Mass spectrometric snapshots for electrochemical reactions.

ABSTRACT: A hybrid ultramicroelectrode containing one micro-carbon electrode and one empty micro-channel was employed to be a micro-electrochemical cell and a mass spectrometric nanospray emitter. This setup can combine MS with an electrode directly and provide in situ information about an electrochemical reaction. The mechanisms proposed by Bard et al. for a Ru(bpy)₃²⁺ (bpy = 2,2'-bipyridine) electrochemiluminescence (ECL) system were confirmed by the MS detection of key intermediates. The short-lived diimine intermediate of electrochemical oxidation of uric acid was also detected, which affirms that the novel technique is able to catch fleeting intermediates. These experimental results demonstrate that this new method is simple, easy to implement and can be coupled with many commercial mass spectrometric instruments to provide very useful information about electrochemical reactions.

Mass spectrometric detection of fleeting neutral intermediates generated in electrochemical reactions.

Project description:Towards the goal of on-line monitoring of transient neutral intermediates during electrochemical reactions, an electrochemistry-neutral reionization-mass spectrometry (EC-NR-MS) technique was developed in this work. The EC-NR setup consisted of a customized EC flow cell, a sonic spray ionization source, a heating tube, an ion deflector and an electrospray ionization source, which were respectively used for the precise control of the electrochemical reaction, solution nebulization, droplet desolvation, ion deflection and neutral intermediate ionization. Based on the EC-NR-MS approach, some long-sought neutral radicals including TPrA˙, DBAE˙ and TEOA˙, which belong to important reductive intermediates in electrochemiluminescence (ECL) reactions, were successfully identified which helps to clarify the previously unproven ECL reaction mechanism. These findings were also supported by spin-trapping experiments and the tandem MS technique. Accordingly, the EC-NR-MS method provides a direct solution for studying complicated electrochemical reactions, especially for detecting short-lived neutral radicals as well as ionic intermediates.

Mass-Spectrometric Imaging of Electrode Surfaces-a View on Electrochemical Side Reactions.

Project description:Electrochemical side reactions, often referred to as "electrode fouling", are known to be a major challenge in electro-organic synthesis and the functionality of modern batteries. Often, polymerization of one or more components is observed. When reaching their limit of solubility, those polymers tend to adsorb on the surface of the electrode, resulting in a passivation of the respective electrode area, which may impact electrochemical performance. Here, matrix-assisted laser-desorption/ionization mass spectrometry (MALDI-MS) is presented as valuable imaging technique to visualize polymer deposition on electrode surfaces. Oligomer size distribution and its dependency on the contact time were imaged on a boron-doped diamond (BDD) anode of an electrochemical flow-through cell. The approach allows to detect weak spots, where electrode fouling may take place and provides insight into the identity of side-product pathways.

Simultaneous determination of different DNA sequences by mass spectrometric evaluation of Sanger sequencing reactions.

Project description:All currently available DNA sequencing protocols rest fundamentally upon the homogeneity of the template. In this paper we describe the parallel DNA sequencing of various templates in one sample by a combination of the Sanger method and MALDI-TOF mass spectrometric analysis of the products. PCR-amplified hypervariable 16S rDNA fragments of the bacterium Escherichia coli DF1020 and cDNA of the 6-phosphofructo-1-kinase isoenzymes (PFK-1, EC 2.7.1.11) in rat brain were chosen as model systems for essentially heterogeneous templates. Avoiding cloning of the inhomogeneous PCR products we were able to read three sequences for both the 16S rDNA fragment of E.coli DF1020 and the cDNA of 6-phosphofructo-1-kinase from the peak lists of the Sanger sequencing reactions. Short sequences with a length between 21 and 25 nt were sufficient to reflect the heterogeneity of the 16S rDNA genes in E.coli and the existence of three isoenzymes of PFK-1 in rat brain.

Snapshots of Iron Speciation: Tracking the Fate of Iron Nanoparticle Drugs via a Liquid Chromatography-Inductively Coupled Plasma-Mass Spectrometric Approach.

Project description:Nanomedicines are nanoparticle-based therapeutic or diagnostic agents designed for targeted delivery or enhanced stability. Nanotechnology has been successfully employed to develop various drug formulations with improved pharmacokinetic characteristics, and current research efforts are focused on the development of new innovator and generic nanomedicines. Nanomedicines, which are often denoted as complex or nonbiological complex drugs, have inherently different physicochemical and pharmacokinetic properties than conventional small molecule drugs. The tools necessary to fully evaluate nanomedicines in clinical settings are limited, which can hamper their development. One of the most successful families of nanomedicines are iron-carbohydrate nanoparticles, which are administered intravenously (IV) to treat iron-deficiency anemia. In the U.S., the FDA has approved six distinct iron-carbohydrate nanoparticles but only one generic version (sodium ferric gluconate for Ferrlecit). There is significant interest in approving additional generic iron-carbohydrate drugs; however, the lack of a direct method to monitor the fate of the iron nanoparticles in clinical samples has impeded this approval. Herein we report a novel liquid chromatography-inductively coupled plasma-mass spectrometry (LC-ICP-MS) method that allows for the direct quantification of the iron-carbohydrate drugs in clinical samples, while simultaneously measuring the speciation of the iron released from the nanoparticles in biological samples. To our knowledge, this is the first time that iron nanoparticles have been observed in clinical samples, opening the door for direct pharmacokinetic studies of this family of drugs. This method has potential applications not only for iron-nanoparticle drugs but also for any nanomedicine with an inorganic component.

Functional group selective ion/molecule reactions: mass spectrometric identification of the amido functionality in protonated monofunctional compounds.

Project description:A mass spectrometric method was developed for the screening of the amido functionality in monofunctional protonated analytes. This method is based on selective gas-phase derivatization of protonated analytes by (N,N-diethylamino)dimethylborane in a Fourier transform ion cyclotron resonance (FT-ICR) and triple quadrupole mass spectrometer. Examination of a series of protonated analytes demonstrated that only the compounds containing the amido functionality react with the aminoborane by the derivatization reaction. The mechanism involves proton transfer from the protonated analyte to the borane, followed by addition of the amide to the boron center, which leads to the elimination of neutral diethylamine. The derivatized analytes are readily identified on the basis of a shift of 40 m/z units relative to the m/z value of the protonated analyte and characteristic boron isotope patterns. Collision-activated dissociation was used to provide support for the structures assigned to the derivatized analytes. The structural information gained from this gas-phase derivatization method will aid in the functional group identification of unknown compounds and their mixtures.

Coupling Liquid Electrochemical TEM and Mass-Spectrometry to Investigate Electrochemical Reactions Occurring in a Na-Ion Battery Anode.

Project description:A novel approach for investigating the formation of solid electrolyte interphase (SEI) in Na-ion batteries (NIB) through the coupling of in situ liquid electrochemical transmission electron microscopy (ec-TEM) and gas-chromatography mass-spectrometry (GC/MS) is proposed. To optimize this coupling, experiments are conducted on the sodiation of hard carbon materials (HC) using two setups: in situ ec-TEM holder and ex situ setup. Electrolyte (NP30) is intentionally degraded using cyclic voltammetry (CV), and the recovered liquid product is analyzed using GC/MS. Solid product (µ-chip) is analyzed using TEM techniques in a post-mortem analysis. The ex situ experiments served as a reference to for insertion of Na+ ions in the HC, SEI size (389 nm), SEI composition (P, Na, F, and O), and Na plating. The in situ TEM analysis reveals a cyclability limitation, this issue appears to be caused by the plating of Na in the form of a "foam" structure, resulting from the gas release during the reaction of Na with DMC/EC electrolyte. The foam structure, subsequently transformes into a second SEI, is electrochemically inactive and reduces the cyclability of the battery. Overall, the results demonstrate the powerful synergy achieved by coupling in situ ec-TEM and GC/MS techniques.

Discovery of true electrochemical reactions for ultrahigh catalyst mass activity in water splitting.

Project description:Better understanding of true electrochemical reaction behaviors in electrochemical energy devices has long been desired. It has been assumed so far that the reactions occur across the entire catalyst layer (CL), which is designed and fabricated uniformly with catalysts, conductors of protons and electrons, and pathways for reactants and products. By introducing a state-of-the-art characterization system, a thin, highly tunable liquid/gas diffusion layer (LGDL), and an innovative design of electrochemical proton exchange membrane electrolyzer cells (PEMECs), the electrochemical reactions on both microspatial and microtemporal scales are revealed for the first time. Surprisingly, reactions occur only on the CL adjacent to good electrical conductors. On the basis of these findings, new CL fabrications on the novel LGDLs exhibit more than 50 times higher mass activity than conventional catalyst-coated membranes in PEMECs. This discovery presents an opportunity to enhance the multiphase interfacial effects, maximizing the use of the catalysts and significantly reducing the cost of these devices.

Mass spectrometric characterization of gelsolin isoforms.

Project description:Gelsolin is the most widely expressed member of the actin capping and severing family of proteins. There are two isoforms of gelsolin: isoform 1, a secretory (plasma) protein that is 51 amino acids longer than isoform 2, a cytosolic protein, at the N-terminus; the first 27 amino acids is a signal sequence. Both isoforms are coded by a single gene and differ as a result of alternative initiation site/splicing. The level of gelsolin in the blood and cerebrospinal fluid (CSF) is altered in many diseases including amyloidoses and other neurodegenerative disorders. Although quantitative analysis of gelsolin has been reported, lack of suitable antibodies makes it impossible to differentiate these two isoforms by immunodetection techniques and no other technique is available. Therefore, ambiguity exists whether gelsolin present in circulation is isoform 1 or also isoform 2 released from lysed cells. We report in this communication a mass spectrometric approach to identify isoform 1 of gelsolin immunopurified from human plasma and CSF. Recombinant isoform 1 was used as reference.

Direct mass spectrometric characterization of disulfide linkages.

Project description:Disulfide linkage is critical to protein folding and structural stability. The location of disulfide linkages for antibodies is routinely discovered by comparing the chromatograms of the reduced and non-reduced peptide mapping with location identification confirmed by collision-induced dissociation (CID) mass spectrometry (MS)/MS. However, CID product spectra of disulfide-linked peptides can be difficult to interpret, and provide limited information on the backbone region within the disulfide loop. Here, we applied an electron-transfer dissociation (ETD)/CID combined fragmentation method that identifies the disulfide linkage without intensive LC comparison, and yet maps the disulfide location accurately. The native protein samples were digested using trypsin for proteolysis. The method uses RapiGest SF Surfactant and obviates the need for reduction/alkylation and extensive sample manipulation. An aliquot of the digest was loaded onto a C4 analytical column. Peptides were gradient-eluted and analyzed using a Thermo Scientific LTQ Orbitrap Elite mass spectrometer for the ETD-triggered CID MS 3 experiment. Survey MS scans were followed by data-dependent scans consisting of ETD MS2 scans on the most intense ion in the survey scan, followed by 5 MS3 CID scans on the 5 most intense ions in the ETD MS2 scan. We were able to identify the disulfide-mediated structural variants A and A/B forms and their corresponding disulfide linkages in an immunoglobulin G2 monoclonal antibody with λ light chain (IgG2λ), where the location of cysteine linkages were unambiguously determined.

Mass Spectrometric Characterization of Epoxidized Vegetable Oils.

Project description:Matrix-assisted laser desorption ionization and electrospray ionization mass spectrometry (MALDI-MS and ESI-MS) were used for the characterization of epoxidized soybean and linseed oils, which are important raw materials in the biopolymer production. The recently invented data mining approach, mass-remainder analysis (MARA), was implemented for the analysis of these types of complex natural systems. Different epoxidized triglyceride mass spectral peak series were identified, and the number of carbon atoms and epoxide groups was determined. The fragmentation mechanisms of the epoxidized triglyceride (ETG) adducts formed with different cations (such as H⁺, Na⁺, Li⁺, and NH₄⁺) were explored. As a novel approach, the evaluation of the clear fragmentation pathways of the sodiated ETG adducts enabled the estimation of the epoxidized fatty acid compositions of these types of oils by MS/MS.
