Single-Step Metal-Free Grafting of Cationic Polymer Brushes on Fluorescent Nanodiamonds.

ABSTRACT: Cationic polymers are often employed in conjugation with nanomaterials, and the resultant hybrids are useful for various bioapplications. Here, a single-step metal-free method for the synthesis of fluorescent nanodiamonds (FNDs) conjugated with cationic polymer brushes is reported. Distinct from the common methods such as atom transfer radical polymerization and reversible addition fragmentation chain transfer, our ring-opening-polymerization-based method is simple and less time consuming and hazardous. Infrared spectroscopy, thermogravimetric analysis, zeta potential, and dynamic light scattering confirmed the synthesis. The produced FND-polymer brushes showed markedly higher cell labeling and internalization efficiency without noticeable cytotoxicity. Our method is general and applicable to other nanoparticles as well for uses in diverse research areas.

Fluorescent Nanodiamonds for Tracking Single Polymer Particles in Cells and Tissues.

Project description:Polymer nanoparticles are widely used in drug delivery and are also a potential concern due to the increased burden of nano- or microplastics in the environment. In order to use polymer nanoparticles safely and understand their mechanism of action, it is useful to know where within cells and tissues they end up. To this end, we labeled polymer nanoparticles with nanodiamond particles. More specifically, we have embedded nanodiamond particles in the polymer particles and characterized the composites. Compared to conventional fluorescent dyes, these labels have the advantage that nanodiamonds do not bleach or blink, thus allowing long-term imaging and tracking of polymer particles. We have demonstrated this principle both in cells and entire liver tissues.

Towards controlled polymer brushes via a self-assembly-assisted-grafting-to approach.

Project description:Precise synthesis of polymer brushes to modify the surface of nanoparticles and nanodevices for targeted applications has been one of the major focuses in the community for decades. Here we report a self-assembly-assisted-grafting-to approach to synthesize polymer brushes on flat substrates. In this method, polymers are pre-assembled into two-dimensional polymer single crystals (PSCs) with functional groups on the surface. Chemically coupling the PSCs onto solid substrates leads to the formation of polymer brushes. Exquisite control of the chain folding in PSCs allows us to obtain polymer brushes with well-defined grafting density, tethering points and brush conformation. Extremely high grafting density (2.12 chains per nm(2)) has been achieved in the synthesized single-tethered polymer brushes. Moreover, polymer loop brushes have been successfully obtained using oddly folded PSCs from telechelic chains. Our approach combines some of the important advantages of conventional 'grafting-to' and 'grafting-from' methods, and is promising for tailored synthesis of polymer brushes.

Grafting Polymer Brushes by ATRP from Functionalized Poly(ether ether ketone) Microparticles.

Project description:Poly(ether ether ketone) (PEEK) is a semi-crystalline thermoplastic with excellent mechanical and chemical properties. PEEK exhibits a high degree of resistance to thermal, chemical, and bio-degradation. PEEK is used as biomaterial in the field of orthopaedic and dental implants; however, due to its intrinsic hydrophobicity and inert surface, PEEK does not effectively support bone growth. Therefore, new methods to modify PEEK's surface to improve osseointegration are key to next generation polymer implant materials. Unfortunately, PEEK is a challenging material to both modify and subsequently characterize thus stymieing efforts to improve PEEK osseointegration. In this manuscript, we demonstrate how surface-initiated atom transfer radical polymerization (SI-ATRP) can be used to modify novel PEEK microparticles (PMP). The hard core-soft shell microparticles were synthesized and characterized by DLS, ATR-IR, XPS and TEM, indicating the grafted materials increased solubility and stability in a range of solvents. The discovered surface grafted PMP can be used as compatibilizers for the polymer-tissue interface.

Surface-Induced ARGET ATRP for Silicon Nanoparticles with Fluorescent Polymer Brushes.

Project description:Well-defined polymer brushes attached to nanoparticles offer an elegant opportunity for surface modification because of their excellent mechanical stability, functional versatility, high graft density as well as controllability of surface properties. This study aimed to prepare hybrid materials with good dispersion in different solvents, and to endow this material with certain fluorescence characteristics. Well-defined diblock copolymers poly (styrene)-b-poly (hydroxyethyl methyl acrylate)-co-poly (hydroxyethyl methyl acrylate- rhodamine B) grafted silica nanoparticles (SNPs-g-PS-b-PHEMA-co-PHEMA-RhB) hybrid materials were synthesized via surface-initiated activators regenerated by electron transfer atom transfer radical polymerization (SI-ARGET ATRP). The SNPs surfaces were modified by 3-aminopropyltriethoxysilane (KH-550) firstly, then the initiators 2-Bromoisobutyryl bromide (BIBB) was attached to SNPs surfaces through the esterification of acyl bromide groups and amidogen groups. The synthetic initiators (SNPs-Br) were further used for the SI-ARGET ATRP of styrene (St), hydroxyethyl methyl acrylate (HEMA) and hydroxyethyl methyl acrylate-rhodamine B (HEMA-RhB). The results indicated that the SI-ARGET ATRP initiator had been immobilized onto SNPs surfaces, the Br atom have located at the end of the main polymer chains, and the polymerization process possessed the characteristic of controlled/"living" polymerization. The SNPs-g-PS-b-PHEMA-co-PHEMA-RhB hybrid materials show good fluorescence performance and good dispersion in water and EtOH but aggregated in THF. This study demonstrates that the SI-ARGET ATRP provided a unique way to tune the polymer brushes structure on silica nanoparticles surface and further broaden the application of SI-ARGET ATRP.

Enthalpic Interactions and Solution Behaviors of Solvent-Free Polymer Brushes

Project description:We performed molecular dynamics simulations to characterize the role of enthalpic interaction in impacting the static and dynamic properties of solvent-free polymer brushes. The intrinsic enthalpic interaction in the simulation was introduced using different attraction strengths between distinct species. Two model systems were considered: one consisting of binary brushes of two different polymer types and the other containing a mixture of homopolymer brushes and free molecules. In the first system, we observed that, when two originally incompatible polymers were grafted to opposing surfaces, the miscibility between them was significantly enhanced. A less favorable intrinsic enthalpic interaction in the brushes resulted in a more stretched chain configuration, a lower degree of inter-brush penetration, and faster segmental relaxation. In the second system, we characterized the solvent capacity of the homopolymer brushes from variations in the energy components of the system as a function of the number of free molecules. We determined that molecular absorption was driven by the release of the entropic frustration for the grafted chains in conjunction with the chemical affinity between the solutes and polymers. The solute distribution function within the inter-wall space showed that solute–polymer mixing in the middle of the gap occurred preferentially when the enthalpic interaction was more favorable. When this was not the case, absorption was predominantly localized near the grafting surface. From the mean square displacement of the solute, we found that the brush profiles restrained the molecular diffusion perpendicular to the grafting wall; the weaker the attraction from the brush, the higher the solute mobility.

Fluorescent Patterns by Selective Grafting of a Telechelic Polymer.

Project description:The preparation of patterned ultrathin films (sub-10 nm) composed of end-anchored fluorescently labeled poly(methyl methacrylate) (PMMA) is presented. Telechelic PMMA was synthesized utilizing activator regenerated by electron transfer atom transfer radical polymerization and consecutively end-functionalized with alkynylated fluorescein by Cu-catalyzed azide-alkyne cycloaddition (CuAAC) "click" chemistry. The polymers were grafted via the α-carboxyl groups to silica or glass substrates pretreated with (3-aminopropyl)triethoxysilane (APTES). Patterned surfaces were prepared by inkjet printing of APTES onto glass substrates and selectively grafted with fluorescently end-labeled PMMA to obtain emissive arrays on the surface.

Aqueous-based initiator attachment and ATRP grafting of polymer brushes from poly(methyl methacrylate) substrates.

Project description:Many polymers, such as PMMA, are very susceptible to swelling or dissolution by organic solvents. Growing covalently attached polymer brushes from these surfaces by atom-transfer radical polymerization (ATRP) is challenging because of the typical requirement of organic solvent for initiator immobilization. We report an unprecedented, aqueous-based route to graft poly(N-isopropylacrylamide), PNIPAAm, from poly(methyl methacrylate), PMMA, surfaces by ATRP, wherein the underlying PMMA is unaffected. Successful attachment of the ATRP initiator, N-hydroxysuccinimidyl-2-bromo-2-methylpropionate, on amine-bearing PMMA surfaces was confirmed by XPS. From this surface-immobilized initiator, thermoresponsive PNIPAAm brushes were grown by aqueous ATRP to yield optically transparent PNIPAAm-grafted PMMA surfaces. This procedure is valuable, as it can be applied for the aqueous-based covalent attachment of ATRP initiator on any amine-functionalized surface, with subsequent polymerization of a variety of monomers.

Independent control of grafting density and conformation of single-stranded DNA brushes.

Project description:We describe self-assembly of ssDNA brushes that exploits the intrinsic affinity of adenine nucleotides (dA) for gold surfaces. The grafting density and conformation of these brushes is deterministically controlled by the length of the anchoring dA sequences, even in the presence of thymine nucleotides (dT). We produce and characterize brushes of model block-oligonucleotides, d(T(m)-A(n)), with systematically varied lengths m and n of the thymine and adenine blocks [denoted d(T(m)) and d(A(n)), respectively]. The hairpin conformation, dominant for self-complementary d(T(m)-A(n)) oligos in solution, is disrupted by the high preferential affinity of dA for gold surfaces. As a result, the d(T(m)-A(n)) oligos adsorb as a brush of d(T) strands immobilized via the d(A) blocks. Quantitative analysis by FTIR spectroscopy and x-ray photoelectron spectroscopy (XPS) reveals a unique feature of DNA immobilization via d(A) blocks: The surface density of dA nucleotides is close to saturation and is nearly independent of d(A) block length. Accordingly, the lateral spacing (grafting density) of the d(T) blocks is determined by the length of the d(A) blocks. The d(T) blocks extend away from the surface in a brush-like conformation at a lateral spacing 2-3 times larger (a grafting density 5-10 times lower) than in analogous films immobilized via standard thiol linkers. This combination of brush-like conformation and low saturation grafting density is expected to increase the efficiency of DNA hybridization at surfaces. Therefore, immobilization via d(A) blocks offers a method of producing DNA brushes with controlled properties for applications in biotechnology and nanotechnology.

Characterization and application of single fluorescent nanodiamonds as cellular biomarkers.

Project description:Type Ib diamonds emit bright fluorescence at 550-800 nm from nitrogen-vacancy point defects, (N-V)(0) and (N-V)(-), produced by high-energy ion beam irradiation and subsequent thermal annealing. The emission, together with noncytotoxicity and easiness of surface functionalization, makes nano-sized diamonds a promising fluorescent probe for single-particle tracking in heterogeneous environments. We present the result of our characterization and application of single fluorescent nanodiamonds as cellular biomarkers. We found that, under the same excitation conditions, the fluorescence of a single 35-nm diamond is significantly brighter than that of a single dye molecule such as Alexa Fluor 546. The latter photobleached in the range of 10 s at a laser power density of 10(4) W/cm(2), whereas the nanodiamond particle showed no sign of photobleaching even after 5 min of continuous excitation. Furthermore, no fluorescence blinking was detected within a time resolution of 1 ms. The photophysical properties of the particles do not deteriorate even after surface functionalization with carboxyl groups, which form covalent bonding with polyL-lysines that interact with DNA molecules through electrostatic forces. The feasibility of using surface-functionalized fluorescent nanodiamonds as single-particle biomarkers is demonstrated with both fixed and live HeLa cells.

Single-step equipment-free extracellular vesicle concentration using super absorbent polymer beads.

Project description:Extracellular vesicles (EVs) contain useful biomarkers for disease diagnosis and are promising biomaterials for the delivery of therapeutic molecules in vivo. Accordingly, an efficient concentration method is necessary for large-scale production or high-throughput isolation of EVs from bulk liquid samples, including culture medium and body fluids, to achieve their clinical application. However, current EV concentration methods, including ultrafiltration, are limited with respect to cost, efficiency, and centrifugation time. In this study, we developed the first single-step, equipment-free EV concentration method using super absorbent polymer (SAP) beads. SAP beads absorb small molecules, including water, via nano-sized channels but expel and thereby concentrate EVs. Consequently, the beads drastically enrich EVs by reducing the solution volume in a single step, without affecting EV characteristics. Moreover, the purity of the concentrated EV solution was high due to the absorption of protein impurities by SAP beads. To further demonstrate the versatility of the method, we showed that SAP beads successfully enrich EVs in human urine samples and culture medium, enabling better isolation performance than conventional ultrafiltration. We believe the newly developed approach and insight gained in this study will facilitate the use of EVs as prominent biomaterials for disease diagnosis and therapy.
