On the identity and typification of Solanum brasilianum Dunal (Solanaceae).

ABSTRACT: Solanum brasilianum Dunal was described by Dunal in 1813 with reference only to an illustration in an 18^th century work by Leonard Plukenet. The plate is difficult to interpret and no explicitly related specimens were available so the name Solanum brasilianum has long been regarded as "unresolved" and has never been used. Material matching the Plukenet plate was discovered in the herbarium of the University of Oxford (OXF) by Stephen Harris during his study of the English privateer William Dampier's Brazilian collection. The specimen is referable to a common Brazilian Solanum that is a member of the Torva clade, Solanum paniculatum L., making Solanum brasilianum Dunal a heterotypic synonym. We lectotypify Solanum brasilianum here, and designate an epitype using the Dampier material from OXF.

Project description:Taxonomic revision of the tropical Asian species of Solanum revealed two names, Solanum poka Dunal and Solanum graciliflorum Dunal, whose identities were uncertain and whose application has always been tentative. Material collected in Java at the beginning of the 19th century by Jean-Baptiste Leschenault de la Tour and used to describe these taxa has not been found, despite extensive searches in European herbaria. We here stabilise use of these names by comparing herbarium specimens and drawings of original material made by the artist Toussaint François Node-Véran. Detailed descriptions with synonymy, preliminary conservation assessments and specimen citations are provided for both species. Lectotypes are designated for all names (including synonyms) and epitypes designated for Solanum poka and Solanum graciliflorum to stabilise usage.

Project description:Solanum umalilaense Manoko sp. nov. (Solanaceae) is described from the Umalila area, in the southern highlands of Tanzania. Its novelty is supported with both morphological and AFLP data. Phenetic and phylogenetic analyses place Solanum umalilaense as a unique and well-supported taxon among tetraploid species of Solanum sect. Solanum from Africa. It can be distinguished from other African species by its extremely developed branching, each branch producing many multi-flowered inflorescences, flowers with short calyx lobes and its persistent, small, light yellowish brown fruits.

Cutin from Solanum Myriacanthum Dunal and Solanum Aculeatissimum Jacq. as a Potential Raw Material for Biopolymers.

Project description:Plant cuticles have attracted attention because they can be used to produce hydrophobic films as models for novel biopolymers. Usually, cuticles are obtained from agroresidual waste. To find new renewable natural sources to design green and commercially available bioplastics, fruits of S. aculeatissimum and S. myriacanthum were analyzed. These fruits are not used for human or animal consumption, mainly because the fruit is composed of seeds. Fruit peels were object of enzymatic and chemical methods to get thick cutins in good yields (approximately 77% from dry weight), and they were studied by solid-state resonance techniques (CPMAS 13C NMR), attenuated total reflection-Fourier transform infrared spectroscopy (ATR-FTIR), atomic force microscopy (AFM) and direct injection electrospray ionization mass spectrometry (DIESI-MS) analytical methods. The main component of S. aculeatissimum cutin is 10,16-dihydroxypalmitic acid (10,16-DHPA, 69.84%), while S. myriacanthum cutin besides of 10,16-DHPA (44.02%); another two C18 monomers: 9,10,18-trihydroxy-octadecanoic acid (24.03%) and 18-hydroxy-9S,10R-epoxy-octadecanoic acid (9.36%) are present. The hydrolyzed cutins were used to produce films demonstrating that both cutins could be a potential raw material for different biopolymers.

On the identity and typification of Violatenuis Bentham (Violaceae).

Project description:Violatenuis Benth. was first described by George Bentham in 1842, without any type specimen designated, but with a collection record by Richard Brinsley Hinds in 1841. Currently, Violatenuis is considered a synonym of another species according to Plants of the World Online (POWO). However, based on a combination of morphological and molecular evidence, we propose reinstating Violatenuis as a distinct species. Our analysis places it within Violasect.Plagiostigmasubsect.Diffusae, where it shows morphological similarities to five other species endemic to Guangdong Province, China. We designated the type of V.tenuis here and assigned additional KFBG specimens to the species.

Selection of reference genes for qPCR normalization in buffalobur (Solanum rostratum Dunal).

Project description:Buffalobur (Solanum rostratum Dunal), which belongs to the Solanaceae family, is a worldwide noxious invasive weed and is listed as one of the top 10 alien invasive species in China. It is harmful to humans and livestock because the entire plant is covered with spines containing toxins. Many studies have analysed the gene expression in this weed species under different stress conditions using quantitative real-time PCR (qPCR). However, until now, there has been no report on suitable reference genes in buffalobur. Herein, 14 candidate reference genes were selected and evaluated for their expression stability in buffalobur in different tissues, at different developmental stages, and in response to several stress conditions using the geNorm, NormFinder, BestKeeper and RefFinder statistical algorithms. The results showed that EF1α, ACT and SAND are suitable reference genes across all samples tested. We recommend the normalization of target gene expression under different experimental conditions using these three genes together. Validation of selected reference genes was achieved by assessing the relative expression levels of P5CS and GI. This work identified the appropriate reference genes for transcript normalization in buffalobur, which will be helpful in future genetic studies of this invasive weed.

New species of Solanum (Solanaceae) from Peru and Ecuador.

Project description:Three new species of "non-spiny" Solanum are described from Peru and Ecuador, and a revised description for Solanum verecundum M. Nee is presented. Solanum kulliwaita S. Knapp, sp. nov. (Dulcamaroid clade) is endemic to the Department of Cuzco in southern Peru, and is most similar to the recently described Solanum sanchez-vegae S. Knapp of northern Peru. Solanum dillonii S. Knapp, sp. nov. (Brevantherum clade) is found in southern Ecuador and northern Peru in the Amotape-Huancabamba phytogeographic zone, and is morphologically similar to the widespread Solanum riparium Ruiz & Pav. Solanum oxapampense S. Knapp, sp. nov., (also of the Brevantherum clade) is endemic to the Oxapampa region (Department of Pasco) of central Peru, and is similar to and segregated from Solanum verecundum M. Nee of Peru and Ecuador. Complete descriptions, distributions and preliminary conservation assessments of all new species are given.

A new species of spiny Solanum (Solanaceae) from Peru.

Project description:A new species of Solanum is described from Peru. Solanum junctum S. Stern & M. Nee, sp nov. is a member of the Torva clade of the spiny solanums (Leptostemonum clade). The narrow corolla lobes and recurved prickles of Solanum junctum are similar to species in the Micracantha clade, but Solanum junctum differs in its branched inflorescences and upright green fruits. These characteristics are shared with other members the Torva clade; within this section Solanum junctum is morphologically most similar to Solanum subinerme and Solanum poinsettiifolium. Solanum subinerme has larger flowers, longer cauline prickles, and often has long straight prickles on the adaxial leaf surface that are lacking in Solanum junctum. Solanum poinsettiifolium has fewer spines, dense white tomentum on the abaxial leaf surfaces, stout unbranched inflorescences, and more extensive interpetalar corolla tissue than Solanum junctum.

In Vitro and In Silico Antidiabetic Efficacy of Solanum lasiocarpum Dunal Fruit Extract.

Project description:Exploring the possibility of familiar dietary sources as additional diabetes treatments is crucial, especially considering the financial difficulties related to diabetes mellitus. Using both in vitro and in silico techniques, this work aims to assess the antidiabetic benefits of extract from Solanum lasiocarpum Dunal. The evaluations encompass the ability to scavenge DPPH radicals, inhibition of α-amylase, α-glucosidase, inhibition of DPP-4, cytotoxicity, and glucose absorption kinetics. With an IC50 value of 0.69 ± 0.14 mg/ml, S. lasiocarpum showed encouraging DPPH inhibition. IC50 values of 2.123 ± 0.14 mg/ml inhibited the enzymes α-amylase, α-glucosidase, and DPP-4. Furthermore, a notable increase (P < 0.05) in glucose uptake by L6 myoblasts was observed with the administration of various combinations. In silico analysis, including XP docking and MM-GBSA, revealed that 10 and 21 compounds within the combination exhibited substantial interactions and stable binding capabilities with α-amylase and DPP-4 proteins, indicating their potential as enzyme inhibitors. Therefore, it can be inferred that S. lasiocarpum represents a promising therapeutic approach for diabetes management.

Mitochondrial genome sequence of tuber-bearing wild potato, Solanum commersonii Dunal.

Project description:We report two complete mitochondrial genome sequences of a tuber-bearing wild potato species (Solanum commersonii). The genomes are circular DNA molecules with lengths of 213,676 bp and 338,427 bp containing 80 nonredundant genes totally, including 34 protein-coding genes, 25 hypothetical open reading frames, 18 tRNA genes, and 3 rRNA genes. Phylogenetic analysis using common protein-coding sequences confirmed that S. commersonii belongs to the Solanoideae subfamily in the Solanaceae family.

Chromosome-level genome assembly and annotation of the prickly nightshade Solanum rostratum Dunal.

Project description:The prickly nightshade Solanum rostratum, an annual malignant weed, is native to North America and has globally invaded 34 countries, causing serious threats to ecosystems, agriculture, animal husbandry, and human health. In this study, we constructed a chromosome-level genome assembly and annotation of S. rostratum. The contig-level genome was initially assembled in 898.42 Mb with a contig N50 of 62.00 Mb from PacBio high-fidelity reads. With Hi-C sequencing data scaffolding, 96.80% of the initially assembled sequences were anchored and orientated onto 12 pseudo-chromosomes, generating a genome of 869.69 Mb with a contig N50 of 72.15 Mb. We identified 649.92 Mb (72.26%) of repetitive sequences and 3,588 non-coding RNAs in the genome. A total of 29,694 protein-coding genes were predicted, with 28,154 (94.81%) functionally annotated genes. We found 99.5% and 91.3% complete embryophyta_odb10 genes in the pseudo-chromosomes genome and predicted gene datasets by BUSCO assessment. The present genomic resource provides essential information for subsequent research on the mechanisms of environmental adaptation of S. rostratum and host shift in Colorado potato beetles.
