Cell volume change through water efflux impacts cell stiffness and stem cell fate.

ABSTRACT: Cells alter their mechanical properties in response to their local microenvironment; this plays a role in determining cell function and can even influence stem cell fate. Here, we identify a robust and unified relationship between cell stiffness and cell volume. As a cell spreads on a substrate, its volume decreases, while its stiffness concomitantly increases. We find that both cortical and cytoplasmic cell stiffness scale with volume for numerous perturbations, including varying substrate stiffness, cell spread area, and external osmotic pressure. The reduction of cell volume is a result of water efflux, which leads to a corresponding increase in intracellular molecular crowding. Furthermore, we find that changes in cell volume, and hence stiffness, alter stem-cell differentiation, regardless of the method by which these are induced. These observations reveal a surprising, previously unidentified relationship between cell stiffness and cell volume that strongly influences cell biology.

Project description:BackgroundHydrogels with tuneable mechanical properties are an attractive material platform for 3D bioprinting. Thus far, numerous studies have confirmed that the biophysical cues of hydrogels, such as stiffness, are known to have a profound impact on mesenchymal stem cell (MSC) differentiation; however, their differentiation potential within 3D-bioprinted hydrogels is not completely understood. Here, we propose a protocol for the exploration of how the stiffness of alginate-gelatin (Alg-Gel) composite hydrogels (the widely used bioink) affects the differentiation of MSCs in the presence or absence of differentiation inducing factors.MethodsTwo types of Alg-Gel composite hydrogels (Young's modulus: 50 kPa vs. 225 kPa) were bioprinted independently of porosity. Then, stiffness-induced biases towards adipogenic and osteogenic differentiation of the embedded MSCs were analysed by co-staining with alkaline phosphatase (ALP) and oil red O. The expression of specific markers at the gene level was detected after a 3-day culture.ResultsConfocal microscopy indicated that all tested hydrogels supported MSC growth and viability during the culture period. Higher expression of adipogenic and osteogenic markers (ALP and lipoprotein lipase (LPL)) in stiffer 3D-bioprinted matrices demonstrated a more significant response of MSCs to stiffer hydrogels with respect to differentiation, which was more robust in differentiation-inducing medium. However, the LPL expression in stiffer 3D-bioprinted constructs was reduced at day 3 regardless of the presence of differentiation-inducing factors. Although MSCs embedded in softer hydrogels to some extent proceeded toward adipogenic and osteogenic lineages within a few days, their differentiation seemed to be slower and more limited. Interestingly, the hydrogel itself (without differentiation-inducing factors) exhibited a slight effect on whether MSCs differentiated towards an adipogenic or an osteogenic fate. Considering that the mechano-regulated protein Yes-associated protein (YAP) is involved in MSC fate decisions, we further found that inhibition of YAP significantly downregulated the expression of ALP and LPL in MSCs in stiffer constructs regardless of the induced growth factors present.ConclusionsThese results demonstrate that the differentiation of MSCs in 3D-bioprinted matrices is dependent on hydrogel stiffness, which emphasizes the importance of biophysical cues as a determinant of cellular behaviour.

Project description:The export of agrochemicals and their transformation products (TPs) following their application in the agricultural fields poses a threat to water quality. Future changes in climatic conditions (e.g. extreme weather events such as heavy rainfall or extended dry periods) could alter the degradation and mobility of agrochemicals. In this research, we use an integrated modelling framework to understand the impact of extreme climate events on the fate and transport of the agrochemical S-Metolachlor and two of its TPs (M-OXA, Metolachlor Oxanilic Acid and M-ESA, Metolachlor Ethyl Sulfonic Acid). This is done by coupling climate model outputs to the Zin-AgriTra agrochemical reactive transport model in four simulation scenarios. 1) Reference (2015-2018), 2) Very dry (2038-2041), 3) Very wet (2054-2057) and 4) High temperature (2096-2099) conditions of a selected RCP8.5 based regional climate scenario. The modelling framework is tested on an agricultural catchment, Wulka, in Burgenland, Austria. The model results indicate that 13-14 % of applied S-Metolachlor is retained in the soil, and around 85 % is degraded into TPs in the different scenarios. In very dry and high-temperature scenarios, degradation is higher, and hence, there is less S-Metolachlor in the soil. However, a large share of formed M-OXA and M-ESA are retained in the soil, which is transported via overland and groundwater flow, leading to a build-up effect in M-OXA and M-ESA river concentrations over the years. Though a small share of S-Metolachlor and TPs are transported to rivers, their river export is affected by the intensity and amount of rainfall. The very wet and high-temperature scenarios show higher S-Metolachlor and TP concentrations at the catchment outlet due to higher river discharge. The reference scenario shows higher river peak concentrations associated with higher overland flow caused by measured hourly rainfall compared to disaggregated daily precipitation data in the other scenarios.

Project description:Osteoclasts ubiquitously participate in bone homeostasis, and their aberration leads to bone diseases, such as osteoporosis. Current clinical strategies by biochemical signaling molecules often perturb innate bone metabolism owing to the uncontrolled management of osteoclasts. Thus, an alternative strategy of precise regulation for osteoclast differentiation is urgently needed. To this end, this study proposed an assumption that mechanic stimulation might be a potential strategy. Here, a hydrogel was created to imitate the physiological bone microenvironment, with stiffnesses ranging from 2.43kPa to 68.2kPa. The impact of matrix stiffness on osteoclast behaviors was thoroughly investigated. Results showed that matrix stiffness could be harnessed for directing osteoclast fate in vitro and in vivo. In particular, increased matrix stiffness inhibited the integrin β3-responsive RhoA-ROCK2-YAP-related mechanotransduction and promoted osteoclastogenesis. Notably, preosteoclast development is facilitated by medium-stiffness hydrogel (M-gel) possessing the same stiffness as vessel ranging from 17.5 kPa to 44.6 kPa by partial suppression of mechanotransduction, which subsequently encouraged revascularization and bone regeneration in mice with bone defects. Our works provide an innovative approach for finely regulating osteoclast differentiation by selecting the optimum matrix stiffness and enable us further to develop a matrix stiffness-based strategy for bone tissue engineering.
