Changes in the allocation of endogenous strigolactone improve plant biomass production on phosphate-poor soils.

ABSTRACT: Strigolactones (SLs) are carotenoid-derived phytohormones shaping plant architecture and inducing the symbiosis with endomycorrhizal fungi. In Petunia hybrida, SL transport within the plant and towards the rhizosphere is driven by the ABCG-class protein PDR1. PDR1 expression is regulated by phytohormones and by the soil phosphate abundance, and thus SL transport integrates plant development with nutrient conditions. We overexpressed PDR1 (PDR1 OE) to investigate whether increased endogenous SL transport is sufficient to improve plant nutrition and productivity. Phosphorus quantification and nondestructive X-ray computed tomography were applied. Morphological and gene expression changes were quantified at cellular and whole tissue levels via time-lapse microscopy and quantitative PCR. PDR1 OE significantly enhanced phosphate uptake and plant biomass production on phosphate-poor soils. PDR1 OE plants showed increased lateral root formation, extended root hair elongation, faster mycorrhization and reduced leaf senescence. PDR1 overexpression allowed considerable SL biosynthesis by releasing SL biosynthetic genes from an SL-dependent negative feedback. The increased endogenous SL transport/biosynthesis in PDR1 OE plants is a powerful tool to improve plant growth on phosphate-poor soils. We propose PDR1 as an as yet unexplored trait to be investigated for crop production. The overexpression of PDR1 is a valuable strategy to investigate SL functions and transport routes.

Project description:Carbon-rich peat soils have been drained and used extensively for agriculture throughout human history, leading to significant losses of their soil carbon. One solution for rewetting degraded peat is wet crop cultivation. Crops such as rice, which can grow in water-saturated conditions, could enable agricultural production to be maintained whilst reducing CO2 and N2O emissions from peat. However, wet rice cultivation can release considerable methane (CH4). Water table and soil management strategies may enhance rice yield and minimize CH4 emissions, but they also influence plant biomass allocation strategies. It remains unclear how water and soil management influences rice allocation strategies and how changing plant allocation and associated traits, particularly belowground, influence CH4-related processes. We examined belowground biomass (BGB), aboveground biomass (AGB), belowground:aboveground ratio (BGB:ABG), and a range of root traits (root length, root diameter, root volume, root area, and specific root length) under different soil and water treatments; and evaluated plant trait linkages to CH4. Rice (Oryza sativa L.) was grown for six months in field mesocosms under high (saturated) or low water table treatments, and in either degraded peat soil or degraded peat covered with mineral soil. We found that BGB and BGB:AGB were lowest in water saturated conditions where mineral soil had been added to the peat, and highest in low-water table peat soils. Furthermore, CH4 and BGB were positively related, with BGB explaining 60% of the variation in CH4 but only under low water table conditions. Our results suggest that a mix of low water table and mineral soil addition could minimize belowground plant allocation in rice, which could further lower CH4 likely because root-derived carbon is a key substrate for methanogenesis. Minimizing root allocation, in conjunction with water and soil management, could be explored as a strategy for lowering CH4 emissions from wet rice cultivation in degraded peatlands.

Project description:In theory, changes in the amount of rainfall can change plant biomass allocation and subsequently influence coupled plant-soil microbial processes. However, testing patterns of combined responses of plants and soils remains a knowledge gap for terrestrial ecosystems. We carried out a comprehensive review of the available literature and conducted a meta-analysis to explore combined plant and soil microbial responses in grasslands exposed to experimental precipitation changes. We measured the effects of experimental precipitation changes on plant biomass, biomass allocation, and soil microbial biomass and tested for trade-offs between plant and soil responses to altered precipitation. We found that aboveground and belowground plant biomass responded asynchronically to precipitation changes, thereby leading to shifts in plant biomass allocation. Belowground plant biomass did not change under precipitation changes, but aboveground plant biomass decreased in precipitation reduction and increased in precipitation addition. There was a trade-off between responses of aboveground plant biomass and belowground plant biomass to precipitation reduction, but correlation wasn't found for precipitation addition. Microbial biomass carbon (C) did not change under the treatments of precipitation reduction. Increased root allocation may buffer drought stress for soil microbes through root exudations and neutralize microbial responses to precipitation reduction. However, precipitation addition increased microbial biomass C, potentially reflecting the removal of water limitation for soil microbial growth. We found that there were positive correlations between responses of aboveground plant biomass and microbial biomass C to precipitation addition, indicating that increased shoot growth probably promoted microbial responses via litter inputs. In sum, our study suggested that aboveground, belowground plant biomass and soil microbial biomass can respond asynchronically to precipitation changes, and emphasizes that testing the plant-soil system as a whole is necessary for forecasting the effects of precipitation changes on grassland systems.

Project description:Soil heterogeneity significantly affects plant dynamics such as plant growth and biomass. Most studies developed soil heterogeneity in two dimensions, i.e. either horizontally or vertically. However, soil heterogeneity in natural ecosystems varies both horizontally and vertically, i.e. in three dimensions. Previous studies on plant biomass and biomass allocation rarely considered the joint effects of soil heterogeneity and species composition. Thus, to investigate such joint effects on plant biomass and biomass allocation, a controlled experiment was conducted, where three levels of soil heterogeneity and seven types of species compositions were applied. Such soil heterogeneity was developed by filling nutrient-rich and nutrient-poor substrates in an alternative pattern in pots with different patch sizes (small, medium or large), and species compositions was achieved by applying three plant species (i.e. Festuca elata, Bromus inermis, Elymus breviaristatus) in all possible combinations (growing either in monoculture or in mixtures). Results showed that patch size significantly impacted plant biomass and biomass allocation, which differed among plant species. Specially, at the pot scale, with increasing patch size, shoot biomass decreased, while root biomass and R:S ratio increased, and total biomass tended to show a unimodal pattern, where the medium patch supported higher total biomass. Moreover, at the substrate scale, more shoot biomass and total biomass were found in nutrient-rich substrate. Furthermore, at the community scale, two of the three target plant species growing in monoculture had more shoot biomass than those growing together with other species. Thus, our results indicate soil heterogeneity significantly affected plant biomass and biomass allocation, which differ among plant species, though more research is needed on the generalization on biomass allocation. We propose that soil heterogeneity should be considered more explicitly in studies with more species in long-term experiments.
