scores declined 53% less than their sham-treated counterparts. However, ADAS-Cog and ADCS-ADL scores were not significantly different between actively-treated and sham-treated mild AD patients, although CDR-Sb scores improved vs. baseline for treated mild AD patients. Patients administered both IVIG and albumin had less reduction in brain glucose metabolism than sham-treated patients. Questions raised by these findings include: what mechanism(s) contributed to slowing of disease progression? Is this approach as effective in mild AD as in moderate AD? Must IVIG be included in the protocol? Does age, sex, or ApoE genotype influence treatment response? Does the protocol increase the risk for amyloid-related imaging abnormalities? How long does disease progression remain slowed post-treatment? A further study should allow this approach to be optimized.

Project description:This study used an advertising campaign focused on the Real ID card to explore the relationship between privacy concerns and information sharing through adoption of the new form of identification. Online surveys were conducted to assess the impact of campaign ads promoting the Real ID, and to determine whether privacy concerns were a barrier to its adoption. At post-test, an ordinal regression analysis of data from registered voters in Montana (n = 302) showed a positive association between the time spent viewing the ads on social media and intent to obtain the Real ID, but no relationship between privacy concerns and Real ID intentions. To date, the states and territories have issued approximately 110 million Real ID-compliant driver’s licenses and identification cards, representing 40 percent of all driver’s licenses and identification card holders. Montana, one of the states originally most resistant to the 2005 security act, lags only slightly behind with 20 percent of drivers and ID card-holders having Real IDs. This research shows that privacy concerns about a new technology or identification system, such as the Real ID, can erode over time. Whether these changes are due to mounting privacy apathy triggered by the public’s perceived lack of control, or a willingness to sacrifice some privacy for national security in the face of rising terrorist threats needs to be researched further. Project description:ObjectivesTo provide a participant's account of the development of a paper commissioned by the tobacco industry examining the reliability of self reported smoking status; to redress the distorted report of this Japanese spousal smoking study which evaluated the reliability and validity of self reported smoking status, and estimated confounding by diet and lifestyle factors.DesignRepeated interviews on smoking status and its verification by environmental and biological markers for environmental tobacco smoke (ETS) exposure.SettingUrban wives in Osaka City and Sizuoka City, JapanParticipantsSemi-random sampling of 200 wives in each city. From the Osaka subjects, 100 non-smoking wives were selected for the validity study.Main outcome measuresKappa coefficient for reliability of self reported smoking status. Correlation coefficients between environmental nicotine concentration, cotinine in saliva and urine, and self reported smoking status.ResultsThe kappa coefficient for the repeated interview was high suggesting sufficient reliability of the response. The proportion of self reported current smokers misclassified as non-smokers was equivalent to the misclassified self reported non-smokers. Ambient concentration of nicotine and personal exposure to nicotine correlated with each other and also with salivary cotinine and self reported ETS exposure but not with urinary cotinine/creatinine ratio (CCR). There was no major difference in diet and lifestyle related to husband's smoking status.ConclusionSelf reported smoking status by Japanese wives shows high reliability. It also shows high validity when verified by both nicotine exposure and salivary cotinine, but not by CCR. A previous report questioning the credibility of self reported smoking status, based on questionable CCR, could thus be of dubious validity. In addition, possible dietary and lifestyle confounding factors associated with smoking husbands were not demonstrable, a finding not reported previously. Using all the data from this project changes the conclusion of the previous published report. In addition to the distortion of scientific findings by a tobacco industry affiliated researcher, anti-smoking campaigners made attempts to intimidate and suppress scientific activities. These distortions of science should be counteracted.

Project description:Since the early nineties countless publications have reported promising medicinal applications for [60]fullerene (C60) related to its unparalleled affinity towards free radicals. Yet, until now no officially approved C60-based drug has reached the market, notably because of the alleged dangers of C60. Nevertheless, since the publication of the effects of C60 on the lifespan of rodents, a myriad of companies started selling C60 worldwide for human consumption without any approved clinical trial. Nowadays, several independent teams have confirmed the safety of pure C60 while demonstrating that previously observed toxicity was due to impurities present in the used samples. However, a purity criterion for C60 samples is still lacking and there are no regulatory recommendations on this subject. In order to avoid a public health issue and for regulatory considerations, a quality-testing strategy is urgently needed. Here we have evaluated several analytical tools to verify the purity of commercially available C60 samples. Our data clearly show that differential scanning calorimetry is the best candidate to establish a purity criterion based on the sc-fcc transition of a C60 sample (Tonset ≥ 258 K, ∆sc-fccH ≥ 8 J g-1).

Project description:Due to their unique physicochemical properties, graphene-based nanoparticles (GPNs) constitute one of the most promising types of nanomaterials used in biomedical research. GPNs have been used as polymeric conduits for nerve regeneration and carriers for targeted drug delivery and in the treatment of cancer via photothermal therapy. Moreover, they have been used as tracers to study the distribution of bioactive compounds used in healthcare. Due to their extensive use, GPN released into the environment would probably pose a threat to living organisms and ultimately to human health. Their accumulation in the aquatic environment creates problems to aquatic habitats as well as to food chains. Until now the potential toxic effects of GPN are not properly understood. Despite agglomeration and long persistence in the environment, GPNs are able to cross the cellular barriers successfully, entered into the cells, and are able to interact with almost all the cellular sites including the plasma membrane, cytoplasmic organelles, and nucleus. Their interaction with DNA creates more potential threats to both the genome and epigenome. In this brief review, we focused on fish, mainly zebrafish (Danio rerio), as a potential target animal of GPN toxicity in the aquatic ecosystem.

Project description:Functionalized fullerene is one of the most advantageous nanotechnologies to develop novel materials for potential biomedical applications. In this study, we applied the ONIOM-GD3 approach to explore the nucleophilic addition reaction mechanism between polyaniline (emeraldine and leucoemeraldine forms) and fullerene. Potential energy surfaces were also analyzed to predict the predominantly formed products of the functionalized reaction. The themoparameters, such as bond dissociation enthalpy (BDE), ionization energy (IE), and electron affinity (EA), characterized by two mechanisms HAT and SET, were used to evaluate the antioxidant activities of the selected compounds. Moreover, the calculated HOMO, LUMO, and DOS results indicate that the electronic structures of polyaniline-fullerene were significantly affected by the presence of fullerene. The computational results show that C60-L1 seems to be the best antioxidant following the SET mechanism.

Project description:This study aims to implement methods for toxicological profiling of engineered nanomaterials using toxicogenomics, proteomics, and metabolomics along with computational analyses. For all three omics layers the human cell lines A549 (lung epithelial cells) and THP1 (monocytes) were separately exposed to the nanomaterials TiO2 NM104, MWNCT NM401, and Ag NM300k. Proteomics and metabolomics samples have been performed as biological triplicates and were taken after 0, 6, 12, 24, and 48 hours. To assess ecotoxic effects C. elegans worms were grown in soil treated with NM300k. Ecotox samples were taken only at 0 and 24 hours. Integrating all three omics layers will enable the identification of (novel) ENM specific modes of action (MoA).

Project description:The effect of utilizing a pure cis-α-dimethoxy carbonyl fulleropyrrolidine C70 (DMEC70) isomer as the electron transporting material (ETM) in inverted perovskite solar cells (PSCs) was evaluated. The as-prepared C70 mono-adduct products are mixtures of regioisomers and the interest was to evaluate them independently as ETMs. Three different cis-DMEC70 isomers (α, β-endo and β-exo) (mix-DMEC70) were synthesized and purified by HPLC. It was found that PSCs based on the pure α-DMEC70 exhibit a substantially enhanced maximum power conversion efficiency (PCE) of 18.6% as compared to devices based on the mixed-DMEC70 isomers that yielded a PCE of 16.4%. A maximum PCE of 15.7% was observed for devices based on [6,6]-phenyl-C71-butyric acid methyl ester (PC71BM). This work points out the importance of using pure fullerene derivative isomers as ETMs to reduce the intrinsic energy disorder, which enhances the overall device performance.
