The Rice Peptide Transporter OsNPF7.3 Is Induced by Organic Nitrogen, and Contributes to Nitrogen Allocation and Grain Yield.

ABSTRACT: Nitrogen use efficiency is important for the development of sustainable agriculture. Plants have different transporters to facilitate nitrogen uptake and internal distribution. This study demonstrates that the peptide transporter OsNPF7.3 enhances nitrogen allocation and increases grain yield in rice. OsNPF7.3 is a member of the nitrate transporter 1/peptide transporter family (NPF) and is localized in the vacuolar membrane. Its expression is higher in the lateral roots and stems. Its transcripts concentrate in the vascular bundle and significantly regulated by organic nitrogen sources. The RNAi lines of OsNPF7.3 affect plant growth and cause amino acids to accumulate in leaf sheaths and decrease in the leaf blades. At later stages of reproductive growth, nitrogen degradation accelerates in the leaves of plants over-expressing OsNPF7.3 and the nitrogen is translocated to grains. The tiller numbers, panicles per plant, filled grain numbers per panicle, and grain nitrogen content of the OsNPF7.3 over-expressing plant were more than that of wide type. The elevated gene expression in OsNPF7.3 could enhance nitrogen utilization efficiency in rice paddy.

Project description:BackgroundRice tiller number is one of the most important factors that determine grain yield, while nitrogen is essential for the crop growth and development, especially for tiller formation. Genes involved in nitrogen use efficiency processes have been identified in the previous studies, however, only a small number of these genes have been found to improve grain yield by promoting tillering.ResultsWe constructed over-expression (OX) lines and RNA-interference (Ri) lines, and selected a mutant of OsNPF7.2, a low-affinity nitrate transporter. Our analyses showed that rice tiller number and grain yield were significantly increased in OX lines, whereas Ri lines and mutant osnpf7.2 had fewer tiller number and lower grain yield. Under different nitrate concentrations, tiller buds grew faster in OX lines than in WT, but they grew slower in Ri lines and mutant osnpf7.2. These results indicated that altered expression of OsNPF7.2 plays a significant role in the control of tiller bud growth and regulation of tillering. Elevated expression of OsNPF7.2 also improved root length, root number, fresh weight, and dry weight. However, reduced expression of OsNPF7.2 had the opposite result on these characters. OsNPF7.2 OX lines showed more significantly enhanced influx of nitrate and had a higher nitrate concentration than WT. The levels of gene transcripts related to cytokinin pathway and cell cycle in tiller bud, and cytokinins concentration in tiller basal portion were higher in OX lines than that in WT, suggesting that altered expression of OsNPF7.2 controlled tiller bud growth and root development by regulating cytokinins content and cell cycle in plant cells. Altered expression of OsNPF7.2 also was responsible for the change in expression of the genes involved in strigolactone pathway, such as D27, D17, D10, Os900, Os1400, D14, D3, and OsFC1.ConclusionOur results suggested that OsNPF7.2 is a positive regulator of nitrate influx and concentration, and that it also regulates cell division in tiller bud and alters expression of genes involved in cytokinin and strigolactone pathways, resulting in the control over rice tiller number. Since elevated expression of OsNPF7.2 is capable of improving rice grain yield, this gene might be applied to high-yield rice breeding.

Project description:IntroductionLong-term application of excessive nitrogen (N) not only leads to low N use efficiency (NUE) but also exacerbates the risk of environmental pollution due to N losses. Substituting partial chemical N with organic fertilizer (SP) is an environmentally friendly and sustainable fertilization practice. However, the appropriate rate of SP in rainfed maize cropping systems in semi-arid regions of China is unknown.MethodsTherefore, we conducted a field experiment between 2021 and 2022 in a semi-arid region of Northern China to investigate the effects of SP on maize growth, carbon and N metabolism (C/NM), and NUE. The following treatments were used in the experiment: no N application (CK), 100% chemical N (SP0, 210 kg N ha-1), and SP substituting 15% (SP1), 30% (SP2), 45% (SP3), and 60% (SP4) of the chemical N. The relationship between these indicators and grain yield (GY) was explored using the Mantel test and structural equation modeling (SEM).Results and discussionThe results found that the SP1 and SP2 treatments improved the assimilates production capacity of the canopy by increasing the leaf area index, total chlorophyll content, and net photosynthetic rate, improving dry matter accumulation (DMA) by 6.2%-10.6%, compared to the SP0 treatment. SP1 and SP2 treatments increased total soluble sugars, starch, free amino acids, and soluble protein contents in ear leaves via increasing the enzymatic reactions related to C/NM in ear leaves during the reproductive growth stage compared with SP0 treatment. The highest plant nitrogen uptake (PNU) and nitrogen recovery efficiency were obtained under the SP2 treatment, and the GY and nitrogen agronomic efficiency were higher than the SP0 treatment by 9.2% and 27.8%. However, SP3 and SP4 treatments reduced DMA and GY by inhibiting C/NM in ear leaves compared to SP0 treatment. Mantel test and SEM results revealed that SP treatments indirectly increased GY and PNU by directly positively regulating C/NM in maize ear leaves. Therefore, in the semi-arid regions, substituting 30% of the chemical N with SP could be considered. This fertilizer regime may avoid GY reduction and improve NUE. This study provides new insights into sustainable cultivation pathways for maize in semi-arid regions.

Project description:Optimized nitrogen (N) management (OPT), with reduced total N input and more N applied during panicle development, has been proved to increase grain yield of rice through panicle enlargement. However, the changes in panicle architecture and source of variation are not well understood. A hybrid rice variety named Tianyou 3618 was subjected to OPT and farmer's fertilizer practice (FFP) in early cropping seasons of 2016 and 2017. With 16.7 % less N input, OPT increased panicle size by 8.6 % and 27.4 %, and grain yield by 13.8 % and 12.3 % for 2016 and 2017, respectively. OPT had greater dry matter accumulation and N uptake from panicle initiation to heading, which bolstered panicle enlargement. The number of surviving florets per branch was quite constant under different N treatments for all primary, secondary, and tertiary branches, implying that panicle size was mainly determined by the number of branches rather than the number of florets per branch. Little change was observed between OPT and FFP in differentiation, degeneration and survival of primary branches and their florets. Surviving secondary and tertiary branches and their florets were significantly more under OPT than those under FFP. The increase in surviving secondary branches under OPT resulted from both enhanced differentiation and reduced degeneration. While the increase in surviving tertiary branches under OPT was merely from enhanced differentiation though their degeneration was also dramatically increased. Among the increased differentiated florets under OPT, 32.4%-36.3 % and 61.6%-67.7 % came from secondary and tertiary branches, respectively. Among the increased surviving florets under OPT, 62.2%-65.2 % and 32.5%-37.8 % came from secondary and tertiary branches, respectively. Both secondary branches and tertiary branches were principal contributors to the increase in panicle size of OPT. To our knowledge, this is the first report on the detailed changes in panicle architecture and their involvement in panicle enlargement and yield gain under OPT.
