5000 years of dietary variations of prehistoric farmers in the Great Hungarian Plain.

ABSTRACT: The development of farming was a catalyst for the evolution of the human diet from the varied subsistence practices of hunter-gatherers to the more globalised food economy we depend upon today. Although there has been considerable research into the dietary changes associated with the initial spread of farming, less attention has been given to how dietary choices continued to develop during subsequent millennia. A paleogenomic time transect for 5 millennia of human occupation in the Great Hungarian Plain spanning from the advent of the Neolithic to the Iron Age, showed major genomic turnovers. Here we assess where these genetic turnovers are associated with corresponding dietary shifts, by examining the carbon and nitrogen stable isotope ratios of 52 individuals. Results provide evidence that early Neolithic individuals, which were genetically characterised as Mesolithic hunter-gatherers, relied on wild resources to a greater extent than those whose genomic attributes were of typical Neolithic European farmers. Other Neolithic individuals and those from the Copper Age to Bronze Age periods relied mostly on terrestrial C3 plant resources. We also report a carbon isotopic ratio typical of C4 plants, which may indicate millet consumption in the Late Bronze Age, despite suggestions of the crop's earlier arrival in Europe during the Neolithic.

Project description:BackgroundHereditary angioedema (HAE) due to C1-inhibitor (C1-INH) deficiency (C1-INH-HAE) is a rare autosomal dominant disorder, characterized by recurrent, unpredictable edematous symptoms involving subcutaneous, and/or submucosal tissue. C1-INH-HAE may be caused by more than 700 different mutations in the gene encoding C1-INH (SERPING1) that may lead to decreased protein synthesis or to functional deficiency.MethodsConcentrations of C1-INH, C4, C1q, and anti-C1-INH antibodies, as well as functional C1-INH activity were determined in subjects suffering from edematous symptoms and admitted to the Hungarian Angioedema Center of Reference and Excellence. In those patients, who were diagnosed with C1-INH-HAE based on the complement measurements, SERPING1 was screened by bidirectional sequencing following PCR amplification and multiplex ligation-dependent probe amplification. For detecting large deletions, long-range PCRs covering the entire SERPING1 gene by targeting 2-7 kb long regions were applied.ResultsAltogether 197 individuals with C1-INH deficiency belonging to 68 families were identified. By applying Sanger sequencing or copy number determination of SERPING1 exons, 48 different mutations were detected in 66/68 families: 5 large and 15 small insertions/deletions/delins, 16 missense, 6 nonsense, and 6 intronic splice site mutations. Two novel variations (p.Tyr199Ser [c.596A>C] and the duplication of exon 7) were shown to cosegregate with deficient C1-inhibitor level and activity, while two other variations were detected in single patients (c.797_800delinsCTTGGAGCTCAAGAACTTGGAGCT and c.812dup). A series of long PCRs was applied in the remaining 2 families without an identified mutation and a new, 2606 bp long deletion including the last 91 bp of exon 6 (c.939_1029+2515del) was identified in all affected members of one pedigree. In the remaining one family, a deep intronic SERPING1 variation (c.1029+384A>G) was detected by a targeted next-generation sequencing panel as reported previously.ConclusionsSequencing and copy number determination of SERPING1 exons uncover most pathogenic variants in C1-INH-HAE patients, and further methods are worth to be applied in cases with unrevealed genetic background. Since knowledge of the genetic background may support the establishment of the correct and early diagnosis of C1-INH-HAE, identification of causative mutations and reporting data supporting the interpretation on the pathogenicity of these variants is of utmost importance.

Project description:Understanding the sources and dynamics of past biomass burning remain a significant challenge due to variations in paleofire combustion patterns across different temporal and spatial scales. This study integrates black carbon and polycyclic aromatic hydrocarbon (PAH) records from the Lubei Plain in the Shandong Peninsula, Lower Yellow River, to reconstruct Holocene fire regimes and their relationship with climatic shifts and human activities over the past 5000 years. During the mid-to-late Holocene (5000-3000 year BP. (calendar years before 1950)), a biomass burning levels were generally low, with a pronounced peak in low-molecular-weight PAHs (3-ring PAHs) and charcoal fluxes between 5000 and 4500 year BP, indicating increased fire activity likely driven by a short-term cold-dry event around 5000 year BP. From 3500 to 1000 year BP, three distinct episodes of low-temperature smoldering fires are identified, coinciding with deforestation and persistent droughts during the Shang Dynasty (3600-3046 year BP), the Qin and Western Han Dynasties (2200-2000 year BP), and the Sui and Tang Dynasties (1400-1100 year BP). In contrast, high-temperature flaming fires are associated with periods of intensified warfare and social upheaval, compounded by cold, arid climates during the Warring States period (2500-2400 year BP), the Eastern Han Dynasty (2000-1800 year BP), and the Wei, Jin, and Southern-Northern Dynasties (1800-1400 year BP). Over the past millennium, anthropogenic biomass burning remained elevated, reflecting sustained human influence on fire regimes. Meanwhile, Pollen and n-alkane records reveal a transition from primary forests to secondary shrubland during the late Holocene, driven by declining seasonal precipitation linked to a weakening East Asian monsoon and increased anthropogenic burning. Principal component analysis indicates that long-term deforestation primarily drove low-temperature smoldering fires, whereas high-temperature fires were more closely linked to periods of conflict. Seasonal precipitation variability, regulated by monsoonal dynamics, emerged as a fundamental control on fire regimes. This integrated analysis of PAHs, black carbon, and charcoal, coupled with multivariate statistical approaches, offers a robust framework for reconstructing fire-climate-human interactions in East Asia. The findings provide new insights into the mechanisms driving fire regimes and their long-term ecological and societal impacts.
