ABSTRACT: Amazon comprises a vast variety of ecosystems, including savannah-like Canga barrens that evolved on iron-lateritic rock plateaus of the Carajás Mountain range. Individual Cangas are enclosed by the rain forest, indicating insular isolation that enables speciation and plant community differentiation. To establish a framework for the research on natural history and conservation management of endemic Canga species, seven chloroplast DNA loci and an ITS2 nuclear DNA locus were used to study natural molecular variation of the red flowered Ipomoea cavalcantei and the lilac flowered I. marabaensis. Partitioning of the nuclear and chloroplast gene alleles strongly suggested that the species share the most recent common ancestor, pointing a new independent event of the red flower origin in the genus. Chloroplast gene allele analysis showed strong genetic differentiation between Canga populations, implying a limited role of seed dispersal in exchange of individuals between Cangas. Closed haplotype network topology indicated a requirement for the paternal inheritance in generation of cytoplasmic genetic variation. Tenfold higher nucleotide diversity in the nuclear ITS2 sequences distinguished I. cavalcantei from I. marabaensis, implying a different pace of evolutionary changes. Thus, Canga ecosystems offer powerful venues for the study of speciation, multitrait adaptation and the origins of genetic variation.

Project description:Ipomoea is a large pantropical genus globally distributed, which importance goes beyond the economic value as food resources or ornamental crops. This highly diverse genus has been the focus of a great number of studies, enriching the plant genomics knowledge, and challenging the plant evolution models. In the Carajás mountain range, located in Eastern Amazon, the savannah-like ferruginous ecosystem known as canga harbors highly specialized plant and animal populations, and Ipomoea is substantially representative in such restrictive habitat. Thus, to provide genetic data and insights into whole plastome phylogenetic relationships among key Ipomoea species from Eastern Amazon with little to none previously available data, we present the complete plastome sequences of twelve lineages of the genus, including the canga microendemic I. cavalcantei, the closely related I. marabaensis, and their putative hybrids. The twelve plastomes presented similar gene content as most publicly available Ipomoea plastomes, although the putative hybrids were correctly placed as closely related to the two parental species. The cavalcantei-marabaensis group was consistently grouped between phylogenetic methods. The closer relationship of the I. carnea plastome with the cavalcantei-marabaensis group, as well as the branch formed by I. quamoclit, I. asarifolia and I. maurandioides, were probably a consequence of insufficient taxonomic representativity, instead of true genetic closeness, reinforcing the importance of new plastome assemblies to resolve inconsistencies and boost statistical confidence, especially the case for South American clades of Ipomoea. The search for k-mers presenting high dispersion among the frequency distributions pointed to highly variable coding and intergenic regions, which may potentially contribute to the genetic diversity observed at species level. Our results contribute to the resolution of uncertain clades within Ipomoea and future phylogenomic studies, bringing unprecedented results to Ipomoea species with restricted distribution, such as I. cavalcantei.

Project description:Background and aimsLow-altitude mountains constitute important centres of diversity in landscapes with little topographic variation, such as the Southwest Australian Floristic Region (SWAFR). They also provide unique climatic and edaphic conditions that may allow them to function as refugia. We investigate whether the Porongurups (altitude 655 m) in the SWAFR will provide a refugium for the endemic Ornduffia calthifolia and O. marchantii under forecast climate change.MethodsWe used species distribution modelling based on WorldClim climatic data, 30-m elevation data and a 2-m-resolution LiDAR-derived digital elevation model (DEM) to predict current and future distributions of the Ornduffia species at local and regional scales based on 605 field-based abundance estimates. Future distributions were forecast using RCP2.6 and RCP4.5 projections. To determine whether local edaphic and biotic factors impact these forecasts, we tested whether soil depth and vegetation height were significant predictors of abundance using generalized additive models (GAMs).Key resultsSpecies distribution modelling revealed the importance of elevation and topographic variables at the local scale for determining distributions of both species, which also preferred shadier locations and higher slopes. However, O. calthifolia occurred at higher (cooler) elevations with rugged, concave topography, while O. marchantii occurred in disturbed sites at lower locations with less rugged, convex topography. Under future climates both species are likely to severely contract under the milder RCP2.6 projection (approx. 2 °C of global warming), but are unlikely to persist if warming is more severe (RCP4.5). GAMs showed that soil depth and vegetation height are important predictors of O. calthifolia and O. marchantii distributions, respectively.ConclusionsThe Porongurups constitute an important refugium for O. calthifolia and O. marchantii, but limits to this capacity may be reached if global warming exceeds 2 °C. This capacity is moderated at local scales by biotic and edaphic factors.

Project description:Background and aimsThe Balkan Peninsula is one of the most important centres of plant diversity in Europe. Here we aim to ﬁll the gap in the current knowledge of the evolutionary processes and factors modelling this astonishing biological richness by applying multiple approaches to the Cyanus napulifer group.MethodsTo reconstruct the mode of diversiﬁcation within the C. napulifer group and to uncover its relationships with potential relatives with x = 10 from Europe and Northern Africa, we examined variation in genetic markers (amplified fragment length polymorphisms [AFLPs]; 460 individuals), relative DNA content (4',6-diamidino-2-phenylindole [DAPI] flow cytometry, 330 individuals) and morphology (multivariate morphometrics, 40 morphological characters, 710 individuals). To elucidate its evolutionary history, we analysed chloroplast DNA (cpDNA) sequences of the genus Cyanus deposited in the GenBank database.Key resultsThe AFLPs revealed a suite of closely related entities with variable levels of differentiation. The C. napulifer group formed a genetically well-defined unit. Samples outside the group formed strongly diversified and mostly species-specific genetic lineages with no further geographical patterns, often characterized also by a different DNA content. AFLP analysis of the C. napulifer group revealed extensive radiation and split it into nine allopatric (sub)lineages with varying degrees of congruence among genetic, DNA-content and morphological patterns. Genetic admixture was usually detected in contact zones between genetic lineages. Plastid data indicated extensive maintenance of ancestral variation across Cyanus perennials.ConclusionThe C. napulifer group is an example of a rapidly and recently diversified plant group whose genetic lineages have evolved in spatio-temporal isolation on the topographically complex Balkan Peninsula. Adaptive radiation, accompanied in some cases by long-term isolation and hybridization, has contributed to the formation of this species complex and its mosaic pattern.
