Morphological and molecular characterization of entomopathogenic nematode, Heterorhabditis baujardi (Rhabditida, Heterorhabditidae) from Mizoram, northeastern India.

ABSTRACT: Isolation of entomopathogenic nematodes from Mizoram, northeastern part of India includes a Heterorhabditis species never been recorded in India. The morphological and multigene (ITS rRNA, 28S rRNA and COI) sequences analysis had revealed that the isolated Heterorhabditis belongs to Heterorhabditis baujardi, originally described from Vietnam. The phylogenetic tree (both MP and ML) revealed that the H. baujardi belongs to H. indica clade, and further, forms a monophyletic clade (99/98% and 94/92% bootstrap support for ITS and 28S respectively) with H. amazonensis, H. floridensis, H. mexicana and H. taysearae forming H. baujardi sub clade. The multigene characterization revealed that both the ITS and 28S rRNA showed similar result in resolving the phylogenetic relationship of the genus Heterorhabditis, with the ITS rRNA being superior based on the strong bootstrap support, whereas, the cytochrome c oxidase I (M1-M6 partition) can be a good supportive tool for species delimitation. This is the first report of H. baujardi from India envisaging its future use as a biological control agent, and further incorporated into the IPM.

Project description:BackgroundHeterorhabditis are important biological control agents in agriculture. Two Heterorhabditis populations, S8 and S10, were isolated from agricultural soils in the United States of America. Molecular analyses, based on mitochondrial and nuclear genes, showed that these populations are conspecific and represent a novel species of the "Bacteriophora" clade. This species was named Heterorhabditis americana n. sp. and is described in this study.MethodsTo describe H. americana n. sp., we carried out phylogenetic reconstructions using multiple genes, characterized their morphology, conducted self-crossing and cross-hybridization experiments, and isolated and identified their symbiotic bacteria.ResultsHeterorhabditis americana n. sp. is molecularly and morphologically similar to H. georgiana. Morphological differences between the males of H. americana n. sp. and H. georgiana include variations in the excretory pore position, the gubernaculum size, the gubernaculum-to-spicule length ratio, the tail length, and the body diameter. Infective juveniles (IJs) of H. americana n. sp. differ from H. georgiana IJs because H. americana n. sp. IJs have an invisible bacterial cell pouch posterior to the cardia and a small posterior phasmid, whereas H. georgiana IJs have a visible bacterial cell pouch and an inconspicuous phasmid. Hermaphrodites of H. americana n. sp. and H. georgiana are differentiated by the body length, the nerve ring distance from the anterior end, the excretory pore distance from the anterior end, the anal body diameter, and the c' ratio. Females of H. americana n. sp. can be differentiated from H. georgiana females by the anal body diameter and the c' ratio. Reproductive isolation was confirmed, as H. americana n. sp. does not produce viable offspring with any of the species of the "Bacteriophora" clade. Heterorhabditis americana n. sp. is associated with the symbiotic bacterium Photorhabdus kleinii.ConclusionsBased on the observed morphological and morphometric differences, the distinct phylogenetic placement, and the reproductive isolation, the nematode isolates S8 and S10 represent a novel species, which we named Heterorhabditis americana n. sp. This study provides a detailed characterization of this novel species, contributing to enhancing our knowledge of species diversity and evolutionary relationships of the Heterorhabditis genus.

Project description:The entomopathogenic nematode Heterorhabditis bacteriophora and its symbiotic bacterium, Photorhabdus luminescens, are important biological control agents of insect pests. This nematode-bacterium-insect association represents an emerging tripartite model for research on mutualistic and parasitic symbioses. Elucidation of mechanisms underlying these biological processes may serve as a foundation for improving the biological control potential of the nematode-bacterium complex. This large-scale expressed sequence tag (EST) analysis effort enables gene discovery and development of microsatellite markers. These ESTs will also aid in the annotation of the upcoming complete genome sequence of H. bacteriophora.A total of 31,485 high quality ESTs were generated from cDNA libraries of the adult H. bacteriophora TTO1 strain. Cluster analysis revealed the presence of 3,051 contigs and 7,835 singletons, representing 10,886 distinct EST sequences. About 72% of the distinct EST sequences had significant matches (E value < 1e-5) to proteins in GenBank's non-redundant (nr) and Wormpep190 databases. We have identified 12 ESTs corresponding to 8 genes potentially involved in RNA interference, 22 ESTs corresponding to 14 genes potentially involved in dauer-related processes, and 51 ESTs corresponding to 27 genes potentially involved in defense and stress responses. Comparison to ESTs and proteins of free-living nematodes led to the identification of 554 parasitic nematode-specific ESTs in H. bacteriophora, among which are those encoding F-box-like/WD-repeat protein theromacin, Bax inhibitor-1-like protein, and PAZ domain containing protein. Gene Ontology terms were assigned to 6,685 of the 10,886 ESTs. A total of 168 microsatellite loci were identified with primers designable for 141 loci.A total of 10,886 distinct EST sequences were identified from adult H. bacteriophora cDNA libraries. BLAST searches revealed ESTs potentially involved in parasitism, RNA interference, defense responses, stress responses, and dauer-related processes. The putative microsatellite markers identified in H. bacteriophora ESTs will enable genetic mapping and population genetic studies. These genomic resources provide the material base necessary for genome annotation, microarray development, and in-depth gene functional analysis.

Project description:Parasitic nematodes constitute one of the major threats to human health, causing diseases of major socioeconomic importance worldwide. Recent estimates indicate that more than 1 billion people are infected with parasitic nematodes around the world. Current measures to combat parasitic nematode infections include anthelmintic drugs. However, heavy exposure to anthelmintics has selected populations of livestock parasitic nematodes that are no longer susceptible to the drugs, rendering several anthelmintics useless for parasitic nematode control in many areas of the world. The rapidity with which anthelmintic resistance developed in response to these drugs suggests that increasing the selective pressure on human parasitic nematodes will also rapidly generate resistant worm populations. Therefore, development of new anthelmintics is of major importance before resistance becomes widespread in human parasitic nematode populations. G-Protein Coupled Receptors (GPCRs) represent an important target for many pharmacological interventions due to their ubiquitous expression in various cell types. GPCRs contribute to numerous physiological processes, and their ligand binding sites located on cell surfaces make them accessible targets and attractive substrates in terms of druggability. In fact, ∼35 % of Food and Drug Administration (FDA) and European Medicines Agency (EMA) approved drugs target GPCRs and their associated proteins, with over 300 additional drugs targeting GPCRs at the clinical trial stage. Nematode Chemosensory GPCRs (NemChRs) are unique to nematodes, and therefore represent ideal substrates for target-based drug discovery. Here we set out to identify NemChRs that are transcriptionally active inside the host, and to use these NemChRs in a reverse pharmacological screen to impede parasitic development. Our data identified several NemChRs, and we focused on one that was expressed in neuronal cells and exhibited the highest fold change in transcription after host activation. Next, we performed homology modelling and molecular dynamics simulations of this NemChR in order to conduct a virtual screening campaign to identify candidate drug targets which were ranked and selected for experimental testing in bioassays. Taken together, our results identify and characterize a candidate NemChR drug target, and provide a chemogenomic pipeline for identifying nematicide substrates.

Project description:The western corn rootworm (WCR), Diabrotica virgifera virgifera LeConte (Coleoptera, Chrysomelidae), is an important insect pest of maize in North America and Central and Eastern Europe. In Central Europe, the larvae emerge in May and its three instars feed intensively on maize roots in June, causing plant lodging that leads to a loss of economic yield. A three-year field experiment (2016-2018) was conducted to compare the effectiveness i) of soil-applied granular insecticide based on the active ingredient tefluthrin, ii) of maize seeds dressed with thiacloprid, and iii) entomopathogenic nematodes Heterorhabditis bacteriophora Poinar (Rhabditida: Heterorhabditidae, product Dianem) against WCR larvae. An additional treatment with alcohol ethoxylate (i.e., soil conditioner) mixed with entomopathogenic nematodes was performed in 2017 and 2018 to check for any increase of entomopathogenic nematodes' effectiveness. Field tests were carried out in two fields infested naturally with a WCR pest population, one in Bu?e?ovci (Eastern Slovenia) and the other in Šmartno pri Cerkljah (northern Slovenia), exhibiting dissimilar pedo-climatic conditions and soil pest densities. The treatments were performed in five replicates per experiment in each year. The efficacy of the treatments was very similar at both locations, despite the approximately five-fold lower WCR soil pest densities in northern than in eastern Slovenia, as well as being constant over time. The largest number of WCR beetles was observed in the negative control, followed by that of beetles subjected to thiacloprid treatment (insignificant decrease taking into account the entire three-year dataset). Treatments with tefluthrin (44.1 ± 11.7%), H. bacteriophora (46.2 ± 7.4%), and H. bacteriophora + alcohol ethoxylate (49.2 ± 1.8%) significantly decreased the numbers of emerging beetles. Treatments of thiacloprid, H. bacteriophora, and H. bacteriophora + alcohol ethoxylate additionally led to significantly increased maize plant weights. Furthermore, entomopathogenic nematodes were able to persist in maize fields for almost five months at both experimental locations in silty and sandy loam soils. It was concluded that the control of WCR larvae in maize using the entomopathogenic nematode H. bacteriophora is as effective as a tefluthrin treatment, and could thus offer a sustainable Diabrotica v. virgifera biological control management option in Europe.

Project description:BackgroundDespite important progress in the field of innate immunity, our understanding of host immune responses to parasitic nematode infections lags behind that of responses to microbes. A limiting factor has been the obligate requirement for a vertebrate host which has hindered investigation of the parasitic nematode infective process. The nematode parasite Heterorhabditis bacteriophora offers great potential as a model to genetically dissect the process of infection. With its mutualistic Photorhabdus luminescens bacteria, H. bacteriophora invades multiple species of insects, which it kills and exploits as a food source for the development of several nematode generations. The ability to culture the life cycle of H. bacteriophora on plates growing the bacterial symbiont makes it a very exciting model of parasitic infection that can be used to unlock the molecular events occurring during infection of a host that are inaccessible using vertebrate hosts.ResultsTo profile the transcriptional response of an infective nematode during the early stage of infection, we performed next generation RNA sequencing on H. bacteriophora IJs incubated in Manduca sexta hemolymph plasma for 9 h. A subset of up-regulated and down-regulated genes were validated using qRT-PCR. Comparative analysis of the transcriptome with untreated controls found a number of differentially expressed genes (DEGs) which cover a number of different functional categories. A subset of DEGs is conserved across Clade V parasitic nematodes revealing an array of candidate parasitic genes.ConclusionsOur analysis reveals transcriptional changes in the regulation of a large number of genes, most of which have not been shown previously to play a role in the process of infection. A significant proportion of these genes are unique to parasitic nematodes, suggesting the identification of a group of parasitism factors within nematodes. Future studies using these candidates may provide functional insight into the process of nematode parasitism and also the molecular evolution of parasitism within nematodes.
