Designing Colloidal Molecules with Microfluidics.

ABSTRACT: The creation of new colloidal materials involves the design of functional building blocks. Here, a microfluidic method for designing building blocks one by one, at high throughput, with a broad range of shapes is introduced. The method exploits a coupling between hydrodynamic interactions and depletion forces that controls the configurational dynamics of droplet clusters traveling in microfluidic channels. Droplet clusters can be solidified in situ with UV. By varying the flow parameters, clusters are prescribed a given size, geometry, chemical and/or magnetic heterogeneities enabling local bonding. Compact structures (chains, triangles, diamonds, tetrahedrons,...) and noncompact structures, such as crosses and T, difficult to obtain with current techniques are produced. Size dispersions are small (2%) and throughputs are high (30 000 h^-1). The work opens a new pathway, based on microfluidics, for designing colloidal building blocks with a potential to enable the creation of new materials.

Designing caps for colloidal Au nanoparticles.

Project description:The plasmonic property of a nanostructure is highly dependent on its morphology, but there are few methods for appending a domain as the "functional group" or modifier. As a means of modulating plasmonic properties, we create and modulate Au hats on Au nanoparticles, including mortarboards, beret hats, helmets, crowns, antler hats and antenna hats. The structural control arises from the active surface growth as a result of dynamic competition between ligand absorption and metal deposition. It allows the continuous tuning of hat morphologies, from the facet-controlled growth of mortarboards, to the spreading-favored growth of beret hats and helmets, and to the vertical growth of pillars in crowns, antler hats and antenna hats. Among these plasmonic nanostructures, the mortarboards show excellent SERS enhancement of 8.1 × 105, which is among the best in colloidal nanostructures; and the antler hats show the photothermal conversion efficiency of 66.2%, which compares favorably with the literature reports.

Designing Splicing Digital Microfluidics Chips Based on Polytetrafluoroethylene Membrane.

Project description:As a laboratory-on-a-chip application tool, digital microfluidics (DMF) technology is widely used in DNA-based applications, clinical diagnosis, chemical synthesis, and other fields. Additional components (such as heaters, centrifuges, mixers, etc.) are required in practical applications on DMF devices. In this paper, a DMF chip interconnection method based on electrowetting-on-dielectric (EWOD) was proposed. An open modified slippery liquid-infused porous surface (SLIPS) membrane was used as the dielectric-hydrophobic layer material, which consisted of polytetrafluoroethylene (PTFE) membrane and silicone oil. Indium tin oxide (ITO) glass was used to manufacture the DMF chip. In order to test the relationship between the splicing gap and droplet moving, the effect of the different electrodes on/off time on the minimum driving voltage when the droplet crossed a splicing gap was investigated. Then, the effects of splicing gaps of different widths, splicing heights, and electrode misalignments were investigated, respectively. The experimental results showed that a driving voltage of 119 V was required for a droplet to cross a splicing gap width of 300 μm when the droplet volume was 10 μL and the electrode on/off time was 600 ms. At the same time, the droplet could climb a height difference of 150 μm with 145 V, and 141 V was required when the electrode misalignment was 1000 μm. Finally, the minimum voltage was not obviously changed, when the same volume droplet with different aqueous solutions crossed the splicing gap, and the droplet could cross different chip types. These splicing solutions show high potential for simultaneous detection of multiple components in human body fluids.

Self-Assembly Dynamics of Reconfigurable Colloidal Molecules.

Project description:Colloidal molecules are designed to mimic their molecular analogues through their anisotropic shape and interactions. However, current experimental realizations are missing the structural flexibility present in real molecules thereby restricting their use as model systems. We overcome this limitation by assembling reconfigurable colloidal molecules from silica particles functionalized with mobile DNA linkers in high yields. We achieve this by steering the self-assembly pathway toward the formation of finite-sized clusters by employing high number ratios of particles functionalized with complementary DNA strands. The size ratio of the two species of particles provides control over the overall cluster size, i.e., the number of bound particles N, as well as the degree of reconfigurability. The bond flexibility provided by the mobile linkers allows the successful assembly of colloidal clusters with the geometrically expected maximum number of bound particles and shape. We quantitatively examine the self-assembly dynamics of these flexible colloidal molecules by a combination of experiments, agent-based simulations, and an analytical model. Our "flexible colloidal molecules" are exciting building blocks for investigating and exploiting the self-assembly of complex hierarchical structures, photonic crystals, and colloidal metamaterials.

Active colloidal molecules assembled via selective and directional bonds.

Project description:The assembly of active and self-propelled particles is an emerging strategy to create dynamic materials otherwise impossible. However, control of the complex particle interactions remains challenging. Here, we show that various dynamic interactions of active patchy particles can be orchestrated by tuning the particle size, shape, composition, etc. This capability is manifested in establishing dynamic colloidal bonds that are highly selective and directional, which greatly expands the spectrum of colloidal structures and dynamics by assembly. For example, we demonstrate the formation of colloidal molecules with tunable bond angles and orientations. They exhibit controllable propulsion, steering, reconfiguration as well as other dynamic behaviors that collectively reflect the bond properties. The working principle is further extended to the co-assembly of synthetic particles with biological entities including living cells, giving rise to hybrid colloidal molecules of various types, for example, a colloidal carrousel structure. Our strategy should enable active systems to perform sophisticated tasks in future such as selective cell treatment.

Chiral colloidal molecules and observation of the propeller effect.

Project description:Chiral molecules play an important role in biological and chemical processes, but physical effects due to their symmetry-breaking are generally weak. Several physical chiral separation schemes which could potentially be useful, including the propeller effect, have therefore not yet been demonstrated at the molecular scale. However, it has been proposed that complex nonspherical colloidal particles could act as "colloidal molecules" in mesoscopic model systems to permit the visualization of molecular phenomena that are otherwise difficult to observe. Unfortunately, it is difficult to synthesize such colloids because surface minimization generally favors the growth of symmetric particles. Here we demonstrate the production of large numbers of complex colloids with glancing angle physical vapor deposition. We use chiral colloids to demonstrate the Baranova and Zel'dovich (Baranova, N. B.; and Zel'dovich, B. Y. Chem. Phys. Lett. 1978, 57, 435) propeller effect: the separation of a racemic mixture by application of a rotating field that couples to the dipole moment of the enantiomers and screw propels them in opposite directions. The handedness of the colloidal suspensions is monitored with circular differential light scattering. An exact solution for the colloid's propulsion is derived, and comparisons between the colloidal system and the corresponding effect at the molecular scale are made.

Flexible Colloidal Molecules with Directional Bonds and Controlled Flexibility.

Project description:Colloidal molecules are ideal model systems for mimicking real molecules and can serve as versatile building blocks for the bottom-up self-assembly of flexible and smart materials. While most colloidal molecules are rigid objects, the development of colloidal joints has made it possible to endow them with conformational flexibility. However, their unrestricted range of motion does not capture the limited movement and bond directionality that is instead typical of real molecules. In this work, we create flexible colloidal molecules with an in situ controllable motion range and bond directionality by assembling spherical particles onto cubes functionalized with complementary surface-mobile DNA. By varying the sphere-to-cube size ratio, we obtain colloidal molecules with different coordination numbers and find that they feature a constrained range of motion above a critical size ratio. Using theory and simulations, we show that the particle shape together with the multivalent bonds creates an effective free-energy landscape for the motion of the sphere on the surface of the cube. We quantify the confinement of the spheres on the surface of the cube and the probability to change facet. We find that temperature can be used as an extra control parameter to switch in situ between full and constrained flexibility. These flexible colloidal molecules with a temperature switching motion range can be used to investigate the effect of directional yet flexible bonds in determining their self-assembly and phase behavior, and may be employed as constructional units in microrobotics and smart materials.

Programmable colloidal molecules from sequential capillarity-assisted particle assembly.

Project description:The assembly of artificial nanostructured and microstructured materials which display structures and functionalities that mimic nature's complexity requires building blocks with specific and directional interactions, analogous to those displayed at the molecular level. Despite remarkable progress in synthesizing "patchy" particles encoding anisotropic interactions, most current methods are restricted to integrating up to two compositional patches on a single "molecule" and to objects with simple shapes. Currently, decoupling functionality and shape to achieve full compositional and geometrical programmability remains an elusive task. We use sequential capillarity-assisted particle assembly which uniquely fulfills the demands described above. This is a new method based on simple, yet essential, adaptations to the well-known capillary assembly of particles over topographical templates. Tuning the depth of the assembly sites (traps) and the surface tension of moving droplets of colloidal suspensions enables controlled stepwise filling of traps to "synthesize" colloidal molecules. After deposition and mechanical linkage, the colloidal molecules can be dispersed in a solvent. The template's shape solely controls the molecule's geometry, whereas the filling sequence independently determines its composition. No specific surface chemistry is required, and multifunctional molecules with organic and inorganic moieties can be fabricated. We demonstrate the "synthesis" of a library of structures, ranging from dumbbells and triangles to units resembling bar codes, block copolymers, surfactants, and three-dimensional chiral objects. The full programmability of our approach opens up new directions not only for assembling and studying complex materials with single-particle-level control but also for fabricating new microscale devices for sensing, patterning, and delivery applications.

Colloidal quantum dot molecules manifesting quantum coupling at room temperature.

Project description:Coupling of atoms is the basis of chemistry, yielding the beauty and richness of molecules. We utilize semiconductor nanocrystals as artificial atoms to form nanocrystal molecules that are structurally and electronically coupled. CdSe/CdS core/shell nanocrystals are linked to form dimers which are then fused via constrained oriented attachment. The possible nanocrystal facets in which such fusion takes place are analyzed with atomic resolution revealing the distribution of possible crystal fusion scenarios. Coherent coupling and wave-function hybridization are manifested by a redshift of the band gap, in agreement with quantum mechanical simulations. Single nanoparticle spectroscopy unravels the attributes of coupled nanocrystal dimers related to the unique combination of quantum mechanical tunneling and energy transfer mechanisms. This sets the stage for nanocrystal chemistry to yield a diverse selection of coupled nanocrystal molecules constructed from controlled core/shell nanocrystal building blocks. These are of direct relevance for numerous applications in displays, sensing, biological tagging and emerging quantum technologies.

Revealing pseudorotation and ring-opening reactions in colloidal organic molecules.

Project description:Colloids have a rich history of being used as 'big atoms' mimicking real atoms to study crystallization, gelation and the glass transition of condensed matter. Emulating the dynamics of molecules, however, has remained elusive. Recent advances in colloid chemistry allow patchy particles to be synthesized with accurate control over shape, functionality and coordination number. Here, we show that colloidal alkanes, specifically colloidal cyclopentane, assembled from tetrameric patchy particles by critical Casimir forces undergo the same chemical transformations as their atomic counterparts, allowing their dynamics to be studied in real time. We directly observe transitions between chair and twist conformations in colloidal cyclopentane, and we elucidate the interplay of bond bending strain and entropy in the molecular transition states and ring-opening reactions. These results open the door to investigate complex molecular kinetics and molecular reactions in the high-temperature classical limit, in which the colloidal analogue becomes a good model.

Interfacing microfluidics with information-rich detection systems for cells, bioparticles, and molecules.

Project description:The development of elegant and numerous microfluidic manipulations has enabled significant advances in the processing of small volume samples and the detection of minute amounts of biomaterials. Effective isolation of single cells in a defined volume as well as manipulations of complex bioparticle or biomolecule mixtures allows for the utilization of information-rich detection methods including mass spectrometry, electron microscopy imaging, and amplification/sequencing. The art and science of translating biosamples from microfluidic platforms to highly advanced, information-rich detection system is the focus of this review, where we term the translation between the microfluidics elements to the external world "off-chipping." When presented with the challenge of presenting sub-nanoliter volumes of manipulated sample to a detection scheme, several delivery techniques have been developed for effective analysis. These techniques include spraying (electrospray, nano-electrospray, pneumatic), meniscus-defined volumes (droplets, plugs), constrained volumes (narrow channels, containers), and phase changes (deposition, freezing). Each technique has been proven effective in delivering highly defined samples from microfluidic systems to the detection elements. This review organizes and presents selective publications that illustrate the advancements of these delivery techniques with respect to the type of sample analyzed, while introducing each strategy and providing historical perspective. The publications highlighted in this review were chosen due to their significance and relevance in the development of their respective off-chip technique. This review highlights advancements of delivery methods off a microfluidic chip for additional information rich detection schemes.
