Induced gamma band activity from EEG as a possible index of training-related brain plasticity in motor tasks.

ABSTRACT: The aim of this study was proposing gamma band activity (GBA) as an index of training-related brain plasticity in the motor cortex. Sixteen controls underwent an experimental session where electroencephalography (EEG) activity was recorded at baseline (resting) and during a motor task (hand movements). GBA was obtained from the EEG data at baseline and during the task. Index of plasticity (IP) was defined as the relationship between GBA at the end of the motor task (GBAM_FIN), divided by GBA at the beginning of the task (GBAM_INI) for movements of both hands. There was a significant increase in GBA at the end of the task, compared to the initial GBA for the motor task (GBAM_FIN > GBAM_INI). No differences were found at baseline (GBAB_FIN ≈ GBAB_INI). Individual IP values had a positive (r = 0.624) and significant correlation with subject's handedness. Due to plastic changes, GBA could indirectly but objectively reveal changes in cerebral activity related to physical training. This method could be used as a future diagnostic test in the follow-up of patients undergoing rehabilitation. It could also have potential applications in the fields of sports medicine.

Project description:Gamma entrainment has been shown to enhance beta amyloid (Aβ) uptake in mouse models of Alzheimer's disease (AD) as well as improve cognitive symptoms of dementia in both humans and mice. Similar improvements have been reported for both invasive and non-invasive brain stimulation in the gamma oscillatory band, with 40 Hz auditory and visual sensory stimulants employed in non-invasive approaches. Non-invasive stimulation techniques possess the clear advantage of not requiring surgical procedures and can hence be applicable to a wider set of patients. The dataset introduced here was acquired with the aim of examining the network-level mechanisms governing the production of the brain's oscillatory activity during non-invasive auditory gamma-band stimulation, and thereby helping to explain the reported therapeutic effects of entrainment in AD patients. Thirteen elderly participants with memory complaints whose conditions were diagnosed as normal aging (non-AD) or mild AD based on the standard criteria for the diagnosis of AD including the mini-mental state exam (MMSE) took part in data collection in which EEG signals were recorded during auditory stimulation of the brain. The data collection session consisted of an initial one-minute rest followed by an alternating set of six stimulation trials interleaved with five rest trials. During each stimulation trial, an auditory stimulant in the form of a 40 Hz chirp was presented to the participant. The collected data from all participants were preprocessed following the full pipeline of Makoto with the use of EEGLAB and posted as a dataset named: Auditory Gamma Entrainment at OpenNeuro repository. The data record for each participant includes the EEG signal represented in standard BIDS format for one-minute rest followed by the auditory task data. A copy of the source EEG data is also provided in .txt format. The dataset can be used to study the characteristics of brain oscillations during entrainment, as well as for studies on auditory perception, analysis of resting state potentials in dementia patients, comparison of auditory evoked potentials with resting state potentials, ERP, ERSP, and SSAVP analysis of auditory response in dementia patients, time series analysis of the stimulation and rest trials, and brain connectivity analysis in dementia patients.

Project description:High gamma activity (HGA) is a crucial biomarker for functional brain mapping, particularly in sensorimotor areas, to preserve functionality after brain surgeries. HGA mapping paradigms typically involve multiple task blocks alternating with resting (R) conditions, where each block comprises consecutive tasks under nonresting (NR) conditions. However, the repetitive nature of these tasks may lead to attenuation due to repetition suppression, potentially compromising the accuracy of HGA mapping. This study tests the hypothesis that repetitive grasping paradigms result in attenuated HGA over time in sensorimotor areas. It explores the temporal and spatial characteristics of this attenuation to optimize electrocorticography (ECoG) HGA protocols and enhance result interpretation. Eleven consecutive patients who underwent surgical treatment of intractable epilepsy or malignant glioma were included in this study. Intracranial electrode locations on the pre- and postcentral gyrus were considered regions of interest (ROI). Each patient performed ten blocks of ten consecutive grasping trials. The mean z-scored HGA (60-170 Hz) across these trials was calculated, and attenuation was analyzed using the Kruskal-Wallis test. Obtained signals were also divided into three grouped periods for R and NR groups to assess short-term attenuation within movement blocks and long-term attenuation over multiple blocks. Electrode locations were mapped to the MNI152 (Montreal Neurological Institute) brain template to investigate the spatial distribution of attenuation. Distances from each electrode to the hand-knob region were compared between attenuated and nonattenuated electrodes. A total of 568 electrodes from 11 patients were analyzed, including 139 electrodes within the ROI. Thus, 60 electrodes demonstrated significant HGAs during the grasping task (p < 0.05). Sensorimotor HGA z-scores significantly attenuated over time during both consecutive grasping trials and repeated blocks. Short-term attenuation (25%, 15/60 electrodes in ROI) was more pronounced than long-term attenuation (15%, 9/60 electrodes in ROI). Notably, three patients undergoing intraoperative mapping demonstrated less short-term attenuation compared to long-term attenuation. Spatially, attenuated electrodes clustered around the hand-knob region of the precentral gyrus and adjacent areas of the postcentral gyrus. However, no significant differences were observed in the distances from electrodes to the hand-knob region between attenuated and nonattenuated electrodes. The present study showed that repetitive grasping tasks attenuated the HGA of significant electrodes in the sensorimotor area over time. Considering the findings with the characteristics can further improve the usability of ECoG mapping in terms of more precise results in the most reasonable mapping time.
