Characteristics of amino acids in the surface sediments of the Changjiang Estuary, China.

ABSTRACT: Amino acids (AAs) constitute the largest reservoir of organic nitrogen in most marine organisms and are a major fraction of characterized carbon in marine particulate matter. AAs are also useful indicators of degradation in the marine environment. This article reports original content data of 17 individual amino acids on 47 surface sediment samples and 18 suspended particulate samples in the water column in Changjiang Estuary. Contents of amino acids in suspended particulates were much higher than that in the surface sediments. The data are supplemental to our research paper, entitled "Organic matter degradation in surface sediments of the Changjiang Estuary: Evidence from amino acids" (Wang et al., 2018) [1]. The data are very useful to researchers and policy makers on estuarine benthic environment and ecology.

Bacterial diversity in the surface sediments of the hypoxic zone near the Changjiang Estuary and in the East China Sea.

Project description:Changjiang (Yangtze River) Estuary has experienced severe hypoxia since the 1950s. In order to investigate potential ecological functions of key microorganisms in relation to hypoxia, we performed 16S rRNA-based Illumina Miseq sequencing to explore the bacterial diversity in the surface sediments of the hypoxic zone near the Changjiang Estuary and in the East China Sea (ECS). The results showed that numerous Proteobacteria-affiliated sequences in the sediments of the inner continental shelf were related to both sulfate-reducing and sulfur-oxidizing bacteria, suggesting an active sulfur cycle in this area. Many sequences retrieved from the hypoxic zone were also related to Planctomycetes from two marine upwelling systems, which may be involved in the initial breakdown of sulfated heteropolysaccharides. Bacteroidetes, which is expected to degrade high-molecular-weight organic matter, was abundant in all the studied stations except for station A8, which was the deepest and possessed the largest grain size. In addition, dissolved organic carbon, water depth, percentage ratio of clay to silt, salinity, and sedimentary grain size were environmental effectors that shaped the sedimentary microbial community structure. Our results showed that putative Gammaproteobacteria-affiliated sulfur-oxidizing bacteria may not only detoxify hydrogen sulfide produced by sulfate-reducing prokaryotes, but also serve as the primary producers in the marine sediments. Specific groups of aerobic Bacteroidetes and Planctomycetes participated in degrading organic matter, which might contribute to the oxygen depletion in the hypoxic zones.

Distribution Characteristics and Environmental Control Factors of Lipophilic Marine Algal Toxins in Changjiang Estuary and the Adjacent East China Sea.

Project description:Marine algal toxins, highly toxic secondary metabolites, have significant influences on coastal ecosystem health and mariculture safety. The occurrence and environmental control factors of lipophilic marine algal toxins (LMATs) in the surface seawater of the Changjiang estuary (CJE) and the adjacent East China Sea (ECS) were investigated. Pectenotoxin-2 (PTX2), okadaic acid (OA), dinophysistoxin-1(DTX1), and gymnodimine (GYM) were detected in the CJE surface seawater in summer, with concentration ranges of not detected (ND)-105.54 ng/L, ND-13.24 ng/L, ND-5.48 ng/L, and ND-12.95 ng/L, respectively. DTX1 (ND-316.15 ng/L), OA (ND-16.13 ng/L), and PTX2 (ND-4.97 ng/L) were detected in the ECS during spring. LMATs formed a unique low-concentration band in the Changjiang diluted water (CJDW) coverage area in the typical large river estuary. PTX2, OA, and DTX1 in seawater were mainly derived from Dinophysis caudate and Dinophysis rotundata, while GYM was suspected to be from Karenia selliformis. Correlation analyses showed that LMAT levels in seawater were positively correlated with dissolved oxygen and salinity, but negatively correlated with temperature and nutrients, indicating that the hydrological condition and nutritional status of seawater and climatic factors exert significant effects on the distribution of LMATs.

Spatiotemporal dynamics of the total and active Vibrio spp. populations throughout the Changjiang estuary in China.

Project description:Vibrio is ubiquitously distributed in marine environments and is the most extensively characterized group within Gammaproteobacteria. Studies have investigated Vibrio spp. worldwide, but mostly focused on pathogenic vibrios and based on cultivation methods. Here, using a combination of molecular and culturing methods, we investigated the dynamics of the total and active Vibrio spp. throughout the Changjiang estuary in China. The total Vibrio abundance was higher in summer (~6.59 × 103 copies ml-1) than in winter (~1.85 × 103 copies ml-1) and increased from freshwater to saltwater (e.g. 8.04 × 101 to 9.39 × 103 copies ml-1 in summer). The ratio of active to total Vibrio (Va/Vt) revealed a high activity of vibrios, with remarkable differences between freshwater and saltwater (p < 0.05). Based on the community compositions of the culturable, total and active Vibrio, Vibrio atlanticus and Vibrio owensii were the dominant and active species in winter and summer, respectively. The distribution of Vibrio was governed by the effects of diverse environmental factors, such as temperature, salinity, pH, dissolved oxygen and SiO3 2-. Our study clearly demonstrates the spatiotemporal dynamics of total and active Vibrio spp. and lays a foundation for fully understanding the ecological roles of marine Vibrio.

Gross Nitrogen Mineralization in Surface Sediments of the Yangtze Estuary.

Project description:Nitrogen mineralization is a key biogeochemical process transforming organic nitrogen to inorganic nitrogen in estuarine and coastal sediments. Although sedimentary nitrogen mineralization is an important internal driver for aquatic eutrophication, few studies have investigated sedimentary nitrogen mineralization in these environments. Sediment-slurry incubation experiments combined with 15N isotope dilution technique were conducted to quantify the potential rates of nitrogen mineralization in surface sediments of the Yangtze Estuary. The gross nitrogen mineralization (GNM) rates ranged from 0.02 to 5.13 mg N kg(-1) d(-1) in surface sediments of the study area. The GNM rates were generally higher in summer than in winter, and the relative high rates were detected mainly at sites near the north branch and frontal edge of this estuary. The spatial and temporal distributions of GNM rates were observed to depend largely on temperature, salinity, sedimentary organic carbon and nitrogen contents, and extracellular enzyme (urease and L-glutaminase) activities. The total mineralized nitrogen in the sediments of the Yangtze Estuary was estimated to be about 6.17 × 10(5) t N yr(-1), and approximately 37% of it was retained in the estuary. Assuming the retained mineralized nitrogen is totally released from the sediments into the water column, which contributed 12-15% of total dissolved inorganic nitrogen (DIN) sources in this study area. This result indicated that the mineralization process is a significant internal nitrogen source for the overlying water of the Yangtze Estuary, and thus may contribute to the estuarine and coastal eutrophication.

The biogeography of fungal communities in wetland sediments along the Changjiang River and other sites in China.

Project description:Whether fungal community structure depends more on historical factors or on contemporary factors is controversial. This study used culture-dependent and -independent (polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE)) methods to assess the influence of historical and contemporary factors on the distributions of fungi in the wetland sediments at 10 locations along the Changjiang River and at 10 other locations in China. The culture-dependent approach detected greater species diversity (177 operational taxonomic units (OTUs)) than PCR-DGGE analysis (145 OTUs), and the species in the genera of Penicillium (relative frequency=16.8%), Fusarium (15.4%), Aspergillus (7.6%), Trichoderma (5.8%) and Talaromyces (4.2%) were dominant. On the basis of DGGE data, fungal diversity along the Changjiang River increased from upstream to downstream; altitude explained 44.8% of this variation in diversity. And based on the data from all 20 locations, the fungal communities were geographically clustered into three groups: Southern China, Northern China and the Qinghai-Tibetan Plateau. Multivariate regression tree analysis for data from the 20 locations indicated that the fungal community was influenced primarily by location (which explained 61.8% of the variation at a large scale), followed by total potassium (9.4%) and total nitrogen (3.5%) at a local scale. These results are consistent with the concept that geographic distance is the dominant factor driving variation in fungal diversity at a regional scale (1000-4000 km), whereas environmental factors (total potassium and total nitrogen) explain variation in fungal diversity at a local scale (<1000 km).

Stratified active archaeal communities in the sediments of Jiulong River estuary, China.

Project description:Here the composition of total and active archaeal communities in a sediment core of Jiulong River estuary at Fujian Province, Southern China was reported. Profiles of CH(4) and SO(2-) (4) concentrations from the sediment core indicated the existence of a sulfate-methane transition zone (SMTZ) in which sulfate reduction-coupled anaerobic oxidation of methane (AOM) occurs. Accordingly, three sediment layers (16-18.5 cm, 71-73.5 cm, and 161-163.5 cm) from the 1.2 m sediment core were sectioned and named top, middle and bottom, respectively. Total DNA and RNA of each layer were extracted and used for clone libraries and sequence analysis of 16S rRNA genes, the reverse transcription (RT)-PCR products of 16S rRNA and methyl CoM reductase alpha subunit (mcrA) genes. Phylogenetic analysis indicated that archaeal communities of the three layers were dominated by the Miscellaneous Crenarchaeotal Group (MCG) whose ecological functions were still unknown. The MCG could be further divided into seven subgroups, named MCG-A, B, C, D, E, F, and G. MCG-A and MCG-G were the most active groups in the estuarine sediments. Known anaerobic methanotrophic archaea (ANMEs) were only found as minor components in these estuarine archaeal communities. This study, together with the studies of deep subsurface sediments, would be a very good start point to target and compare the specific active archaeal groups and their roles in the dark, deep subsurface sediment environments.

Characteristics and Distribution of Phosphorus in Surface Sediments of Limnetic Ecosystem in Eastern China.

Project description:Phosphorus (P) is an essential nutrient for aquatic organisms; however, excessive P inflow to limnetic ecosystems can induce eutrophication. P concentrations in the rivers, wetlands and lakes of Eastern China have been amplified by fertilizer and sewage inputs associated with the development of industry and agriculture. Yet, knowledge of the distribution and speciation of P is lacking at the regional scale. We determined the distribution and speciation of P in limnetic ecosystems in Eastern China using Standards, Measurements and Testing (SMT) and phosphorus nuclear magnetic resonance (31P-NMR). The results indicate that P pollution in surface sediments was serious. Inorganic P (Pi) was the primary drive of variation in total P (TP) among different river systems, and Pi accounted for 71% to 90% of TP in surface sediment in Eastern China. Also, the concentrations of TP and Pi varied among watersheds and Pi primarily drove the variation in TP in different watersheds. Sediments less than 10-cm deep served as the main P reservoir. Environmental factors affect the speciation and origin of P. NaOH-Pi, HCl-Pi and organic P (Po) were related to pH accordingly at the regional scale. The physicochemical properties of sediments from different limnetic ecosystems affect the P speciation. HCl-Pi was higher in wetland sediments than in riverine and lake sediments in Eastern China. Conversely, NaOH-Pi was lowest in wetland sediments. Total Po concentration was lower in riverine sediments than in other sediments, but Mono-P was higher, with an average concentration of 48 mg kg-1. Diesters-P was highest in lake sediments. By revealing the regional distribution of TP, Pi and Po, this study will support eutrophication management in Eastern China.

Abiotic racemization kinetics of amino acids in marine sediments.

Project description:The ratios of d- versus l-amino acids can be used to infer the sources and composition of sedimentary organic matter. Such inferences, however, rely on knowing the rates at which amino acids in sedimentary organic matter racemize abiotically between the d- and the l-forms. Based on a heating experiment, we report kinetic parameters for racemization of aspartic acid, glutamic acid, serine, and alanine in bulk sediment from Aarhus Bay, Denmark, taken from the surface, 30 cm, and 340 cm depth below seafloor. Extrapolation to a typical cold deep sea sediment temperature of 3°C suggests racemization rate constants of 0.50×10(-5)-11×10(-5) yr(-1). These results can be used in conjunction with measurements of sediment age to predict the ratio of d:l amino acids due solely to abiotic racemization of the source material, deviations from which can indicate the abundance and turnover of active microbial populations.
