A closer look at neuron interaction with track-etched microporous membranes.

ABSTRACT: Microporous membranes support the growth of neurites into and through micro-channels, providing a different type of neural growth platform to conventional dish cultures. Microporous membranes are used to support various types of culture, however, the role of pore diameter in relation to neurite growth through the membrane has not been well characterised. In this study, the human cell line (SH-SY5Y) was differentiated into neuron-like cells and cultured on track-etched microporous membranes with pore and channel diameters selected to accommodate neurite width (0.8?µm to 5?µm). Whilst neurites extended through all pore diameters, the extent of neurite coverage on the non-seeded side of the membranes after 5 days in culture was found to be directly proportional to channel diameter. Neurite growth through membrane pores reduced significantly when neural cultures were non-confluent. Scanning electron microscopy revealed that neurites bridged pores and circumnavigated pore edges - such that the overall likelihood of a neurite entering a pore channel was decreased. These findings highlight the role of pore diameter, cell sheet confluence and contact guidance in directing neurite growth through pores and may be useful in applications that seek to use physical substrates to maintain separate neural populations whilst permitting neurite contact between cultures.

Project description:Actinoporins are a group of soluble toxic proteins that bind to membranes containing sphingomyelin (SM) and oligomerize to form pores. Sticholysin II (StnII) is a member of the actinoporin family produced by Stichodactyla helianthus. Cholesterol (Chol) is known to enhance the activity of StnII. However, the molecular mechanisms behind this activation have remained obscure, although the activation is not Chol specific but rather sterol specific. To further explore how bilayer lipids affect or are affected by StnII, we have used a multiprobe approach (fluorescent analogs of both Chol and SM) in combination with a series of StnII tryptophan (Trp) mutants to study StnII/bilayer interactions. First, we compared StnII bilayer permeabilization in the presence of Chol or oleoyl-ceramide (OCer). The comparison was done because both Chol and OCer have a 1-hydroxyl, which helps to orient the molecule in the bilayer (although OCer has additional polar functional groups). Both Chol and OCer also have increased affinity for SM, which StnII may recognize. However, our results show that only Chol was able to activate StnII-induced bilayer permeabilization; OCer failed to activate it. To further examine possible Chol/StnII interactions, we measured Förster resonance energy transfer between Trp in StnII and cholestatrienol, a fluorescent analog of Chol. We could show higher Förster resonance energy transfer efficiency between cholestatrienol and Trps in position 100 and 114 of StnII when compared to three other Trp positions further away from the bilayer binding region of StnII. Taken together, our results suggest that StnII was able to attract Chol to its vicinity, maybe by showing affinity for Chol. SM interactions are known to be important for StnII binding to bilayers, and Chol is known to facilitate subsequent permeabilization of the bilayers by StnII. Our results help to better understand the role of these important membrane lipids for the bilayer properties of StnII.

Project description:Effective removal of toxic inorganic and organic pollutants is one of the current leading challenges of wastewater treatment. In this study, the decomposition of methylene blue (MB) under UV light irradiation was investigated in the presence of copper nanoclusters (NCs)-deposited polyethylene terephthalate (PET) track-etched hybrid membranes. PET track-etched membranes (TeMs) with an average pore size of ~400 nm were grafted by functional acrylic acid (AA) monomer under electron beam irradiation after oxidation with H2O2/UV system. The radiation dose varied between 46 and 200 kGy. For the deposition of copper NCs, poly(acrylic acid) (PAA)-grafted membranes saturated with Cu(II) ions were irradiated either by electron beam or γ-rays to obtain copper-based NCs for the catalytic degradation of MB. Irradiation to 100 kGy with accelerated electrons resulted in the formation of small and uniform copper hydroxide (Cu(OH)2) nanoparticles homogeneously distributed over the entire volume of the template. On the other hand, irradiation under γ-rays yielded composites with copper NCs with a high degree of crystallinity. However, the size of the deposited NCs obtained by γ-irradiation was not uniform. Nanoparticles with the highest uniformity were obtained at 150 kGy dose. Detailed analysis by X-ray diffraction (XRD) and scanning electron microscopy (SEM) confirmed the loading of copper nanoparticles with an average size of 100 nm on the inner walls of nanochannels and on the surface of PET TeMs. Under UV light irradiation, composite membranes loaded with NCs exhibited high photocatalytic activity. It was determined that the highest catalytic activity was observed in the presence of Cu(OH)2@PET-g-PAA membrane obtained at 250 kGy. More than 91.9% of the initial dye was degraded when this hybrid membrane was employed for 180 min, while only 83.9% of MB was degraded under UV light using Cu@PET-g-PAA membrane. Cu(OH)2@PET-g-PAA membranes obtained under electron beam irradiation demonstrated a higher photocatalytic activity compared to Cu@PET-g-PAA membranes attained by γ-rays.
