Project description:Even though it is known that sleep benefits declarative memory consolidation, the role of sleep in the storage of temporal sequences has rarely been examined. Thus we explored the influence of sleep on temporal order in an episodic memory task followed by sleep or sleep deprivation. Thirty-four healthy subjects (17 men) aged between 19 and 28 years participated in the randomized, counterbalanced, between-subject design. Parameters of interests were NREM/REM cycles, spindle activity and spindle-related EEG power spectra. Participants of both groups (sleep group/sleep deprivation group) performed retrieval in the evening, morning and three days after the learning night. Results revealed that performance in temporal order memory significantly deteriorated over three days only in sleep deprived participants. Furthermore our data showed a positive relationship between the ratios of the (i) first NREM/REM cycle with more REM being associated with delayed temporal order recall. Most interestingly, data additionally indicated that (ii) memory enhancers in the sleep group show more fast spindle related alpha power at frontal electrode sites possibly indicating access to a yet to be consolidated memory trace. We suggest that distinct sleep mechanisms subserve different aspects of episodic memory and are jointly involved in sleep-dependent memory consolidation.

Project description:Episodic memories are contextual experiences ordered in time. This is underpinned by associative binding between events within the same contexts. The role of prediction errors in declarative memory is well established but has not been investigated in the time dimension of complex episodic memories. Here we combine these two properties of episodic memory, extend them into the temporal domain and demonstrate that prediction errors in different naturalistic contexts lead to changes in the temporal ordering of event structures in them. The wrongly predicted older sequences were weakened despite their reactivation. Interestingly the newly encoded sequences with prediction errors, seen once, showed accuracy as high as control sequences which were viewed repeatedly without change. Drift-diffusion modelling revealed a lower decision threshold for the newer sequences than older sequences, reflected by their faster recall. Moreover, participants' adjustments to their decision threshold significantly correlated with their relative speed of sequence memory recall. These results suggest a temporally distinct and adaptive role for prediction errors in learning and reorganizing episodic temporal sequences.

Project description:The ability to generalize from and distinguish between aversive memories and novel experiences is critical to survival. Previous research has revealed mechanisms underlying generalization of threat-conditioned defensive responses, but little is known about generalization of episodic memory for threatening events. Here we tested if aversive learning influences generalization of episodic memory for threatening events in human adults. Subjects underwent Pavlovian threat-conditioning in which objects from one category were paired with a shock and objects from a different category were unpaired. The next day, subjects underwent a recognition memory test that included old, highly similar, and entirely novel items from the shock-paired and shock-unpaired object categories. Results showed that items highly similar to those from the object category previously paired with shock were mistaken for old items more often than items from the shock-unpaired category. This finding indicates that threat learning promotes generalization of episodic memory, and is consistent with the idea that threat generalization is an active process that may be adaptive for avoiding a myriad of potential threats following an emotional experience. Enhanced generalization of aversive episodic memories may be maladaptive, however, when old threat memories are inappropriately reactivated in harmless situations, exemplified in a number of stress- and anxiety-related disorders. Project description:A necessary condition for forming episodic memories is the construction of specific episodes demarcated from other episodes in space and time. Evidence from studies of episodic memory formation using rodent models suggest that the medial temporal lobe (MTL) supports the representation of boundary information. Building on recent work using human microelectrode recordings as well, we hypothesized of human MTL neurons with firing rates sensitive to episodic boundary information. We identified 27 episodic boundary neurons out of 736 single neurons recorded across 27 subjects. Firing of these neurons increased at the beginning and end of mnemonically relevant episodes in the free recall task. We distinguish episodic boundary neurons from a population of ramping neurons (n = 58), which are time-sensitive neurons whose activity provides complementary information during episodic representation. Episodic boundary neurons exhibited a U-shaped activity pattern demonstrating increased activity after both beginning and end boundaries of encoding and retrieval epochs. We also describe evidence that the firing of boundary neurons within episodic boundaries is organized by hippocampal theta oscillations, using spike-field coherence metrics.

Project description:The ability to synthesize information across multiple senses is known as multisensory integration and is essential to our understanding of the world around us. Sensory stimuli that occur close in time are likely to be integrated, and the accuracy of this integration is dependent on our ability to precisely discriminate the relative timing of unisensory stimuli (crossmodal temporal acuity). Previous research has shown that multisensory integration is modulated by both bottom-up stimulus features, such as the temporal structure of unisensory stimuli, and top-down processes such as attention. However, it is currently uncertain how attention alters crossmodal temporal acuity. The present study investigated whether increasing attentional load would decrease crossmodal temporal acuity by utilizing a dual-task paradigm. In this study, participants were asked to judge the temporal order of a flash and beep presented at various temporal offsets (crossmodal temporal order judgment (CTOJ) task) while also directing their attention to a secondary distractor task in which they detected a target stimulus within a stream visual or auditory distractors. We found decreased performance on the CTOJ task as well as increases in both the positive and negative just noticeable difference with increasing load for both the auditory and visual distractor tasks. This strongly suggests that attention promotes greater crossmodal temporal acuity and that reducing the attentional capacity to process multisensory stimuli results in detriments to multisensory temporal processing. Our study is the first to demonstrate changes in multisensory temporal processing with decreased attentional capacity using a dual task paradigm and has strong implications for developmental disorders such as autism spectrum disorders and developmental dyslexia which are associated with alterations in both multisensory temporal processing and attention.

Project description:Episodic memory is a critical capacity that involves remembering past events along with their spatial and temporal contexts. Relatively little is known about the relations between spatial and temporal information in long-term memory in children or adults. The present research examined the influence of the mental timeline (linear horizontal display extending from the left to right direction for English speakers) on memory for events and their spatial and temporal features in 7-year-olds, 9-year-olds, 11-year-olds and young adults (N = 146). During encoding, participants studied triplets of objects, varying on two dimensions of the mental timeline: linearity (whether objects were presented in linear succession or not) and direction (whether objects were presented from left-to-right or right-to-left). After a delay, during retrieval, participants were tested on their memory for individual objects, and either the spatial location or temporal order of the objects. We found that overall accuracy for spatial location was higher than accuracy for temporal order, and there was a parallel developmental trajectory for both these aspects of memory. Across age groups we found that memory for temporal order, but not spatial location, was influenced by linearity and direction (i.e., match to mental timeline). Thus, in both children and adults the spatiotemporal mental timeline supported memory for temporal order, converging with predictions generated within domains of language and thought and enhancing our understanding of how space and time are represented in the mind.

Project description:Temporal order judgment (TOJ) measures the ability to correctly perceive the order of consecutive stimuli presented rapidly. Our previous research suggested that the major predictor of auditory dichotic TOJ threshold, a paradigm that requires the identification of the order of two tones, each of which is presented to a different ear, is the time separating the onset of the first tone from the onset of the second tone (stimulus-onset-asynchrony, SOA). Data supporting this finding, however, was based on a young adult population and a tone duration range of 10-40 msec. The current study aimed to evaluate the generalizability of the earlier finding by manipulating the experimental model in two different ways: a) extending the tone duration range to include shorter stimulus durations (3-8 msec; Experiment 1) and b) repeating the identical testing procedure on a different population with temporal processing deficits, i.e., older adults (Experiment 2). We hypothesized that the SOA would predict the TOJ threshold regardless of tone duration and participant age. Experiment 1 included 226 young adults divided into eight groups (each group receiving a different tone duration) with duration ranging from 3-40 msec. Experiment 2 included 98 participants aged 60-75 years, divided into five groups by tone duration (10-40 msec). The results of both experiments confirmed the hypothesis, that the SOA required for performing dichotic TOJ was constant regardless of stimulus duration, for both age groups: about 66.5 msec for the young adults and 33 msec longer (100 msec) for the older adults. This finding suggests that dichotic TOJ threshold is controlled by a general mechanism that changes quantitatively with age. Clinically, this has significance because quantitative changes can be more easily remedied than qualitative changes. Theoretically, our findings show that, with dichotic TOJ, tone duration affects threshold by providing more time between the onsets of the consecutive stimuli to the two ears. The findings also imply that a temporal processing deficit, at least among older adults, does not elicit the use of a different mechanism in order to judge temporal order.

Project description:Space and time are both essential features of episodic memory. However, while spatial tasks have been used effectively to study the behavioral relevance of place cells, the behavioral paradigms utilized for the study of time cells have not used time duration as a variable that animals need to be aware of to solve the task. In order to evaluate how time flow is coded into memory, time duration needs to be a variable that animals use to solve the behavioral task. This protocol describes a novel behavioral paradigm, the time duration discrimination (TDD) task, which is designed to directly investigate the neurological mechanisms that underlie temporal processing. During the TDD task, rats navigate around a Figure-8 Maze, which contains a rectangular track with a central arm and a delay box at the end of the central arm. While confined to the delay box, rats experience a 10- or 20-second time delay, during which a tone will play for the duration of the 10- or 20-second delay. When the delay box opens, the rat will choose whether to turn left or right out of the delay box and receive a reward for the correct choice (e.g., 10 seconds = left turn; 20 seconds = right turn). By directly manipulating elapsed time, we can better explore the behavioral relevance of hippocampal time cells and whether the time-dependent activity seen in physiological recordings of hippocampal neurons reflects a neuronal representation of time flow that can be used by the animal for learning and storing memories. Project description:Moderate doses of stimulant drugs are known to enhance memory encoding and consolidation, but their effects on memory retrieval have not been explored in depth. In laboratory animals, stimulants seem to improve retrieval of emotional memories, but comparable studies have not been carried out in humans. In the present study, we examined the effects of dextroamphetamine (AMP) on retrieval of emotional and unemotional stimuli in healthy young adults, using doses that enhanced memory formation when administered before encoding in our previous study. During 3 sessions, healthy volunteers (n = 31) received 2 doses of AMP (10 and 20 mg) and placebo in counterbalanced order under double-blind conditions. During each session, they first viewed emotional and unemotional pictures and words in a drug-free state, and then 2 days later their memory was tested, 1 hour after AMP or placebo administration. Dextroamphetamine did not affect the number of emotional or unemotional stimuli remembered, but both doses increased recall intrusions and false recognition. Dextroamphetamine (20 mg) also increased the number of positively rated picture descriptions and words generated during free recall. These data provide the first evidence that therapeutic range doses of stimulant drugs can increase memory retrieval errors. The ability of AMP to positively bias recollection of prior events could contribute to its potential for abuse.

Project description:Integrating visual and tactile information in the temporal domain is critical for active perception. To accomplish this, coordinated timing is required. Here, we study perceived duration within and across these two modalities. Specifically, we examined how duration comparisons within and across vision and touch were influenced by temporal context and presentation order using a two-interval forced choice task. We asked participants to compare the duration of two temporal intervals defined by tactile or visual events. Two constant standard durations (700 ms and 1,000 ms in 'shorter' sessions; 1,000 ms and 1,500 ms in 'longer' sessions) were compared to variable comparison durations in different sessions. In crossmodal trials, standard and comparison durations were presented in different modalities, whereas in the intramodal trials, the two durations were presented in the same modality. The standard duration was either presented first (<sc>) or followed the comparison duration (<cs>). In both crossmodal and intramodal conditions, we found that the longer standard duration was overestimated in <cs> trials and underestimated in <sc> trials whereas the estimation of shorter standard duration was unbiased. Importantly, the estimation of 1,000ms was biased when it was the longer standard duration within the shorter sessions but not when it was the shorter standard duration within the longer sessions, indicating an effect of temporal context. The effects of presentation order can be explained by a central tendency effect applied in different ways to different presentation orders. Both crossmodal and intramodal conditions showed better discrimination performance for <sc> trials than <cs> trials, supporting the Type B effect for both crossmodal and intramodal duration comparison. Moreover, these results were not dependent on whether the standard duration was defined using tactile or visual stimuli. Overall, our results indicate that duration comparison between vision and touch is dependent on presentation order and temporal context, but not modality.
