Cannabis and the Anxiety of Fragmentation-A Systems Approach for Finding an Anxiolytic Cannabis Chemotype.

ABSTRACT: Cannabis sativa is a medicinal herb with a diverse range of chemotypes that can exert both anxiolytic and anxiogenic effects on humans. Medical cannabis patients receiving organically grown cannabis from a single source were surveyed about the effectiveness of cannabis for treating anxiety. Patients rated cannabis as highly effective overall for treating anxiety with an average score of 8.03 on a Likert scale of 0 to 10 (0 = not effective, 10 = extremely effective). Patients also identified which strains they found the most or least effective for relieving their symptoms of anxiety. To find correlations between anxiolytic activity and chemotype, the top four strains voted most and least effective were analyzed by HPLC-MS/MS to quantify cannabinoids and GC-MS to quantify terpenes. Tetrahydrocannabinol (THC) and trans-nerolidol have statistically significant correlations with increased anxiolytic activity. Guiaol, eucalyptol, ?-terpinene, ?-phellandrene, 3-carene, and sabinene hydrate all have significant correlations with decreased anxiolytic activity. Further studies are needed to better elucidate the entourage effects that contribute to the anxiolytic properties of cannabis varieties.

Project description:This study aimed to investigate the anxiolytic effects of GABA administration and its underlying molecular mechanisms in a chronic restraint stress (CRS)-induced anxiety model in C57BL/6J mice. CRS mice were established by placing the animals in well-ventilated 50 ml centrifuge tubes for 2 hours daily over 14 consecutive days, while control mice were housed under standard conditions. Behavioral assessments, including the elevated plus maze (EPM) test and open field test (OFT), were performed on days 15 and 16, respectively. GABA was administered intragastrically at various doses throughout the experiment, and prefrontal cortex (PFC) and hippocampal tissues were harvested on day 17 for further proteomic and biochemical analyses. Behavioral results demonstrated that GABA treatment exerted dose-dependent anxiolytic effects, as evidenced by increased time and entries in the open arms in the EPM and increased time and distance spent in the center area in the OFT. Furthermore, GABA treatment significantly elevated reduced GABA levels in the PFC. To elucidate the molecular mechanisms of GABA’s anxiolytic effects, a 4D-label-free quantitative proteomics approach was employed. Principal component analysis (PCA) and differential protein analyses revealed that CRS induced substantial proteomic alterations in the hippocampus, while GABA treatment partially reversed these changes. Functional clustering (Mfuzz) and gene ontology (GO) enrichment analysis identified critical pathways involved in the “humoral immune response,” “complement activation,” and “acute inflammatory response.” Key proteins such as C3, Cfh, C4b, and Cfi were found to overlap across these pathways, highlighting their involvement in CRS-induced neuroimmune dysfunction and GABA-mediated rescue effects. This integrated analysis provides valuable insights into the behavioral and neuroimmune mechanisms through which GABA alleviates anxiety-like behaviors, shedding light on its potential as a therapeutic agent for stress-related disorders.

Project description:Background and objectivesAnxiety sensitivity, or fear of anxious arousal, may be an important risk factor for problematic cannabis use. Specifically, anxiety sensitivity may motivate cannabis use to cope with distress, particularly among trauma-exposed individuals. The current study tested associations among anxiety sensitivity, its subdomains, and cannabis use motives in a sample of trauma-exposed cannabis users. We hypothesized elevated anxiety sensitivity, particularly cognitive concerns, would be associated with increased maladaptive coping use motives, after covarying for the number of traumas and cannabis use quantity.MethodHypotheses were tested in a cross-sectional study of trauma-exposed young adult cannabis users (N = 56) (Mage = 20.7 years, 59% women, 73% white). Participants completed self-report measures, and a clinical interview assessing cannabis use quantity.ResultsMultiple regression analyses indicated that elevated anxiety sensitivity was associated with increased cannabis use coping motives, after covarying for the number of traumas experienced and cannabis use quantity. Specifically, higher levels of anxiety sensitivity cognitive and social concerns were associated with coping motives for cannabis use.Discussion and conclusionsAnxiety sensitivity, particularly concerns about cognitive dyscontrol and negative social evaluations of anxious arousal, may motivate cannabis use to cope with stress among trauma-exposed cannabis users. Future research should include prospective studies with diverse samples to replicate results and determine whether intervening on anxiety sensitivity could reduce maladaptive coping motives for cannabis use.Scientific significanceThe current study replicates prior research indicating anxiety sensitivity, particularly cognitive concerns, is associated with maladaptive cannabis use. Expanding on prior research, findings indicated anxiety sensitivity is associated with coping motives for cannabis use among trauma-exposed cannabis users.
