Reversible gel-sol photoswitching with an overcrowded alkene-based bis-urea supergelator.

ABSTRACT: A new type of low-molecular-weight gelator (LMWG), i.e. overcrowded alkene-based bis-ureas, can be switched effectively between cis and trans isomers using light as demonstrated by ¹H NMR and UV-Vis spectroscopy. Gelation studies reveal that one of the synthesized trans compounds forms stable gels in aromatic hydrocarbon solvents down to a critical concentration of 0.4 mg mL^-1. Transmission electron microscopy (TEM) shows that this gel consists of an entangled fibrous network. For the trans isomer of this LMWG intermolecular urea hydrogen bonding is observed in the solid state, whereas density functional theory (DFT) geometry optimization of the cis isomer indicates the possible formation of an intramolecular hydrogen bond. Irradiation of the gel triggers trans-to-cis isomerization and consequently, a gel-sol phase transition. This process can be fully reversed by altering the irradiation wavelength.

Liquid-Liquid Phase-Separated Systems from Reversible Gel-Sol Transition of Protein Microgels.

Project description:Liquid-liquid phase-separated biomolecular systems are increasingly recognized as key components in the intracellular milieu where they provide spatial organization to the cytoplasm and the nucleoplasm. The widespread use of phase-separated systems by nature has given rise to the inspiration of engineering such functional systems in the laboratory. In particular, reversible gelation of liquid-liquid phase-separated systems could confer functional advantages to the generation of new soft materials. Such gelation processes of biomolecular condensates have been extensively studied due to their links with disease. However, the inverse process, the gel-sol transition, has been less explored. Here, a thermoresponsive gel-sol transition of an extracellular protein in microgel form is explored, resulting in an all-aqueous liquid-liquid phase-separated system with high homogeneity. During this gel-sol transition, elongated gelatin microgels are demonstrated to be converted to a spherical geometry due to interfacial tension becoming the dominant energetic contribution as elasticity diminishes. The phase-separated system is further explored with respect to the diffusion of small particles for drug-release scenarios. Together, this all-aqueous system opens up a route toward size-tunable and monodisperse synthetic biomolecular condensates and controlled liquid-liquid interfaces, offering possibilities for applications in bioengineering and biomedicine.

Coordination-Assisted Reversible Photoswitching of Spiropyran-Based Platinum Macrocycles.

Project description:Control over the stimuli-responsive behavior of smart molecular systems can influence their capability to execute complex functionalities. Herein, we report the development of a suite of spiropyran-based multi-stimuli-responsive self-assembled platinum(II) macrocycles (5-7), rendering coordination-assisted enhanced photochromism relative to the corresponding ligands. 5 showed shrinking and swelling during photoreversal, while 6 and 7 are fast and fatigue-free supramolecular photoswitches. 6 turns out to be a better fatigue-resistant photoswitch and can retain an intact photoswitching ability of up to 20 reversible cycles. The switching behavior of the macrocycles can also be precisely controlled by tuning the pH of the medium. Our present strategy for the construction of rapid stimuli-responsive supramolecular architectures via coordination-driven self-assembly represents an efficient route for the development of smart molecular switches.

Reversible fluorescence photoswitching in DNA.

Project description:We describe the engineering of reversible fluorescence photoswitching in DNA with high-density substitution, and its applications in advanced fluorescence microscopy methods. High-density labeling of DNA with cyanine dyes can be achieved by polymerase chain reaction using a modified DNA polymerase that has been evolved to efficiently incorporate Cy3- and Cy5-labeled cytosine base analogues into double-stranded DNA. The resulting biopolymer, "CyDNA", displays hundreds of fluorophores per DNA strand and is strongly colored and highly fluorescent, although previous observations suggest that fluorescence quenching at such high density might be a concern, especially for Cy5. Herein, we first investigate the mechanisms of fluorescence quenching in CyDNA and we suggest that two different mechanisms, aggregate formation and resonance energy transfer, are responsible for fluorescence quenching at high labeling densities. Moreover, we have been able to re-engineer CyDNA into a reversible fluorescence photoswitchable biopolymer by using the properties of the Cy3-Cy5 pair. This novel biopolymer constitutes a new class of photoactive DNA-based nanomaterial and is of great interest for advanced microscopy applications. We show that reversible fluorescence photoswitching in CyDNA can be exploited in optical lock-in detection imaging. It also lays the foundations for improved and sequence-specific super-resolution fluorescence microscopy of DNA.

Reversible Photoswitching of Carbon Dots.

Project description:We present a method of reversible photoswitching in carbon nanodots with red emission. A mechanism of electron transfer is proposed. The cationic dark state, formed by the exposure of red light, is revived back to the bright state with the very short exposure of blue light. Additionally, the natural on-off state of carbon dot fluorescence was tuned using an electron acceptor molecule. Our observation can make the carbon dots as an excellent candidate for the super-resolution imaging of nanoscale biomolecules within the cell.

Reversible photo-responsive gel-sol transitions of robust organogels based on an azobenzene-containing main-chain liquid crystalline polymer.

Project description:Stimuli-responsive supramolecular gels have been widely investigated, but the construction of a liquid crystalline gel with a high mechanical property and reversible photo-response still remains a challenge. This is due to the difficulty of designing gelators with liquid crystal properties and gelation abilities in organic solvents simultaneously. In this study, an azobenzene-containing main-chain polyester (Azo-mLCP) with a pendant amide group was synthesized. The organogel of Azo-mLCP via a hydrogen bond in dioxane possessed reversible thermal- and photo-responsive behaviours. The organogel exhibited a good self-supporting ability when the concentration of the gelator was more than 7.5 wt%. The rapid trans-to-cis isomerization of Azo-mLCP in solution was studied via UV-Vis absorption spectra. In addition, the gel-to-sol transition of the organogel could be triggered efficiently by an incomplete trans-to-cis conversion strategy. This study opens a way for the main-chain liquid crystalline polymers to serve in potential applications in photo-responsive robust actuators, electro-optical devices, and so on.

Experimental data of inorganic gel based smart window using silica sol-gel process.

Project description:In this article experimental data are presented for inorganic gel based smart window fabricated using silica sol-gel process. Parallel beam transmittances were measured as functions of voltages for samples fabricated with different concentrations of nitric acid. Spectroscopic transmittance data at different driving voltages for samples fabricated with different LC concentrations are shown. Transmittance spectra of the Si-Ti based gel-based-liquid-crystal (GDLC) device measured as different driving voltages were compared with those of PDLC. GDLC showed much lower operating voltages, 10-15 V, for on-state. Formation of the LC droplet in gelation process is illustrated. The methyl organic group surrounds LC droplets. Demonstration of GDLC based smart window showed the successful operation with low driving voltages. GDLC window shows clear color, even at off-state, compared with PDLC.

Dilution-driven gel-sol-gel-sol transitions by the sequential evolution of surfactant micelles.

Project description:Here are described the dilution-driven gel-sol-gel-sol transitions in aromatic hydrotrope/zwitterionic surfactant bi-component aqueous mixtures. Long and entangled wormlike micelles (WLMs) of concentrated erucyl dimethyl amidopropyl betaine (EAPB) solutions sequentially transform into hexagonal liquid crystal phase (H1), highly concentrated spherical micelles (HCSMs), and finally hexagonal close packed micellar phase (HCP) upon gradual introducing an aromatic hydrotrope, sodium salicylate (NaSal). The HCP then successively converts into HCSMs, WLMs, and spherical micelles (SMs) upon further dilution with water, corresponding to a gel-sol-gel-sol transition. This dilution-induced HCSMs to WLMs transition results in an interesting thickening process, which is caused by a slow kinetic process, from the agglomeration and recombination of SMs to long and rigid WLMs via the intermediately formed rodlike micelles. In this work, the sustained adjustment of the competitive self-sorting and co-assembly between hydrotropes and surfactants by dilution was identified, benefitting the creation and application of WLMs fluids with switchable viscosity.

Structural basis for reversible photoswitching in Dronpa.

Project description:Dronpa is a novel GFP-like fluorescent protein with exceptional light-controlled switching properties. It may be reversibly switched between a fluorescent on-state and a nonfluorescent off-state by irradiation with light. To elucidate the molecular basis of the switching mechanism, we generated reversibly switchable Dronpa protein crystals. Using these crystals we determined the elusive dark-state structure of Dronpa at 1.95-A resolution. We found that the photoswitching results in a cis-trans isomerization of the chromophore accompanied by complex structural rearrangements of four nearby amino acid residues. Because of this cascade of intramolecular events, the chromophore is exposed to distinct electrostatic surface potentials, which are likely to influence the protonation equilibria at the chromophore. We suggest a comprehensive model for the light-induced switching mechanism, connecting a cascade of structural rearrangements with different protonation states of the chromophore.

Reversible photoswitching of encapsulated azobenzenes in water.

Project description:Efficient molecular switching in confined spaces is critical for the successful development of artificial molecular machines. However, molecular switching events often entail large structural changes and therefore require conformational freedom, which is typically limited under confinement conditions. Here, we investigated the behavior of azobenzene-the key building block of light-controlled molecular machines-in a confined environment that is flexible and can adapt its shape to that of the bound guest. To this end, we encapsulated several structurally diverse azobenzenes within the cavity of a flexible, water-soluble coordination cage, and investigated their light-responsive behavior. Using UV/Vis absorption spectroscopy and a combination of NMR methods, we showed that each of the encapsulated azobenzenes exhibited distinct switching properties. An azobenzene forming a 1:1 host-guest inclusion complex could be efficiently photoisomerized in a reversible fashion. In contrast, successful switching in inclusion complexes incorporating two azobenzene guests was dependent on the availability of free cages in the system, and it involved reversible trafficking of azobenzene between the cages. In the absence of extra cages, photoswitching was either suppressed or it involved expulsion of azobenzene from the cage and consequently its precipitation from the solution. This finding was utilized to develop an information storage medium in which messages could be written and erased in a reversible fashion using light.

Reversible photoswitching of spiropyran-conjugated semiconducting polymer dots.

Project description:Semiconducting polymer dots (Pdots) recently have emerged as a new class of ultrabright fluorescent probes with promising applications in biological detection and imaging. We developed photoswitchable Pdots by conjugating photochromic spiropyran molecules onto poly[9,9-dioctylfluorenyl-2,7-diyl)-co-1,4-benzo-{2,1'-3}-thiadiazole)] (PFBT). The modulation of fluorescence was achieved by ultraviolet irradiation, which converted spiropyran into its visible-absorbing merocyanine form. The merocyanine efficiently quenched the fluorescence of PFBT via Förster resonance energy transfer (FRET). We then reversed the quenching by subsequent irradiation with visible light to get back the fluorescence of PFBT. This FRET-based photomodulation of Pdot fluorescence could be repeated multiple times. We next conjugated biomolecules onto the surface of these photoswitchable Pdots and demonstrated their specific cellular and subcellular labeling to different types of cells without any noticeable nonspecific binding. We anticipate these photoswitchable and biocompatible Pdots will be useful in developing bioimaging techniques in the future.
