A general strategy for the ultrafast surface modification of metals.

ABSTRACT: Surface modification is an essential step in engineering materials that can withstand the increasingly aggressive environments encountered in various modern energy-conversion systems and chemical processing industries. However, most traditional technologies exhibit disadvantages such as slow diffusion kinetics, processing difficulties or compatibility issues. Here, we present a general strategy for the ultrafast surface modification of metals inspired by electromigration, using aluminizing austenitic stainless steel as an example. Our strategy facilitates the rapid formation of a favourable ductile surface layer composed of FeCrAl or β-FeAl within only 10 min compared with several hours in conventional processes. This result indicates that electromigration can be used to achieve the ultrafast surface modification of metals and can overcome the limitations of traditional technologies. This strategy could be used to aluminize ultra-supercritical steam tubing to withstand aggressive oxidizing environments.

General synthetic strategy for regioselective ultrafast formation of disulfide bonds in peptides and proteins.

Project description:Despite six decades of efforts to synthesize peptides and proteins bearing multiple disulfide bonds, this synthetic challenge remains an unsolved problem in most targets (e.g., knotted mini proteins). Here we show a de novo general synthetic strategy for the ultrafast, high-yielding formation of two and three disulfide bonds in peptides and proteins. We develop an approach based on the combination of a small molecule, ultraviolet-light, and palladium for chemo- and regio-selective activation of cysteine, which enables the one-pot formation of multiple disulfide bonds in various peptides and proteins. We prepare bioactive targets of high therapeutic potential, including conotoxin, RANTES, EETI-II, and plectasin peptides and the linaclotide drug. We anticipate that this strategy will be a game-changer in preparing millions of inaccessible targets for drug discovery.

BSA modification of bacterial surface: a promising anti-cancer therapeutic strategy.

Project description:Attenuated live bacterial therapy and medical BSA materials have their own advantages in anti-cancer research, and their combination is expected to overcome some of the disadvantages of conventional anti-cancer therapeutics. Utilizing the high affinity between biotin and streptavidin, BSA modification on the surface of Escherichia coli (E. coli) was achieved. Then, the adhesion and targeting abilities of BSA modified E. coli was explored on different bladder cancer cells, and the underlying mechanism was also investigated. BSA modification on the surface of E. coli enhances its ability to adhere and target cancer cells, and we speculate that these characteristics are related to the expression of SPARC in different bladder cancer cell lines. BSA and live bacteria have their own advantages in anti-cancer research. In this study, we found that E. coli surface-modified by BSA had stronger adhesion and targeting effects on bladder cancer cells with high expression of SPARC. These findings pave the way for the future studies exploring the combination of BSA combined with live bacteria for cancer therapy.

Surface Modification of Super Arborized Silica for Flexible and Wearable Ultrafast-Response Strain Sensors with Low Hysteresis.

Project description:Conductive hydrogels exhibit high potential in the fields of wearable sensors, healthcare monitoring, and e-skins. However, it remains a huge challenge to integrate high elasticity, low hysteresis, and excellent stretch-ability in physical crosslinking hydrogels. This study reports the synthesis of polyacrylamide (PAM)-3-(trimethoxysilyl) propyl methacrylate-grafted super arborized silica nanoparticle (TSASN)-lithium chloride (LiCl) hydrogel sensors with high elasticity, low hysteresis, and excellent electrical conductivity. The introduction of TSASN enhances the mechanical strength and reversible resilience of the PAM-TSASN-LiCl hydrogels by chain entanglement and interfacial chemical bonding, and provides stress-transfer centers for external-force diffusion. These hydrogels show outstanding mechanical strength (a tensile stress of 80-120 kPa, elongation at break of 900-1400%, and dissipated energy of 0.8-9.6 kJ m-3 ), and can withstand multiple mechanical cycles. LiCl addition enables the PAM-TSASN-LiCl hydrogels to exhibit excellent electrical properties with an outstanding sensing performance (gauge factor = 4.5), with rapid response (210 ms) within a wide strain-sensing range (1-800%). These PAM-TSASN-LiCl hydrogel sensors can detect various human-body movements for prolonged durations of time, and generate stable and reliable output signals. The hydrogels fabricated with high stretch-ability, low hysteresis, and reversible resilience, can be used as flexible wearable sensors.

Surface Modification Strategy for Enhanced NO<sub>2</sub> Capture in Metal-Organic Frameworks.

Project description:The interaction strength of nitrogen dioxide (NO2) with a set of 43 functionalized benzene molecules was investigated by performing density functional theory (DFT) calculations. The functional groups under study were strategically selected as potential modifications of the organic linker of existing metal-organic frameworks (MOFs) in order to enhance their uptake of NO2 molecules. Among the functional groups considered, the highest interaction energy with NO2 (5.4 kcal/mol) was found for phenyl hydrogen sulfate (-OSO3H) at the RI-DSD-BLYP/def2-TZVPP level of theory-an interaction almost three times larger than the corresponding binding energy for non-functionalized benzene (2.0 kcal/mol). The groups with the strongest NO2 interactions (-OSO3H, -PO3H2, -OPO3H2) were selected for functionalizing the linker of IRMOF-8 and investigating the trend in their NO2 uptake capacities with grand canonical Monte Carlo (GCMC) simulations at ambient temperature for a wide pressure range. The predicted isotherms show a profound enhancement of the NO2 uptake with the introduction of the strongly-binding functional groups in the framework, rendering them promising modification candidates for improving the NO2 uptake performance not only in MOFs but also in various other porous materials.

Protein surface functionalisation as a general strategy for facilitating biomimetic mineralisation of ZIF-8.

Project description:The durability of enzymes in harsh conditions can be enhanced by encapsulation within metal-organic frameworks (MOFs) via a process called biomimetic mineralisation. Herein we show that the surface charge and chemistry of a protein determines its ability to seed MOF growth. We demonstrate that chemical modification of amino acids on the protein surface is an effective method for systematically controlling biomimetic mineralisation by zeolitic imidazolate framework-8 (ZIF-8). Reaction of surface lysine residues with succinic (or acetic) anhydride facilitates biomimetic mineralisation by increasing the surface negative charge, whereas reaction of surface carboxylate moieties with ethylenediamine affords a more positively charged protein and hinders the process. Moreover, computational studies confirm that the surface electrostatic potential of a protein is a good indicator of its ability to induce biomimetic mineralisation. This study highlights the important role played by protein surface chemistry in encapsulation and outlines a general method for facilitating the biomimetic mineralisation of proteins.

Ultrafast optical modification of exchange interactions in iron oxides.

Project description:Ultrafast non-thermal manipulation of magnetization by light relies on either indirect coupling of the electric field component of the light with spins via spin-orbit interaction or direct coupling between the magnetic field component and spins. Here we propose a scenario for coupling between the electric field of light and spins via optical modification of the exchange interaction, one of the strongest quantum effects with strength of 10(3) Tesla. We demonstrate that this isotropic opto-magnetic effect, which can be called inverse magneto-refraction, is allowed in a material of any symmetry. Its existence is corroborated by the experimental observation of terahertz emission by spin resonances optically excited in a broad class of iron oxides with a canted spin configuration. From its strength we estimate that a sub-picosecond modification of the exchange interaction by laser pulses with fluence of about 1 mJ cm(-2) acts as a pulsed effective magnetic field of 0.01 Tesla.

Ultrafast Demagnetization Control in Magnetophotonic Surface Crystals.

Project description:Magnetic memory combining plasmonics and magnetism is poised to dramatically increase the bit density and energy efficiency of light-assisted ultrafast magnetic storage, thanks to nanoplasmon-driven enhancement and confinement of light. Here we devise a new path for that, simultaneously enabling light-driven bit downscaling, reduction of the required energy for magnetic memory writing, and a subtle control over the degree of demagnetization in a magnetophotonic surface crystal. It features a regular array of truncated-nanocone-shaped Au-TbCo antennas showing both localized plasmon and surface lattice resonance modes. The ultrafast magnetization dynamics of the nanoantennas show a 3-fold resonant enhancement of the demagnetization efficiency. The degree of demagnetization is further tuned by activating surface lattice modes. This reveals a platform where ultrafast demagnetization is localized at the nanoscale and its extent can be controlled at will, rendering it multistate and potentially opening up so-far-unforeseen nanomagnetic neuromorphic-like systems operating at femtosecond time scales controlled by light.

Subsurface Carbon: A General Feature of Noble Metals.

Project description:Carbon moieties on late transition metals are regarded as poisoning agents in heterogeneous catalysis. Recent studies show the promoting catalytic role of subsurface C atoms in Pd surfaces and their existence in Ni and Pt surfaces. Here energetic and kinetic evidence obtained by accurate simulations on surface and nanoparticle models shows that such subsurface C species are a general issue to consider even in coinage noble-metal systems. Subsurface C is the most stable situation in densely packed (111) surfaces of Cu and Ag, with sinking barriers low enough to be overcome at catalytic working temperatures. Low-coordinated sites at nanoparticle edges and corners further stabilize them, even in Au, with negligible subsurface sinking barriers. The malleability of low-coordinated sites is key in the subsurface C accommodation. The incorporation of C species decreases the electron density of the surrounding metal atoms, thus affecting their chemical and catalytic activity.

General strategy for broadening adenovirus tropism.

Project description:In spite of its broad host range, adenovirus type 5 (Ad5) transduces a number of clinically relevant tissues and cell types inefficiently, mostly because of low expression of the coxsackievirus-adenovirus receptor (CAR). To improve gene transfer to such cells, we modified the Ad5 fiber knob to recognize novel receptors. We expressed a functional Ad5 fiber knob domain on the capsid of phage lambda and employed this display system to construct a large collection of ligands in the HI loop of the Ad5 knob. Panning this library on the CAR-negative mouse fibroblast cell line NIH 3T3 resulted in the identification of three clones with increased binding to these cells. Adenoviruses incorporating these ligands in the fiber gene transduced NIH 3T3 cells 2 or 3 orders of magnitude better than the parent vector. The same nonnative tropism was revealed in other cell types, independently of CAR expression. These Ad5 derivatives proved capable of transducing mouse and human primary immature dendritic cells with up to 100-fold increased efficiency.

Ultrafast generation and decay of a surface metal.

Project description:Band bending at semiconductor surfaces induced by chemical doping or electric fields can create metallic surfaces with properties not found in the bulk, such as high electron mobility, magnetism or superconductivity. Optical generation of such metallic surfaces on ultrafast timescales would be appealing for high-speed electronics. Here, we demonstrate the ultrafast generation of a metal at the (10-10) surface of ZnO upon photoexcitation. Compared to hitherto known ultrafast photoinduced semiconductor-to-metal transitions that occur in the bulk of inorganic semiconductors, the metallization of the ZnO surface is launched by 3-4 orders of magnitude lower photon fluxes. Using time- and angle-resolved photoelectron spectroscopy, we show that the phase transition is caused by photoinduced downward surface band bending due to photodepletion of donor-type deep surface defects. The discovered mechanism is in analogy to chemical doping of semiconductor surfaces and presents a general route for controlling surface-confined metallicity on ultrafast timescales.
