Association between CD40 rs1883832 and immune-related diseases susceptibility: A meta-analysis.

ABSTRACT:

Background/objective

It has been reported that CD40 rs1883832 might be associated with immune-related diseases susceptibility. Owing to mixed and inconclusive results, we conducted a meta-analysis of case-control studies to summarize and clarify this association.

Methods/main results

A systematic search of studies on the association between CD40 rs1883832 and immune-related diseases susceptibility was conducted in databases. Odds ratios and 95% confidence intervals were used to pool the effect size. 40 articles were included in our meta-analysis.

Conclusions

CD40 rs1883832 is associated with decreased risk of Graves' disease, especially in Asian; CD40 rs1883832 is associated with increased risk of multiple sclerosis; CD40 -1C>T (rs1883832) is not associated with the susceptibility of Hashimoto's thyroiditis, systemic sclerosis or Asthma; there is insufficient data to fully confirm the association between CD40 rs1883832 and systemic lupus erythematosus (SLE), rheumatoid arthritis (RA), Behçet's disease (BD), myasthenia gravis (MG), Crohn's disease (CD), ulcerative colitis (UC), Sarcoidosis, Fuch uveitis syndrome (FUS), Vogt-Koyanagi-Harada syndrome (VKH), Kawasaki disease (KD), giant cell arteritis (GCA) or Immune thrombocytopenia (ITP).

Project description:BackgroundAllergic diseases are type I hypersensitivity reactions mediated by various allergens. The most common allergic diseases include allergic rhinitis, allergic asthma, allergic dermatitis, and allergic conjunctivitis. The incidence of allergic diseases has been increasing in the recent past, and allergen avoidance and adoption of desensitization treatment can significantly decrease the incidence of allergic diseases. Previous studies have explored the association between vitamin A supplementation and allergic diseases; however, the results are inconsistency. The aim of the present study was to evaluate the association between vitamin A supplementation and allergic diseases, with a focus on atopy and wheezing.MethodsArticles reporting randomized controlled trials (RCTs) on the association of vitamin A supplementation and allergic diseases were retrieved from PubMed, Embase, Web of science, and China National Knowledge Infrastructure database from inception of to November 15, 2021. STATA 12.0 software was used for meta-analysis, sensitivity analysis and analysis of publication bias.ResultsSeven studies comprising 2201 participants met the inclusion criteria and were included in the meta-analysis. The findings showed that vitamin A supplementation was associated with increased risk of atopy in young females compared with the placebo [RR = 1.70, 95% confidence interval (1.20, 2.41), P = 0.171, I 2 = 43.4% fixed effect model]. The frequency of delayed atopy among adults was associated with vitamin A supplementation (MD = 0.46, 95% CI = 0.04, 0.88). Analysis showed no significant association between vitamin A supplementation with incidence of wheezing in children [RR = 1.40, 95% CI (0.49, 3.98), P = 0.018, I 2 = 82.1% random effect model]. Sensitivity and publication bias analysis showed that each individual study did not affect the combined results and there was no significant publication bias among the studies.ConclusionThe findings showed that vitamin A supplementation is associated with increased risk of atopy but no correlation was observed with the incidence of wheezing. The results of this meta-analysis provide evidence for effective management of fibrosis. More studies should be conducted to verify the results.

Project description:IntroductionThe most prevalent head and neck cancer type is laryngeal cancer. Laryngeal cancer susceptibility is increased by a combination of genetic variables and environmental factors. Genetic predispositions that influence the functioning of the immune system can affect tumor development. Our study investigates the impact of alterations in CD40 (rs1883832) and CD40L (rs1126535) genes and the levels of their proteins on the development of laryngeal cancer.Materials and methodsThe PCR-RFLP method was used for genotyping SNPs in 96 patients with laryngeal cancer and 127 healthy individuals. Additionally, ELISA was utilized to measure circulating levels of sCD40 and sCD40L.ResultsWe identified a significant difference in the genotype distribution of CD40 (rs1883832) between laryngeal cancer patients and healthy individuals (p = 0.05). The C allele was dominant, and the CC genotype was more frequently observed in patients with laryngeal cancer (OR: 2.34, 95% CI: 0.98-5.54). In contrast, no statistically significant difference in the genotypes of CD40L (rs1126535) was detected between laryngeal cancer patients and the control group (p = 0.12). Additionally, no significant differences in serum sCD40 or sCD40L levels were observed between the groups (p = 0.48 and p = 0.15, respectively). However, a moderate positive correlation was found between sCD40 and sCD40L levels in the laryngeal cancer group (r = 0.52, p<0.01), a relationship that was not observed in the control group.DiscussionAccording to the current findings, it is suggested that the CD40 (rs1883832) gene variation found in patients may indicate an individual's susceptibility to developing laryngeal cancer. On the other hand, CD40L (rs1126535) seems to not play a significant role. While serum sCD40 and sCD40L levels did not show significant differences between patients and controls, the correlation in cancer patients suggests that these markers may be relevant in tumor progression. Further research is required to clarify the functional implications of these genetic variants and their potential use as biomarkers for laryngeal cancer.
