Growth and Grazing Kinetics of the Facultative Anaerobic Nanoflagellate, Suigetsumonas clinomigrationis.

ABSTRACT: The functional and numerical responses of the facultative anaerobic heterotrophic nanoflagellate, Suigetsumonas clinomigrationis NIES-3647 to prey density were examined under oxic and anoxic conditions. S. clinomigrationis grew at temperatures between 10 and 30°C and in the salinity range of 3.9-36.9 psu. The maximum specific growth and ingestion rates of S. clinomigrationis were lower under anoxic conditions than under oxic conditions. Half-saturation constants for the growth of S. clinomigrationis were within or greater than the range of bacterial densities in the water column of Lake Suigetsu, suggesting that its growth rate is limited by bacterial prey densities in natural environments.

Project description:Haloferax volcanii is a facultative anaerobic haloarchaeon that can grow using nitrate or dimethyl sulfoxide (DMSO) as a respiratory substrate under anaerobic conditions. Comparative transcriptome analysis of denitrifying and aerobic cells of H. volcanii indicated extensive changes in gene expression involving the activation of denitrification, suppression of DMSO respiration, and conversion of the heme biosynthetic pathway under denitrifying conditions. The anaerobic growth of H. volcanii by DMSO respiration was inhibited at nitrate concentrations of <1 mM, whereas nitrate-responsive growth inhibition was not observed in the ΔnarO mutant. A reporter assay demonstrated that the transcription of the dms operon was suppressed by nitrate. In contrast, the anaerobic growth of the ΔdmsR mutant by denitrification was little affected by the addition of DMSO. NarO has been identified as an activator of denitrification-related genes in response to anaerobic conditions, and here, we found that NarO is also involved in nitrate-responsive suppression of the dms operon. Nitrate-responsive suppression of DMSO respiration is known in several bacteria such as Escherichia coli and photosynthetic Rhodobacter species. This is the first report to show that a regulatory mechanism that suppresses DMSO respiration in response to nitrate exists not only in bacteria but also in haloarchaea. IMPORTANCE Haloferax volcanii can grow anaerobically by denitrification (nitrate respiration) or DMSO respiration. In facultative anaerobic bacteria that can grow by both nitrate respiration and DMSO respiration, nitrate respiration is preferentially induced when both nitrate and DMSO are available as the respiratory substrates. The results of transcriptome analysis, growth phenotyping, and reporter assays indicated that DMSO respiration is suppressed in response to nitrate in H. volcanii. The haloarchaeon-specific regulator NarO, which activates denitrification under anaerobic conditions, is suggested to be involved in the nitrate-responsive suppression of DMSO respiration.

Project description:Understanding how livestock grazing strategies of native warm season grasses (NWSG) can impact facultative grassland bird nesting can provide insight for conservation efforts. We compared pre and post treatment effects of rotational grazing (ROT) and patch-burn grazing (PBG) for facultative grassland bird species nest success and nest-site selection on NWSG pastures at three Mid-South research sites. We established 14, 9.7 ha NWSG pastures and randomly assigned each to either ROT or PBG and monitored avian nest-site selection and nest success, 2014-2016. We collected nesting and vegetation data in 2014, before treatment implementation, as an experimental pre-treatment. We implemented treatments across all research sites in spring 2015. We used a step-wise model selection framework to estimate treatment effect for ROT or PBG on avian nest daily survival rate (DSR) and resource selection function (RSF) at the temporal scale and within-field variables. Daily survival rates were 0.93% (SE = 0.006) for field sparrow (Spizella pusilla), 0.96% (SE = 0.008) for red-winged blackbird (Agelaius phoeniceus), and 0.92% (SE = 0.01) for indigo bunting (Passerina cyanea). Model support for PBG treatment and vegetation height were indicated as negative and positive influences for field sparrow DSR, respectively. Red-winged blackbirds' DSR were negatively influenced by ROT while vegetation height positively affected DSR, and DSR for indigo bunting did not differ among treatments. Combined RSF models indicated nest-site selection for all species was positively related to vegetation height and only weakly associated with other within-field variables. We provide evidence that ROT and/or PBG effects vary by species for DSR for these three facultative grassland birds, and vegetation characteristics affected their nest-site selection in the Mid-South USA. A lack of disturbance in Mid-South grasslands can lead to higher successional stages (i.e., mix shrub-grassland), but some combination of ROT, PBG, and unburned/ungrazed areas can provide adequate nesting habitat on small pasture lands (∼1.8 -7.8 ha) for various facultative grassland birds and potentially offer the opportunity to simultaneously maintain livestock production and grassland bird nesting habitat.

Project description:Microbial reduction is an effective way to deal with hexavalent chromium [Cr(VI)] contamination in the environment, which can significantly mitigate the biotoxicity and migration of this pollutant. The present study investigated the influence of environmental factors on aqueous Cr(VI) removal by a newly isolated facultative anaerobic bacterium, Exiguobacterium sp. PY14, and revealed the reduction mechanism. This strain with a minimum inhibitory concentration of 400 mg/L showed the strongest Cr(VI) removal capacity at pH 8.0 because of its basophilic nature, which was obviously depressed by increasing the Cr(VI) initial concentration under both aerobic and anaerobic conditions. In contrast, the removal rate constant for 50 mg/L of Cr(VI) under anaerobic conditions (1.82 × 10-2 h-1) was 3.3 times that under aerobic conditions. The co-existence of Fe(III) and Cu(II) significantly promoted the removal of Cr(VI), while Ag(I), Pb(II), Zn(II), and Cd(II) inhibited it. Electron-shuttling organics such as riboflavin, humic acid, and anthraquinone-2,6-disulfonate promoted the Cr(VI) removal to varying degrees, and the enhancement was more significant under anaerobic conditions. The removal of aqueous Cr(VI) by strain PY14 was demonstrated to be due to cytoplasmic rather than extracellular reduction by analyzing the contributions of different cell components, and the end products existed in the aqueous solution in the form of organo-Cr(III) complexes. Several possible genes involved in Cr(VI) metabolism, including chrR and chrA that encode well-known Chr family proteins responsible for chromate reduction and transport, respectively, were identified in the genome of PY14, which further clarified the Cr(VI) reduction pathway of this strain. The research progress in the influence of crucial environmental factors and biological reduction mechanisms will help promote the potential application of Exiguobacterium sp. PY14 with high adaptability to environmental stress in Cr(VI) removal in the actual environment.
