Pulsed Corona Discharge Induced Hydroxyl Radical Transfer Through the Gas-Liquid Interface.

ABSTRACT: The highly energetic electrons in non-thermal plasma generated by gas phase pulsed corona discharge (PCD) produce hydroxyl (OH) radicals via collision reactions with water molecules. Previous work has established that OH radicals are formed at the plasma-liquid interface, making it an important location for the oxidation of aqueous pollutants. Here, by contacting water as aerosol with PCD plasma, it is shown that OH radicals are produced on the gas side of the interface, and not in the liquid phase. It is also demonstrated that the gas-liquid interfacial boundary poses a barrier for the OH radicals, one they need to cross for reactive affinity with dissolved components, and that this process requires a gaseous atomic H scavenger. For gaseous oxidation, a scavenger, oxygen in common cases, is an advantage but not a requirement. OH radical efficiency in liquid phase reactions is strongly temperature dependent as radical termination reaction rates increase with temperature.

Optimum Operating Frequency for Electrocoalescence Induced by Pulsed Corona Discharge.

Project description:Current oil-water separation methods require a significant power, a high processing time, and costly equipment, which typically yield low treatment efficiency. Pulsed direct current (dc) electric fields and recently nonuniform electric fields caught considerable attention in the petroleum industry research in order to address the most common oil-water separation issues such as chain formation, partial coalescence, and low efficiency in either the energy consumption or coalescence rate. Here, a contact-less charge injection method induced by corona discharge is utilized to investigate the impacts of nonuniform and pulsed dc electric fields on the coalescence of water droplets inside an oil medium. The operating process parameters were experimentally calibrated and optimized with the goal of increasing the effectiveness and energy consumption efficiency of the coalescence process. High-speed imaging and image processing techniques were used in order to investigate the effect of different active forces (i.e., dipole-dipole interaction, migratory coalescence, or electrophoresis, and dielectrophoresis) during the coalescence process. Different pulsed dc frequencies and pure dc waveforms were utilized and their impact on the coalescence of water droplets was investigated. An optimal coalescence recipe was proposed by continuous measurement of the distance, velocity, and acceleration of the coalescing water droplets. The results of this study suggest use of pulsed dc and pure dc electric fields for coalescence of water droplets in concentrated and dispersed emulsions, respectively.

Pulsed electron beam water radiolysis for submicrosecond hydroxyl radical protein footprinting.

Project description:Hydroxyl radical footprinting is a valuable technique for studying protein structure, but care must be taken to ensure that the protein does not unfold during the labeling process due to oxidative damage. Footprinting methods based on submicrosecond laser photolysis of peroxide that complete the labeling process faster than the protein can unfold have been recently described; however, the mere presence of large amounts of hydrogen peroxide can also cause uncontrolled oxidation and minor conformational changes. We have developed a novel method for submicrosecond hydroxyl radical protein footprinting using a pulsed electron beam from a 2 MeV Van de Graaff electron accelerator to generate a high concentration of hydroxyl radicals by radiolysis of water. The amount of oxidation can be controlled by buffer composition, pulsewidth, dose, and dissolved nitrous oxide gas in the sample. Our results with ubiquitin and beta-lactoglobulin A demonstrate that one submicrosecond electron beam pulse produces extensive protein surface modifications. Highly reactive residues that are buried within the protein structure are not oxidized, indicating that the protein retains its folded structure during the labeling process. Time-resolved spectroscopy indicates that the major part of protein oxidation is complete in a time scale shorter than that of large scale protein motions.

Gas-phase amination of aromatic hydrocarbons by corona discharge-assisted nitrogen fixation.

Project description:This paper reports on the gas-phase amination reaction of aromatic hydrocarbons occurring under corona discharge conditions with N2 gas as the nitrogen source. The corona discharge device within an atmospheric pressure chemical ionization source was employed to achieve the plasma-assisted N2 fixation, and the coupled ion trap mass spectrometer (IT-MS) was used to detect positively charged product ions. In the model case, under APCI conditions, unusual product ions, [M + 16]+ and [M + 14]+, were observed. Based on the high resolution MS data and tandem mass spectrometric information, [M + 16]+ was confirmed to be protonated p-toluidine and [M + 14]+ was confirmed to be p-methylphenylnitrenium ion. According to the experimental results of the isotopic labelling and substituent effect, one feasible mechanism is proposed as follows. Firstly, N2 is activated by plasma caused via the corona discharge and then electrophilically attacks toluene, yielding the key intermediate, p-methylphenylnitrenium; secondly, the intermediate undergoes double-hydrogen transfer reaction to give rise to the final product ion, protonated p-toluidine. This study may provide a novel idea to explore new and green method for the synthesis of anilines.

Hydroxyl Radical-Coupled Electron-Transfer Mechanism of Flavin-Dependent Hydroxylases.

Project description:Class A flavin-dependent hydroxylases (FdHs) catalyze the hydroxylation of organic compounds in a site- and stereoselective manner. In stark contrast, conventional synthetic routes require environmentally hazardous reagents and give modest yields. Thus, understanding the detailed mechanism of this class of enzymes is essential to their rational manipulation for applications in green chemistry and pharmaceutical production. Both electrophilic substitution and radical intermediate mechanisms have been proposed as interpretations of FdH hydroxylation rates and optical spectra. While radical mechanistic steps are often difficult to examine directly, modern quantum chemistry calculations combined with statistical mechanical approaches can yield detailed mechanistic models providing insights that can be used to differentiate reaction pathways. In the current work, we report quantum mechanical/molecular mechanical (QM/MM) calculations on the fungal TropB enzyme that shows an alternative reaction pathway in which hydroxylation through a hydroxyl radical-coupled electron-transfer mechanism is significantly favored over electrophilic substitution. Furthermore, QM/MM calculations on several modified flavins provide a more consistent interpretation of the experimental trends in the reaction rates seen experimentally for a related enzyme, para-hydroxybenzoate hydroxylase. These calculations should guide future enzyme and substrate design strategies and broaden the scope of biological spin chemistry.

High structural resolution hydroxyl radical protein footprinting reveals an extended Robo1-heparin binding interface.

Project description:Interaction of transmembrane receptors of the Robo family and the secreted protein Slit provides important signals in the development of the central nervous system and regulation of axonal midline crossing. Heparan sulfate, a sulfated linear polysaccharide modified in a complex variety of ways, serves as an essential co-receptor in Slit-Robo signaling. Previous studies have shown that closely related heparin octasaccharides bind to Drosophila Robo directly, and surface plasmon resonance analysis revealed that Robo1 binds more tightly to full-length unfractionated heparin. For the first time, we utilized electron transfer dissociation-based high spatial resolution hydroxyl radical protein footprinting to identify two separate binding sites for heparin interaction with Robo1: one binding site at the previously identified site for heparin dp8 and a second binding site at the N terminus of Robo1 that is disordered in the x-ray crystal structure. Mutagenesis of the identified N-terminal binding site exhibited a decrease in binding affinity as measured by surface plasmon resonance and heparin affinity chromatography. Footprinting also indicated that heparin binding induces a minor change in the conformation and/or dynamics of the Ig2 domain, but no major conformational changes were detected. These results indicate a second low affinity binding site in the Robo-Slit complex as well as suggesting the role of the Ig2 domain of Robo1 in heparin-mediated signal transduction. This study also marks the first use of electron transfer dissociation-based high spatial resolution hydroxyl radical protein footprinting, which shows great utility for the characterization of protein-carbohydrate complexes.

Oxyl and hydroxyl radical transfer in mitochondrial amidoxime reducing component-catalyzed nitrite reduction.

Project description:A combination of electron paramagnetic resonance (EPR) spectroscopy and computational approaches has provided insight into the nature of the reaction coordinate for the one-electron reduction of nitrite by the mitochondrial amidoxime reducing component (mARC) enzyme. The results show that a paramagnetic Mo(V) species is generated when reduced enzyme is exposed to nitrite, and an analysis of the resulting EPR hyperfine parameters confirms that mARC is remarkably similar to the low-pH form of sulfite oxidase. Two mechanisms for nitrite reduction have been considered. The first shows a modest reaction barrier of 14 kcal/mol for the formation of ·NO from unprotonated nitrite substrate. In marked contrast, protonation of the substrate oxygen proximal to Mo in the Mo(IV)-O-N-O substrate-bound species results in barrierless conversion to products. A fragment orbital analysis reveals a high degree of Mo-O(H)-N-O covalency that provides a π-orbital pathway for one-electron transfer to the substrate and defines orbital constraints on the Mo-substrate geometry for productive catalysis in mARC and other pyranopterin molybdenum enzymes that catalyze this one-electron transformation.

Interactions between gas-liquid mass transfer and bubble behaviours.

Project description:Interactions between gas-liquid mass transfer and bubble behaviours were investigated to improve the understanding of the relationship between the two sides. The CO2/N2-water system was applied to study the bubble behaviours based on the volume-of-fluid (VOF) model. The mass transfer conditions were taken into consideration when the fluid field was analysed. The bubble behaviours were compared with and without mass transfer. The results show that the absolute slopes of the curves for mass fraction inside the single rising bubbles, with diameters from 3 to 6 mm, decrease from 0.09325 to 0.02818. It means that small single bubbles have higher mass transfer efficiency. The daughter bubbles of cutting behaviour and initial side-by-side bubbles of coalescence behaviour also perform better than the initial large bubbles and coalesced bubbles, respectively. The bubble behaviours affect the mass transfer process. However, the latter also reacts upon the former. The critical intervals between the side-by-side bubbles decrease from 2.0 to 0.9 mm when the bubble diameter changes from 3 to 7 mm. For the coalescence behaviour without mass transfer, the critical intervals are larger because there is no influence of concentration around the bubbles on the bubble motion. The coalescence of cut daughter bubbles is also influenced by the concentration. It was suggested that the interaction between the gas-liquid mass transfer and bubble behaviours cannot be ignored.

Quantifying protein interface footprinting by hydroxyl radical oxidation and molecular dynamics simulation: application to galectin-1.

Project description:Biomolecular surface mapping methods offer an important alternative method for characterizing protein-protein and protein-ligand interactions in cases in which it is not possible to determine high-resolution three-dimensional (3D) structures of complexes. Hydroxyl radical footprinting offers a significant advance in footprint resolution compared with traditional chemical derivatization. Here we present results of footprinting performed with hydroxyl radicals generated on the nanosecond time scale by laser-induced photodissociation of hydrogen peroxide. We applied this emerging method to a carbohydrate-binding protein, galectin-1. Since galectin-1 occurs as a homodimer, footprinting was employed to characterize the interface of the monomeric subunits. Efficient analysis of the mass spectrometry data for the oxidized protein was achieved with the recently developed ByOnic (Palo Alto, CA) software that was altered to handle the large number of modifications arising from side-chain oxidation. Quantification of the level of oxidation has been achieved by employing spectral intensities for all of the observed oxidation states on a per-residue basis. The level of accuracy achievable from spectral intensities was determined by examination of mixtures of synthetic peptides related to those present after oxidation and tryptic digestion of galectin-1. A direct relationship between side-chain solvent accessibility and level of oxidation emerged, which enabled the prediction of the level of oxidation given the 3D structure of the protein. The precision of this relationship was enhanced through the use of average solvent accessibilities computed from 10 ns molecular dynamics simulations of the protein.

Reactivity of hydropersulfides toward the hydroxyl radical unraveled: disulfide bond cleavage, hydrogen atom transfer, and proton-coupled electron transfer.

Project description:Hydropersulfides (RSSH) are highly reactive as nucleophiles and hydrogen atom transfer reagents. These chemical properties are believed to be key for them to act as antioxidants in cells. The reaction involving the radical species and the disulfide bond (S-S) in RSSH, a known redox-active group, however, has been scarcely studied, resulting in an incomplete understanding of the chemical nature of RSSH. We have performed a high-level theoretical investigation on the reactions of the hydroxyl radical (?OH) toward a set of RSSH (R = -H, -CH3, -NH2, -C(O)OH, -CN, and -NO2). The results show that S-S cleavage and H-atom abstraction are the two competing channels. The electron inductive effect of R induces selective ?OH substitution at one sulfur atom upon S-S cleavage, forming RSOH and ?SH for the electron donating groups (EDGs), whereas producing HSOH and ?SR for the electron withdrawing groups (EWGs). The H-Atom abstraction by ?OH follows a classical hydrogen atom transfer (hat) mechanism, producing RSS? and H2O. Surprisingly, a proton-coupled electron transfer (pcet) process also occurs for R being an EDG. Although for RSSH having EWGs hat is the leading channel, S-S cleavage can be competitive or even dominant for the EDGs. The overall reactivity of RSSH toward ?OH attack is greatly enhanced with the presence of an EDG, with CH3SSH being the most reactive species found in this study (overall rate constant: 4.55 × 1012 M-1 s-1). Our results highlight the complexity in RSSH reaction chemistry, the extent of which is closely modulated by the inductive effect of the substituents in the case of the oxidation by hydroxyl radicals.

Study by Optical Spectroscopy of Bismuth Emission in a Nanosecond-Pulsed Discharge Created in Liquid Nitrogen.

Project description:Time-resolved optical emission spectroscopy of nanosecond-pulsed discharges ignited in liquid nitrogen between two bismuth electrodes is used to determine the main discharge parameters (electron temperature, electron density and optical thickness). Nineteen lines belonging to the Bi I system and seven to the Bi II system could be recorded by directly plunging the optical fibre into the liquid in close vicinity to the discharge. The lack of data for the Stark parameters to evaluate the broadening of the Bi I lines was solved by taking advantage of the time-resolved information supported by each line to determine them. The electron density was found to decrease exponentially from 6.5 ± 1.5 × 1016 cm-3 200 ns after ignition to 1.0 ± 0.5 × 1016 cm-3 after 1050 ns. The electron temperature was found to be 0.35 eV, close to the value given by Saha's equation.
